Molecular characterization of an α-N-acetylgalactosaminidase from Clonorchis sinensis.

<P>The α-N-acetylgalactosaminidase (α-NAGAL) is an exoglycosidase that selectively cleaves terminal α-linked N-acetylgalactosamines from a variety of sugar chains. A complementary DNA (cDNA) clone encoding a novel Clonorchis sinensis α-NAGAL (Cs-α-NAGAL) was identified in the expressed sequence tags database of the adult C. sinensis liver fluke. The complete coding sequence was 1,308 bp long and encoded a 436-residue protein. The selected glycosidase was manually curated as α-NAGAL (EC 3.2.1.49) based on a composite bioinformatics analysis including a search for orthologues, comparative structure modeling, and the generation of a phylogenetic tree. One orthologue of Cs-α-NAGAL was the Rattus norvegicus α-NAGAL (accession number: NP_001012120) that does not exist in C. sinensis. Cs-α-NAGAL belongs to the GH27 family and the GH-D clan. A phylogenetic analysis revealed that the GH27 family of Cs-α-NAGAL was distinct from GH31 and GH36 within the GH-D clan. The putative 3D structure of Cs-α-NAGAL was built using SWISS-MODEL with a Gallus gallus α-NAGAL template (PDB code 1ktb chain A); this model demonstrated the superimposition of a TIM barrel fold (α/β) structure and substrate binding pocket. Cs-α-NAGAL transcripts were detected in the adult worm and egg cDNA libraries of C. sinensis but not in the metacercaria. Recombinant Cs-α-NAGAL (rCs-α-NAGAL) was expressed in Escherichia coli, and the purified rCs-α-NAGAL was recognized specifically by the C. sinensis-infected human sera. This is the first report of an α-NAGAL protein in the Trematode class, suggesting that it is a potential diagnostic or vaccine candidate with strong antigenicity.</P>

Expression of ER stress and autophagy‐related molecules in human non‐small cell lung cancer and premalignant lesions

<P><B>Abstract</B></P><P>Stress that impairs endoplasmic reticulum (ER) function leads to an accumulation of unfolded or misfolded proteins in the ER (ER stress). Autophagy is a lysosomal pathway involved in the turnover of cellular macromolecules and organelles, which emerging data indicate that ER stress is also a potent inducer of autophagy. ER stress and autophagy are involved in human cancer. We examined the expression of ER stress‐related proteins [GRP78 and C/EBP homologous protein (CHOP)] and autophagic proteins (Beclin‐1 and LC3) in non‐small cell lung carcinomas (NSCLCs), bronchioloalveolar carcinomas (BACs) and atypical adenomatous hyperplasias (AAHs) to understand their role in the NSCLC pathogenesis. The expression of GRP78 and CHOP, Beclin‐1 and LC3 were analyzed using immunohistochemistry on tissue sections from 133 NSCLC (69 squamous cell carcinomas, 56 adenocarcinomas (AC) and eight other NSCLCs), 21 BAC and 9 AAH. Expression of GRP78 and Beclin‐1 was correlated with low tumor stage (<I>p</I> < 0.001 and <I>p</I> = 0.019, respectively) and longer survival (<I>p</I> = 0.007 and <I>p</I> <0.001, respectively) by Kaplan–Meier analysis. However, CHOP was correlated with high tumor stage (<I>p</I> = 0.038) and shorter survival (<I>p</I> = 0.012). Expression of GRP78 and Beclin‐1 was positively correlated (<I>p</I> = 0.006). Our study showed that the expression of GRP78, CHOP, Beclin‐1 and LC3 in lung cancer and its relation with clinicopathologic factors and patients survival. These results suggest that GRP78, CHOP and Beclin‐1 may play an important role in tumorigenesis of lung AC and may serve as new prognostic indicators for outcome of the patients with NSCLC.</P>

간세포암의 예후인자에 대한 연구: p53과 Angiopoietin 포함

저자들은 간 절제술을 받은 49명의 환자를 대상으로 Ang, p53, CD34를 포함한 여러 임상병리학적인 인자들이 환자의 예후에 미치는 영향을 연구하였다. 14종류의 임상 병리학적인 인자들을 분석하였으며 생존율 계산은 Kaplan-Meier 방법을 이용하였다. 단일변수 분석을 하였을 때, 간경변을 동반 하지 않는 경우, 수술 전 측정한 AFP의 혈 청치가 200 ng/ml 이하인 경우, Child-Pugh`s 본류법상 A 또는 B 등급인 경우, 수술 후 병기가 1 혹은 2 인 겨우 및 와 복수를 동반하지 않았을 때 간세포암의 절 체 후 환자의 생존율이 높았다. 그러나 종양 내 혈관 밀도 측정의 지표인 CD34에 대한 면역조직화학 염색결과에 따라 분류한 환자군과, 암 억제 물질인 p53에 대한면역염색결과에 따라 분류한 환자군 에서 생존율의 통계학적 유의성은 없었다. 본 연구는 간경변의 유무, 수술 전 혈청 내 AFP 수치 및 복수의 유무는 간 절제술을 받은 환자의 장기적인 예후와 관련이 있는 반면 Ang, p53 또는 CD34는 예후적 지표가 될 수 없었다. 또한 간세포암의 파라핀 포매 조직에서 in situ hybridization이 잘 수행돤 점은 간세포암 및 여러 인체에서 발생하는 종양을 연구하는데 in situ hybridization이 유용한 기법으로 적용돨 수 있음을 보여주고 있다. Purpose: Factors affecting outcome after resection of hepatocellular carcinoma (HCC) in Korean patients are not clearly defined. This study evaluated the prognostic influence of various clinicopathologic factors in HCC. Methods: Fourteen clinicopathological factors, including expression of angiopoietin mRNA, and p53 and CD34 proteins were evaluated retrospectively in 49 patients undergoing curative resection. Survival rates were calculated by the Kaplan-Meier method. Results: By univariate analysis, survival was significantly higher in patients with Child-Pugh`s classification A, TNM stage 1 and stage 2, an absence of liver cirrhosis, a low serum α-fetoprotein level ≤200 ng/ml, and an absence of ascites. However, angiopoietin 1 (Ang1), angiopoietin 2 (Ang2), CD34 expression pattern, and p53 expression did not influence the survival rate. Conclusions: This study provides information that can be used to predict the prognosis of patients with HCC after undergoing partial hepatectomy. Although expression of Ang1 and Ang2 detected by in situ hybridization, and the immuno-histochemical detection of p53 and CD34 expression are not prognostic predictors of patients with HCC, this study shows that in situ hybridization is performed well with paraffin embedded sections in HCC.

Neurilemmomas arising in lymph nodes are exceedingly rare with a few cases described hitherto. We experienced a case of intranodal neurilemmoma on the perirenal adipose tissue. The lesions were characterized by interlacing fascicles of spindle cells with wavy nuclei and were surrounded by a rim of lymphoid tissue, which in turn formed a fibrous capsule. The lymphoid tissue was comprised of follicles with germinal center, paracortical tissue with high endothelial venules and interdigitating cells. Immunohistochemically, the tumor cells were positive for S-100 protein.

Telomerase activity is required for bleomycin-induced pulmonary fibrosis in mice.

<P>In addition to its well-known expression in the germline and in cells of certain cancers, telomerase activity is induced in lung fibrosis, although its role in this process is unknown. To identify the pathogenetic importance of telomerase in lung fibrosis, we examined the effects of telomerase reverse transcriptase (TERT) deficiency in a murine model of pulmonary injury. TERT-deficient mice showed significantly reduced lung fibrosis following bleomycin (BLM) insult. This was accompanied by a significant reduction in expression of lung alpha-SMA, a marker of myofibroblast differentiation. Furthermore, lung fibroblasts isolated from BLM-treated TERT-deficient mice showed significantly decreased proliferation and increased apoptosis rates compared with cells isolated from control mice. Transplantation of WT BM into TERT-deficient mice restored BLM-induced lung telomerase activity and fibrosis to WT levels. Conversely, transplantation of BM from TERT-deficient mice into WT recipients resulted in reduced telomerase activity and fibrosis. These findings suggest that induction of telomerase in injured lungs may be caused by BM-derived cells, which appear to play an important role in pulmonary fibrosis. Moreover, TERT induction is associated with increased survival of lung fibroblasts, which favors the development of fibrosis instead of injury resolution.</P>

중장년의 ‘삶의 사다리’ 인식에 대한 영향요인 분석 : 60-70대를 대상으로

본 연구는 고령사회에 접어든 우리나라의 인구구조를 제고 하여, 중장년층의 저소득층들이 느끼는 삶에 대한 인식을 고찰하였다. 분석을 위하여, 보건복지부에서 제공하는 2019년 복지패널자료를 토대로 필요로 하는 변수를 선정하였고, 총 14,918명의 패널표본으로부터 ‘저소득층’의 ‘60대-70대’ 중에서, 무응답 및 불성실한 표본을 제거하여, 최종 1,684명의 표본을 선정하였다. 특히, 본 연구에서는 종속변수로써, 패널자료에서 측정한 ‘삶의 사다리’로 정의하였다. 이에, ‘삶의 사다리’에 미치는 개인적 특성 및 생활만족도의 영향요인을 분석하였다. 분석결과, 종속변수인 「삶의 사다리」변수에 대한 영향요인으로, 개인적 특성만을 고려하였을 경우에는, 대부분의 개인적 특성이 「삶의 사다리」의 영향요인으로 나타났다. 또한, 경제상태변수에 대한 상호작용 효과를 살펴본 결과, 유의미한 상호작용 효과를 나타내지 않았다. 이는 중년노인들의 삶의 사다리에 미치는 영향요인은 경제상태보다는 건강상태가 유의미한 영향요인임을 반증해주고 있다. 따라서, 노인의 ‘삶의 사다리’에 대한 인식은 노인 개인적인 여건과 환경보다는 노인의 내적 및 외적인 다양한 ‘환경요인’이 노인의 삶에 대한 인식에 영향을 미침을 제고 해야 한다. 즉, 그들의 삶에 대한 인식에 긍정적인 대안을 제시할 수 있는 방안이 필요할 것이다. This study examined the perception of life felt by low-income people in middle-aged and elderly people by enhancing the population structure of Korea, which is entering an aging society. For the analysis, the variables that need from the 2019 welfare panel data provided by the Ministry of Health and Welfare were selected. The final 1,684 samples were selected from the panel samples of 14,918 people, from the 60-70s of low income class, by removing the unresponsive and insincere samples. In particular, this study defined as a dependent variable as a ladder of life measured in panel data, and analyzed the personal characteristics and life satisfaction factors affecting ladder of life. As a result of the analysis, most of the personal characteristics were found to be influential factors of the ladder of life when considering only personal characteristics as an influential factor for the dependent variable ladder of life. Also, as a result of examining the interaction effect on the economic state variable, there was no significant interaction effect. This shows that the health condition is more important than the economic condition as the influential factor on the ladder of middle-aged elderly people. Therefore, the recognition of the elderly ladder of life should improve the awareness of the elderlys life by various environmental factors, both internal and external, rather than the personal conditions and environment of the elderly. In other words, a plan to present a positive alternative to their perception of life will be needed.

고대 그리스 미술에 나타난 신들의 특징과 헤어스타일에 관한 연구

This study is carried out to analogize ancient hair style by investigating gods feature in ancient Greece arts. Modem designers required rapid and mass communication collect existing informations and reassemble selected data out of informations. That's because needs to be communicated in society are required symbol or symbol system aleady made by imagination of group. This imagination and symbol is continuing from past to now so we can think there are very little difference between hair style of now and that of gods in ancient Greece arts. On that score I studied data related tp the myth of paintings on ancient Greece china through previous papers and documentaries. Also I studied hair styles of goddess in myth by example features of them. The feature and hair style should be studied because they give us unlimited imagination. This study's purpose is it can encourage the possibility of development on hair design and hair designer's modelling works in hair arts.

공장에 단독으로 생긴 결절성 다발성 동맥염

Polyarteritis nodosa(PAN) is a systemic vasculitis affecting mainly small and medium sized arteries. Over 50% of the cases, PAN may cause gastrointestinal symptoms. Main gastrointestinal symptoms are abdominal pain, vomiting, nausea, diarrhea and melena. A very rare case of isolated PAN affecting the jejunum, manifested as stricture, is reported. A 55-year-old woman with prior history of hypertension presented herself complaining of abdominal pain and weight loss. Radiologic studies showed a segmental narrowing of the jejunum. At the time of surgery, 3cm length narrowed, thickened fibrotic intestinal wall was found at 110cm distal to the Treitz ligament. Pathological finding showed acute necrotizing vasculitis of a medium sized artery. (Korean J Gastroenterol 1997; 30:411-414)
