        한국인에서 5,10-Methylenetetrahydrofolate Reductase 유전자 다형성 분석 및 임신의 결과에 미치는 영향

        목적: 본 연구는 한국인 임신 여성에서 MTHFR 유전자형 변이와 혈청내 호모시스테인 및 엽산의 농도와 이들이 임신에 미치는 영향을 알아보고자 하였다. 방법: 임신부 600명에게서 혈액을 채취하여 얻어진 DNA로 MTHFR C677T와 A1298C 유전자의 다형성을 PCR-RELP로 분석하였고 혈청내 호모시스테인은 HPLC 방법으로, 엽산은 RIA 방법으로 측정하였다. 결과: 혈청내 호모시스테인은 MTHFR C677T 유전자형이 C/C형이나 C/T인 군에 비해 T/T인 군에서 유의하게 높았으며 MTHFR A1298C 유전자형이 A/C형이나 C/C인 군에 비해 A/A인 군에서 역시 유의하게 높았다( p<0.05). 혈청 호모시스테인은 모든 MTHFR 유전자형에서 혈청 엽산농도와 역상관관계를 보였으나, 변이형 유전자형(T/T와 A/A유전자형)에서 더 큰 역 상관관계를 보였다. 15?mol/L 이상의 고호모시스테인혈증의 임신부에서 임신 주수와 출생체중이 36.1주, 3053.8 g으로 15 ?mol/L미만인 군의 38.3 주, 3215.3 g 보다 유의하게 낮았다( p<0.05). 결론: 혈청 호모시스테인의 농도는 MTHFR C677T와 A1298T 유전자의 다형성에 의해 영향을 받으며, MTHFR C677T와 A1298T 유전자의 다형성과 혈청 호모시스테인은 모두 임신주수와 신생아의 체중에 영향을 미친다. Objective: The purpose of this study was to analyze MTHFR polymorphism among the Korean population and to evaluate the relationship between serum levels of homocysteine and MTHFR polymorphism and also to investigate the effect on pregnancy outcomes. Methods: DNA was extracted from whole blood of 600 pregnant women. All samples were genotyped for the C677T and A1298C polymorphisms in MTHFR gene by PCR-RELP assay. Serum levels of homocysteine and folate were measured by high performance liquid chromatography for homocysteine and radioassay for folate. Pregnancy outcomes were estimated by gestational weeks and birth weights of newborns. Results: Serum homocysteine was higher in women with the T/T genotype than those with the C/T or C/C genotype of the MTHFR C677T polymorphism (p<0.05). And also serum homocysteine was higher in women with the A/A genotype than those with the A/C or C/C genotype of the MTHFR A1298C polymorphism (p<0.05). Serum homocysteine was negatively correlated with serum folate in all MTHFR genotypes, especially prominent in T/T genotype of MTHFR C677T polymorphism and A/A genotype of MTHFR A1298C polymorphism. Gestational age and the birth weight of infant from hyperhomocysteinemic mothers whose homocysteine levels higher than 15 μmol/L were 36.1 weeks, 3053.8 g, respectively, which were significant lower than those from normohomocysteinemic mothers (38.3 weeks, 3,215.3 g) (p<0.05). Conclusion: Serum homocysteine was influenced significantly by MTHFR C677T polymorphism and MTHFR A1298C polymorphism. MTHFR C677T and A1298C polymorphism and serum homocysteine levels affect pregnancy outcomes, although not mainly by serum folate level.

        Changes and distributional pattern of microorganism in compost and fruiting body for button mushroom cultivation

        Agaricus bisporus grows on a substrate known as compost, which is a product of aerobic fermentation by various microorganisms. These organisms convert and degrade the straw and form lignin humus complex which is utilized later on by the population of organisms. Theses microflora play a key role in the process of composting and can be regarded as the active agents in the preparation of nutrient medium as many of them may ultimately contribute themselves to the nutrition of A. bisporus. The diversity of microflora according to growing farmhouse and fruiting body of Agaricus bisporus were investigated. The aerobic bacteria and Bacillus as longer of turning stage of compost pile were increased. And, thermophilic actinomycetes and fluorescent Pseudomonas sp. showed high density after the pasteurization stage. But Tricoderma sp. was decreased toward the end of turning stage of compost pile. Ten mushroom farms was selected to research of microflora of fruiting body of button mushroom. The microflora showed significant difference according to mushroom farms. The bacteria density was 0.4~41.6×105 cfu/ml and the fungus was 1.3~3.9×103 cfu/ml. But The microorganism density was not significant change for the storage periods. These isolates were classified into Chryseobacterium indologenes(6 strains), Pseudomonas agarici(5 strains), Sphingobacterium multivorum(2 strains), Flavobacterium anhuiense(2 strains), Microbacterium sp.(10 strains), Pseudomonas sp.(13 strains) on the basis of 16 rDNA analysis. The most dominants of these species were Chryseobacterium indologenes and Pseudomonas agarici.

        The gastrointestinal involvement in rheumatoid arthritis may be broad and includes many causes: drug induced colitis, colitis associated with infectious agents, vasculitis in gut, autoimmune disease located in gut. Rheumatoid arthritis is a chronic multisystemic disease of unknown cause. Although a number of mixed autoimmue disease exist, there were rare reports about Ulcerative colitis with Rheumatoid arthritis associated. We report one case of Rheumatoid arthritis patient who diagnosed as Ucerative colitis by colonoscopic and histologic finding with review of literature.

      • Runoff Losses of Suspended Sediment, Nitrogen, and Phosphorus from a Small Watershed in Korea

        <P>Nutrients and sediments in runoff lead to the degradation of water quality of lakes and streams. The development of schemes to mitigate such degradation requires a characterization of the underlying transport processes. The objectives of this study were to develop annual and seasonal load-discharge relationships for suspended sediment (SS), total nitrogen (TN), and total phosphorus (TP) losses from a small mixed land use watershed and to use these relationships to explicate the annual and monthly patterns of losses of these species. Data from 1996 to 2004 were used to develop load-discharge relationships for SS, TN, and TP at the HP#6 watershed, a subwatershed of the Balhan reservoir watershed located in Bongdam-myun and Paltan-myun, Gyeonggi-do, Korea. Standard least squares curve fitting and S-estimation procedures were used to fit power functions to the data collected over this time period. The fitted load-discharge relationships are indicative of seasonal variations in SS and TN and of TP losses from HP#6. The exponents of the fitted power functions for TN and TP in the fall, for TP in summer season, and for SS in all seasons are >1, indicating that the concentrations of these species increase as flow rate increases. Most of the SS, TN, and TP transported in runoff left the watershed between April and September; thus, cost-efficient strategies can be established by focusing on this period. Further study of the seasonal variations is required for a better characterization of seasonal losses of SS, TN, and TP in runoff from the HP#6 watershed.</P>

        Acute bulbar palsy as a variant of Guillain-Barré syndrome

        <P>Objective: To categorize a syndrome manifesting as prominent acute bulbar palsy (ABP) without limb motor weakness as a variant form of Guillain-Barre syndrome (GBS) and differentiate it from Miller Fisher syndrome (MFS) and pharyngeal-cervical-brachial (PCB) variants. Methods: We analyzed cases of ABP without limb motor weakness based on a dataset containing clinical information and the results of antiganglioside antibodies assays for acute immune-mediated neuropathies. Results: Eleven cases with an age at onset ranging from 18 to 65 years (mean 33.8 years) were identified as ABP-plus syndrome. All of the enrolled cases manifested with ABP as the predominant symptom, and with no limb weakness. The following features accompanied ABP in order of decreasing frequency: ophthalmoplegia (n = 9, 82%), ataxia (n = 9, 82%), and facial palsy (n = 6, 55%). An enzyme-linked immunosorbent assay study disclosed that immunoglobulin G (IgG) anti-GT1a antibodies were the most frequent (n = 11), followed by IgG anti-GQ1b antibodies (n = 6). Conclusions: We propose that ABP-plus syndrome without neck or limb weakness is a variant of GBS that is distinct from the MFS and PCB variants. The presence of IgG anti-GT1a antibodies can explain the relationships between the distinct clinical characteristics and the underlying pathomechanisms.</P>

      • Association of SLC6A12 variants with aspirin-intolerant asthma in a Korean population : SLC6A12 polymorphisms and AIA

        <P>Aspirin-intolerant asthma (AIA) occurs from asthma exacerbation after exposure to aspirin. However, the underlying mechanisms of AIA occurrence are still unclear. The critical role of the solute carrier family 6 (neurotransmitter transporter, betaine/GABA) member 12 (SLC6A12) gene in GABAergic transmission, which is associated with mucus production in asthma, makes it a candidate gene for AIA association study. Eight single nucleotide polymorphisms (SNPs) in SLC6A12 were genotyped in 163 aspirin-intolerant asthma (AIA) and 429 aspirin-tolerant asthma (ATA) patients of Korean ethnicity. Associations between polymorphisms of SLC6A12 and AIA were analysed using multivariate logistic analysis. Results showed that two polymorphisms and a haplotype in SLC6A12, rs499368 (P= 0.005; P(corr)= 0.03), rs557881 (non-synonymous C10R, P= 0.007; P(corr)= 0.04), and SLC6A12_BL1_ht1 (P= 0.009; P(corr)= 0.05) respectively, were significantly associated with AIA after multiple testing corrections. In addition, SNPs of SLC6A12 were significantly associated with the fall rate of FEV(1) by aspirin provocation suggesting that SLC6A12 could affect reversibility of lung function abnormalities in AIA patients. Although these results are preliminary and future replications are needed to confirm these findings, this study showed evidence of association between variants in SLC6A12 and AIA occurrence among asthmatics in a Korean population.</P>

        A Preliminary Study on the Potential Protective Role of the Antioxidative Stress Markers of Cognitive Impairment: Glutathione and Glutathione Reductase

        Objective: To investigate the relationship between reduced glutathione (GSH), a key molecule of the antioxidant defense system in the blood, and glutathione reductase (GR), which reduces oxidized glutathione (glutathione disulfide [GSSG]) to GSH and maintains the redox balance, with the prevalence of Alzheimer’s dementia and cognitive decline. Methods: In all, 20 participants with Alzheimer’s dementia who completed the third follow-up clinical evaluation over 6 years were selected, and 20 participants with normal cognition were selected after age and sex matching. The GSH and GR concentrations were the independent variables. Clinical diagnosis and neurocognitive test scores were the dependent variables indicating cognitive status. Results: The higher the level of GR, the greater the possibility of having normal cognition than of developing Alzheimer’s dementia. Additionally, the higher the level of GR, the higher the neurocognitive test scores. However, this association was not significant for GSH. After 6 years, the conversion rate from normal cognition to cognitive impairment was significantly higher in the lower 50th percentile of the GR group than in the upper 50th percentile. Conclusion: The higher the GR, the lower the prevalence of Alzheimer’s dementia and incidence of cognitive impairment and the higher the cognitive test scores. Therefore, GR is a potential protective biomarker against Alzheimer’s dementia and cognitive decline.

        Quality of Head and Chalky Rice and Deterioration of Eating Quality by Chalky Rice

        Chalk, an opaque area in the rice grain, is an important quality characteristic in rice and occurs most commonly when grains are exposed to high temperatures during development. Chalky rice decreases the value of rice because of its undesirable appearance and eating quality for consumers. We investigated the chemical composition, morphological structure, cooking, texture properties of cooked rice, and pasting and gelatinization properties to evaluate the reason for the deterioration in eating quality of chalky rice. The ultra-structure of chalky kernels differed greatly from that of head rice. Chalky grain had more air spaces, a disordered cellular structure, and rounder amyloplasts than head rice. Chalky rice absorbed water more rapidly during cooking and had a slightly larger expansion volume than head rice, confirming its loose starch granule structure. The lower amylose content of chalky kernels caused a more soluble solid and higher iodine index during cooking indicating their lower eating quality compared to translucent kernels. The lower gelatinization temperature of chalky kernels was suggested to be related to a more short-branch chain amylopectin structure. Chalky kernels resulted in a harder and less adhesive (or sticky) texture of cooked rice, requiring more time for chewing than those of head kernels, caused by its low protein content. The palatability of cooked rice showed a linear decrease with increasing chalky rice proportion in the sensory evaluation. In amylography, the peak and final viscosities greatly decreased when the chalky rice proportion increased from 0 to 5%. It indicated that chalky rice kernels induce quality deterioration even at 5%.

        알코올중독자의 대인관계와 자살생각 간의 관계: 우울의 매개효과를 중심으로

        본 연구의 목적은 알코올중독자의 대인관계와 자살생각 간의 관계에서 우울이 어떤 역할을 하는지를 파악하는 것이다. 연구대상자는 서울 및 경기 지역에 위치한 정신병원 6곳에 알코올중독 또는 알코올남용으로 입원한 환자 총 151명이며, 다중회귀분석과 Baron&Kenny(1986)의 방식을 통하여 매개효과를 검증하였다. 본 연구의 결과는 다음과 같다. 첫째, 연구대상자들의 자살시도 횟수는 일반성인의 자살 시도율과 비교하면 약 13배 높았고, 자살생각은 평생 유병률보다 약 3배 높게 나타났다. 둘째, 우울과 대인 관계, 자살시도횟수, 음주심각성, 소득, 연령은 자살생각에 직접적인 영향을 미치는 것으로 나타났다. 셋째, 매개효과 검증에서 우울은 대인관계와 자살생각 간의 관계에서 완전매개역할을 하는 것으로 나타났다. 이러한 연구결과를 토대로 알코올중독자의 자살 위험성을 사정하고 개입할 수 있는 실천가들의 교육 및 훈련, 자살 고위험군에 대한 관리체계구축 등과 같은 사회복지영역에서의 실천적 함의를 논하였다. The purpose of this study was to investigate the role of depression in the relationship between interpersonal relationships and suicide ideation among alcoholics. The study subjects included a total of 151 patients hospitalized at six psychiatric hospitals in Seoul and Gyeonggi Province for alcoholism or alcohol abuse. For data analysis, mediating effects were tested with multiple regression analysis and the Baron&Kenny (1986) method. The study findings were as follows: first, the suicide attempt rate of the subjects was approximately 13 times higher than that of non-alcoholic adults, while the subjects` suicide ideation rate was about three times higher. Secondly, depression, interpersonal relationships, number of suicide attempts, alcohol problem, income, and age had direct influence on suicide ideation. Finally, the test results of mediating effects showed that depression had full mediating effect between interpersonal relationships and suicide ideation. Based on these findings, the study discusses practice implications for the field of social welfare such as the need for educating and training practitioners capable of considering suicide risk of alcoholics, as well as the need for tailored intervention programs and their management systems for the high suicide risk group.

        Outreach results of domestic mushroom varieties for mushroom farms

        This study was carried out to decrease the royalties through the dissemination of domestic mushroom varieties by following UPOV (Union Internationale Pour la Protaection des Obtentions Vegetablue: the international union for the protection of new varieties of plants). Although RDA has bred various mushrooms of 25 species of 78 varieties since 1990, most of these were not cultivated in mushroom farms due to two reasons. One is the weaknees of spawn supply system caused by increasing own spawn production and the other is the difficulty in replacing a new variety due to the year-round production in bottle cultivation. Therefore, we made a program to disseminate domestic mushroom varieties. In 2010, five species of 14 varieties were provided as a form of spawn for 43 farms that do not have their own cultivation facility and 11 species of 18 varieties were provided for 45 farms that were equipped with their own cultivation facility. In addition, the breeders explained the cultural characteristics of their own varieties and consulted cultural farms. Since these activities were helpful to mushroom farmers, we plan to open the presentation for mushroom instructor in February 2011 with the consideration of the opinion of mushroom farmers to fully disseminate the spawn of domestic mushroom varieties for the bottle cultivation farms.
