        Flavone Attenuates Vascular Contractions by Inhibiting RhoA/Rho Kinase Pathway

        Our previous study demonstrated that flavone inhibits vascular contractions by decreasing the phosphorylation levelof the myosin phosphatase target subunit (MYPT1). In the present study, we hypothesized that flavone attenuates vascular contractions through the inhibition of the RhoA/Rho kinase pathway. Rat aortic rings were denuded of endothelium, mounted in organ baths, and contracted with either 30 nM U46619 (a thromboxane A2 analogue) or 8.0 mM NaF 30 min after pretreatment with either flavone (100 or 300 $({\mu}M$) or vehicle. We determined the phosphorylation level of the myosin light chain ($MLC_{20}$), the myosin phophatase targeting subunit 1 (MYPT1) and the protein kinase C-potentiated inhibitory protein for heterotrimeric myosin light chain phophatase of 17-kDa (CPI17) by means of Western blot analysis. Flavone inhibited, not only vascular contractions induced by these contractors, but also the levels of $MLC_{20}$ phosphorylation. Furthermore, flavone inhibited the activation of RhoA which had been induced by either U46619 or NaF. Incubation with flavone attenuated U46619 or NaF-induced phosphorylation of $MYPT1^{Thr855}$ and $CPI17^{Thr38}$, the downstream effectors of Rho-kinase. In regards to the $Ca^{2+}$-free solution, flavone inhibited the phosphorylation of $MYPT1^{Thr855}$ and $CPI17^{Thr38}$, as well as vascular contractions induced by U 46619. These results indicate that flavone attenuates vascular contractions, at least in part, through the inhibition of the RhoA/Rho-kinase pathway.

        A new pregnane hexaglycoside from Adonis multiflora

        A new pregnane hexaglycoside, named amurensioside L (1), was isolated from the whole plant of Adonis multiflora. Amurensioside L (1) emerged to be new pregnane hexaglycoside which we fully spectroscopically characterized. The chemical structure of compound 1 was established as $14{\beta}$,20R-epoxy-$3{\beta}$-hydroxypregna-4,6-dien-18-oic acid-20-lactone 3-O-${\beta}$-$\small{D}$-glucopyranosyl-($1{\rightarrow}4$)-O-${\beta}$-$\small{D}$-diginopyranosyl-($1{\rightarrow}4$)-O-${\beta}$-$\small{D}$-cymaropyranosyl-($1{\rightarrow}4$)-O-${\beta}$-$\small{D}$-cymaropyranosyl-($1{\rightarrow}4$)-O-${\beta}$-$\small{D}$-oleandropyranosyl-($1{\rightarrow}4$)-O-${\beta}$-$\small{D}$-oleandropyranoside on the basis of spectroscopic analyses including nuclear magnetic resonance spectrometry, high-resolution fast atom bombardment mass spectrometry, and infrared spectroscopy.

        Prevalence of feline calicivirus in Korean cats determined by an improved real-time RT-PCR assay

        Feline calicivirus (FCV) is considered the main viral pathogen of feline upper respiratory tract disease (URTD). The frequent mutations of field FCV strains result in the poor diagnostic sensitivity of previously developed molecular diagnostic assays. In this study, a more sensitive real-time reverse transcription-polymerase chain reaction (qRT-PCR) assay was developed for broad detection of currently circulating FCVs and comparatively evaluated the diagnostic performance with previously developed qRT-PCR assay using clinical samples collected from Korean cat populations. The developed qRT-PCR assay specifically amplified the FCV p30 gene with a detection limit of below 10 copies/ reaction. The assay showed high repeatability and reproducibility, with coefficients of intra-assay and inter-assay variation of less than 2%. Based on the clinical evaluation using 94 clinical samples obtained from URTD-suspected cats, the detection rate of FCV by the developed qRT-PCR assay was 47.9%, which was higher than that of the previous qRT-PCR assay (43.6%). The prevalence of FCV determined by the new qRT-PCR assay in this study was much higher than those of previous Korean studies determined by conventional RT-PCR assays. Due to the high sensitivity, specificity, and accuracy, the new qRT-PCR assay developed in this study will serve as a promising tool for etiological and epidemiological studies of FCV circulating in Korea. Furthermore, the prevalence data obtained in this study will contribute to expanding knowledge about the epidemiology of FCV in Korea.

        Development and evaluation of a triplex real-time quantitative reverse transcription-polymerase chain reaction for rapid and differential detection of three feline respiratory viral pathogens

        In this study, a new triplex real-time quantitative reverse transcription polymerase chain reaction (tqRT-PCR) assay was developed for the rapid and differential detection of three feline viral pathogens including feline calicivirus (FCV), feline herpesvirus 1 (FHV-1), and influenza A virus (IAV) in a single reaction. The assay specifically amplified three targeted viral genes with a detection limit of below 10 copies/reaction. The assay showed high repeatability and reproducibility, with intra- and inter-assay coefficients of variation of less than 1%. Based on the diagnostic results of the assay using 120 clinical samples obtained from cats with feline respiratory disease complex (FRDC)-suspected signs, the prevalence of FCV, FHV-1, or IAV was 43.3%, 22.5%, or 0%, respectively, indicating that the diagnostic sensitivity was comparable or superior to those of previously reported monoplex qRT-PCR/qPCR assays. The dual infection rate for FCV and FHV-1 was 8.3%. These results indicate that FCV and FHV-1 are widespread and that co-infection with FCV and FHV-1 frequently occur in the Korean cat population. The developed tqRT-PCR assay will serve as a promising tool for etiological and epidemiological studies of these three bacterial pathogens, and the prevalence data for three feline viruses obtained in this study will contribute to expanding knowledge about the epidemiology of FRDC in the current Korean cat population.

      • Application of temperature-controlled metabolic chamber in mice

        Recently, research on various metabolic diseases such as obesity and diabetes has been in the spotlight. A representative example of this is the study of metabolic function analysis using Metabolic cage. It is believed that these studies will greatly contribute to the prevention and treatment of various metabolic diseases in humans. This study focuses on the study of obesity in mice. Mouse metabolic phenotyping has been widely used for detecting metabolic disorder in genetically engineered mouse. Korea Mouse Phenotyping Center (KMPC), nation-wide program for mouse production and phenotyping in Korea has been establishing several pipelines for disease-specific mouse phenotyping to support the mouse research. KMPC has been providing mouse metabolic phenotyping including high fat diet, exercise and cold challenges. Multiple parameter including energy expenditure (EE), O2/CO2 consumption (RER), heat generation and activity has been provided with histology service and body composition. Also temperature can be measured during metabolic chamber with telemetric system. Brown adipogenesis is one of key mechanism for developing new candidate drug target for curing obesity and type 2 diabetes. Several factors including cold, adrenergic b3 agonist and exercise are well known factor leading to brown adipogenesis. Here we introduce mouse metabolic phenotyping services with environmental control chamber at KMPC. Temperature-controlled metabolic cage, the climate chambers can control temperatures from 4 °C to 35 °C, while measuring energy consumption using oxygen consumption, carbon dioxide production, and respiratory exchange ratio and food intake. Using this, exposing the mouse to a low temperature environment can activate BAT by browning WAT, which causes various diseases such as heart disease, diabetes, and obesity. The changes of metabolic parameter was finely measured during cold exposure with temperature-controlled metabolic cage.

      • 음성인식에 기반한 스마트폰 게임 인터페이스

        스마트폰에서 사용되는 게임의 인터페이스로 터치스크린과 음성인식 인터페이스를 사용하여 실험하고 그 타당성을 확인해 본다. 음성인식의 경우 별도의 훈련 없이 사용 될 수 있는 차별화된 강점을 보유하고 있으므로 자연스러운 인터페이스 기술로 활용 될 수 있으며 이동 중이거나 다른 작업을 할 때 음성을 통한 입력이 가능하므로 모바일 기기에 적합한 인터페이스이다. 이를 위하여 간단한 2D 게임을 제작하고 터치스크린 방식의 버튼과 음성인식을 사용하여 공격과 방어를 위한 실험을 하였다. 실험을 통하여 음성인식을 통한 인터페이스 가능성을 확인 할 수 있었다. 제안한 방법은 스마트폰 인터페이스를 위한 기본 연구로 활용 될 수 있다. In this paper, we refer to the validity of touch screen and voice recognition interface of smartphone by the experiments and the result. Voice recognition is unique to human information delivery and does not require special training. It is suitable for mobile gadgets when the user is on the move or on other work while inputting information. We made 2D smartphone game for test of defence and attack using touch screen and voice recognition interface. The results demonstrate that the proposed approach is desirable and robust as well as the results can be used interface of smartphone.

        유아 채소섭취 증진을 위한 사회인지이론에 기초한 영양 교육프로그램 개발 및 효과평가

        Purpose: The purpose of this study was to develop and evaluate a nutrition education program based on social cognitive theory. Education topics focused on improving vegetable intake in preschoolers. Methods: The program was developed based on social cognitive theory, which focuses on self-efficacy, teacher modeling, rewarding and sensory activity. Program development proceeded in five stages (draft development, social cognitive theory application, preliminary investigation, program revision, program implementation). A non-equivalent control group, pretest-posttest design was used in this study. Participants were recruited from one childcare facility; 35 preschoolers (treatment group, n=15 and control group, n=20) participated. The treatment group received 3 lessons. Intake surveys were conducted for each group before and after the nutrition education program. The data were analyzed using a t-test, paired t-test, chi Square-test (Fisher's exact test) and ANOVA. Results: In the treatment group, post-education, there was significantly higher intake in side dishes (vegetable dishes) by children aged between four and five (p<0.01). Among the vegetable dishes, there was a significantly higher intake in broccoli and spinach (p<0.001). Conclusion: This study showed that improving preschoolers' vegetable intake could be achieved via a nutrition education program based on social cognitive theory which focuses on behavior change. It is necessary to develop nutrition education program for teachers to increase the vegetables intake of children.

        전자기 연속 주조법을 이용한 의료용 타이타늄 합금 제작에 관한 연구

        Electromagnetic continuous casting (EMCC) was used to fabricate Ti-6Al-4V alloys with properties suitable for medical applications. Ti-6Al-4V alloy ingots fabricated by EMCC were subjected to heat treatment, such as residual stress removing (RRS), furnace cooling after solution treatment (ST-FC) and water-cooling after solution treatment (ST-WC), in order to obtain characteristics suitable for the standard. After component analysis, the microstructure and mechanical properties (tensile strength and elongation) were evaluated by ICP, gas analysis, OM, SEM, a Rockwell hardness tester and universal testing machine. The Ti-6Al-4V alloy ingot fabricated by EMCC was fabricated without segregation, and the lamellar structure was observed in the RRS and ST-FC specimens. The ST-WC specimen showed only martensite structure. As a result of evaluating the mechanical properties based on the microstructure results, we found that the water-cooled heat treatment condition after the solution treatment was most suitable for the Ti-6Al-4V ELI standard.

        <P>The collagen-stimulated generation of reactive oxygen species (ROS) regulates signal transduction in platelets, although the mechanism is unclear. The major targets of ROS include protein tyrosine phosphatases (PTPs). ROS-mediated oxidation of the active cysteine site in PTPs abrogates the PTP catalytic activity. The aim of this study was to elucidate whether collagen-induced ROS generation leads to PTP oxidation, which promotes platelet stimulation.</P>
