유아교사의 임파워먼트와 자기개발 욕구와의 관계에서 교사 자긍심의 매개 효과 분석

본 연구는 유아교사의 임파워먼트와 자기개발 욕구와의 관계를 매개하는 변인으로서 교사 자긍심의 매개효과 검증을 목적으로 한다. 연구대상은 본 설문조사에 자발적 참여의사를 밝힌 서울과 경기지역 어린이집과 유치원의 현직유아교사 254명이다. 수집된 자료는 spss 25.0 통계프로그램을 활용하여 신뢰도 분석, 기술통계 분석, 상관관계 분석, 위계적 회귀분석을 실시하였고, Sobel Test를 통해 매개효과의 유의성을 확인하였다. 연구결과는 다음과 같다. 첫째, 유아교사의 임파워먼트와 자기개발 욕구, 교사 자긍심은 양호한 수준이며 유아교사의 근무기관 유형 및 근무시간에 따라 모두 유의한 차이가 나타났다. 둘째. 유아교사의 임파워먼트와 자기개발 욕구, 교사 자긍심 간에는 유의미한 정적 상관관계가 나타났다. 셋째, 유아교사의 임파워먼트가 자기개발 욕구에 미치는 영향에서 교사 자긍심이 이들의 관계를 부분적으로 매개하고 있음이 확인되었다. 본 연구의 의의는 교사 자율성, 전문성과 관련하여 오늘날관심이 높아지고 있는 임파워먼트가 어떻게 교사의 자기개발 욕구로 이어지는가라는 물음에 대한 답으로서 이들의 관계를 매개하는 교사 자긍심의 가치를 입증하였다는 것이다. 본 결과를 토대로 우리사회에서 교사 자긍심을 향상시킬수 있는 근무여건 개선 및 교육의 대상자가 아닌 학습공동체의 주체로서 교사교육의 방향 전환을 제안하는 바이다. The purpose of this study is to verify mediated effect of early childhood teachers’ self-esteem as a variable that mediates the relationship between early childhood teachers’ empowerment and self-development desire. Subjects to be studied were 254 early childhood teachers currently work for child care centers and kindergartens located in Seoul and Gyeonggi regions who would like to voluntarily take part in this questionnaire. The results are as follows: First, the early childhood teachers’ empowerment, self-development desire, and self-esteem were observed to be good and all showed a significant difference according to the type of institutions they work for and how long they work. Second, the early childhood teachers’ empowerment, self-development desire, and self-esteem showed a significant positive correlation among them. Third, the early childhood teachers’ self-esteem was observed to partially mediate the effects of empowerment on self-development desire. This study has significance in that it answers how the empowerment, to which growing attention being paid today, leads to the early childhood teachers’ self-development desire and that it proved the value of early childhood teachers’ self-esteem in terms of mediating the relationship between them. Based on the results, improvement of work environments that would improve early childhood teachers’ self-esteem and switching teacher education strategies as the main agent of learning community are proposed.

In inflammation-associated carcinogenesis, COX-2 is markedly overexpressed, resulting in accumulation of various prostaglandins. 15-deoxy-Δ12,14-prostaglandin J2 (15d-PGJ2) is one of the terminal products of COX-2-catalyzed arachidonic acid catabolism with oncogenic potential. Epithelial-to-mesenchymal transition (EMT) is a process by which epithelial cells lose their polarity and adhesiveness, and thereby gain migratory and invasive properties. Treatment of human breast cancer MCF-7 cells with 15d-PGJ2 induced EMT as evidenced by increased expression of Snail and ZEB1, with concurrent down-regulation of E-cadherin. Nuclear extract from 15d-PGJ2-treated MCF-7 cells showed the binding of Snail and ZEB1 to E-box sequences present in the E-cadherin promoter, which accounts for repression of E-catherin expression. Unlike 15d-PGJ2, its non-electrophilic analogue 9,10-dihydro- 15d-PGJ2 failed to induce EMT, suggesting that the α,β-unsaturated carbonyl group located in the cyclopentenone ring of 15d-PGJ2 is essential for its oncogenic function. Notably, the mRNA level of interleukin-8 (IL-8)/CXCL8 was highly elevated in 15d-PGJ2-stimulated MCF-7 cells. 15d-PGJ2-induced up-regulation of IL-8/CXCL8 expression was abrogated by silencing of Snail short interfering RNA. Treatment of normal fibroblast with conditioned medium obtained from cultures of MCF-7 cells undergoing EMT induced the expression of activated fibroblast marker proteins, α-smooth muscle actin and fibroblasts activation protein-α. Co-culture of normal fibroblasts with 15d-PGJ2-stimulated MCF-7 cells also activated normal fibroblast cells to cancer associated fibroblasts. Taken together, above findings suggest that 15d-PGJ2 induces EMT through up-regulation of Snail expression and subsequent production of CXCL8 as a putative activator of fibroblasts, which may contribute to tumor-stroma interaction in inflammatory breast cancer microenvironment.

Purpose: It is important to discover predictive factors that can identify rectal cancer patients who will respond well to neoadjuvant concurrent chemoradiotherapy (CCRT) to develop management strategies, preserve sphincter and avoid over- treatment. This study explored clinical factors that would predict the adequacy of nonradical management after CCRT in patients with middle or low rectal cancer. Methods: We retrospectively evaluated 447 patients with middle or low rectal cancer who were treated with curative surgery after neoadjuvant CCRT between January 2010 and December 2019. The good response group comprised patients with stages ypT0–1N0 on resection after CCRT; the remaining patients were included in the poor response group. Results: Of 447 patients (mean age, 60.37 ± 11.85 years), 108 (24.2%) had ypT0–1N0 (71.3% with ypT0N0, 4.6% with ypTisN0, and 24.1% with ypT1N0). Overall, 19 patients with cT1–2 (50.0% . 21.8% with cT3–4, P < 0.001), 22 with well-differentiated tumors (51.2% . 21.3% with moderately/poorly differentiated tumors, P < 0.001), 16 with fungating tumors (47.1% . 22.3% with other types, P = 0.001), and 66 with anterior/posterior circumference direction (28.9% . 19.2% with lateral/ encircling direction, P = 0.016) had stage ypT0–1N0. On multivariable analysis, cT1–2 (P = 0.021) and well-differentiated tumor (P = 0.001) were independent predictors of ypT0–1N0. Fungating tumors were not significantly associated with ypT0– 1N0 (P = 0.054). Conclusion: Stage cT1–2 and well differentiation are predictors of ypT0–1N0, while fungating tumors could be considered clinically meaningful, possibly identifying candidates for nonradical treatment post-CCRT.

BACKGROUND/OBJECTIVES: In this study, we determined the anti-inflammatory activities and the underlying molecular mechanisms of the methanol extract from Erigeron Canadensis L. (ECM) in LPS-stimulated RAW264.7 macrophage cells. MATERIALS/METHODS: The potential anti-inflammatory properties of ECM were investigated by using RAW264.7 macrophages. We used western blot assays and real time quantitative polymerase chain reaction to detect protein and mRNA expression, respectively. Luciferase assays were performed to determine the transactivity of transcription factors. RESULTS: ECM significantly inhibited inducible nitric oxide synthase (iNOS)-derived NO and cyclooxygenase-2 (COX-2) derived PGE2 production in LPS-stimulated RAW264.7 macrophages. These inhibitory effects of ECM were accompanied by decreases in LPS-induced nuclear translocations and transactivities of NFκB. Moreover, phosphorylation of mitogen-activated protein kinase (MAPKs) including extracellular signal-related kinase (ERK1/2), p38, and c-jun N-terminal kinase (JNK) was significantly suppressed by ECM in LPS-stimulated RAW264.7 macrophages. Further studies demonstrated that ECM by itself induced heme oxygenase-1 (HO-1) protein expression at the protein levels in dose-dependent manner. However, zinc protoporphyrin (ZnPP), a selective HO-1 inhibitor, abolished the ECM-induced suppression of NO production. CONCLUSIONS: These results suggested that ECM-induced HO-1 expression was partly responsible for the resulting anti-inflammatory effects. These findings suggest that ECM exerts anti-inflammatory actions and help to elucidate the mechanisms underlying the potential therapeutic values of Erigeron Canadensis L.
