<P>The use of colorimetric bioassays for protein detection is one of the most interesting diagnostic approaches, but their relatively poor detection limits have been a critical issue. In this study, we developed an efficient colorimetric bioassay based on switchable linkers (SLs) for the detection of prostate-specific antigen (PSA), which is one of the most widely used protein biomarkers for the diagnosis of prostate and breast cancers. SLs can cross-link gold nanoparticles (AuNPs) to generate large-scale aggregates and thereby induce precipitation to achieve visual signal amplification. In addition, when SLs are occupied by target proteins (referred to as ‘switch-off’), highly sensitive detection is enabled. To maximize sensitivity, we adjusted the total surface area of AuNPs by controlling their concentration. As a result, PSA was detected at an ultralow concentration of 100 fg mL<SUP>−1</SUP>. This SL-based assay is shown to be simple, easy to handle and visualize, and highly sensitive. Therefore, in addition to PSA, the proposed SL-based assay could be used to detect other protein biomarkers.</P>

Instant Messenger is a communication tool which allows instant real-time connection between parties through Internet. Compared to the most popular Internet communication tool, email, the messenger allows people to check network presence of other parties and to connect in real time among multiple parties. Mostly due to the convenience it provides, use of instant messengers has increased tremendously not only for personal purpose but also for business purpose. However, firms are agonizing about the real impact of instant messenger usage among employees, whether the impact is positive or negative in terms of the productivity. Applying the theory of reasoned action (TRA) and technology acceptance model (TAM), this study sets UP perceived ease of use, perceived usefulness and cultural orientation of individuals as antecedents of adoption and use of the messenger application. Also, the impacts of messenger usage are measured as task productivity and intimacy among colleagues. This study presents several findings about the instant messenger. First, perceived ease of use, and usefulness play important roles in raising the adoption level of instant messenger. Also, culture propensity also maintains strong influence towards the actual usage of the messenger. Among cultural variables, innovativeness, supportiveness and outcome orientation reveal strong positive impacts on adoption and use of the messenger. On the dependent side, use of instant messenger seems to have a strong direct impact on task productivity as well as indirect impact through intimacy. Implications are discussed.
클러스터링 기법을 활용한 중소기업 지원 지식서비스의 성과유형 분류

많은 제조기반의 중소기업들은 프로세스나 제품을 혁신하기 위해 공공 및 사기업의 지식서비스를 아웃소싱하고 있다. 본 연구의 사례인 K 연구원은 여러 지식서비스를 다양한 조합형태로 제공하는데, 이러한 제공형태의 복잡성으로 인해 지식서비스 사업성과를 정확히 분석 한다는 것이 어려운 상황이다. 본 연구는 기존에 성과평가 항목을 상향식으로 선정하는 방식이 아닌 하향식 관점에서 성과항목을 도출하였다. K 연구원 지식서비스 수혜기업인 82개 기업사례에서 74개의 성과항목이 도출되었고, 최종적으로 17개 항목으로 정제하였다. 이후 사례-성과 행렬을 구조화하여 기업별 성과의 유무를 조사하여 이진 데이터로 입력하였다. K-means 클러스터링 분석을 통해 3개의 군집을 각각 ‘핵심 경쟁력 강화(제품 및 특허)’, ‘국내 및 해외시장 확대’, ‘운영 효율성 제고’로 식별할 수 있었다. In recent years, many small and medium-sized manufacturing companies are making process innovation and product innovation through the public knowledge services. K Research institute provides different types of knowledge services in combination and due to this complexity, it is difficult to analyze the performance of knowledge service programs precisely. In this study, we derived performance items from bottom-up viewpoints, rather than top-down approaches selecting those items as in previous performance analysis. As a result, 74 items were finded from 82 companies in the K Research Institute case book, and the final result was refined to 17 items. After that a case-performance matrix was constructed, and binary data was entered to analyze. As a result, three clusters were identified through K-means clustering as ‘enhancement of core competitiveness (product and patent),’ ‘expansion of domestic and overseas market,’ and ‘improvement of operational efficiency.’
Screening and Application of the Natural Pigment for Meat Analogues

Recently, a lots of studies have been conducted to imitate real meat with the factors including texture, aroma, taste, and color of meat analogues. Since the color component of meat is diverse and complex, as well as large of degree of change according to cooking, the color imitating has been still challenging. In this study, we selected up a representative meat analogue (tteok-galbi), tracked the color change at three making points as steps of batter, steaming, and cooking. In order to realize the color at these points, three plant-based natural pigments including prickly pear cactus, sweet pumpkin, and mugwort were selected, and the color with optimized ratio was applied to the tteok-galbi. As a result, it was found that 1:3:1 in the batter, 2:1:2 in steaming, and 2:1:2 in cooking were optimal ratios. The results in this study were significant in that it could apply the colors to meat analogues according to the combination of natural pigments derived from plants.
Bifunctional linker-based immunosensing for rapid and visible detection of bacteria in real matrices

<P><B>Abstract</B></P> <P>Detection of pathogens present in food and water is essential to help ensure food safety. Among the popular methods for pathogen detection are those based on culture and colony-counting and polymerase chain reaction (PCR). However, the time-consuming nature and/or the need for sophisticated instrumentation of those methods limit their on-site applications. We have developed a rapid and highly sensitive immunosensing method for visible detection of bacteria in real matrices based on the aggregation of AuNPs without requiring any readout device. We use biotinylated anti-bacteria antibodies as bifunctional linkers (BLs) to mediate the aggregation of streptavidin-functionalized gold nanoparticles (st-AuNPs) to produce visually recognizable color change, due to surface plasmon resonance (SPR), which occurs in about 30min of total assay time when the sample is mildly agitated or within three hours in quiescent conditions. The aggregation of st-AuNPs, which produces the indication signal, is achieved very differently than in visual detection methods reported previously and hence affords ultrahigh sensitivity. While BLs can both bind to the target and crosslink st-AuNPs, their latter function is essentially disabled when they bind to the target bacteria. By varying the amount of st-AuNPs used, we can tailor the assay effectiveness improving limit of detection (LOD) down to 10CFUmL<SUP>−1</SUP> of <I>E. coli</I> and <I>Salmonella</I>. Test results obtained with tap water, lake water and milk samples show that assay performance is unaffected by matrix effects. Further, in a mixture of live and autoclaved <I>E. coli</I> cells our assay could detect only live cells. Therefore, our BL-based immunosensor is suitable for highly sensitive, rapid, and on-site detection of bacteria in real matrices.</P> <P><B>Highlights</B></P> <P> <UL> <LI> An immunosensor was developed for the detection of bacterial targets. </LI> <LI> The indication signal is visual color change due to AuNPs aggregating away from target. </LI> <LI> The detection sensitivity is very high, down to 10CFU/mL. </LI> <LI> Total assay time is about 30min, without needing sample pretreatment or readout device. </LI> <LI> pH and constituents of real matrix has no effect of sensor effectiveness. </LI> <LI> The sensor can detect only live <I>E. coli</I> (ATCC 11775T) in a mixture of live and autoclaved cells. </LI> </UL> </P>
