Life history responses of the cabbage beetle Colaphellus bowringi to temperature change

Temperature is considered one of the most important mediators of phenotypic plasticity in ectotherms. Here, we investigated life history traits of the cabbage beetle, Colaphellus bowringi Baly (Coleoptera: Chrysomelidae), at a wide range of temperatures (16, 19, 22, 24, 26 and 28°C). The larval and pupal times were significantly decreased with increasing rearing temperature and growth rate was positively correlated with temperature. However, the relationship between body size and rearing temperature in C. bowringi did not follow the temperature–size rule; both males and females reached the highest body weight at 19°C. Females were significantly larger than males at all temperatures. Male pupae lost significantly more weight at metamorphosis compared to females. However, diapausing males gained significantly higher weight after feeding compared to diapausing females at higher temperatures of 22, 24, 26 and 28°C. Body weight tended to decrease with increasing rearing temperature, whereas sexual size dimorphism (SSD) tended to increase with increasing rearing temperature; thus, Rensch's rule is upheld. The degree to which SSD changed with temperature varied with different development stages. SSD was lowest in pupae, highest in newly emerged adults and intermediate in diapausing adults.

SHP1 negatively regulates the Janus kinase 2/signal transducer and activator of transcription (JAK2/STAT) signaling pathway, which is constitutively activated in myeloproliferative neoplasms (MPNs) and leukemia. Promoter hypermethylation resulting in epigenetic inactivation of SHP1 has been reported in myelomas, leukemias and other cancers. However, whether SHP1 hypermethylation occurs in MPNs, especially in Chinese patients, has remained unclear. Here, we report that aberrant hypermethylation of SHP1 was observed in several leukemic cell lines and bone marrow mononuclear cells from MPN patients. About 51 of 118 (43.2%) MPN patients including 23 of 50 (46%) polycythaemia vera patients, 20 of 50 (40%) essential thrombocythaemia and 8 of 18 (44.4%) idiopathic myelofibrosis showed hypermethylation by methylation-specific polymerase chain reaction. However, SHP1 methylation was not measured in 20 healthy volunteers. Hypermethylation of SHP1 was found in MPN patients with both positive (34/81, 42%) and negative (17/37, 45.9%) JAK2V617F mutation. The levels of SHP1 mRNA were significantly lower in hypermethylated samples than unmethylated samples, suggesting SHP1 may be epigenetically inactivated in MPN patients. Furthermore, treatment with 5-aza-2'-deoxycytidine (AZA) in K562 cells showing hypermethylation of SHP1 led to progressive demethylation of SHP1, with consequently increased reexpression of SHP1. Meanwhile, phosphorylated JAK2 and STAT3 were progressively reduced. Finally, AZA increased the expression of SHP1 in primary MPN cells with hypermethylation of SHP1. Therefore, our data suggest that epigenetic inactivation of SHP1 contributes to the constitutive activation of JAK2/STAT signaling. Restoration of SHP1 expression by AZA may contribute to clinical treatment for MPN patients.

A new triterpenoid, 20(R),22( ξ),24(S)-dammar-25(26)-ene-3 β,6 α,12 β,20,22,24-hexanol (1), and three known triterpenoids, β-D-glucopyranoside,(3 β,12 β)-12,20-dihydroxydammar-24-en- 3-yl,6-acetate (2), 20(R)-ginsenoside Rg3 (3), and 20(R)-ginsenoside Rh2 (4), were isolated from the leaves of Panax ginseng. Their structures were determined by chemical analysis and spectral methods (IR, 1D and 2D NMR, HR-ESI-MS). Compounds 1-4 were exhibited various degrees of cytotoxicity in the human hepatoma cell line, HepG2. Compound 1 had the highest cytotoxic potency, with an IC50 value of 20.1 µM, by stimulating p53-mediated cell cycle arrest at the G1 to S phase transition, leading to apoptosis via activation of the caspase signaling pathway.

The Hemipteran predator, Cyrtorhinus lividipennis, feeds on the eggs and nymphs of rice planthoppers and leafhoppers. We previously demonstrated that sublethal concentrations of imidacloprid stimulated the reproduction of C. lividipennis. Considering the essential roles of juvenile hormone (JH) in insect reproduction, we speculated that sublethal concentrations of imidacloprid may stimulate the reproduction of C. lividipennis by regulating JH level. To test this, we cloned C. lividipennis JH acid methyl transferase (ClJHAMT) and JH esterase (ClJHE), which are responsible for JH biosynthesis and degradation genes, respectively. We then knocked down ClJHAMT by injecting dsRNA into C. lividipennis nymphs and found that emerging female adults exhibited 88.8% lower expression of the vitellogenin gene (ClVg) and the number of eggs was reduced by 41.5% as compared with controls. Silencing ClJHE increased ClVg mRNA expression by 275.0% but did not affect fecundity. A sublethal concentration of imidacloprid (LC 20 ) increased the JH titer in females by 35.3% and 60.6% at 24 and 48 h post-emergence, respectively. In treatments containing both imidacloprid and dsJHAMT, the silencing of CLJHAMT reduced the number of eggs produced by adult females by 21.4% as compared to the control (imidacloprid + dsGFP). Our results indicated that sublethal concentration of imidacloprid may induce C. lividipennis reproduction by upregulating JH level via JHAMT. The finding could provide valuable information for improved integration of C. lividipennis and insecticides in pest management.
Bulk Synthesis of Fe3Al Intermetallic Compound Nanoparticles by Flow-Levitation Method

Metallic nanopowders have an increasing application in magnetic materials, catalysts and chemical and metallic industries. In this research, a novel bulk synthesis method for preparing high pure intermetallic Fe3Al nanoparticles was developed by flow-levitation (FL) method. The Fe and Al vapors ascending from the high-temperature levitated droplet were condensed by cryogenic argon gas under atmospheric pressure. X-ray diffraction (XRD) and selected-area electron diffraction (SAED) were used to identify and characterize the prepared nanopowders exhibiting a Fe3Al phase. Measurement of transmission electron microscopy (TEM) indicated that the Fe3Al particles are nearly spherical, and the particle size of the compound ranges from 10 nm to 200 nm in diameter. The chemical composition of the nanoparticles were determined with energy dispersive spectrometer. The magnetic properties of the nanopowder indicate that Fe3Al intermetallic compound is a soft magnet at room temperature, with coercivity of 24.2 Oe and saturation magnetization of 173.2 emu/g. The production rate of Fe3Al nanoparticles was estimated to be about 4 g/h in a continuous manner, by using the FL method. This method as great potential in mass production of Fe3Al nanoparticles.

Ecdysteroids play an important role in the regulation of molting process in crustaceans. The suppression subtractive hybridization (SSH) technique was employed to search for 20-hydroxyecdysone-responsive genes in the hepatopancreas of the crayfish Procambarus clarkii. Ninety-three putative expressed sequence tags were identified by SSH, including three immune response-related genes, two cell cycle and apoptosis genes, four respiration and energy metabolism genes, four transport-related genes, six metabolism-related genes, two stress response genes, and eight transcription and translation regulation genes. The expression levels of the examined genes were regulated by 20-hydroxyecdysone and varied in the hepatopancreas during the molting stages; data were confirmed by real-time PCR. RNAi of the ecdysteroid receptor had significant effects on the expression levels of 20-hydroxyecdysone- responsive genes. These differentially expressed genes identified by SSH will provide insight into the ecdysteroid signaling pathway in P. clarkii.

Bifurcations of A Third-Order Common-Base Transistor Circuit

This paper reports the dynamics and a great variety of one parameter bifurcation phenomena observed in the laboratory and confirmed by extensive computer simulations for the circuit reported firstly in [2]. The circuit consists of five elements, where the only nonlinear element is a common-base transistor pair with the characteristics of a current-controlled nonlinear resistor. Several strange attractors, the intermittent attractor, periodic windows, cascades of period doubling, period plus two and period minus one phenomena are observed by adjusting the value of L₁. The computer simulation results agree with the experiment results quite nicely. Hopf bifurcation is proved mathematically and the Lyapunov dimensions of several chaotic attractors are derived to confirm the existence of chaos.
