        Streptomyces subrutilus P5가 생산하는 철 함유 superoxide dismutase의 분비

        본 연구에서는 Streptomyces subrutilus P5의 생장과 세포내 외 철 함유 superoxide dismutase 활성을 비교 분석하여 철함유 superoxide dismutase의 분비 시점을 확인하고 분자 수준에서 이 효소의 분비에 관여하는 유전정보를 확인하고자 하였다. Streptomyces subrutilus P5의 균체 생장은 건체 중량을 측정하여 결정하였다. Glucose는 log phase에서 급격히 소모되어 24시간 후에 이르러 완전히 고갈되었다. 세포내의 철 함유 superoxide dismutase는 배양 후 3시간에 나타나며 세포외 철 함유 superoxide dismutase는 배양 후 7.5시간부터 나타난다. 따라서 superoxide dismutase는 용균에 의해서가 아니라 능동적인 분비기작에 의해서 세포 외로 분비된 것으로 추측할 수 있다. Streptomyces subrutilus P5의 sodF에는 signal peptide 유전정보가 존재하지 않았다. 그러나 sodF의 상류지역에서 다른 세균의 type III 분비단백질 유전자와 유사한 type III 분비상자가 발견되었다. Streptomyces 균주에서 type III 분비단백질이 존재할 가능성이 있음을 처음으로 제시하였다. We tried to analyze the growth time for secretion of the iron containing superoxide dismutase by comparing the intra-and extracellular enzyme activity from Streptomyces subrutilus P5 and analyze possible genetic information for this enzyme secretion. The mycelial dry weights and glucose concentrations in culture filtrates were determined during growth. Glucose was consumed rapidly during logarithmic growth phase and almost exhausted at 24 h of cultivation. While the intracellular activity of iron containing superoxide dismutase was first appeared at three hours, the extracellular activity of this enzyme appeared from 7.5 h of cultivation, early logarithmic growth phase. This early presence of the superoxide dismutase might not be the result of cell lysis but active secretion pathway. There was no information for signal peptide responsible for the enzyme secretion in sodF. However, we found a type three secretion box in the promoter region of sodF that has been known for the genes of type III secreted proteins in other bacteria. This is the first report on the possible existence of type III secretion in Streptomyces.

        중국어 유의어 교육 방법 개선안 연구

        This work was motivated by the need for an efficient item-weight-assignment and the need that a test result makes a sense to an examinee which well accords with the purpose of a given test. In this paper, we showed empirically that classical test theory served the needs up to some level and in the same context, we pointed out that score approach may not suffice the needs. Vocabulary is the most fundamental element to get the abilities of using chinese correctly and effectively. The most basic element which composes an individual's vocabularies is to know the meaning of each word clearly. One word has its own basic meaning and also has a relative meaning that depends on the relation of another word. Therefore the right vocabulary teaching should be like this : Learners are able to know the word's basic meaning and related meaning correctly enough to use it in a context in a right way. Each result was presented in numerical value as a statistical data and was analyzed. As the result I confirmed this synonym teaching method is more effective one to grasp the difference of the meaning existed between synonyms. As a matter of facts, the study in this thesis doesn't get to suggesting a perfect vocabulary teaching model, for this study was done in restricted conditions. But by proposing the point of views and the method the synonym teaching has to get, it seems to obtain some results which raise the necessities to reconsider the directions and the ways the future Chinese vocabulary teaching has to pursue.

        중한 번역기 시스템에 관한 연구

        The purpose of this study is to establish the chinese-korean machine translation system. The study of the machine translation has received relatively little attention from korean scholarship in general, what was worse, that is much neglected branch of chinese linguistics in korea. So far. very little has been done in this direction. In China, this topic of the study has been studied extensively during the past five decades. The generation of sentence. till now. has mainly examined in syntactic dimension. That is to say, throughout these studies runs a common theme: the algorithm of machine translation system must be based on Chom`s rewriting rules. However, it cannot be satisfactorily explicated by syntax analysis. Because the sentence is not generated from the combinations between the syntactic categories. The structure of sentence is strong association with cooccurrence restrictions. Therefrore, we need more concrete and delicate the cooccurrence restrictions such as collocational restrictions. My approach in this paper is based on collocational restrictions and semantic informations. First, we pointed out the importance of punctuation in analyzing the structure of chinese sentence. Second. in considering language dependent phenomena. we would rather use morphologidcal alalysis than use word segmentation. This kind of processing may all the more possible because Chinese sentence has no word delimiter. Third Qi hu YAng`s (2000) study introduces the established researches on disambiguation. His approach is based on the principle of "preference". This principle seems plausible, yet it requires further examination. THe purpose of my paper is to an alternative approach which might present the method in solving the question. Four, we introduce some kind of method for establishing machine readable dictionary. we find out the method for dealing with equivocation. name and classify the collocational constructions. This study is only a first step. we believe it offers some insight into the problems. There are, however, a number of problems that remain to be explpred. The study concludes by mentioning some of the further problems raised by this approach to spreading activation model.

        의무 양태 범주의 재구성

        This paper has reorganized the obligation modality category and we developed the method of analyzing the distinctive functions of ‘必须,要,得,应该’ in each functional category. As a first step, we set up a basic characteristic shared by the members of the category. Distinguish between ′Subjective and objective′ and ′Speaker′s behavior control power′, which is used as a concept in the existing research. In the second step, we presented ′可以′ which is selected as the boundary mark of categories, is how to be classified into ′the obligation modality category′ and ′permission modality category′. As a third step, We presented the distinctive function of the members of the obligation modality category.

        문항 변별도를 기반으로 한 문항 설계 제안

        Beginning with the analysis of the problems that the chinses proficiency testing has brought about in recent years. the purpose of this paper is explore the rather reliability and test validity of FLEX developed by Hankuk University of foreign studies. This study investigates the potential roles in the Generalizability theory in the validation of a performance-based chinses test. Data for this study come from Hankuk University of foreign studies`s FLEX. There is no doubt that the FLEX reform will change the traditional chinese way of teaching, teachers` dominating the classes by explaining the language points. Therefore, teachers should pay more attention to the formative test, such as diagnostic tests as the various learning periods, so that the teaching plans and methods can be adjusted in time.

        ATP가 마우스 담관세포의 세포크기 조절에 미치는 영향

        ATP 분비 경로는 담관세포종 세포주를 포함한 일부 세포의 RVD 조절에 있어서 주요한 조절 역할을 수행할 것으로 생각되나, 정상담관세포의 RVD에 있어서의 역할은 잘 알려져 있지 않다. 정상 담관세포의 RVD에 있어서 세포 외 ATP와 ATP 경로의 역할을 알아보기 위하여, 정상마우스에서 담관세포를 분리하여 연구에 이용하였다. BDCCs의 크기 변화는 정량측정 비디오현미경을 이용하여 횡단면을 간접 측정하였다. 정상마우스의 BDCCs의 횡단면은 저장 액에서 노출한 후 10분에 1.20±0.02 (n=20)이었으나, 40분에는 1.06±0.03로 감소하였다. ATP와 ATP 가수분해 효소인 아피라아제 또는 P2 수용체 차단제인 슈라민 RVD 과정 중에 처리 하였으나 처리하지 않은 대조군과 중요한 차이를 보이지 않았다. 더 나아가, PKC 저해제인 비신돌아마이드 I 과 Ro 31-8220을 처리하여 RVD에 미치는 영향을 알아 보았으나 대조군과 중요한 차이를 보이지 않았다. 이러한 결과들에서 정상 마우스 담관세포는 담관세포종 세포주와 다른 결과를 보였으며, 정상 마우스의 RVD에 있어서 ATP는 중요한 역할을 하지 않음을 알 수 있었다. Although the adenosine triphosphate (ATP) efflux pathway is thought to play a major regulatory role in RVD in some cells, including cholangiocarcinoma cells, the role of ATP in regulatory volume decrease (RVD) of normal cholangiocytes is not well defined. Thus, this study was conducted to investigate the role of extra cellular ATP and ATP pathways of BDCCs isolated from normal mice. Changes in cell volume of BDCCs were indirectly assessed by measurement of the cross-sectional area (CSA) by quantitative videomicroscopy. The relative CSA of BDCCs from normal mice increased with hypotonic maneuver to 1.20±0.02 (n=20) within 10 min, but decreased to 1.06±0.03 at 40 min. Administration of ATP, ATP hydrolase apyrase or the P2 receptor blocker suramin during RVD had no significant effects compared with untreated controls. In addition, treatment with the PKC inhibitors, Bisindolamide I and Ro 31-8220, during RVD had no significant effects when compared with untreated controls. These results indicate that unlike the results from cholangiocarcinoma cells, ATP plays no significant role in the RVD of normal mouse cholangiocytes.
