      • Spring emergence pattern and its forecasting of Grapholita molesta (Busck) (Lepidoptera: Tortricidae) in stone fruit orchards

        The study was conducted to investigate the spring emergence pattern of G. molesta and to forecast the emerging time of overwintering G. molesta on tree fruit orchards. G. molesta is one of major insect pests on fruit trees in Korea. The host range of G. molesta includes many economically important tree fruit plants such as apple, pear, peach and plum. The overwintering G. molesta emerge from late March as an adult lay eggs on the shoot of peach or fruits of apple, plum and peach. Therefore, it is important to understand the biofix and to forecast the emerging peak period of overwintering G. molesta for establishing the pest management strategy. The pheromone trap of G. molesta has been utilized to monitor the population density in apple orchard. The commercial stick trap (GreenAgroTech) and lure (Z8-12:AC, E8-12:Ac, Z8-12:OH, 95:5:1) was set to monitor the population density of G. molesta on each place (56 different fruit orchards). The record of temperature was received from meteorological center close to monitoring orchards. The parameters for forecasting the emerging time and peak period of overwintering G. molesta were calculated from the results of Yang et al (1997 and 2001). Although the estimated biofix of G. molesta was not fitted well, the peak period of overwintering G. molesta was explained by linear regression model. The spring emergence pattern of G. molesta was presented differently related to host plant and geographical location. The peak period of G. molesta at the same mornitoring county was presented differently according to host plant. The synchronization between host plant and G. molesta may be studied to figure out the spring emerging time of overwintering G. molesta.

      • Forecasting the phenology of important lepidopterous pests, Grapholita molesta, Grapholita dimorpha and Carposina sasakii: II. Case study of apple

        Grapholita molesta, G. dimorpha and C. sasakii as “internal feeders” are important apple pests in Korea. Three species overwinters around and in apple orchards. New young larvae of three species bore into new shoots or fruits and then feed inside apple. When mature larvae escape from fruits they make holes that reduces the commercial value of fruit. Therefore, understanding the phenological distribution of three species is critical to establish the precise management system for reducing three species population. The study was conducted to investigate the adult emergence of G. molesta, G. dimorpha and C. sasakii using pheromone traps and to forecast the cumulative proportion of each population. This study is second part of consecutive experiment. Data collection was carried out on three commercial apple orchards and one experimental orchard of Giran in 2010 and 2011. The experimental process was same in the study of plum. More than 50% of G. molesta male was occurred in spring season (within 500 degree-days), 2010 and 2011. The adult emergence of G. dimorpha and C. sasakii was linear and sigmoidal pattern in each year. The phenology of C. sasakii was explained well by nonlinear functions and the equation 3, 6, 8 and 11 were selected based on AICc and BIC. The selected equations were validated by the data of present year (2011) in each region. The performance of G. molesta and G. dimorpha was analyzed well by bimodal functions. The importance of phenological model is discussed to develop and maintain a more precise system for multiple pest management on apple orchard.

        Plant Protective and Growth Promoting Effects of Seed Endophytes in Soybean Plants

        Seed-borne diseases reduce not only the seed germination and seedling growth but also seed quality, resulting in the significant yield loss in crop production. Plant seed harbors diverse microbes termed endophytes other than pathogens inside it. However, their roles and application to agricultures were rarely understood and explored to date. Recently, we had isolated from soybean seeds culturable endophytes exhibiting in-vitro antagonistic activities against common bacterial and fungal seed-borne pathogens. In this study, we evaluated effects of seed treatment with endophytes on plant growth and protection against the common seed-borne pathogens: four fungal pathogens (Cercospora sojina, C. kikuchii, Septoria glycines, Diaporthe eres) and two bacterial pathogens (Xanthomonas axonopodis pv. glycines, Pseudomonas syringae pv. tabaci). Our experiments showed that treatment of soybean seeds with seed endophytes clearly offer protection against seed-borne pathogens. We also found that some of the endophytes promote plant growth in addition to the disease suppression. Taken together, our results demonstrate agricultural potential of seed endophytes in crop protection.

        Dual effects of isoflavonoids from <i>Pueraria lobata</i> roots on estrogenic activity and anti-proliferation of MCF-7 human breast carcinoma cells

        <P><B>Abstract</B></P> <P> <I>Pueraria lobata</I> root (PLR), well known as Kudzu root, has recently become commercially available in Western dietary supplements for menopausal symptoms. The scientific basis for its action has been attributed to the action of phytoestrogens. This study aimed to investigate the estrogen-like activity of isoflavonoids isolated from <I>P. lobata</I> root and their safety with respect to their effect on breast cancer cell proliferation. In an E-screen assay, crude MeOH extract of PLR significantly increased the proliferation of MCF-7 cells in a concentration-dependent manner. Among the four fractions obtained by solvent fractionation of MeOH extract, the <I>n</I>-BuOH fraction had significant estrogen-like activities at all concentrations tested. Phytochemical analysis of the <I>n</I>-BuOH fraction led to the isolation of 10 isoflavones (<B>1</B>–<B>10</B>), among which genistein (<B>10</B>) had significant estrogen-like activities at all concentrations tested. These activities were significantly enhanced by treatment with genistein and 17β-estradiol compared with 17β-estradiol alone, and this effect was mediated by decreased expression of estrogen receptor (ER)α and phospho-ERα in MCF-7 cells. In a cell cytotoxicity assay, genistein (<B>10</B>) exhibited significant cytotoxicity in both ER-positive MCF-7 and ER-negative MDA-MB-231 breast cancer cells. This cytotoxicity was characterized by the induction of apoptotic cells stained with annexin V conjugated with Alexa Fluor 488 and involved activation of mitochondria-independent and -dependent apoptosis pathways in MCF-7 cells.       • Urinary Metabolomic Profiling to Identify Potential Biomarkers for the Diagnosis of Behcet’s Disease by Gas Chromatography/Time-of-Flight−Mass Spectrometry

        <P>Diagnosing Behcet’s disease (BD) is challenging because of the lack of a diagnostic biomarker. The purposes of this study were to investigate distinctive metabolic changes in urine samples of BD patients and to identify urinary metabolic biomarkers for diagnosis of BD using gas chromatography/time-of-flight–mass spectrometry (GC/TOF−MS). Metabolomic profiling of urine samples from 44 BD patients and 41 healthy controls (HC) were assessed using GC/TOF−MS, in conjunction with multivariate statistical analysis. A total of 110 urinary metabolites were identified. The urine metabolite profiles obtained from GC/TOF−MS analysis could distinguish BD patients from the HC group in the discovery set. The parameter values of the orthogonal partial least squared-discrimination analysis (OPLS-DA) model were <I>R</I><SUP>2</SUP><I>X</I> of 0.231, <I>R</I><SUP>2</SUP><I>Y</I> of 0.804, and <I>Q</I><SUP>2</SUP> of 0.598. A biomarker panel composed of guanine, pyrrole-2-carboxylate, 3-hydroxypyridine, mannose, <SMALL>L</SMALL>-citrulline, galactonate, isothreonate, sedoheptuloses, hypoxanthine, and gluconic acid lactone were selected and adequately validated as putative biomarkers of BD (sensitivity 96.7%, specificity 93.3%, area under the curve 0.974). OPLS-DA showed clear discrimination of BD and HC groups by a biomarker panel of ten metabolites in the independent set (accuracy 88%). We demonstrated characteristic urinary metabolic profiles and potential urinary metabolite biomarkers that have clinical value in the diagnosis of BD using GC/TOF−MS.</P>

        Rheumatoid Factor Positivity is Associated with Lower Bone Mass in Korean Male Health Examinees without Clinically Apparent Arthritis

        Objective. This cross-sectional study aimed to investigate the association between rheumatoid factor (RF) positivity and bone mineral density (BMD) in male Korean subjects without any history of joint disease. Methods. Of 84,344 males who had undergone a comprehensive health checkup program in 2012, 1,390 male health examinees were recruited, whose BMD and RF results were available. A RF titer ≥20 IU/mL was considered positive. BMD was measured at lumbar spine (L1∼L4) or hip (femoral neck and total hip) by dual-energy X-ray absorptiometry. Results. The association between RF positivity and BMD was assessed by multiple linear regression analysis. The mean age was 52.7±10.9 years (range 19∼88 years), and RF was detected in 64 subjects (4.6%). Demographics and laboratory data were not different between RF-positive and -negative subjects except hepatitis B surface antigen (HBsAg), which was more frequently seen in RF-positive subjects (15.6% vs. 4.3%, p=0.001). RF-positive subjects had significantly lower BMD compared to RF-negative subjects in lumbar spine but not in total hip regardless of the existence of HBsAg (1.17±0.16 g/㎠ vs. 1.10±0.18 g/㎠, p=0.002 in total subjects; 1.17±0.16 g/㎠ vs. 1.10±0.18 g/㎠, p=0.004 in HBsAg-negative subjects). After adjusting for multiple confounders, RF positivity was negatively associated with lumbar spine BMD (B=-0.088 and standard error=0.035, p=0.011). Conclusion. Our results show that the presence of RF could have an unfavorable impact on bone density in apparently normal males. Additional studies to elucidate the osteoimmunological mechanism of rheumatoid factor are warranted. (J Rheum Dis 2019;26:31-40)

      • Potential metabolomic biomarkers for reliable diagnosis of Behcet's disease using gas chromatography/ time-of-flight-mass spectrometry

        <P><B>Abstract</B></P> <P><B>Objectives</B></P> <P>Although many diagnostic criteria of Behcet's disease (BD) have been developed and revised by experts, diagnosing BD is still complicated and challenging. No metabolomic studies on serum have been attempted to improve the diagnosis and to identify potential biomarkers of BD. The purposes of this study were to investigate distinctive metabolic changes in serum samples of BD patients and to identify metabolic candidate biomarkers for reliable diagnosis of BD using the metabolomics platform.</P> <P><B>Methods</B></P> <P>Metabolomic profiling of 90 serum samples from 45 BD patients and 45 healthy controls (HCs) were performed via gas chromatography with time-of-flight mass spectrometry (GC/TOF-MS) with multivariate statistical analyses.</P> <P><B>Results</B></P> <P>A total of 104 metabolites were identified from samples. The serum metabolite profiles obtained from GC/TOF-MS analysis can distinguish BD patients from HC group in discovery set. The variation values of the partial least squared-discrimination analysis (PLS-DA) model are R<SUP>2</SUP>X of 0.246, R<SUP>2</SUP>Y of 0.913 and Q<SUP>2</SUP> of 0.852, respectively, indicating strong explanation and prediction capabilities of the model. A panel of five metabolic biomarkers, namely, decanoic acid, fructose, tagatose, linoleic acid and oleic acid were selected and adequately validated as putative biomarkers of BD (sensitivity 100%, specificity 97.1%, area under the curve 0.998) in the discovery set and independent set. The PLS_DA model showed clear discrimination of BD and HC groups by the five metabolic biomarkers in independent set.</P> <P><B>Conclusions</B></P> <P>This is the first report on characteristic metabolic profiles and potential metabolite biomarkers in serum for reliable diagnosis of BD using GC/TOF-MS.</P>
