The effects of the citrus bioflavonoid naringin were tested by using as a supplement in a high-cholesterol diet. Male rats were fed for 42 days with a 1%(wt/wt) high cholesterol diet either with or without naringin-supplementation (0.1%, wt/wt) to study the effect on plasma lipid levels, hepatic lipid contents, hepatic enzyme activity, and the excretion of fecal neutral sterols. Naringin did not significantly alter the levels of plasma triglycerides, however, the levels of plasma cholesterol (3.80±0.31 mmol/L vs. 2.61±0.30 nmol/L, mean±SE;p<0.05) and hepatic cholesterol (70.3±4.3 mg/g vs. 54.3±3.8mg/g, mean±SD;p<0.05) were significantly lowered compared to those of the control. HMG-CoA reductase (2487.0±210.0 pmole/min/mg vs. 1879.0±236.0 pmole/min/mg, mean±SE; p<0.05) activities were both substantially lower in the naringin-supplemented group than in the control. The naringin supplementation markedly decreased the excretion of fecal neutral sterols (204.7±28.5 mg/day) compared to the control (521.9±53.9 ,g/day). The combination of the inhibited HMG-Coa reductase (-24.45) and ACAT (-20.2%) activities as a result of naringin supplementation could account for the decrease of fecal neutral sterols.

<P>Umbilical cord blood-derived mesenchymal stem cells are a promising source of cells for regeneration therapy due to their multipotency, high proliferative capacity, relatively noninvasive collection, and ready availability. However, extended cell culture inevitably triggers cellular senescence-the irreversible arrest of cell division-thereby limiting the proliferative lifespan of adult stem cells. Wnt/β-catenin signaling plays a functional role as a key regulator of self-renewal and differentiation in mesenchymal stem cells (MSCs), and thus Wnt/β-catenin signaling and cellular senescence might be closely connected. Here, we show that the expression levels of canonical Wnt families decrease as MSCs age during subculture. Activation of the Wnt pathway by treatment with Wnt3a-conditioned medium or glycogen synthase kinase 3β inhibitors, such as SB-216763 and 6-bromoindirubin-3'-oxime, delays the progression of cellular senescence as shown by the decrease in the senescence effectors p53 and pRb, lowered senescence-associated β-galactosidase activity, and increased telomerase activity. In contrast, suppression of the Wnt pathway by treatment with dickkopf-1 (an antagonist of the Wnt coreceptor) and β-catenin siRNA transfection promotes senescence in MSCs. Interestingly, the magnitude of the response to enhanced Wnt3a/β-catenin signaling appears to depend on the senescent state during extended culture, particularly after multiple passages. These results suggest that Wnt3a signaling might be a predominant factor that could be used to overcome senescence in long-term cultured MSCs by directly intervening in the proliferative capacity and MSC senescence. The functional role of Wnt3a/β-catenin signaling in hedging cellular senescence may allow the development of new approaches for stem cell-based therapies.</P>
Korean red ginseng extract does not cause embryo-fetal death or abnormalities in mice

<P>BACKGROUND: Ginseng has been used for a long time and is well tolerated in humans. However, recent studies have shown that ginsenosides Rb1, Rg1, and Re exert embryotoxicity in in vitro culture systems. We investigated the effects of Korean red ginseng extract (KRGE) on embryonic implantation and fetal development in mice. METHODS: Mice were orally administered KRGE (20, 200, or 2,000 mg/kg/day) from 2 weeks before mating to gestational day (GD) 18, and implantation rate, fetal mortality, body weights, as well as external, visceral, and skeletal abnormalities were determined by Caesarean section on GD18. Ginsenosides in KRGE and in the blood of dams were identified and quantified by HPLC analysis. RESULTS: KRGE did not affect embryonic implantation and mortality as well as fetal body weights up to 2,000 mg/kg/day (≈200 times clinical doses), the upper-limit dose recommended by the Korea Food and Drug Administration (KFDA). Although the prevalence of supernumerary ribs increased at the medium dose (200 mg/kg/day), no dose-dependent increases in external, visceral, and skeletal abnormalities were observed. Major ginsenosides such as Rb1, Rg1, and Re were not detected in the blood of dams based on their chromatographic profiles. CONCLUSIONS: Considerable developmental toxicities of KRGE, even at the upper-limit dose, were not observed in mice. These results might be due to the negligible blood concentrations of ginsenosides in their original forms following oral administration, suggesting that in vitro experiments to assess the effects of ginsenosides on embryotoxicity may not reliably explain the risks of ginsenosides to in vivo embryo-fetal development. Licorice Extract Does Not Impair the Male Reproductive Function of Rats

<P>The effect of water extract of licorice (<I>Glycyrrhiza uralensis</I>), one of the most widely used medicinal plants in Oriental nations and in Europe, on male reproductive function was investigated in rats. Licorice extract was prepared as in Oriental clinics and orally administered at doses of 500, 1,000 or 2,000 mg/kg, the upper-limit dose (2,000 mg/kg) recommended in the Toxicity Test guideline of the Korea Food and Drug Administration, to 6-week-old male rats for 9 weeks. Licorice extract neither induced clinical signs, nor affected the daily feed consumption and body weight gain. There were no significant changes in testicular weights, gross and microscopic findings, and daily sperm production between vehicle- and licorice-treated animals, in spite of slight decreases in prostate weight and daily sperm production at the high dose (2,000 mg/kg). In addition, licorice did not affect the motility and morphology of sperm, although the serum testosterone level tended to decrease without significant difference, showing a 28.6% reduction in the high-dose (2,000 mg/kg) group. The results suggest that the no observed adverse-effect level of licorice extract is higher than 2,000 mg/kg, the upper-limit dose, and that long-term exposure to licorice might not cause profound adverse effects.</P>
Antifibrotic effect of TRP-2 Loaded Micelles (Prodrug of BMP7) in Bleomycin Induced Fibrosis Model

Background Idiopathic pulmonary fibrosis is a usually lethal parenchymal lung disease which only has a life expectancy of 3-5 years after diagnosis. There are only two approved oral drugs pirfenidone and nintedanib, and those are not effective than lung transplantation in end-stage treatment of IPF. It is necessary to further investigate the development of novel, targeted therapeutic options. We evaluate anti-fibrotic effect of TRP-2, a prodrug of BMP7, via inhalation Method in bleomycin induced fibrosis model. TRP-2 is recombinant growth factor which consist of BMP-7 and protein transduction domain, a novel platform technique that overcome conventional growth factor drawbacks such as rapid clearance, proteolytic cleavage with poor transport to target origin. Method Thirty-two mice (C57BL/6) were divided to three groups including control (intranasal inhalation of vehicle 3 times weekly after PBS instillation), bleomycin group (intranasal inhalation of vehicles 3 times weekly after bleomycin instillation), and bleomycin + BMP-7 group (intranasal inhalation of TRP-2, 3 times weekly after bleomycin instillation) for 3 weeks. The body weight change, cell counts in the bronchoalveolar lavage fluid (BALF), changes in histopathology, Ashcroft score, Western blots of TGF-b, survival outcome have compared among the 3 groups. Results Median baseline body weight was similar between three groups at the beginning of experiment. After 3 weeks, Bleomycin group showed sever weight loss than other groups. [Control vs. Bleomycin vs. TRP2, 24.5(23.7, 26.0) vs. 15.0(13.75, 20.0) vs. 21.5 (16.5, 23.0) P<0.004] In Kaplan-meier analysis, TRP-2 showed lower mortality rate than bleomycin group [23.1% (n=4) vs. 66.7% (n=10), P<0.003). Also, in histopathology analysis, TRP2 group has lower Ashcroft score than bleomycin group (3.25 vs 5.5, P<0.003) Conclusion TRP-2 transport well to pulmonary parenchymal via form of micelle, and showed anti fibrotic effect Resulting in survival benefit.

The present study was designed to characterize the modulatory effects of the constltuents of Gastrodia elate and their analogues on the GABAergic neurotransmission. 4-Hydroxybenzaldehyde (1) and 4-hydroxy-3-methoxybenzaldehyde (4) inhibited potently the activity of GABA transaminase (IC_50 = 4.1 and 5.4 ㎍/ml,respectively), while the activity of another constituent, 4-hydroxybenzyl alcohol (2), was very weak. Further investigation with 10 analogues revealed a structureactivity correlation, suggesting that the aldehyde group and the hydroxy group at C-4 are necessary for the inhibitory effect on the enzyme activity. Some potent enzyme inhibitors were examined for the effcet on the radioligands to the GABA_A receptor complexes of rat cerebral cortices. Among them, the component 4 dose-dependently lncreased (20-30%) the binding of [^3H]flunitrazepam in the presence of GABA.

Placental abnormality is the important predisposing cause of intrauterine growth retardation. Massive subchorionic hematoma is defined as a large size of maternal blood clot that separates the chorionic plate from the villous chorion and can result in serious obstetrical complications. We report a case of massive subchorionic hematoma diagnosed prenatally, and propose an additional peculiar finding detectable on both the ultrasound and magnetic resonance images: a large hematoma in the subchorionic region at 17 weeks gestation. At 18 weeks 2 days gestation, the fetus was miscarried. The clinical and pathological findings were compatible with massive subchorionic hematoma. Recurrent massive subchorionic hematoma without thrombophilic finding was observed at the next pregnancy in 17 weeks 5 days by ultrasound. The patient was managed conservatively and had successful outcome at term. So we report the case with the brief review of literatures.

본 연구의 목적은 현상학적인 방법을 통해 발달장애 아동이 있는 어머니의 양육 경험의 의미가 무엇인지 이해하여 본질을 추구하고 그 현상의 의미를 심층적으로 기술하고자 함이다. 본 연구의 대상자는 C 시에 있는 장애인복지관에서 치료를 받고 있는 발달장애 아동을 돌보고 있는 어머니 11명을 편의표본 추출하여 선정하였다. 연구방법은 심층 인터뷰와 녹음테이프를 사용하여 2018년 2월에서 2018년 4월까지 자료를 수집하였다. 인터뷰 자료는 Giorgi의 현상학적 분석방법으로 분석하였다. 분석결과는 다음과 같은 일곱 가지 구성요소가 도출되었다: 장애진단이 늦어짐을 후회함, 장애자녀 수용이 어려움, 주변의 참기 어려운 시선, 장애자녀로 인한 가족 간의 불화, 장애자녀 양육을 수용하고 전념함, 장애자녀 양육에 지쳐감, 장애자녀를 받아들임. 결론으로 발달장애 아동 어머니의 양육태도는 발달장애 아동뿐만 아니라 어머니의 정서적 안정을 포함한 일상생활과 대인관계의 사회적 접촉기회, 사회적 고립 여부까지도 영향을 줄 수 있기 때문에 발달장애자녀를 돌보는 어머니들을 위한 신체적, 심리적, 정서적인 지원을 위한 의료나 복지가 체계적으로 도입되어야 할 것으로 사료된다. The purpose of this study is to understand and describe in depth the meaning of parenting experiences of mothers with children with developmental disabilities through phenomenological methods. Participants of this study were selected by convenience sampling 11 mothers who were taking care of children with developmental disabilities who were being treated at the Welfare Center for the Disabled in C city. Data were collected from February to April, 2018 by using in-depth interview and tape-recording. The interview data were analyzed by the phenomenological analysis method of Giorgi. As the result of analysis, the following seven constituents were emerged; Regreting to delay the uncertain diagnosis, Difficulty in accommodating children with disabilities, Unbearable gaze around, Family disagreements due to children with disabilities, Adopting and dedicated to child care for disabled childre, Tiring of raising children with disabilities, Accepting children with disabilities. In conclusion, the parenting attitude of mother with developmental disability is influenced not only by the children with developmental disabilities but also by the social contact opportunities of daily life and interpersonal relationship including mothers" emotional stability and social isolation, It is considered that medical and welfare should be systematically introduced for physical, psychological and emotional support for mothers.
