      • Pituitary Adenoma Biomarkers Identified Using Proteomic Fingerprint Technology

        Objective: To determine whether pituitary adenomas can be diagnosed by identifying protein biomarkers in the serum. Methods: We compared serum proteins from 65 pituitary adenoma patients and 90 healthy donors using proteomic fingerprint technology combining magnetic beads with matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MS). Results: A total of 42 M/Z peaks were identified as related to pituitary adenoma (P<0.01). A diagnostic model established based on three biomarkers (3382.0, 4601.9, 9191.2) showed that the sensitivity of diagnosing pituitary adenoma was 90.0% and the specificity was 88.3%. The model was further tested by blind analysis showing that the sensitivity was 88.0% and the specificity was 83.3%. Conclusions: These results suggest that proteomic fingerprint technology can be used to identify pituitary adenoma biomarkers and the model based on three biomarkers (3382.0, 4601.9, 9191.2) provides a powerful and reliable method for diagnosing pituitary adenoma.

      • Meta-analysis of the MDM2 T309G Polymorphism and Gastric Cancer Risk

        Background: Mdm2 binds to the amino-terminus of p53 to induce its degradation and a single nucleotide polymorphism in the MDM2 promoter region (T309G) has been reported to increase the risk of several carcinomas, such as gastric cancer. However, the results of published studies to analyze the association between MDM2 T309G and gastric cancer havve often conflicted. Methods: To better illustrate the filiation between MDM2 T309G and gastric cancer, we performed a meta-analysis. Odds ratios (ORs) and 95% confidence intervals (CIs) were used to evaluate the strength of the relationship. The pooled ORs were performed for 4 models, additive, recessive, co-dominant model, and dominant. Results: Nine published case-control studies including 3,225 gastric cancer cases and 4,118 controls were identified. The MDM2 T309G polymorphism was associated with a significantly increased risk of gastric cancer risk when all studies were pooled into the meta-analysis (GG versus TT, OR=1.57; 95%CI=1.57-2.12; p=0.003) and GG versus GT/TT, OR=1.52; 95%CI=1.217-1.90; p<0.001). Furthermore, Egger's test did not show any evidence of publication bias (P = 0.608 for GG versus TT). Conclusion: Our results suggest that the MDM2 T309G polymorphism is indeed associated with a significantly increased risk of gastric cancer.

      • An Estimation Method of Consolidation and Settlement of Soil Mass in Dredger Fill Area Based on SBAS-InSAR

        The land subsidence poses a nonnegligible threat to production and life, especially in dredger fill area. The essence of land subsidence is soil consolidation. Better evaluation of the degree of soil consolidation presents significant progress for predicting land subsidence. In this study, SBAS-InSAR was conducted to Chenjia town, the typical dredger fill area in Chongming East Shoal, Shanghai. SAR data and precise Orbit data were collected from the ESA satellite Sentinel-1 in recent 2 years with approximately one month intervals. the relationship curve of cumulative subsidence with time is obtained after processing. On this basis, the final settlement and soil consolidation can be estimated by applying the method proposed by Professor Huang Wenxi of China. The results show that: The subsidence rate is faster than 20 mm/a; The estimated final settlement is 26.6-52.9 mm, and the corresponding consolidation degree is 54.6 % on average; Soil mass has not yet been fully consolidated and it still takes a long time to achieve stability. In conclusion, the method proposed in this study can provide a clearer understanding of the consolidation degree, and a new reference for the evaluation and prevention of land subsidence. Future research will focus on longer monitoring time to enrich and improve the method.         Cyanidin-3-Glucoside-Rich Extract from Chinese Bayberry Fruit Protects Pancreatic b Cells and Ameliorates Hyperglycemia in Streptozotocin-Induced Diabetic Mice

        Chinese bayberry fruit is a rich source of anthocyanins, especially cyanidin-3-glucoside (C3G). The present study investigated the protective effects of C3G-rich bayberry fruit extract (CRBFE) against pancreatic b cells against oxidative stress–induced injury as well as its hypoglycemic effect in diabetic mice. Bayberry extract from ‘‘Biqi’’ was used for both in vitro and in vivo testing because of its high C3G content and high antioxidant capacity. Pretreatment of b cells with CRBFE (containing 0.5 lmol/L C3G) prevented cell death, increased cellular viability, and decreased mitochondrial reactive oxygen species production and cell necrosis induced by 800 or 1200 lmol/L H2O2. CRBFE dose-ependently up-regulated pancreatic duodenal homeobox 1 gene expression, contributing to increased insulin-like growth factor II gene transcript levels and insulin protein in INS-1 cells. In addition, adminitration of CRBFE (150 lg of C3G/10 g of body weight twice per day)significantly reduced blood glucose in streptozotocin-induced diabetic ICR mice and increased the glucose tolerance in an oral glucose tolerance test (P < .05). Such results indicated that CRBFE might be useful in prevention and control of diabetes mellitus and diabetes-associated complications.

      • Evaluation of Transient Motion During Gadoxetic Acid–Enhanced Multiphasic Liver Magnetic Resonance Imaging Using Free-Breathing Golden-Angle Radial Sparse Parallel Magnetic Resonance Imaging

        <P>Objectives: The aims of this study were to observe the pattern of transient motion after gadoxetic acid administration including incidence, onset, and duration, and to evaluate the clinical feasibility of free-breathing gadoxetic acid-enhanced liver magnetic resonance imaging using golden-angle radial sparse parallel (GRASP) imaging with respiratory gating.& para;& para;Materials and Methods: In this institutional review board-approved prospective study, 59 patients who provided informed consents were analyzed. Free-breathing dynamic T1-weighted images (T1WIs) were obtained using GRASP at 3 T after a standard dose of gadoxetic acid (0.025 mmol/kg) administration at a rate of 1 mL/s, and development of transient motion was monitored, which is defined as a distinctive respiratory frequency alteration of the self-gating MR signals. Early arterial, late arterial, and portal venous phases retrospectively reconstructed with and without respiratory gating and with different temporal resolutions (nongated 13.3-second, gated 13.3-second, gated 6-second T1WI) were evaluated for image equality and motion artifacts. Diagnostic performance in detecting focal liver lesions was compared among the 3 data sets.& para;& para;Results: Transient motion (mean duration, 21.5 +/- 13.0 seconds) was observed in 40.0% (23/59) of patients, 73.9% (17/23) of which developed within 15 seconds after gadoxetic acid administration. On late arterial phase, motion artifacts were significantly reduced on gated 13.3-second and 6-second T1WI (3.64 +/- 0.34, 3.61 +/- 0.36, respectively), compared with nongated 13.3-second T1WI (3.12 +/- 0.51, P < 0.0001). Overall, image quality was the highest on gated 13.3-second T1WI (3.76 +/- 0.39) followed by gated 6-second and nongated 13.3-second T1WI (339 +/- 0.55, 2.57 +/- 0.57, P < 0.0001). Only gated 6-second T1WI showed significantly higher detection performance than nongated 13.3-second TIWI (figure of merit, 0.69 [0.63-0.76]) vs 0.60 [0.56-0.65], P = 0.004).& para;& para;Conclusions: Transient motion developed in 40% (23/59) of patients shortly after gadoxetic acid administration, and gated free-breathing T1WI using GRASP was able to consistently provide acceptable arterial phase imaging in patients who exhibited transient motion.</P>

        Mutation of IPO13 causes recessive ocular coloboma, microphthalmia, and cataract

        Ocular coloboma is a developmental structural defect of the eye that often occurs as complex ocular anomalies. However, its genetic etiology remains largely unexplored. Here we report the identification of mutation (c.331C>T, p. R111C) in the IPO13 gene in a consanguineous family with ocular coloboma, microphthalmia, and cataract by a combination of whole-exome sequencing and homozygosity mapping. IPO13 encodes an importin-B family protein and has been proven to be associated with the pathogenesis of coloboma and microphthalmia. We found that Ipo13 was expressed in the cornea, sclera, lens, and retina in mice. Additionally, the mRNA expression level of Ipo13 decreased significantly in the patient compared with its expression in a healthy individual. Morpholinooligonucleotide- induced knockdown of ipo13 in zebrafish caused dose-dependent microphthalmia and coloboma, which is highly similar to the ocular phenotypes in the patient. Moreover, both visual motor response and optokinetic response were impaired severely. Notably, these ocular phenotypes in ipo13-deficient zebrafish could be rescued remarkably by full-length ipo13 mRNA, suggesting that the phenotypes observed in zebrafish were due to insufficient ipo13 function. Altogether, our findings demonstrate, for the first time, a new role of IPO13 in eye morphogenesis and that loss of function of IPO13 could lead to ocular coloboma, microphthalmia, and cataract in humans and zebrafish.

        EFFECTS OF INJECTION TIMING ON TRANSIENT PERFORMANCE OF A REGULATED TWO-STAGE TURBOCHARGED DIESEL ENGINE WITH TURBINE BYPASS VALVE

        The object of this paper is to reduce soot emissions under typical 5s transient conditions of constant speed and increasing torque. And effects of fuel injection timing on combustion and emissions parameters were experimentally and numerically studied in a regulated two-stage turbocharged diesel engine with a turbine bypass valve (TBV). The test results indicated that: the smaller TBV opening could improve deterioration of smoke emissions and BSFC at medium and heavy loads. Afterward, the full-stage injection timing (FSIT) strategies (delaying injection timing during the entire transient process) could reduce soot and NOX emissions simultaneously. However, when TBV opening became larger, smoke emissions and BSFC were deteriorated gradually. Moreover, the sectional-stage injection timing (SSIT) strategies (advancing injection timing from 10 % load to a preset load and delaying injection timing from the preset load to 100 % load) could markedly reduce soot emissions by 75.8 % with TBV opening 20 %; the degradation of fuel consumption could be effectively suppressed. Finally, coupling the SSIT strategies with the TBV control strategies could significantly improve the transient performance.
