        Preparation, characterization and in vitro release properties of pectin-based curcumin film

        Curcumin is a kind of natural polyphenol whose functions are anti-tumor, anti-oxidant, anti-inflammatory and anti-microbial with low side effects. Solubility of Cur in water is poor, which results in low bioavailability, rapid metabolism, instability and poor absorption in vivo. Based on the crosslinking mechanism of pectin-calcium, the curcumin- pectin film (PT-Curf) was prepared by casting method, where chitosan and sodium alginate were added to the film, forming a system for blending modification to improve the performance of PT-Curf. Furthermore, the release of the curcumin films in vitro was studied and the effects of natural polysaccharides on the drug delivery system were discussed. According to our results, the natural polysaccharides involving pectin, chitosan and sodium alginate were excellent carriers of the fat-soluble drug Cur, and Cur was uniformly dispersed in the composite films in different crystal forms. Out of different drug delivery media, the three films exhibited different release rate and sustained release effect in vitro. However, the pectin-sodium alginate curcumin film (PT-SA-Curf) exhibited the most sustained release effect, the pectin-chitosan curcumin film (PT-CS-Curf) was second.

      • Use of an Artificial Neural Network to Predict Risk Factors of Nosocomial Infection in Lung Cancer Patients

        Statistical methods to analyze and predict the related risk factors of nosocomial infection in lung cancer patients are various, but the results are inconsistent. A total of 609 patients with lung cancer were enrolled to allow factor comparison using Student's t-test or the Mann-Whitney test or the Chi-square test. Variables that were significantly related to the presence of nosocomial infection were selected as candidates for input into the final ANN model. The area under the receiver operating characteristic (ROC) curve (AUC) was used to evaluate the performance of the artificial neural network (ANN) model and logistic regression (LR) model. The prevalence of nosocomial infection from lung cancer in this entire study population was 20.1% (165/609), nosocomial infections occurring in sputum specimens (85.5%), followed by blood (6.73%), urine (6.0%) and pleural effusions (1.82%). It was shown that long term hospitalization (${\geq}22days$, P= 0.000), poor clinical stage (IIIb and IV stage, P=0.002), older age (${\geq}61days$ old, P=0.023), and use the hormones were linked to nosocomial infection and the ANN model consisted of these four factors. The artificial neural network model with variables consisting of age, clinical stage, time of hospitalization, and use of hormones should be useful for predicting nosocomial infection in lung cancer cases.

        Three-dimensional Fe2O3–TiO2–graphene aerogel nanocomposites with enhanced adsorption and visible light-driven photocatalytic performance in the removal of RhB dyes

        A series of Fe2O3 and TiO2 co-doped graphene aerogel (Fe2O3–TiO2–GA) nanocomposites with a unique 3D architecture are assembled by a one-pot hydrothermal method combined with freeze-drying. The one with 30 wt% Fe2O3 exhibits the highest adsorptivity (95.0%) and the another with 25 wt% Fe2O3 displays the highest total efficiency (97.7%) including adsorption and visible light-driven photocatalytic degradation for rhodamine B dyes (20.0 mg L−1, 25 mL) within 1 h over 5.0 mg of the nanocomposites. The materials also display satisfactory cyclic stability and the removal efficiency can be 81.8% even after four runs. Additionally, a related photocatalytic mechanism is proposed.

      • PU.1 Is Identified as a Novel Metastasis Suppressor in Hepatocellular Carcinoma Regulating the miR-615-5p/IGF2 Axis

        Invasion and metastasis is the major cause of tumor recurrence, difficulty for cure and low survival rate. Excavating key transcription factors, which can regulate tumor invasion and metastasis, are crucial to the development of therapeutic strategies for cancers. PU.1 is a master hematopoietic transcription factor and a vital regulator in life. Here, we report that, compared to adjacent non-cancerous tissues, expression of PU.1 mRNA in metastatic hepatocellular carcinoma (HCC), but not primary HCC, was significantly down-regulated. In addition, levels of PU.1 mRNA in metastatic hepatoma cell lines MHCC97L and MHCC97H were much lower than in non-metastatic Hep3B cells. Transwell invasion assays after PU.1 siRNA transfection showed that the invasion of hepatoma cell lines was increased markedly by PU.1 knockdown. Oppositely, overexpression of PU.1 suppressed the invasion of these cells. However, knockdown and overexpression of PU.1 did not influence proliferation. Finally, we tried to explore the potential mechanism of PU.1 suppressing hepatoma cell invasion. ChIP-qPCR analysis showed that PU.1 exhibited a high binding capacity with miR-615-5p promoter sequence. Overexpression of PU.1 caused a dramatic increase of pri-, pre- and mature miR-615-5p, as well as a marked decrease of miR-615-5p target gene IGF2. These data indicate that PU.1 inhibits invasion of human HCC through promoting miR-615-5p and suppressing IGF2. These findings improve our understanding of PU.1 regulatory roles and provided a potential target for metastatic HCC diagnosis and therapy.

        Examining the Economic Effects of Logistics Infrastructure: The Case of New Western Land-Sea Corridor in China

        Purpose: To examine the economic effects of logistics under the influence of policies. Research design, data and methodology: This study is the first to use the panel data of 31 provinces and municipalities in China from 2012 to 2021, and use the OLS and DID models to evaluate whether the New Western Land-Sea Corridor (NWLSC) has promoted the economic development of the regions along the corridor. Results: The NWLSC has stimulated local economic growth by promoting the development of transportation, postal, and telecommunications industries along the corridor. Further, considering the locational differences of the regions along the NWLSC, we examined the differences in economic effects between regions along the Yangtze River and those not along the Yangtze River under the background of NWLSC implementation. We found that waterway and airway transport located along the NWLSC and in the Yangtze River Economic Belt (YREB) region can significantly promote economic growth. However, for regions located along the NWLSC but not in the YREB region, the impact of roadway, railway, and airway transport in these regions on economic growth is more significant. Conclusions: This study has important reference value on how to use logistics to promote the economic and cross-border commerce development of landlocked countries or regions.

        Expression Profiles of the Insulin-like Growth Factor System Components in Liver Tissue during Embryonic and Postnatal Growth of Erhualian and Yorkshire Reciprocal Cross F<sub>1</sub> Pigs

        In Erhualian and Yorkshire reciprocal cross $F_1$ pig populations, we examined the mRNA expression characteristic of liver-derived IGF-1, IGF-1R, IGF-2, IGF-2R and IGFBP-3 during the embryonic and postnatal developmental periods (E50, E70, E90, D1, D20, D70, D120 and D180). Our results demonstrated that the IGF-system genes mRNA levels exhibited an ontogenetic expression pattern, which was potentially associated with the porcine embryonic development, postnatal growth, organogenesis and even the initiation and acceleration of puberty. The expression pattern of IGF-system genes showed variation in the reciprocal cross ($F_1$ YE and EY pigs). This study also involved the expression features of imprinted genes IGF-2 and IGF-2R. The parent-of-origin effect of imprinted genes was reflected by their differential expression between the reciprocal crosses populations. The correlation analysis also indicated that the regulatory network and mechanisms involved in the IGF system were a complex issue that needs to be more fully explored. A better understanding of IGF system components and their interactive mechanisms will enable researchers to gain insights not only into animal organogenesis but also into somatic growth development and even reproduction.

        In silico Discovery of Genes Expressed in Liver, Kidney, Spleen and Small Intestine of Pigs

        An in silico approach was developed to survey the genes expressed in four internal organs of pig: liver, kidney, spleen and small intestine. The major procedures of the approach included: (1) BLAST searching against GenBank "est_others" database using human cDNA sequences as queries to screen the porcine orthologous expressed sequence tags (ESTs), (2) classifying the porcine ESTs records by resources according to certain criteria and (3) analyzing data for ESTs specifically expressed in each organ. In order to do so, four Java programs were developed. Based on the ESTs available in the GenBank database, it was found that there were at least 2,100 genes expressed in these four organs, including 128 in the liver, 81 in the kidney, 780 in the spleen, and 1,423 in the small intestine respectively (a few genes co-expressed in these tissues). Gene expression patterns, such as co-expressed genes, preferentially expressed genes and basic active genes were also compared and characterized among these organs. This study provides a comprehensive model on how to use the bioinformatics approach and Genbank databases to facilitate the discovery of new genes in livestock species.

        A Novel Quality Evaluation Index and Strategies to Identify Scenting Quality of Jasmine Tea Based on Headspace Volatiles Analysis

        Jasmine tea is quite popular in daily consumption and beverage industry, and scenting quality and synthetic artificial oil are crucial issues of its quality control. The headspace volatile profiles of jasmine tea samples were analyzed by headspace-solid phase microextraction (HSSPME)coupled with GC-MS. Twenty-nine volatile compounds were determined as ‘common components’ that should exist in jasmine scented teas. Potential correlation was discovered between quality grade of jasmine tea and the ratio of peak area percentage of α-farnesene, (Z)-3-hexenyl benzoate, methyl anthranilate, and indole to that of linalool. This ratio was denoted as ‘jasmine tea flavor’ (JIF) index,a novel index for quality evaluation of jasmine tea. JTF index and common components could provide a good identification of well-scented, poor-scented, and not-scented jasmine teas. Moreover, carrier compounds might be a clear evidence for identifying fake jasmine teas that were added with synthetic fragrance oil.

        Dynamic Changes of Transcriptome and Metabolites During Ripening of Alpinia Oxyphylla Fruit (AOF)

        Alpinia oxyphylla (Yizhi) is an economically valuable plant, not only used as edible fruit, but also as an important traditional medicinal material. However, there are few studies on fruit development and metabolic pathways. To elucidate the dynamic changes of metabolites and transcription levels during fruit development of A. oxyphylla, the four parts of A. oxyphylla fruit from different periods, including early fruit (EF), middle fruit (MF), late pericarp (LP) and late seed (LS) were collected. KEGG pathway analysis shows that DEGs in EF, MF, LP are involved in amino sugar and nucleotide sugar metabolism, starch and sucrose metabolism, and plant hormone signal transduction. However, at the late part of fruit development (LS), most of the DEGs between LS and LP were enriched in pyruvate metabolism, glycolysis/gluconeogenesis, tryptophan metabolism. We found that in the early and middle parts of fruit development (EF, MF), many differential metabolites were enriched in piperidine tropane and biosynthesis of secondary metabolites, pyridine alkaloid biosynthesis. In addition, important active components, such as nootkatone in A. oxyphylla fruits accumulated in the LS part, but did not show significant differences in the first three parts. Some key transcription factors that are significantly positively correlated with the maturation of AOF have also been identified. Together, our research provides new insights into the accumulation of metabolites and the dynamic changes of the transcriptome during fruit development.
