An artificial network which can accomplish recombinant protein synthesis guided by cell population in E. coli was constructed. The successful functioning of this network requires two plasmids, pWNB and pET. pWNB is responsible for production of T7 RNA polymerase, which controls pET; pET, in turn, regulates the production of target proteins. Several model proteins were tested and the results show that this E. coli system can be used to efficiently express various recombinant proteins. Since system contains T7 RNA polymerase production elements, it is transferable and applicable to well-characterized E. coli strains. Compared to the IPTG-induced system, an equal or greater amount of target protein can be obtained using this auto-inducible expression system in flasks and bioreactors. Our results suggest that it is a competitive alternative to other expression systems used in labs or for industrial applications.

We report on experimental studies of an Al$_{0.15}$Ga$_{0.85}$N/GaN high-electron-mobility transistor structure grown on a $p$-type Si (111) substrate. This structure is compatible with complementary metal-oxide-semiconductor (CMOS) technology and, thus, has great potential device applications. The low-temperature magnetoresistivity shows a parabolic dependence on the applied perpendicular magnetic field. This effect is ascribed to electron-electron interaction (EEI) effects in a weakly-disordered two-dimensional system. Our experimental results agree with the EEI theory when the correction term in the ballistic region has been subtracted.
Structural dynamic optimization with probability constraints of frequency and mode

The structural dynamic optimization problem based on probability is studied. Considering the randomness of structural physical parameters and the given constraint values, we develop a dynamic optimization mathematical model of engineering structures with the probability constraints of frequency, forbidden frequency domain and the vibration mode. The sensitivity of structural dynamic characteristics based on probability is derived. Two examples illustrate that the optimization model and the method applied are rational and efficient.

Lung cancer is the most common causes of cancer-related deaths worldwide, and a lack of effective methods for early diagnosis has greatly impacted the prognosis and survival rates of the affected patients. Tumor-initiating cells (TICs) are considered to be largely responsible for tumor genesis, resistance to tumor therapy, metastasis, and recurrence. In addition to representing a good potential treatment target, TICs can provide clues for the early diagnosis of cancer. MicroRNA (miRNA) alterations are known to be involved in the initiation and progression of human cancer, and the detection of related miRNAs in TICs is an important strategy for lung cancer early diagnosis. As Hsa-miR-155 (miR-155) can be used as a diagnostic marker for non-small cell lung cancer (NSCLC), a smart molecular beacon of miR-155 was designed to image the expression of miR-155 in NSCLC cases. TICs expressing CD133 and CD338 were obtained from A549 cells by applying an immune magnetic bead isolation system, and miR-155 was detected using laser-scanning confocal microscopy. We found that intracellular miR-155 could be successfully detected using smart miR-155 molecular beacons. Expression was higher in TICs than in A549 cells, indicating that miR-155 may play an important role in regulating bio-behavior of TICs. As a non-invasive approach, molecular beacons could be implemented with molecular imaging to diagnose lung cancer at early stages.

Exosomes derived from mesenchymal stem cells (MSCs) could protect against myocardial infarction (MI). TLR4 is reported to play an important role in MI, while microRNA-182-5p (miR-182-5p) negatively regulates TLR4 expression. Therefore, we hypothesize that MSCs-derived exosomes overexpressing miR-182-5p may have beneficial effects on MI. We generated bone marrow mesenchymal stem cells (BM-MSCs) and overexpressed miR-182-5p in these cells for exosome isolation. H2O2-stimulated neonatal mouse ventricle myocytes (NMVMs) and MI mouse model were employed, which were subjected to exosome treatment. The expression of inflammatory factors, heart function, and TLR4 signaling pathway activation were monitored. It was found that miR-182-5p decreased TLR4 expression in BM-MSCs and NMVMs. Administration of exosomes overexpressing miR-182-5p to H2O2-stimulated NMVMs enhanced cell viability and suppressed the expression of inflammatory cytokines. In addition, they promoted heart function, suppressed inflammatory responses, and de-activated TLR4/NF-κB signaling pathway in MI mice. In conclusion, miR-182-5p transferred by the exosomes derived from BM-MSCs protected against MI-induced impairments by targeting TLR4.
Association Between XRCC1 Gene Polymorphisms and Risk of Glioma Development: A Meta-analysis

Objective: Previous studies of the association between X-ray cross-complementing group 1 (XRCC1) gene polymorphisms and the gliomas risk have yielded conflicting results, and thus a meta-analysis was performed to provide a more accurate estimation. Methods: A computerized literature search of 5 electronic databases was conducted to identify the relevant studies. Fixed or random effect models were selected based on the heterogeneity test. Publication bias was estimated using Begg's funnel plots and Egger's regression test. Results: A total of 11 studies (3,810 cases and 6,079 controls), 7 studies (2,928 cases and 5,048 controls), and 4 studies (1,461 cases and 2,593 controls) were finally included in the analyses of the association between XRCC1 Arg399Gln, Arg194Trp, and Arg280His polymorphisms and glioma risk, respectively. The pooled results showed that GlnGln carriage was associated with moderately increased risk of gliomas in Asians (GlnGln vs. ArgArg, OR=1.490, 95%CI 1.031-2.153; GlnGln/ArgGln vs. ArgArg, OR=1.321, 95%CI 1.037-1.684), whereas a marginal association was revealed in Caucasians. For the Arg194Trp polymorphism, although a significant association was shown in the homozygous genotype comparisons (TrpTrp vs. ArgArg, OR = 2.209, 95%CI 1.398-2.945), no significant link was found on subgroup analysis stratified by ethnicity. With regard to the Arg280His polymorphism, no significant association was found in each comparison. No particular study was found to significantly influence the pooled results, and no potential publication bias was detected. Conclusions: This meta-analysis suggested that the XRCC1 Arg399Gln polymorphism is moderately associated with increased risk of gliomas in Asians, while Arg194Trp and Arg280His polymorphisms demonstrated no significant influence. Due to the limited studies and the potential confounders, further studies are needed to confirm these results.

This study presents a robust model predictive control (MPC) strategy to handle the trajectory tracking problem for a underactuated two-wheeled inverted pendulum (WIP) vehicle, in addition to taking various physical constraints into account. To begin with, a saturated trajectory generator is proposed to produce the desired velocities by which all posture tracking errors converge to the compact sets as well as the saturation of velocities being guaranteed. In addition, a MPC approach is put up forward after the approximate feedback linearization is performed to decrease the burden of computation and increase the realtime performance of the control system. Particularly, various physical constraints can be readily assured by the presented MPC method although the equilibrium of WIP vehicle is unstable. Meanwhile, to validate the robustness and availability of the proposed approach, initial errors, pulse disturbance and random noise are introduced to test the control performance of the closed-loop system in the simulation environment. The results from both theoretical analysis and simulation show that the proposed control strategy are effective and feasible for practical implementation.

A model based on inter-diffusion theory was established to predict growth kinetics of the intermetallic compounds (IMCs)formed in roll bonded Al/Cu sheets during annealing process. The model can give good prediction of both IMC layers growthkinetics and element concentration distribution at the interface of roll bonded Cu/Al sheets with different thickness underdifferent annealing conditions with certain roll reduction range. It is revealed that the calculated results of the proposedmodel are depended on the inter-diffusion coefficient of each components, which could be related to the interface bondingstate, i.e., bonding methods or rolling reductions for roll bonding. And there was big difference in inter-diffusion coefficientof Al and Cu matrix under different bonding state, while the inter-diffusion coefficient of IMCs changed slightly.
Identification of a Cancer Stem-like Population in the Lewis Lung Cancer Cell Line

Objective: Although various human cancer stem cells (CSCs) have been defined, their applications are restricted to immunocompromised models. Developing a novel CSC model which could be used in immunocompetent or transgenic mice is essential for further understanding of the biomolecular characteristics of tumor stem cells. Therefore, in this study, we analyzed murine lung cancer cells for the presence of CSCs. Methods: Side population (SP) cells were isolated by fluorescence activated cell sorting, followed by serum-free medium (SFM) culture, using Lewis lung carcinoma cell (LLC) line. The self-renewal, differentiated progeny, chemosensitivity, and tumorigenic properties in SP and non-SP cells were investigated through in vitro culture and in vivo serial transplantation. Differential expression profiles of stem cell markers were examined by RT-PCR. Results: The SP cell fraction comprised 1.1% of the total LLC population. SP cells were available to grow in SFM, and had significantly enhanced capacity for cell proliferation and colony formation. They were also more resistant to cisplatin in comparison to non-SP cells, and displayed increased tumorigenic ability. Moreover, SP cells showed higher mRNA expression of Oct-4, ABCG2, and CD44. Conclusion: We identified SP cells from a murine lung carcinoma, which possess well-known characteristics of CSCs. Our study established a useful model that should allow investigation of the biological features and pharmacosensitivity of lung CSCs, both in vitro and in syngeneic immunocompetent or transgenic/knockout mice.

A purified water-soluble polysaccharide was isolated from the fruiting bodies of sanghuang mushroom (Phellinus baumii Pilát) using hot water extraction, DEAE Sepharose Fast Flow anion exchange and High-Resolution Sephacryl S-1000 gel-filtration chromatography. The purified polysaccharide was a complex β-D-glucan, with a molecular mass of 230 kDa. Fourier-transform infrared (FT-IR), methylation analysis, and NMR spectroscopy of sanghuang mushroom polysaccharide (PBF3) indicated that the polysaccharide contained (1→3)-β-D-, (1→4)-β-D-,and branched (1→3,6)-β-D-glucopyranosyl residues. On the basis of hydroxyl radical assay, superoxide radical assay, and DPPH radical assay, its antioxidant activities were investigated. PBF3 had significant effect on scavenging hydroxyl radicals, an equivalent inhibiting ability to vitamin C on superoxide radical, and a little lower scavenging activity on DPPH radical than vitamin C, and should be explored as a novel potential antioxidant.
