        Immediate Vitrectomy for Acute Endophthalmitis in Patients with a Visual Acuity of Hand Motion or Better

        Purpose: To determine the efficacy of immediate pars plana vitrectomy as the primary treatment for acute endophthalmitis in patients with a visual acuity (VA) of hand motion (HM) or better. Methods: A total of 149 patients who were referred to a single center for acute endophthalmitis after cataract surgery over the 13-year study period were retrospectively analyzed. Only patients presenting with a VA of at least HM were included. Patients were initially treated with either primary vitrectomy or intravitreal antibiotic injection alone, and their visual outcomes and reintervention rates after initial treatment were compared. Results: There was no significant difference in the proportion of good (final VA ≥20 / 40) and poor (VA ≤ counting finger) visual outcomes between the groups. However, subgroup analysis of patients with a VA of HM (92 eyes) showed that the incidence of reintervention (14 of 72 eyes [19.4%] vs. 9 of 20 eyes [45.0%]) and poor visual outcomes (10 of 72 eyes [13.9%] vs. 8 of 20 eyes [40.0%]) were lower after prompt vitrectomy than after intravitreal antibiotic injection alone (p = 0.019 and p = 0.022, respectively). For those with a VA of at least counting finger, no significant difference was observed between the groups. Conclusions: For patients with endophthalmitis presenting with a VA of HM, performing a prompt vitrectomy reduced the incidence of reintervention and poor visual outcomes than the administration of intravitreal antibiotics alone. Our results suggest that primary vitrectomy for patients with endophthalmitis presenting with a VA of HM could be more beneficial than intravitreal antibiotic injection alone.

        Characterization of the Lsi1 Homologs in Cucurbita moschata and C. ficifolia for Breeding of Stock Cultivars Used for Bloomless Cucumber Production

        Bloomless cucumber fruits are commercially produced by grafting onto the pumpkin stocks (Cucurbita moschata) to restricted silicon ($SiO_2$) absorption. Inhibition of silicon absorption in bloomless stocks is conferred by a mutant allele of the CmLsi1 homologous to Lsi1 in rice. In this study, we characterized the Lsi1 homologs in pumpkin (C. moschata) and its cold-tolerant wild relative C. ficifolia ('Heukjong') in order to develop a DNA marker for selecting a bloomless trait and to establish the molecular basis for breeding bloomless stock cultivars of C. ficifolia. A Cleaved amplified polymorphic sequence (CAPS) marker (CM1-CAPS) was designed based on a non-sysnonymous single nucleotide polymorphism (SNP, C>T) of the CmLsi1 mutant-type allele, and its applicability for Marker-assisted selection (MAS) was confirmed by evaluating three bloom and five bloomless pumpkin stock cultivars. Quantitative RT-PCR of the CmLsi1 for these stock cultivers implied that expression level of the CmLsi1 gene does not appear to be associated with the bloom/bloomless trait and may differ depending on plant species and tissues. A full length cDNA of the Lsi1 homolog [named CfLsi1($B^+$)] of 'Heukjong' (C. ficifolia), was cloned and sequence comparison between CmLsi1($B^+$) and CfLsi1($B^+$) revealed that there exists total 24 SNPs, of which three were non-synonymous. Phylogenetic analysis of CfLsi1($B^+$) and Lsi1 homologs further revealed that CfLsi1($B^+$) is closesly related to Nodulin 26-like intrinsic proteins (NIPs) and most similar to CpNIP1 of C. pepo than C. moschata.

      • FRASH : Exploiting storage class memory in hybrid file system for hierarchical storage

        <P>In this work, we develop a novel hybrid file system, FRASH, for storage-class memory and NAND Flash. Despite the promising physical characteristics of storage-class memory, its scale is an order of magnitude smaller than the current storage device scale. This fact makes it less than desirable for use as an independent storage device. We carefully analyze in-memory and on-disk file system objects in a log-structured file system, and exploit memory and storage aspects of the storage-class memory to overcome the drawbacks of the current log-structured file system. FRASH provides a hybrid view storage-class memory. It harbors an in-memory data structure as well as a on-disk structure. It provides nonvolatility to key data structures which have been maintained in-memory in a legacy log-structured file system. This approach greatly improves the mount latency and effectively resolves the robustness issue. By maintaining on-disk structure in storage-class memory, FRASH provides byte-addressability to the file system object and metadata for page, and subsequently greatly improves the I/O performance compared to the legacy log-structured approach. While storage-class memory offers byte granularity, it is still far slower than its DRAM counter part. We develop a copy-on-mount technique to overcome the access latency difference between main memory and storage-class memory. Our file system was able to reduce the mount time by 92% and file system I/O performance was increased by 16%.</P>

      • <monospace> nvramdisk</monospace>: A Transactional Block Device Driver for Non-Volatile RAM

        <P>In this work, we developed nvramdisk, a transactional block device driver for byte-addressable NVRAM. nvramdisk effectively addresses the key technical challenges in using a section of NVRAM as a transactional persistent block device. nvramdisk adopts (i) shadow block, (ii) mapping table journaling, and (iii) type-dependent ordering guarantee to provide atomicity, consistency, integrity and durability in write operations on nvramdisk imposed block device. We fully implemented nvramdisk device driver on Linux OS and port it on the desktop computer as well as Android smartphones. In memcachedb, locating the database table in nvramdisk brings x 1.9 insertions/sec and updates/sec performance gain against locating the database table in a high-end SSD (FusionIO ioDrive2). SQLite performance increases by x2.9, from 743 ins/sec to 2,184 ins/sec, in smartphone(Samsung Galaxy S4) and x15, from 730 ins/sec to 12390 ins/sec in PC. nvramdisk yields 26 percent higher random write performance against Persistent Memory Block Driver. The overhead of supporting transaction accompanies 6 percent performance penalty in memcachedb operations.</P>

        Assessment of the current infraspecifi c classifi cation scheme in melon ( Cucumis melo L.) based on genome-wide single nucleotide polymorphisms

        Current nomenclature for the taxonomic classifi cation of melon cultivars ( Cucumis melo L.) at the horticultural group levelrelies on morphological variation in certain key characters. However, the reliability of current infraspecifi c classifi cationscheme in considering horticultural groups as botanical taxa was not fully understood. In the present study, the informationof horticultural group classifi cation in melon was assessed at the molecular level using genome-wide single nucleotide polymorphisms(SNPs). A total of 143 melon accessions of 15 horticultural groups in two subspecies, subsp. melo and subsp. agresti were collected and genotyped by using Genotyping-By-Sequencing (GBS). From the fi ltering of resultant sequencevariants, 10,949 SNPs were selected and used for downstream genetic analysis including population structure, principlecomponent analysis, and hierarchical clustering of 143 melon accessions. Our genetic analyses indicated that the distributionof accessions at the molecular level generally matched the subspecies classifi cation and no substantial contradictions existedbetween the division of accessions based on horticultural group information and genetic relatedness revealed by the GBS. However, the distinction between horticultural groups was not clear-cut, implying the limitation of considering horticulturalgroups as botanical taxa. To improve the resolution of horticultural group classifi cation in melon, our SNP data may be usefulas supporting information in conjunction with morphological characters.

      • enomenological Analysis for Water Exchange in Bentonite Porewater and Suggestion on the Performance Assessment on the Domestic Disposal Facility for High-level Radioactive Waste

        Bentonite, a material mainly used in buffer and backfill of the engineering barrier system (EBS) that makes up the deep geological repository, is a porous material, thus porewater could be contained in it. The porewater components will be changed through ‘water exchange’ with groundwater as time passes after emplacement of subsystems containing bentonite in the repository. ‘Water exchange’ is a phenomenon in which porewater and groundwater components are exchanged in the process of groundwater inflow into bentonite, which affects swelling property and radionuclide sorption of bentonite. Therefore, it is necessary to assess conformity with the performance target and safety function for bentonite. Accordingly, we reviewed how to handle the ‘water exchange’ phenomenon in the performance assessment conducted as part of the operating license application for the deep geological repository in Finland, and suggested studies and/or data required for the performance assessment of the domestic disposal facility on the basis of the results. In the previous assessment in Finland, after dividing the disposal site into a number of areas, reference and bounding groundwaters were defined considering various parameters by depth and climate change (i.e. phase). Subsequently, after defining reference and bounding porewaters in consideration of water exchange with porewater for each groundwater type, the swelling and radionuclides sorption of bentonite were assessed through analyzing components of the reference porewater. From the Finnish case, it is confirmed that the following are important from the perspective of water exchange: (a) definition of reference porewater, and (b) variations in cation concentration and cation exchange capacity (CEC) in porewater. For applying items above to the domestic disposal facility, the site-specific parameters should be reflected for the following: structure of the bedrock, groundwater composition, and initial components of bentonite selected. In addition, studies on the following should be required for identifying properties of the domestic disposal site: (1) variations in groundwater composition by subsurface depth, (2) variations in groundwater properties by time frame, and (3) investigation on the bedrock structure, and (4) survey on initial composition of porewater in selected bentonite The results of this study are presumed to be directly applied to the design and performance assessment for buffer and backfill materials, which are important components that make up the domestic disposal facility, given the site-specific data.

        <i>In vitro</i> and <i>in vivo</i> imaging and tracking of intestinal organoids from human induced pluripotent stem cells

        <P>Human intestinal organoids (hIOs) derived from human pluripotent stem cells (hPSCs) have immense potential as a source of intestines. Therefore, an efficient system is needed for visualizing the stage of intestinal differentiation and further identifying hIOs derived from hPSCs. Here, 2 fluorescent biosensors were developed based on human induced pluripotent stem cell (hiPSC) lines that stably expressed fluorescent reporters driven by intestine-specific gene promoters Kruppel-like factor 5 monomeric Cherry (KLF5(mCherry)) and intestine-specific homeobox enhanced green fluorescence protein (ISXeGFP). Then hIOs were efficiently induced from those transgenic hiPSC lines in which mCherry- or eGFP-expressing cells, which appeared during differentiation, could be identified in intact living cells in real time. Reporter gene expression had no adverse effects on differentiation into hIOs and proliferation. Using our reporter system to screen for hIO differentiation factors, we identified DMH1 as an efficient substitute for Noggin. Transplanted hIOs under the kidney capsule were tracked with fluorescence imaging(FLI) and confirmed histologically. After orthotopic transplantation, the localization of the hIOs in the small intestine could be accurately visualized using FLI. Our study establishes a selective system for monitoring the in vitro differentiation and for tracking the in vivo localization of hIOs and contributes to further improvement of cell-based therapies and preclinical screenings in the intestinal field.</P>
