      • Induction of heme oxygenase-1 protects against podocyte apoptosis under diabetic conditions

        Heme oxygenase-1 (HO-1) is an anti-oxidant enzyme normally upregulated in response to oxidant injury. Here we determined the role of HO-1 in podocyte apoptosis in glomeruli of streptozotocin-treated rats and in immortalized mouse podocytes cultured in media containing normal or high glucose. HO-1 expression, its activity, the ratio of Bax/Bcl-2 protein, and active caspase-3 fragments were all significantly higher in isolated glomeruli of diabetic rats and in high glucose–treated podocytes. These increases were inhibited by zinc protoporphyrin treatment of the rats or by HO-1 siRNA treatment of the podocytes in culture. The number of apoptotic cells was also significantly increased in the glomeruli of diabetic rats and in high glucose–treated podocytes. Inhibition of HO-1 accentuated the increase in apoptotic cells both in vivo and in vitro. Our findings suggest that HO-1 expression protects against podocyte apoptosis under diabetic conditions.

        Heme oxygenase-1 induced by desoxo-narchinol-A attenuated the severity of acute pancreatitis via blockade of neutrophil infiltration

        <P><B>Abstract</B></P> <P>Heme oxygenase-1 (HO-1) has an anti-inflammatory action in acute pancreatitis (AP). However, its mechanism of action and natural compounds/drugs to induce HO-1 in pancreas are not well understood. In this study, we investigated the regulatory mechanisms of HO-1 during AP using desoxo-narchinol-A (DN), the natural compound inducing HO-1 in the pancreas. Female C57/BL6 Mice were intraperitoneally injected with supramaximal concentrations of cerulein (50 μg/kg) hourly for 6 h to induce AP. DMSO or DN was administered intraperitoneally, then mice were sacrificed 6 h after the final cerulein injection. Administration of DN increased pancreatic HO-1 expression through activation of activating protein-1, mediated by mitogen-activated protein kinases. Furthermore, DN treatment reduced the pancreatic weight-to-body weight ratio as well as production of digestive enzymes and pro-inflammatory cytokines. Inhibition of HO-1 by tin protoporphyrin IX abolished the protective effects of DN on pancreatic damage. Additionally, DN treatment inhibited neutrophil infiltration into the pancreas via regulation of chemokine (C-X-C motif) ligand 2 (CXCL2) by HO-1.       • Curcumin protects retinal pigment epithelial cells against oxidative stress via induction of heme oxygenase-1 expression and reduction of reactive oxygen

        <P><B>Purpose</B></P><P>To determine whether curcumin induces expression of the defensive enzyme heme oxygenase-1 (HO-1) and protects cells against oxidative stress in cultured human retinal pigment epithelial cells.</P><P><B>Methods</B></P><P>Effective concentrations and toxicities of curcumin were determined after 3 h of curcumin treatment with the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. Confluent human retinal pigment epithelium cell lines (ARPE-19) were preincubated with curcumin and oxidatively challenged with H<SUB>2</SUB>O<SUB>2</SUB>. HO-1 expression was determined with western blot analysis. To confirm the protective role of HO-1 in oxidative stress, small interfering RNA (siRNA) against HO-1 or inhibitor of HO-1 was treated with curcumin in retinal pigment epithelium cells. Intracellular levels of reactive oxygen species (ROS) were measured with flow cytometry and confocal microscopy. Apoptosis was evaluated with Annexin V-fluoroscein isothiocyanate staining.</P><P><B>Results</B></P><P>Curcumin had little cytotoxicity at concentrations less than 30 μM, and HO-1 expression was the highest at the 15 μM concentration. At this concentration, curcumin also increased the cytoprotective effect against the oxidative stress of H<SUB>2</SUB>O<SUB>2</SUB> through the reduction of ROS levels in human retinal pigment epithelial cells. Curcumin’s effect on the reduction of ROS was mediated by the increase in HO-1 expression.</P><P><B>Conclusions</B></P><P>Curcumin upregulated the oxidative stress defense enzyme HO-1 and may protect human retinal pigment epithelial cells against oxidative stress by reducing ROS levels.</P>

      • 동하중을 받는 FRAME 구조물의 확률유한요소해석 및 신뢰성해석

        Most dynamic systems have various random properties in excitation and system parameters. Therefore, in this paper, reliability analysis for structural response is proposed fro the linear dynamic system with random properties in both excitation and system parameters. The system parameters and responses with random properties are modeled by the perturbation technique. A displacement and bending moment of frame structure is acquired in Stochastic Finite Element Method Program(SFEMP) and Dynamic Stochastic Finite Element Method Program(DSFEMP) using perturbation method, and Direct Monte Carlo Simulation Program(DMCSP) is used to verify and compare previous results. Comparing outputs of SFEMP and DSFEMP with them of DMCSP, it show that they are similar to each other. Also DMCSP need much time to analyze a complicated structure, but SFEMP and DSFEMP can reduce quite time. In this study, it is assumed that excitations, system parameters and response are Gaussian. The results of response for the analysis are compared to those of numerical simulation. Also the probability of failure are calculated for using Advanced First Order Second Moment(AFOSM) of the general reliability analysis method.

        우수방사성의약품 생산시설 개발

        Manufacturing facilities of the pharmaceuticals must meet certain level of the cleanness required so that foreign substances such as dust, moisture, heat, microorganism, or virus do not contaminate the product. In case of radiopharmaceuticals for medical treatment and diagnosis, not only should the operators and environment be protected from radiation but also need to be isolated from the foreign contaminant. Therefore, manufacturing facilities for radiopharmaceuticals must satisfy the design standards of both hot cell and clean room which are specified by GNP. However, standards of maintaining negative pressure for preventing spread of radioactive contaminant in isolated facilities conflict with the standards of maintaining positive pressure for keeping cleanness. To solve this problem, are pressure of hot cell was designed lower than in the adjacent area to meet standards of the radiation safety. To keep higher cleanness in certain part of the hot cell for filling, minimal relative positive pressure allows. In order to effectively maintain the cleanness that is required for production of Tc-99m generator, which takes 70% of whole demand of radiopharmaceuticals, the rooms placed in each side of production room are used as a buffer area and three lead hot cells are installed in production room. In this research, we established the appropriate engineered design concept for Tc-99m generator manufacturing facility, which satisfies both GMP cleanness standard for preventing particles, bacteria, other contaminants and the regulations of radiation safety for supervising and controlling the amount of radiation exposure and exhausted radioactivity. And the concept of multi-buffer zones is introduced to apply negative air pressure for hot cell with first priority and to continue relative positive air pressure for clean room.

        당근을 이용한 식초 및 음료수의 개발

        당근파쇄물을 Termamyl, Dextrozyme 및 Viscozyme과 같은 당질 가수분해 효소로 가수분해시킨 당화물을 Saccharomyces cerevisiae로 발효시켜 얻어진 알코올 용액을 기질로하여 Acetobacter aceti 배양에 의하여 식초를 제조하고, 이와같은 제조한 식초를 이용하여 여러 형태의 식음료를 조제하여 관능검사를 실시하였다. 먼저, 당질가수분해효소로 당근파쇄물을 가수분해아여 환원당 농도가 0.6%이던 당근 파쇄물의 환원당 농도가 7.0%로 증가된 당근 당화물을 제조할 수 있었다. 이어서, 당근당화물에 포도당을 첨가하여 환원당 농도를 20%로 조절한 후, 알코올 발효시켜 알코올농도를 6.0%로 조절한 후, 종초(Acetobacer aceti의 전 배양액)를 가해 배지의 초기산도가 1.0% 부근이 되도록 조절하여 30℃에서 정치배양 및 진탕 배양시켰다. 진탕 배양의 경우, 발효 22일 경에 최대 산도 7.0%에 도달하였고, 정치배양의 경우에는 발효 30일 경에 최대 산도 6.8%에 도달하였다. 이와같이 조제한 당근식초를 저온살균하고 이를 이용하여 제조한 식음료로는 액상과당을 10%가량 첨가하여 제조한 음료 시제품이 당근과즙과 각종 부원료를 첨가하여 제조한 시제품보다 높은 관능 평가 결과를 얻었다. To develop vinegar and beverage products from carrots, carbohydrates in carrots were hydrolyzed with hydrolyzing enzymes such as Termamyl, Dextrozyme, and Viscozyme. The hydrolysate was used as the substrates of alcohol and acetic acid fermentation by Saccharomyces cerevisiae and Acetobacter aceti, respectively. The sensory evaluation for the prepared vinegar and vinegar-containing beverages was carried out. After hydrolyzing crushed carrots, the saccharified product could be manufactured in the 7.0% ratio of deoxidized saccharification, whose concentration was originally 0.6%. After the concentration of reducing sugars was made up to 20% by adding glucose to the saccharified product, the alcohol fermentation was done at 30℃. After alcohol fermentation, 6.0% of alcohol content was composed in the liquid medium precultivated by acetobacter aceti and the acidity in the early stage was reached around 1.0%. While it arrived at the maximum acidity of 7.0% at the 22th day of fermentation in the case of shaking cultivation, the alcohol concentration in the culture was the maximum acidity of 6.8% at the 30th day of fermentation in the case of the static cultivation. In the sensory evaluation for the various kinds of beverage prepared with the carrot vinegar, the testing beverage manufactured with adding 10% of liquid fructose to the culture showed best results.

      • 유비쿼터스 센서와 ZigbeX를 이용한 독거노인 모니터링 시스템 구현

        The number of elderly people living alone increases with the trend of nuclear family in recent aging society and advances of health and medical technologies, where the safety of the elderly people becomes a big social issue. One of the safety system for them these days is that security guards regularly visit homes of the elderly living alone to check their safety. However, it is an inefficient system since it costs a great deal. So, a new efficient system with low cost using modern advanced technologies needs to be developed. In this paper, we implement a monitoring system for elderly living alone using ubiquitous sensor and zigbeX. The system can remotely determine the health status of elderly people and report to their hospitals. Since the system can be implemented with low cost and do the same job as security guards do, we expect that it should replace the existing expensive monitoring system.

      • Underactuated 유영하는 우주 매니퓰레이터에 대한 강인한 적응 직교 제어

        A free-floating space robot manipulator system is a robotic system that has a robot manipulator mounted on a space station or satellite which freely moves in reaction to the robot manipulator due to the dynamic coupling. In usual, a robot manipulator that has fewer number of joint actuators than the number of its total joints is so-called an underactuated manipulator. For the control in Cartesian space in which tasks of robot manipulators are planned, we assume that the number of active joints is greater than or equal to the number of configurational dimension of the manipulator's end-effector in Cartesian space. In this paper, a robust adaptive Cartesian control scheme of an underactuated free-floating space manipulator is proposed in the presence of the parametric uncertainty and disturbances. The proposed scheme does not need a prior knowledge of the exact dynamic parameters and uncertainty bounds. To show the feasibility and robustness of the proposed control scheme, simulation results for a six-link PUMA560 arm with three passive joints mounted on a free-floating space vehicle are presented.

      • 아산시 일대 수계의 수환경 요인 변화

        To understand the changes of water quality according to watershed, the four physiochemical characters of water quality, including water temperature, pH, electric conductivity and dissolved oxygen, were investigated from 3 July 1997 to 22 August 1997 at 122 sites in Asan city, Chungnam, Korea. Water temperature ranged from 16.8℃ to 35℃, pH from 6.4 to 10.6, dissolved oxygen from 3.5 mg/l to 19.4 mg/l, and electric conductivity from 6.2 mS/m to 66.3 mS/m. Among 4 characters, the electric conductivity did not show the significant differences between running water and still waters, like reservoir or lakes. only. These results suggest that the water quality around Asan city were worsened by the regional output of pollutant.
