      • 환자에서 분리한 Klebsiella pneumoniae의 장독소 생성능 및 활성에 대하여

        임상에서 가져온 99주에 대한 재 실험 결과 40주의 분리균은 생물, 화학적 성상이 Klebsiella pneumoniae ATCC 13883과 동일한 성상을 나타내었으며, K99 균주의 장독소 생성능을 CYE 배지와 CAYE 배지에서 비교하였을 때, CYE 배지에서의 독소 생성능이 높았다. 또한 균주의 배양시간별 장독소 생성능 측정을 위해 배양 후 12시간부터 24시간까지 3시간 간격으로 측정해 본 결과, 21시간 배양했을 때 장독소 생성능이 가장 높았고 마우스 접종시간별 장 독성능을 알아보기 위해 접종 후 시간별로 장 독성능을 측정해 본 결과, 접종 후 4시간후에 장 독성능을 측정했을 때의 효과가 가장 높았다. 따라서 이상의 결과를 종합하면, 환자로부터 분리한 Klebsiella pneumoniae K99 균주는 기존에 보고된 것과는 다소 차이가 있는 생물 및 생화학적 특징을 나타내는 장독소를 생산하는 것을 알 수 있었다. In this study, the enterotoxin of Klebsiella pneumoniae isolated from clinical patients in Korea was characterized. Enterotoxin of Kelbsiella pneumoniae has been known to be secreted in the water and electrolytes in the small intestines of human resulted in diarrhea. For this reason, Klebsiella pneumoniae strains were isolated from patients at hospitals in Seoul, Korea and were investigated for their ability for enterotoxin production by the method of suckling mouse assay. The results were as follows; 1. For their ability of enterotoxin production by the culture media, the casamino acid yeast extract medium was more effective than the casamino acid yeast extract salt medium. 2. For their ability of enterotoxin production by the culture time, enterotoxin their peaked at 21 hrs of incubation. 3. Maximal cytotoxicity was achieved at 4 hrs after the administration of the enterotoxin into the mouse.

      • 척수를 손상시킨 후 꼬리정맥에 주입한 사람탯줄혈액세포가 뇌줄기에 미치는 영향

        Background and Objectives: Stem cells are a valuable resource for treatment of many disease, but limited access to stem cells in some organs such as brain restricts their utility. Many approaches have been attempted to restore the function following brain stem injury (BSI) and spinal cord injury (SCI). The use of the human umbilical cord blood cells (hUCB) - a rich source of nonembryonic or adult stem cells - has recently been reported to ameliorate the behavioral consequences of stroke. Mateiials and Methods: Forty rats were divided into 8 groups: (1) SCI l+hUCB (infused 1 day post injury); (2) SCI 2+hUCB (infused 2 days post injury); (3) SCI 3+hUCB (infused 3 days post injury); (4) SCI 4+hUCB (infused 4 days post injury); (5) SCI 5+hUCB (mfusedt 5 days post injury); (6) SCI 6+hUCB (infused 6 days post injury); (7) LO+hUCB (laminectomy+hUCB); and (8) LO (laminectomy only). SCI was produced by compressing the spinal cord for one minute with an aneurysm clip calibrated to a closing pressure of 50 g. We report here that immunhistotochemical identification of fluorescent hUCB positive cells in the brain stem after compressed spinal cord injury using mouse anti-human mitochondria monoclonal antibody (MAB1273). Results: All SCI+hUCB (1-8) groups contained fluorescent hUCB positive cells in the all area of the brain stem. But especially a large number of fluorescent hUCB positive cells were observed in the whole area of the brain stem of the experimental 5 (SCI 5+hUCB) and 6 (SCI 6+hUCB)groups. No hUCB positive cells were found in the brain stem of group with non-injured spinal cord of these animals and group with laminectomy only. Conclusion: These results suggest that hUCB are potentially useful as a vector for treating a variety of the central nervous system disorders, and we are sure that continuous of stem cell study will give an best opportunity to treat the uncurable disorders in the future.

      • 흰쥐 척수 손상후 Parvalbumin과 Calbindin D-28K 함유신경세포에 관한 면역세포화학적 연구

        Background and Objectives: This study was done to compared the distribution of the two calcium-binding proteins immunoreactive neurons, parvalbumin immunoreactiv (PV-IR) and calbindin D-28K immunoreactive (CB-IR) neurons in the spinal cord after transection. Materials and Methods: Twenty rats were divided into 3 groups (control, complete spinal cord injury (SCI), and right and left spinal cord hemisection). SCI was produced by cutting the spinal cord use blades 11 with scalpel handles. Results: In this experiment, CB-IR neurons were mainly found in many pyramidal cells distributed in the brain stem and spinal cord of rats. PV-IR Neurons were demonstrated in all lamina of the gray matter of the spinal cord. These immunoreactive cells had the highest density in the layer I and II of dorsal horn and several nuclei of the ventral horn of the all the segments of the spinal cord. CB-IR neuropil labeling was strongly noted in all the segments of the spinal cord. In contrast PV-IR neurons were different in distribution, size and morphology in the spinal cord. The number of PV-IR neurons were greater than in the spinal cord compared with the CB-IR neurons. CB-IR and PV-IR somata were round, oval, spindle and polygonal in shape, and were unipolar, bipolar, multipolar and horizontal in types. The diameters of the somata of the PV-IR and CB-IR neurons were 40-50 ㎛, respectively. Also dendrites of PV-IR and CB-IR neurons were densely arrayed in network.

      • 서울市內一部 動物病院에서 臨床診療된 愛玩犬의 病類別 分布에 對한 醫學統計學的 考察

        우리 나라의 소동물 수의임상분야(獸醫臨床分野)에서 주류를 이루고 있는 애완견의 병류별 질병분포를 파약하여 수의임상반야의 기초자료로 제시하고자 시도하고 서울시내 중견 동물병원 5곳을 무작위선택방법에 의하여 선정하여 1993년도부터 1995년도 말까지 3개년간의 임상의료철을 수집, 의학통계학적 방법을 이용 분석 및 관찰한바 몇 자기 결과를 얻어 다음과 같이 적요 한다. 진료과별 진료건수의 분포를 보면, 전체진료건수 8,360건 중에서 내과질환이 53.6%를 점유하고, 외과질환은 13.01%, 비뇨생식기질환1.96%, 피부과질환 15.47%, 안과질환 2.68% 그리고 이(耳)과질환 13.28% 등으로 관찰되었다. 조사 관찰된 내과질환은 총 19종의 질병과 4,481건의 진료건수가 있었으며, 이중에서 위발증(胃發症)이 가장 많아 21.38%를 점유하고 있었고, 외과질환은 성형외과와 정형외과 질환을 포함하여 역시 총 19종의 질병과 1,088건의 진료건수가 있었는데, 이중에서 외상(外傷)이 가장 많아 19.21%, 비뇨 생식기질환은 6종류의 질병과 164건의 진료건수가 있었으며, 그 중에서 비뇨기 감염증이 가장 많아 37.0%를, 피부질환은 5종의 질병에 1,293건의 진료가 있었고, 이중에서 체외기생충의 감염이 37.74% 그리고 안이과(眼耳科)질환은 7종에 1,332건으로 외이염(外耳炎)이 가장 많아 59.49%를 점유하고 있었다. 애완견에서 빈발하는 10대 질환의 분포를 관찰하바, 위염 (11.46%), 위장염(10.28%), 외이염(9.74%), 기관기관지염(8.49%), 대장염(6.41%), 체외기생충증(5.84%), 체내기생충증(4.35%), 이내(耳內) 진드기(3.8%), 피부진균증(3.53%) 및 피지누증 (3.12%) 등의 순으로 관찰되었다. 끝으로 애완견의 전염병으로는 distemper가 44.8%, parvoviral infection 39.37%, coronaviral infection 15.83% 그리고 본조사기간중 서울시내에서의 rabies 발생건수는 전혀 없었다. The purpose of this study was to a retrospective and medical statistical observation on the clinical cases of pet-dogs by veterinary hospital in Seoul City, during the period from 1993 to 1995, which important not only in veterinary clinic but also pet animal health information system development for veterinary practitioner of Korea. The results were summarized as follows: In the clinical cases determined by the international classification of diseases, during the period of three years, 8,358 cases of morbidity of pet-dogs were shown by five veterinary hospital for small animals. Among them, the disease of internal medicine, including medicine, including digestive, respiratory, circulatory, metabolic and parasitosis were 53.6% of total cases, the surgical disease, including orthopedic and plastic surgery were 13.01%, the disease of urogenital was 1.96%, the disease of dermatology was 15.47%, the disease of ophthalmic was 2.68% and the disease of otiatric was 13.28%, respectively. The gastric crisis of 21.38% among 4,481 cases of 19 kinds on the disease of internal medicine showed the highest morbidity. The trauma of 19.21% among 1,088 cases of 19 kinds in the surgery, including orthopedic and plastic surgery, the case of urogenic infection of 37.0% among 164 cases of 6 kinds in the urogenital organe, the ectoparasitosis of 37.24% among 1,293 cases of 5 kinds in the disease of dermatology, the otitis externa of 59.49% among 1,332 cases of 7 kinds n the opthalmatic and otiatric diseases showed the highest morbidity in each part of veterinary clinics. Observation on the morbidity of the main disease of pet-dogs were gastritis (11.46%), gastroenteritis (10.28%), ectoparasitosis (9.64%) including ear-mite, otitis externa (9.47%), trachobronchitis (8.49%), enterocolitis (6.41%), endoparasitosis (4.35%), dermatophytosis (3.53%) and seborrhea (3.12%), respectively. Finally, distributive observation on the incidence case of the main infections disease in pet-dogs which that distemper, parvoviral infection and coronaviral infection were 44.80%, 39.37% and 15.83, respectively.

      • 延日, 鶴田洞地域 玄武岩類의 岩石學 및 그 風化特性

        For the petrology and weathering characteristics of basalts in the vicinity of Yoˇnil, main joint patterns randomly developed in the basalts, microscopic observation, and chemical analysis of the fresh and weathered rocks were studied and got the following conclusions. 1. The main joint patterns randomly developed in the basalts are divided into 4 patterns, i.e. EW, 80°∼90°N or S; N50°∼60°W, 85°∼90°SW or NE; N30°∼40°E, 70°∼80°NW and N35°∼55°E, 60°∼70°SE. These patterns are seemed to be some direction of columnar joints, which are remarkably developed in the basalts. They generally show vertical dips in the core but they are slightly oblique in the marginal part of basalts. 2. The basalts in this area belong to alkali olivine basalt and are porphyritic texture, of which about 35∼40 percent is phenocryst. 3. The relationship between the released quantities of constituents through weathering process is as follows: (Na_2O, K_2O)>(MgO, FeO+Fe_2O_3); MgO>FeO+Fe_2O_3; Na_2O.K_2O 4. Released quantity of 'Al_2O_3' is neoformed into chemical compoud of 'Al_2O_3' in the immediate vicinity of dissolving area, 'H_2O' content is increased by dint of hydration, 'SiO_2' is released by small amounts, and the amount of 'CaO' increases subsequently after decreasing. 5. Sequence of released constituent cations in the early stage of weathering can be summarized as follows; Ca, Na, Si, K, Mg, Fe, Al. And those in the last stage of weathering are Na, Si, Mg, K, Fe, Ca, Al. In process of weathering, decreasing rate of 'Co' is higher than that of 'Cr'.

      • 고장식염수가 토끼 빈창자 점막 산호성 과립세포의 미세구조에 미치는 영향

        This experiment was performed to study the influence of hypertonic saline on the production of the secretory granules by the cellular organelles of the jejunal Paneth cells of the rabbit fed on the 3 % hypertonic saline. The jejunal mucosae of the experimental rabbits were removed partially in the upper one third of the jejunum and fixed in 2.0% glutaraldehyde for 2 hours, followed by 1% osmium tetroxide fixative for 2 hours. Then they were embedded in epon mixture, cut with ultramicrotome and done uranyl acetate-lead citrate double stain, and observed with JEM100 CX Ⅱ electron microscope. Observed results were as follows: 1. The number of secretory granules of a higher electron density was gradually increased as a whole in the course of time after feeding on the hypertonic saline. 2. After feeding on the hypertonic saline, secretory granules of various sizes with a higher electron density mostly occupied the supranuclear region of Paneth cells, but a few secretory granules of various sizes with low electron density were observed. 3. After feeding on the hypertonic saline, the Goigi complex, the mitochondria and the rough surfaced endoplasmic reticula were gradually figured, especially the cisternae of the rough surfaced endoplasmic reticula displayed polyhedral shapes. 4. Considering the above findings, following the feeding on the hypertonic saline, the function of the jejunal Paneth cells was influenced and secretory granules of various sizes with a higher electron density were more abundant as compared with those of a lower electron density. From the above results, it was considered that the Golgi complex, the mitochondria and especially the rough surfaced endoplasmic reticula were activated to maintain homeostasis. As a result of that, the production and concentration of secretory granules were prominently observed.

      • 흰쥐 편도체에서 칼슘결합단백인 calbindin-D28K함유 신경세포의 생후 발달에 관한 면역조직학적 연구

        Background and Objectives : The calcium-binding protein, calbindin-D28K(CB) is widely distribution of CB in rat amygdala during postnatal maturation, in order to provide an insight into the developmental role it may play in the amygdaloid neurons. Materials and Methods : The present study is based on 28 postnatal cases ranging from the day of birth, postnatal day 0 (P0) to 30 days (P5, P10, P15, P20, P30), and these cases were compared with adult rat amygdala. Cryostat sections processed free-floating with monoclonal antibody by immunohistochemistry. Results : CB-immunoreactive neurons were already found at birth in all amygdaloid nuclei except the intercalated nucleus. The intensity and number of immunoreactivity of CB-containing neurons increased during the first 10 days of postnatal life but dramatically decreased at P15. Mature patterns of CB-immunoreactive neurons were achieved at P20. In the adult amygdala, CB-immunoreactive neurons were distributed in almost all amygdaloid nuclei except the intercalated nucleus. Conclusion : The present results show early appearance of CB immunoreactivity in the amygdala. It may be related to the early appearance of CB immunoreactivity in many other brain regions, and suggests that CB may play a functional role during the development of the nervous system including amygdala.
