        Assessment of toxicity and genotoxic safety profile of novel fisetin ruthenium-p-cymene complex in mice

        Throughout the last decades flavonoids have been considered as a powerful bioactive molecule. Complexation of these flavonoids with metal ions demonstrated the genesis of unique organometallic complexes which provide improved pharmacological and therapeutic activities. In this research, the fisetin ruthenium-p-cymene complex was synthesized and characterized via different analytical methods like UV–visible spectroscopy, Fourier-transform infrared spectroscopy, mass spectroscopy, and scanning electron microscope. The toxicological profile of the complex was evaluated by acute and sub-acute toxicity. Additionally, the mutagenic and genotoxic activity of the complex was assessed by Ames test, chromosomal aberration test, and micronucleus based assay in Swiss albino mice. The acute oral toxicity study exhibited the LD50 of the complex at 500 mg/ kg and subsequently, the sub-acute doses were selected. In sub-acute toxicity study, the hematology and serum biochemistry of the 400 mg/kg group showed upregulated white blood cells, aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, creatinine, glucose and cholesterol. However, there was no treatment related alteration of hematological and serum biochemical parameters in the 50, 100, and 200 mg/kg group. In the histopathological analysis, the 50, 100, and 200 mg/kg groups were not associated with any toxicological alterations, whereas the 400 mg/kg group showed prominent toxicological incidences. Nevertheless, the treatment with fisetin ruthenium-p-cymene complex did not exhibit any mutagenic and genotoxic effect in Swiss albino mice. Thus, the safe dose of this novel organometallic complex was determined as 50, 100, and 200 mg/kg without any toxicological and genotoxic potential.

      • Computational characterization of moonlighting proteins.

        <P>Moonlighting proteins perform multiple independent cellular functions within one polypeptide chain. Moonlighting proteins switch functions depending on various factors including the cell-type in which they are expressed, cellular location, oligomerization status and the binding of different ligands at different sites. Although an increasing number of moonlighting proteins have been experimentally identified in recent years, the quantity of known moonlighting proteins is insufficient to elucidate their overall landscape. Moreover, most moonlighting proteins have been identified as a serendipitous discovery. Hence, characterization of moonlighting proteins using bioinformatics approaches can have a significant impact on the overall understanding of protein function. In this work, we provide a short review of existing computational approaches for illuminating the functional diversity of moonlighting proteins.</P>

      • Prevalence of Human Papillomavirus and Co-Existent Sexually Transmitted Infections among Female Sex Workers, Men having Sex with Men and Injectable Drug abusers from Eastern India

        Background: Human papillomavirus (HPV) is a very common sexually transmitted disease affecting both men and women and is responsible for different ano-genital cancers in either sex. Co-existing sexually transmitted infections (STI) including HIV have been considered as important co-factors for carcinogenesis induced by HPV. The purpose of this study was to determine the prevalence of any HPV, HPV 16 and HPV 18 and also concomitant STIs among female sex workers (FSW), men having sex with men (MSM) and injectable drug users (IDU). Material and Method: This cross-sectional study was conducted among 45 FSWs, 26 MSMs and 58 IDUs who attended the STI or de-addiction clinics. Genital scrape samples collected from glans penis and coronal sulcus in males and cervical squamo-columnar junction in females were tested for HPV DNA by PCR using HPV L1 consensus primer. Type specific PCR to detect HPV 16 and 18 was done on the samples positive on consensus PCR. All participants were tested for associated STIs including HIV and hepatitis B and cervical cytology was done on all females. Results: Among the FSWs, HPV was detected in 73.3% and HPV 16 and 18 was detected in 25.7%. Though the HPV prevalence was similarly high among MSMs (69.2%) and IDUs (72.4%), the prevalence of HPV 16 and 18 was much lower in these groups compared to the FSWs. Prevalence of cervico-vaginal infection with Trichomonas vaginalis and syphilis was significantly higher in the HPV positive women compared to the HPV negative women. There was no statistically significant difference in the prevalence of other STIs among HPV positive and negative women and men. Conclusion: HPV infection is highly prevalent among FSW, MSM and IDUs. Trichomonas vaginalis infection is more frequent in HPV positive women.

        Coumarins from Angelica decursiva inhibit lipopolysaccharideinduced nitrite oxide production in RAW 264.7 cells

        Angelica decursiva has long been used in Korean traditional medicine as an antitussive, analgesic, antipyretic, and cough remedy. In this study, the anti-inflammatory activity of 9 coumarin derivatives isolated from a 90 % methanol fraction was evaluated via inhibition of production of nitric oxide (NO) and tumor necrosis factor-a (TNF-a), as well as the expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX- 2) in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. Among the tested compounds, edulisin II (1) exhibited the most potent NO production inhibitory activity, followed by decursidin (2), Pd–C-III (3), 4-hydroxy Pd–CIII (4), Pd–C-I (5), and Pd–C-II (6). In contrast, (?)-transdecursidinol (7) did not exhibit NO suppressive effects on LPS-stimulated RAW 264.7 cells. Structure-activity relationships revealed that esterification of the hydroxyl at C-30 or C-40 of 7 with an angeloyl/senecioyl/acetyl group is essential for its inhibitory activity against NO production, while the number of angeloyl or senecioyl groups, and their positions greatly affect the potency of these coumarins. Coumarins 1-6 also inhibited TNF-a production and iNOS protein expression, while compounds 1-4 inhibited COX-2 protein expression in LPS-stimulated RAW 264.7 cells. These results suggest that coumarins isolated from A. decursiva might be used as potential leads for the development of therapeutic agents for inflammation-associated disorders.

        『겨울연가(일본에서는 겨울 소나타(冬のソナタ)로 번역』(韓?KBS制作, 2002年, 以下 겨울 소나타로」と 略) 만큼 다종다양한 사람들의 관심을 모으고, 많은 서적이 출판된 드라마가 있었을까? 작금의 일본TV 프로그램은 시청자가 세대별ㆍ성별로 세분화되어 있기 때문에, 금새 소비되고 잊혀져 간다. 서서히 화제가 되기 시작하여 2년 이상에 걸쳐 봄을 일으킨 「겨울 소나타」는 그러한 의미 에서도 특별한 TV프로그램이었다. 또한 한류 붐이 되면서, 단순한 드라마붐이나 아이들(스타) 현상을 넘어 한일 문화교류 이벤트나 학술심포지엄이 다수 개최되었다. 이 글에서는 ‘왜 「겨울 소나타」에 인기가 집중되었는가’의 문제가 아니라 ‘왜 이렇게 많이 「겨울 소나타」 및 「한류 붐」을 이야기하게끔 되었는가’의 문제에 주목하고자 한다. 한류 붐을 이야기하는 사람들은 어떠한 사람들이었으며, 매체들이었을까? 먼저 「겨울 소나타」 현상으로 알려진 TV 드라마의 인기가 점차 「한류」붐으로 발전(等値)하면서 그 성질을 바꾸어 간 경위를 살펴 보자. 다음으로 키워드가 되는 〈타자표상(他者表象)〉과 분석의 틀을 정리한다. 그리고 TV뉴스에서 사용되는 말에 있어서 〈타자표상(他者表象)〉 구축의 구체적인 예(例)에 관하여 분석하고자 한다. 그런 후에 한류 붐에 관하여 다양한 레벨에서의 이야기를 구별하면서〈타자표상(他者表象)〉을 구축하는 언설공간(言說空間)을 추출하고자 한다. 대중문화의 글로벌화가 진행됨에 따라 문화내셔널리즘의 기운이 높아지고 있으며, 성공한 문화상품 의 경우 특정 지역에 기원을 갖는 본질주의적 문화로 생각하는 경향이 강해지고 있다. 문화적 다양성에 관한 논의는 문화의 글로벌화에 대항하는 움직임이나 각 지역의 전통문화로의 회귀를 촉진하는 움직임 과 관련을 갖는 경우가 많다. 그러나 E. 모란이 주장하듯이 어떠한 문화에도 기원에 관해서는 접촉 (接觸)이나 결합(結合), 혼휴(混淆)가 있으며, 순수한 문화란 있을 수 없다. 한류 붐은 대중문화가 국경을 넘어 확대되어 가는 경우 제기되는 일반 문제로서 독해(讀解)될 필요가 있다.

        In the present study, we explore the role of different cluster techniques on the relative role of different cross-sections. The cluster techniques include the simple computer algorithm minimum spanning tree method based on spatial constraint and the one based on the metropolis procedure simulated annealing clusterization algorithm. We find that the relative role of different cross-sections is sensitive toward the clusterization technique used to construct complex multi-bound structures.

        Microwave-assisted polycrystalline Ag/AgO/AgCl nanocomposites synthesis using banana corm (rhizome of Musa sp.) extract: Characterization and antimicrobial studies

        The rapid emergence of multidrug-resistant microbes has turned into the main threat to public healthworldwide. The preparation and application of nanomaterial-based antimicrobials have increased toaddress this problem with great significance. This study has investigated the bio-fabrication of polycrystallineAg-NCs using BCE as a reducer and stabilizer under microwave irradiation. Ag-NCs formationswere optimized using varying concentrations of BCE and silver acetate. The XRD confirmed the incidenceof phases of nanostructured crystals of Ag/AgO/AgCl. The average crystallite sizes were calculated to be22.19, 36.65, and 36.65 nm for Ag, AgO, and AgCl, respectively. SERS spectra revealed the vibrationalmodes of the Ag and Ag-O stretch bands. FTIR spectra have indicated the incidence of chemical fingerprintsfor Ag-NCs and BCE biomolecules. SEM and HR-TEM micrographs exhibited the almost sphericalto the oval shape of 50 nm. SAED pattern attested to silver crystallinity in agreement with XRD analysis. The MIC values of the Ag-NCs against Escherichia coli, Staphylococcus aureus, Aspergillus brasiliensis, andRhizopus stolonifer were observed to be 120, 60, 200, and 100 mg/ml, respectively. This study has demonstratedthe facile method to prepare polycrystalline Ag-NCs with appropriate structural and morphologicalcharacteristics for antimicrobial applications.

        An Overview on Taxol Production Technology and Its Applications as Anticancer Agent

        Cancer is undeniably a major health risk factor that is growing rapidly globally despite promising medicinal and pharmaceutical development. Treatment of cancer remains an issue due to financial constrains in most of the developing countries. The majority of todayʼs medicines are derived from medicinal plants, as we all know. Taxol is such a plant-based chemotherapeutic agent which has blown the anticancer research field. The extinction of Taxus spp., which is the main natural source of taxol synthesis, required the development of novel alternative approaches in the oncology field. Many novel discoveries have been in trend in which endophytic microbial fermentation process is more focused as this area is cost-efficacious, less time consumable, and eco-friendly. Endophytes are one of another alternative approach for the taxol production, preserving the natural resources of Taxus plants worldwide. Endophytic microorganisms can be exploited in the taxol industry because most of the endophytic microbes have yet to be explored for the eco-friendly and economical production of this wonder drug. The aim of this review article is to cover everything from the discovery of taxol through the development of new natural resource-based biotechnology techniques. Furthermore, the current review has emphasised the use of taxol in fields other than anti-cancer properties, as well as pointing to some future opportunities.
