Radiological Assessment of the Effect of Congenital C3–4 Synostosis on Adjacent Segments

Study Design: Retrospective case series. Purpose: To assess the effect of non-kyphotic aligned congenital C3–4 synostosis on the adjacent segment in 10 patients. Overview of Literature: In the cervical spine, fusion disease at the adjacent motion segments may be a risk factor for potential neurological compromise and death. Methods: Radiograms of 10 patients 13 to 69 years of age presenting with neck/shoulder discomfort or pain with or without trauma history were examined. C3–4 synostosis was found incidentally in all patients on routine examination radiographs of cervical spine. Results: Adjacent segment disease (ASD) was not found in the three patients younger than 39 years of age. Five of the 10 (50%) patients, including a 67-year-old man, did not develop spondylosis in any of the cervical mobile segments. Spondylosis was observed only in the caudal 1–2 mobile segments in the remaining five patients. The youngest was a 40-year-old male who had spondylosis in the two caudal mobile segments (C4–5 and C5–6). Spondylosis was limited to the two close caudal mobile segments and was not in the cranial segments. Flaring of the lower part of synostotic vertebra associated with advanced narrowed degenerate disc was evident in five patients. Conclusions: Mobile segment spondylosis in the individuals with congenital monosegment C3–4 synostosis over age of 40 years may be a natural manifestation of aging and is not solely an adjacent segment disease directly and fully related with congenital C3–4 synostosis.
Eclosion hormone functions in larva-pupa-adult ecdysis in Leptinotarsa decemlineata

In Drosophila melanogaster, ecdysis-triggering hormone (ETH) and eclosion hormone (EH) mutually enhance the release of each other through a positive feedback loop. Both are involved in the stimulation of ecdysis behavior series and molting-related physiological preparations. However, whether 20-hydroxyecdysone (20E) controls eh expression and whether EH plays a similar role in non-Drosophilid insects remain to be determined. In Lep tinotarsa decemlineata, we found that Ldeh mRNA levels were high in the early and/or late stages and low in the intermediate period within each larval instar, indicating that the mRNA peaks positively correlate with 20E pulses. Consistent with the indication, we confirmed that 20E signaling stimulated the transcription of Ldeh. RNA interference (RNAi)-aided knockdown of Ldeh at the final instar stage slightly impaired pupation and signifi cantly affected wing expansion. Around 20% of the Ldeh RNAi larvae remained as prepupae, completely wrapped in the old larval cuticles. These prepupae gradually darkened, dried and withered, and finally died. The remaining (about 80%) Ldeh hypomorphs became pupae and emerged as abnormal adults, bearing smaller and wrinkle elytra and hindwings. The adult tracheae in the Ldeh hypomorphs were full of liquid. In addition, the low pupation rate and high deformed adult proportion cannot be rescued by ingestion of either 20E or juvenile hormone. Thus, EH is critical for ecdysis and trachea clearance in L. decemlineata. The failure of trachea clearance may disenable air-swallowing during larva-pupa-adult transition and thus impacts wing growth and expansion.
Active mass driver control system for suppressing wind-induced vibration of the Canton Tower

In order to suppress the wind-induced vibrations of the Canton Tower, a pair of active mass river (AMD) systems has been installed on the top of the main structure. The structural principal directions n which the bending modes of the structure are uncoupled are proposed and verified based on the rthogonal projection approach. For the vibration control design in the principal X direction, the simplified odel of the structure is developed based on the finite element model and modified according to the field measurements under wind excitations. The AMD system driven by permanent magnet synchronous linear otors are adopted. The dynamical models of the AMD subsystems are determined according to the pen-loop test results by using nonlinear least square fitting method. The continuous variable gain feedback VGF) control strategy is adopted to make the AMD system adaptive to the variation in the intensity of wind excitations. Finally, the field tests of free vibration control are carried out. The field test results of AMD control show that the damping ratio of the first vibration mode increases up to 11 times of the original value without control.
Cat NPC2, a Newly Identified Allergen, With High Cross-Reactivity to Can f 7

Purpose: Pet-derived allergens are the common indoor inhalant allergens. Among them, cat and dog allergens constitute more than 80% of animal allergic patients, which greatly affect the quality-of-life of patients and increase the burden of social health care. The aim of this study was to identify Cat-Niemann pick type C2 (NPC2) protein, a homologue of Can f 7, as a new allergen. Methods: Cat-NPC2 complementary DNA (cDNA) was cloned and optimized for amplification and expression in Escherichia coli. Then, recombinant Cat-NPC2 (rCat-NPC2) was purified by Ni2+ affinity chromatography. The allergenicity was assessed by enzyme-linked immunosorbent assay (ELISA), western blot and basophil activation test (BAT). Based on the sequence similarity, the cross-reactivity between Cat-NPC2 and Can f 7 was investigated by inhibition ELISA. Circular dichroism spectroscopy and homology modeling were used to characterize the structure of Cat-NPC2. Results: The cDNA sequence of Cat-NPC2 was cloned with a 450-bp open reading frame coding for 149 amino acids (GenBank MN_737596). The condon-optimized NPC2 gene was subcloned and expressed in E. coli with a molecular weight of 18.9 kDa. The native Cat-NPC2 was detected in cat dander extracts. The allergenicity determined by ELISA, western blot and BAT suggested at least 14.5% cat-allergic patients displayed high specific immunoglobulin E (IgE) recognition of Cat-NPC2. The predicted structure of Cat-NPC2 was found to consist of 7 β-strands arranged in 2 β-sheets. An ELISA based assay showed that rCat-NPC2 bound to cholesterol in a dose dependent manner. Based on the structure and sequence similarities, IgE cross-reactivity was demonstrated between Cat-NPC2 and Can f 7/Der f 2. Conclusions: In the study, a novel cat allergen, belonging to the NPC2 protein family, was identified and characterized at both molecular and immunological levels. The study will offer a deeper understanding of cat allergens and improve a component-resolved diagnosis in pet allergy.

Graphene-based biosensor and chiral sensor have made significant progress in recent years. Because of the similarity of the optical enantiomers, enantioseparation is perhaps the most subtle to achieve. Developing more effectively chiral separation techniques based on graphene is highly desirable. Herein, pristine graphene was prepared and then applicated to assist resolution of two racemic drugs of propranolol and ofloxacin using thin-layer chromatography (TLC). In comparison with TLC chiral separation by only using pure D-(-)-tartaric acid as a selector with relatively low degree of separation, a mixture of graphene and D-(-)-tartaric acid is more attractive, efficient and ready available. The high specific resolution ability for racemic compounds aided by graphene might allow its potential application in future chiral separation technologies.
Activation of Akt as a mechanism for tumor immune evasion.

<P>Immune evasion is an important reason why the immune system cannot control tumor growth. To elucidate the mechanism for tumor immune evasion, we generated an immune-resistant human papillomavirus type 16 (HPV-16) E7-expressing tumor cell line by subjecting a susceptible tumor cell line to multiple rounds of in vivo immune selection with an E7-specific vaccine. Comparison of parental and immune-resistant tumors revealed that Akt is highly activated in the immune-resistant tumors. Retroviral transfer of a constitutively active form of Akt into the parental tumor significantly increased its resistance against E7-specific CD8(+) T-cell mediated apoptosis. The observed resistance against apoptosis was found to be associated with the upregulation of antiapoptotic molecules. We also observed that intratumoral injection of an Akt inhibitor enhanced the therapeutic efficacy of E7-specific vaccine or E7-specific CD8(+) T-cell adoptive transfer against the immune-resistant tumors. Thus, our data indicate that the activation of PI3K/Akt pathway represents a new mechanism of immune escape and has important implications for the development of a novel strategy in cancer immunotherapy against immune-resistant tumor cells.</P>
