        TAp73 and ΔNp73 Have Opposing Roles in 5-aza-2'-Deoxycytidine-Induced Apoptosis in Breast Cancer Cells

        The p73 gene contains an extrinsic P1 promoter and an intrinsic P2 promoter, controlling the transcription of the pro-apoptotic TAp73 isoform and the anti-apoptotic ΔΝp73 isoform, respectively. The DNA methylation status of both promoters act equally in the epigenetic transcriptional regulation of their relevant isoforms. The aim of this study was to analyze the different effects of these p73 isoforms in 5-aza-2'-deoxycytidine (5-aza-dC)-induced apoptosis in breast cancer cells. We investigated the effects of the DNA demethylation agent, 5-aza-dC, on the T-47D breast cancer cell line, and evaluated the methylation status of the p73 promoters and expression of TAp73 and ΔNp73. Furthermore, we assessed the expression of p53 and p73 isoforms in 5-aza-dC-treated T-47D cells and p53 knockout cells. 5-aza-dC induced significant anti-tumor effects in T-47D cells, including inhibition of cell viability, G1 phase arrest and apoptosis. This was associated with p73 promoter demethylation and a concomitant increase in TAp73 mRNA and protein expression. In contrast, the methylation status of promoter P2 was not associated with ΔNp73 mRNA or protein levels. Furthermore, demethylation of P2 failed to inhibit the expression of ΔNp73 with 5-aza-dC in the p53 knockdown cell model. Our study suggests that demethylation of the P1 and P2 promoters has opposite effects on the expression of p73 isoforms, namely up-regulation of TAp73 and down-regulation of ΔΝp73. We also demonstrate that p53 likely contributes to 5-aza-dC-induced ΔNp73 transcriptional inactivation in breast cancer cells.

        Design of Drug-Delivery Micro-Wheel and Auto-Rolling Control by Change of Gravity Center

        A micro-dosing system of overall size 5×5×4.2 mm3 for drug delivery is proposed and presented. The drug delivery system mainlyconsists of a micro-wheel and a micro-drug release mechanism. The motion of the micro-wheel is controlled by its center of gravityvia a running disk, which is placed within the hollow micro-wheel and attracted by the actuated micro-solenoids fabricated on theinner wall of micro-wheel in shift. In addition, the micro-wheel is controlled to roll forwards/backwards to the designated locationby two sliding mode control strategies: one for long-distance motion (to transport the drug to the vicinity of the spots under disease)and the other for short-distance motion (to decelerate down and stop at the exact drug-release location). On the other hand, the microdrugrelease mechanism is composed by a cantilever beam and a chamber filled up by medicine. The pyramid tip of the cantileverbeam deflected by the applied electrostatic force is designed to penetrate the micro-film which seals the chamber so that the medicinecan be released at the specified spot.

      • Insulin Resistance Reduces Sensitivity to Cis-Platinum and Promotes Adhesion, Migration and Invasion in HepG2 Cells

        The liver is normally the major site of glucose metabolism in intact organisms and the most important target organ for the action of insulin. It has been widely accepted that insulin resistance (IR) is closely associated with postoperative recurrence of hepatocellular carcinoma (HCC). However, the relationship between IR and drug resistance in liver cancer cells is unclear. In the present study, IR was induced in HepG2 cells via incubation with a high concentration of insulin. Once the insulin-resistant cell line was established, the stability of HepG2/IR cells was further tested via incubation in insulin-free medium for another 72h. Afterwards, the biological effects of insulin resistance on adhesion, migration, invasion and sensitivity to cis-platinum (DDP) of cells were determined. The results indicated that glucose consumption was reduced in insulin-resistant cells. In addition, the expression of the insulin receptor and glucose transportor-2 was downregulated. Furthermore, HepG2/IR cells displayed markedly enhanced adhesion, migration, and invasion. Most importantly, these cells exhibited a lower sensitivity to DDP. By contrast, HepG2/IR cells exhibited decreased adhesion and invasion after treatment with the insulin sensitizer pioglitazone hydrochloride. The results suggest that IR is closely related to drug resistance as well as adhesion, migration, and invasion in HepG2 cells. These findings may help explain the clinical observation of limited efficacy for chemotherapy on a background of IR, which promotes the invasion and migration of cancer cells.

        Effects of co-ion initial concentration ratio on removal of Pb2+ from aqueous solution by modified sugarcane bagasse

        A modified sugarcane bagasse (SCB) fixed bed column was used to remove Pb2+ from aqueous solution. To determine the optimal condition for Pb2+ separation, Ca2+ was chosen as the model interfering ion, and effects of Ca2+ and Pb2+ initial concentration ratio (C0 Ca : C0 Pb) on the adsorption of Pb2+ were investigated. Results showed that adsorption amount ratio of Ca2+ and Pb2+ (qe Ca : qe Pb) had a good linear relationship with C0 Ca : C0 Pb. Mass ratio of Pb2+ absorbed on the modified SCB was higher than 95% at C0 Ca : C0 Pb<1.95, illustrating that Pb2+ could be selectively removed from aqueous solution. To verify that, simulated waste water containing co-ions of K+, Na+, Cd2+ and Ca2+ was treated, and results showed that the equilibrium amount of Pb2+, K+, Na+, Cd2+ and Ca2+ adsorbed was 134.14, 0.083, 0.058, 1.28, and 1.28mg g−1, respectively, demonstrating that the modified SCB could be used to remove Pb2+ from aqueous solution in the investigated range.

        Investigation of in-train stability and safety assessment for railway vehicles during braking

        In-train stability of railway vehicles has becoming a major concern for railway vehicles, which refers to the jackknifing behavior of couplers under large in-train forces. For the train to train rescue scenario, braking induced impacts from couplers can adversely affect the dynamic performance of the coupled train. It is indicated from field tests that in-train forces if combined with large rotational angles of couplers can produce vertical components, which will further lead to the interference of adjacent carbodies and structural damages. In this paper, the dynamic model of the train and coupler system is developed. The model verifications are conducted by comparing the calculated responses with the tested results. The safety indices are formulated on the basis of which the running safety of the coupled train is evaluated. The propelling test in the laboratory is conducted to reproduce the coupler jackknifing behavior. The quasi-static analysis and anti-jackknifing mechanism under compressing in-train forces are analysed. Parametric studies are then conducted to propose some limitations for the application of train to train rescue. It is indicated from numerical and testing results that the decrease of the braking deceleration or a limitation of the free rotational angle of couplers is beneficial to lower the amplitude of braking induced impacts.

      • Selecting and Identifying Lactic Acid Bacteria in Sourdough and Their Effect on Sourdough Physical and Chemical Properties

        Sourdough is traditionally used as the leavening agent for bread making and a potential source of probiotics. In this study, sourdoughs from seven restaurants in Taiwan were collected. The lactic acid bacteria strains were isolated and identified by 16S rDNA sequencing from these sourdoughs. The following lactic acid bacteria species were identified: Lactobacillus pentosus, Lactobacillus paracasei, Lactobacillus casei, Lactobacillus plantarum, Lactobacillus sanfranciscensis. Five lactic acid bacteria strains, Lactobacillus paracasei LYC1242, Lactobacillus casei LYC1244, Lactobacillus plantarum LYC1245 and Lactobacillus casei LYC1246, were selected and singly used to ferment wheat flour. The pH value of various sourdoughs decreased to pH3 from pH5 and kelp at pH3. The titratable acidity and viscosity of various sourdoughs increased significantly compared to spontaneous fermented flour. However, there is no significant difference in titratable acidity between various sourdoughs. The sourdough fermented with L. casei LYC1244 had highest cell count. These studies provided preliminary results for choosing the best lactic acid bacteria starter strain to develop probiotic sourdough bread.

      • A BIST RAM Architecture With Parallel Testing in a Microprogram ROM

        Symmetrically simple test patterns for parallel testing of row/column pattern-sensitive faults in RAMS are proposed in the paper. Based on the concept of the maximum leakage current among RAM cells, the worst case testing becomes the critically efficient way of the RAM testing. Due to the simplexity of generating test patterns, we can implement a BIST RAM with test procedures stored in a microprogram ROM to reduce the cost and time of testing. The test complexity of row/column pattern-sensitive faults is reduced to O(N) in an N bits RAM as compared with O(N^(3/2)) of [5].

        CagL polymorphisms between East Asian and Western Helicobacter pylori are associated with different abilities to induce IL-8 secretion

        Helicobacter pylori colonizes human gastric mucosa. Its infection is associated with gastric diseases including gastric cancer. CagA is one of the most important toxins produced by H. pylori. It is related to gastric cancer which can be injected into host cells via a type IV secretion system (T4SS). CagL is a structural component of T4SS apparatus, which triggers host cell signaling pathway. It has been reported that CagL polymorphisms may influence the severity of disease development. To explore the contribution of CagL polymorphisms between East Asian and Western H. pylori in pathogenesis, cagL gene in G27 H. pylori was swapped by K74 cagL which is identical to East Asian CagL consensus sequence and by Western 26695 H. pylori, resulting in G27ΔcagL/cagLK74 and G27ΔcagL/cagL26695, respectively. Intriguingly, G27ΔcagL/ cagLK74 showed significantly less ability of IL-8 induction than G27ΔcagL/cagL26695 while displayed similar abilities of CagA phosphorylation, and cell elongation. Taken together, this study suggests that the CagL polymorphism may influence IL-8 induction, and K74 CagL has less ability to induce IL-8 secretion than G27 or 26695 CagL. Further research should address how the different capabilities of IL-8 induction between intraspecies-CagL are associated with the large differences of the incidence of gastric cancer between East Asian and Western countries.
