        Immobilization of the Hyperthermophilic Archaeon Thermococcus onnurineus Using Amine-coated Silica Material for H<sub>2</sub> Production

        초고온성 고세균 Thermococcus onnurineus NA1은 개미산, 일산화탄소, 또는 전분 등을 이용해서 수소를 생산하는 것으로 알려져 있다. 본 연구에서는 T. onnurineus NA1의 고정화 세포를 이용한 수소생산을 고찰하였다. 고정화 실험결과, T. onnurineus NA1은 표면에 아민기가 코팅된 규조토 담체에 정전기적 인력에 의해 효과적으로 고정화되었고, 1 g의 담체에 고정화 될 수 있는 최대 세포의 양은 71.7 mg-dcw로 확인되었다. 고정화 세포를 이용한 세 번의 반복회분식 배양을 통해 개미산으로부터 수소생산 특성을 고찰하였고, 그 결과 배양이 반복됨에 따라 고정화 세포 농도의 증가에 기인하여 초기수소생산속도가 2.3 에서 4.0 mmol l<sup>−1</sup> h<sup>−1</sup>로 상당량 증가됨이 관찰되었다. 따라서, T. onnurineus NA1의 고정화세포 시스템은 수소생산을 위한 좋은 대안이 될 수 있을 것으로 사료된다. 본 연구는 초고온성 고세균의 고정화세포를 수소생산에 적용한 첫 번째 사례이다. Previously we reported that the hyperthermophilic archaeon, Thermococcus onnurineus NA1 is capable of producing hydrogen (H<sub>2</sub>) from formate, CO or starch. In this study, we describe the immobilization of T. onnurineus NA1 as an alternative means of H<sub>2</sub> production. Amine-coated silica particles were effective in immobilizing T. onnurineus NA1 by electrostatic interaction, showing a maximum cell adsorption capacity of 71.7 mg-dried cells per g of particle. In three cycles of repeated-batch cultivation using sodium formate as the sole energy source, immobilized cells showed reproducible H<sub>2</sub> production with a considerable increase in the initial production rate from 2.3 to 4.0 mmol l<sup>−1</sup> h<sup>−1</sup>, mainly due to the increase in the immobilized cell concentration as the batch culture was repeated. Thus, the immobilized-cell system of T. onnurineus NA1 was demonstrated to be feasible for H<sub>2</sub> production. This study is the first example of immobilized cells of hyperthermophilic archaea being used for the production of H<sub>2</sub>.

        Furosemide에 의한 소디움 운반체 발현의 변화

        배경 : 임상에서 흔히 사용하는 이뇨제 furosemide는 비후상행각에서 Na+-K+-2CI- cotranspoter (NKCC2)를 억제하여 NaCl 재흡수를 차단하여 이뇨작용을 나타낸다. Furosemide를 장기간 투여하면 내성과 대사성 알카리증의 부작용이 발생할 수 있는데, 이는 집합관에 도달하는 소디움 증가와 관련 있을 가능성이 있다. 방법 : 저자들은 furosemide의 내성이나 부작용이 집합관 상피 소디움 통로 (ENaC) 단백발현의 변화와 관련이 있는지를 확인하고자, Sprague-Dawley rat에서 farosemide (12 mg/day)을 7일간 지속적 피하 주입한 후 반정량적 immunoblotting과 면역조직화학법을 이용하여 NKCC2, Na +-CL- cotransporter (NCC), ENaC 단백의 발현을 관찰하였다. 실험기간 동안 수분과 전해질 용액 (0.8% NaCl & 0.1% KCl)을 자유롭게 섭취하도록 하여 체액 감소를 방지하였다. 결과 : 부형약 (vehicle)을 투여한 대조군에 비하여, furosemide를 투여한 군에서 각각 요량과 요 소디움 배설이 증가하였으나, 체중, 혈청 알도스테론치 및 크레아티닌 청소율은 차이가 없었다. Furosemid 투여 후 NKCC2 단백은 피질 (151±10 vs. 100±10%, p<0.05)과 외수질 (122±5 vs. 100±3%, p<0.01)에서 증가해 있었다. ENaC 단백은 세 가지 subunit 모두 furosemide 투여 후 대조군에 비하여 피질 (α:187±25 vs. 100±22%, p<0.05; β:155±8 vs. 100±15%, p<0.05; γ:168±16 vs. 100±9%, p<0.05)과 외수질 (α:171±27 vs. 100±17%, p<0.05; β :986±91 vs. 100±33%, p<0.01; γ :242±24 vs. 100±22%, p<0.01)에서 증 가하였다. 면역조직화학법에서도 furosemide를 투여한 군의 집합관 주세포에서 ENaC β-subunit가 더 강하게 염색되었다. 결론 : 이상에서 장기간 furosemide 투여시 집합관 ENaC 발현이 증가하며, 이러한 원위부네프론의 적응 반응이 이뇨제 내성을 유발하는데 기여할 것으로 생각한다. Background : Furosemide inhibit NaCl absorption in the thick ascending limb and produce an increase in distal delivery of Na+. We carried out semiquantitative immunoblotting and immunohistochemistry of rat kidneys to investigate whether chronic furosemide infusion is associated with compensatory increases in the abundance of Na+ transporters in distal nephron. Methods : Osmotic minipumps were implanted into Sprague-Dawley rats to deliver 12 mg/day of furosemide(n=6) with simultaneous administration of 0.8% NaCl and 0.1% KCl in drinking water for 7days. Results : Compared with vehicle infused controls, urine volume and urine sodium amount were increased. However, there were no differences in body weight, serum aldosterone, and creatinine clearance. The abundance of Na+-K+-2CI - cotransporter after furosemide infusion was increased in cortex (151±10 vs. 100±10%, p<0.05) and outer medulla (122±5 vs. 100±3%, p<0.01). In furosemide infusion group, the abundance of all three subunits of epithelial sodium channel (ENaC) was increased both in cortex (α:187±25 vs. 100±17%, p<0.05; β:155±8 vs. 100±15%, p<0.05; γ :168±16 vs. 100±9%, p<0.05) and outer medulla (α:171±27 vs. 100±17%, p<0.05; β :986±91 vs. 100±33%, p<0.01; γ :242±24 vs. 100±22%, p<0.01). Consistent with these results, ENaC β-subunit immunohistochemistry showed a remarkable increase in immunoreactivity in the principal cells of collecting ducts with furosemide treatment. Conclusion : These increases in the abundance of ENaC protein may account for the generation of diuretic tolerance.

      • Facile and fast Na-ion intercalation employing amorphous black TiO<sub>2-x</sub>/C composite nanofiber anodes

        <P><B>Abstract</B></P> <P>Structural and electronic modification of titanium oxide (TiO<SUB>2</SUB>) nanomaterials induced by the co-introduction of fully disordered glass phase and oxygen vacancies can lead to remarkable advances in the electrode performance in emerging energy storage systems. We report on the effective co-creation of fully amorphous nanofibers (NFs) composed of black TiO<SUB>2-x</SUB> and conductive carbons throughout the NF structure, and evaluate the materials as potential anodes in sodium-ion batteries. The black TiO<SUB>2-x</SUB> nanofiber is successfully fabricated by electrospinning a precursor solution followed by a two-step sequential thermal treatment in an air and reducing atmosphere. The NF electrode could deliver approximately two-fold higher 2nd discharge capacity and an excellent kinetic performance even under high rates compared to that delivered by anatase-structured white TiO<SUB>2</SUB> NFs used as reference, because of (i) an inherent free volume in the glass phase corresponding to the enlarged Na<SUP>+</SUP> sites, (ii) increased electrical conductivity (low bandgap) resulting from the presence of Ti<SUP>3+</SUP>, (iii) introduction of conductive carbon agents around the TiO<SUB>2-x</SUB> domain, and (iv) one-dimensional NF feature allowing numerous Na<SUP>+</SUP> reaction sites at the electrochemical interface.         효소분해와 염과 당 및 항산화 작용 상승제의 첨가에 의한 계피 추출액의 특성 변화

        The dried cinnamon was extracted with enzymes, salts, sugars and additives in order to find the most effective extraction material. Enzymatic hydrolysis of cinnamon suspension with cellulase, hemicellulase, pectinase, β-1.4-glucosidase, tannase and lipase showed a little increase of their cinnamic aldehyde contents. Solid yield, antioxidant activity and degree of browning were increased in hemicellulase treatment. Acid and alkali extraction of cinnamon showed a some increase in solid yields and antioxidant activity was increased by addition of glucose and Na-ascorbate. Cinnamic aldehyde contents and degree of browning were increased in extraction with Na-citrate addition.

        가정연계 신체활동이 유아의 기초체력에 미치는 영향

        The awareness of the decreasing physical activities of children today introduced the application of Home-Based physical activities as one of the solutions to the problem. This Home-Based physical activity program provided the families with training materials and wall posters for physical activities as well as the eurhythmics videos accessible through provided QR codes in attempt to assist the families to practice the physical activities taken in children educational institutes from homes. 4910 children of both sexes across the nation partook in this research while the researchers were determined to test the change on children’s physical strength before/after the application of the program. In order to track the change, the researchers have adjusted and supplemented the research tools of Na-young An, Eun-jung Kim, Tae-seop Park (2013) to measure the endurance, muscle strength, flexibility, agility, adaptation, and balance of the children. The research revealed that the children that partook in the program for household physical activities showed an overall superiority on all 6 fields - endurance (scissor jump for 30 seconds), muscle strength (grasping power), flexibility (sitting trunk flexion), agility (30meters shuttle run), adaptation (standing high jump), balance (standing on one foot) - compared to the children who haven’t engaged in the program. Also the researchers found that the general physical strength of the children in the program to have increased periodically during the test. Most of all, there was a significant difference on overall body endurance and right-left balance from which the deduction of the relatedness between the two and the household physical activities was made. In conclusion, Home-Based physical activities have shown to have positive influence on the general physical strength of the children.

        퍼머넌트와 염색 연결시술시 정향트리트먼트 적용시점에 따른 모발보호효과

        This study attempted to determine the effects of hair protection by investigating the characteristics and possibilities of hair cosmetics using the hot water extracts of Syzygium aromaticum and then hair-protection effects were confirmed through pre-treatment and post-treatment at consecutive chemical treatments. At the consecutive chemical treatments, the general treatment was divided into pretreatment(N1) and general treatment post-process(N2). Then, the Syzygium aromaticum-based treatment was divided into pre-treatment(J1) and post processing(J2). According to the analysis, tensile strength was the highest in the Syzygium aromaticum extract treatment group. At consecutive treatments, ‘J2’ which processed treatment for Syzygium aromaticum, was the highest. In terms of hair thickness, the Syzygium aromaticum group was greater than the general treatment group. When measured to determine the morphological characteristics of ‘Hair’ at 20,000 magnifications using SEM, the Syzygium aromaticum extract post-treatment group (J2) revealed the highest value (373.98±10.83㎚). Regarding hair composition under the EDS, Syzygium aromaticum extract post-treatment group was the highest with 07.77% interms of sulfur content. The results confirmed that Syzygium aromaticum extract treatment is extremely effective in easing hair damage which is caused by consecutive dyeing after permanent. Therefore, the Syzygium aromaticum extract treatment would be effective in preventing hair damage and keeping hair healthy and glossy.

        De novo Hyponatremia in Patients Undergoing Peritoneal Dialysis: A 12-month Observational Study

        Purpose: Hyponatremia occurs infrequently in patients undergoing peritoneal dialysis (PD). Nevertheless, one must understand its pathophysiology, since the therapeutic strategy differs from that of non-PD-related hyponatremia. This study examined the clinical features of hyponatremia in PD and evaluated the factors that may contribute to its development. Methods: We retrospectively enrolled 51 normonatremic PD patients at Gachon University Gil Hospital, South Korea. Using the plasma sodium levels at month 13, the patients were divided into hyponatremia (Na+<135mEq/L) and normonatremia (Na+≥135mEq/L) groups. Then, the clinical variables of these patients were examined, including peritoneal function and adequacy tests, and biochemical parameters. Results: The de novo hyponatremia (n=8) and normonatremia (n=43) groups had no significant differences in baseline characteristics. At month 1, the serum albumin was lower in the hyponatremia group (p=0.022). In the peritoneal equilibration test analysis, the dialysate-to-plasma ratio for creatinine (D/Pcr) measured after 13 months differed significantly between the two groups (p=0.007), while the maximum dip in sodium did not differ. No significant differences were observed in the normalized protein equivalent of nitrogen appearance, Kt/V, or residual renal function. Conclusion: Our data suggest that the development of hyponatremia is associated with a lower initial serum albumin level and increased D/Pcr in patients undergoing PD. Therefore, the serum sodium levels should be monitored more carefully in these patients.

        Structural basis for the ATP-independent proteolytic activity of LonB proteases and reclassification of their AAA+ modules.

        <P>Lon proteases degrade defective or denature proteins as well as some folded proteins for the control of cellular protein quality. There are two types of Lon proteases, LonA and LonB. Each consists of two functional components: a protease component and an ATPase associated with various cellular activities (AAA+ module). Here, we report the 2.03 -resolution crystal structure of the isolated AAA+ module (iAAA+ module) of LonB from Thermococcus onnurineus NA1 (TonLonB). The iAAA+ module, having no bound nucleotide, adopts a conformation virtually identical to the ADP-bound conformation of AAA+ modules in the hexameric structure of TonLonB; this provides insights into the ATP-independent proteolytic activity observed in a LonB protease. Structural comparison of AAA+ modules between LonA and LonB revealed that the AAA+ modules of Lon proteases are separated into two distinct clades depending on their structural features. The AAA+ module of LonB belongs to the -H2 & Ins1 insert clade (HINS clade)- defined for the first time in this study, while the AAA+ module of LonA is a member of the HCLR clade.</P>

        원위부 신세뇨관성 산증에서 산-염기 운반체의 결손

        The purpose of this study was to elucidate whether the molecular defect of acid-base transporters in renal tubules is related to the functional defect of urinary acidification in distal renal tubular acidosis(RTA). We performed NH₄Cl, furosemide, or bicarbonate loading test to evaluate renal acidification function, and immunohistochemistry using antibodies to H^+ -ATPase, Cl^-/HCO₃^- exchanger(band-3 protein), and Na^+/K^+ -ATPase in kidney tissue in 6 patients with RTA and renal cell carcinoma patients as normal controls. Kidney tissue was obtained either by percutaneous needle biopsy(RTA) or nephrectomy(NC). The results were as follows; 1) In all six RTA patients, proton secretory defect of distal acidification was shown by a failure to lower the urine pH after NHC1 loading or furosemide test or abnormally low urine-blood pCO₂ difference during bicarbonate loading. In two patients with RTA, proximal acidification defect was combined, which was demonstrated by increased fractional excretion of bicarbonate. 2) In mal control, intense H^+ -ATPase and band-3 protein staining was observed in collecting ducts. 3) In distal RTA patients, H6+ -ATPase and band-3 protein staining was not demonstrable or markedly decreased in the intercalated cells of distal nephron. 4) In two patients who had both proximal and distal RTA, H^+ -ATPase staining was markedly decreased in the brush border of proximal tubules as well as the distal nephron. In conclusion, the defect of acid-base transporters in renal tubule was related with the functional defect of urinary acidification in distal RTA.

        Gitelman 증후군 환자에서 면역조직화학법으로 확인한 원위세관 sodium-chloride cotransporter (NCCT)의 결손

        목적 : Gitelman 증후군은 저포타시움혈증, 대사성 알칼리증, 고레닌혈증, 고알도스테론혈증, 요 중 칼슘 배설의 저하 및 저마그네슘혈증을 특징으로 하는 유전성 질환이다. 이는 원위세관 sodium-chloride cotransporter (NCCT)의 유전자 돌연변이에 의하여 발생하는 것으로 알려져 있으나 사람의 신조직에서 NCCT 결손이 증명된 바는 없었다. 방법 : 저자들은 임상적으로 Gitelman 증후군이 의심되는 환자에서 이뇨제를 이용한 신청소율 검사와 유전자 검사를 시행하였고, 이를 통하여 감별진단한 Gitelman 증후군 환자의 신조직에서 인간 NCCT에 대한 토끼 다클론 항체를 이용한 면역조직화학법을 시행하였다. 신세포암으로 신적출술을 시행 받은 환자의 정상 신조직과 전해질 이상이 없었던 사구체신염 환자의 신조직을 각각 정상 대조군과 질환 대조군으로 하였다. 결과 : 대상 환자는 저포타시움혈증과 대사성 알칼리증, 저마그네슘혈증 및 요 중 칼슘 배설의 저하를 보였다. Bartter 증후군과 감별을 위하여 furosemide 및 hydrochlorothiazide를 이용한 신청소율 검사를 시행하였다. 수분 부하를 시행한 기저치(86.1%)에 비해서 furosemide를 투여한 후 distal fractional chloride reabsorption이 감소하였으나(9.7%) hydrochlorothiazide 투여 후에는 변화가 없었다(81.4%). 유전자 검사 결과 SLC12A3 유전자의 돌연변이(S967F)가 발견되었다. 신조직에서 면역조직화학법을 시행한 결과 정상 및 질환 대조군에서는 원위세관 세포의 내강 막 쪽에 NCCT가 뚜렷이 염색되었으나, Gitelman 증후군에서는 원위세관 세포의 NCCT에 대한 면역 반응성이 관찰되지 않았다. 반면에 Na/K-ATPase, Na-K-2Cl cotransporter, calbindin-D28K는 대조군과 대상 환자의 신조직에서 모두 관찰되었다. 결론 : 기능적 검사로 진단된 Gitelman 증후군 환자의 신조직에서 NCCT의 결함을 면역조직화학법으로 확인하였다. Background : Gitelman`s syndrome is an autosomal recessive renal tubular disorder characterized by hypokalemic metabolic alkalosis, hypomagnesemia, and hypocalciuria. It is known to be caused by a mutation of SLC12A3 gene coding the sodium-chloride cotransporter (NCCT) in the distal tubule. The defect of NCCT in human renal tissues has not been investigated, and we tested whether the defect of NCCT can be detected in renal tissue of a patient with Gitelman`s syndrome by using immunohistochemistry. Methods : In an adult patient with Gitelman`s syndrome, blood and urine samples were collected for measurement of biochemical parameters. Renal clearance study and gene analysis were performed. Immunohistochemistry was performed on the renal tissue of the patient using a rabbit polyclonal antibody directed against a synthetic peptide corresponding to a portion in the amino terminal tail for human NCCT. Normal human renal tissues from surgical nephrectomy due to renal cell carcinoma and renal biopsy tissues from patients with glomerulonephritis but without any electrolyte disturbance were used as controls. Results : The patient had hypokalemic metabolic alkalosis, hypocalciuria and hypomagnesemia. Renal clearance study revealed a decrease in distal fractional chloride reabsorption after the administration of furosemide. SLC12A3 gene mutation (S967F) was found by direct sequencing method. Immunohistochemistry showed the absence of NCCT staining in the renal tissue of the patient. On the other hand, the immunostaining of other transporters was all positive in renal tissues from both Gitelman`s syndrome patients and controls. Conclusions : We report the absence of intact NCCT in the renal tissue of a Gitelman`s syndrome patient.(Korean J Med 69:642-650, 2005)
