      • eIF2α is a novel O-GlcNAcylated protein under ER stress condition

        O-GlcNAcylation is highly involved in cellular stress responses including the endoplasmic reticulum (ER) stress response. However, it is largely unknown which component(s) of the unfolded protein response (UPR) is directly regulated by O-GlcNAcylation. In this study, eukaryotic translation initiation factor 2α (eIF2α), a major branch of the UPR, was O-GlcNAcylated at Ser 219, Thr 239, and Thr 241. Upon ER stress, eIF2α is phosphorylated at Ser 51 by PERK and this inhibits global translation initiation, except for that of specific mRNAs that induce stress-responsive genes such as CHOP. Hyper-O-GlcNAcylation hindered phosphorylation of eIF2α at Ser 51. The level of O-GlcNAcylation of eIF2α was changed by DTT dependent on its phosphorylation at Ser 51. Point mutation of the O-GlcNAcylation sites of eIF2α increased its phosphorylation at Ser 51 and CHOP expression and resulted in increased apoptosis upon ER stress. This O-GlcNAcylation of eIF2α was reproduced in thiamet-G-injected mouse liver. In conclusion, proper regulation of O-GlcNAcylation and phosphorylation of eIF2α is important to maintain cellular homeostasis upon ER stress.

      • Selective Autophagy Receptors Interact with EDEM1 During itsDegradation

        EDEM1 is an endoplasmic reticulum-associated protein degradation (ERAD) component. ERAD is a cellular pathway that targets misfolded and misassembled glycoproteins for degradation. EDEM1 is involved in the recognition of misfolded luminal glycoproteins and in routing them for dislocation to the cytosol, followed by their degradation. EDEM1 interacts with substrate glycoproteins after their exit from the calnexin/calreticulin cycle and after processing by ER-mannosidase I. Although EDEM1 was proposed to be lectin–like and to react with Man8GlcNAc2 oligosaccharides, itsmechanism of action and its fate are still largely unknown. In a previous report, we found that EDEM1 becomes rapidly degraded and that this occurs by basal autophagy. Here, we show that EDEM1 forms complexes with the selective autophagy receptor p62, NBR1 and Alfy. This was demonstrated in HepG2 cells by double immunogold electron microscopy. Furthermore, we show the interaction between p62 and EDEM1 by immunoprecipitation- Western blot experiments. By serial section analysis, the origin of the phagophore for selective autophagy of EDEM1 could be identified as modified parts of rough ER cisternae. Hence, we provide new insight into the details of EDEM1 degradation process.

        간호학생을 위한 격리실-시뮬레이션 기반 감염관리 교육의 효과

        Purpose: This study aimed to analyze the effects of infection control education (ICE) prospectively, based on isolation room simulation for nursing college students. Methods: From September 21 to 30 2020, a total of 97 students in the 4th grade of nursing college located in D city, were enrolled (48 in the experimental group and 49 in the control group). The experimental group received an isolation room simulation for ICE. The control group conducted an objective structured clinical evaluation for the same education. The effects after education were evaluated using the items of awareness of standard precaution, knowledge of wearing personal protective equipment (PPE), and confidence in wearing it. Results: There was no statistically significant difference in the awareness of standard precautions between the two groups, and the knowledge of wearing PPE was significantly higher in the experimental group (t=-3.41, p<.001) after education. The confidence score for wearing PPE was also significantly higher in the experimental group (t=-2.23, p=.028). Conclusion: The ICE using the isolation room simulation can be effective for knowledge and confidence regarding the aspect of wearing PPE for nursing students, during and after the pandemic.

        C. S. 루이스의 『마법사의 조카』와 성경에 나타난 창조의 비교 연구

        The purpose of this paper is to compare the creation in the Bible and The Magician’s Nephew. This dissertation suggests that the biblical world can be shown in much abundance and gives readers pleasure through literature and allusion. This thesis suggests that the Biblical world can be delineated in much abundance and pleasure through literature technique such as allusion by comparing the creation stories revealed in Lewis’s work with those in the Bible. Despite of the fact that there are undeniable substantial differences between the creation stories of the Bible and those in The Magician’s Nephew, this study shows an imaginary world of literary portrayal and essential concepts of the Bible are effectively conceived through allusions in the work. The possibilities of more pleasant and vivid way of understanding of the real world created by God through literary works are also discussed.

        병원 간호사의 코로나바이러스감염증-19 감염관리 수행에 영향을 미치는 요인

        본 연구는 병원 간호사를 대상으로 감염관리 조직문화, 자원의 지원, 코로나19 지식, 회복탄력성이 COVID-19 감염관리 수행에 영향을 미치는 요인을 파악하고자 시도되었다. 2022년 7월 27일부터 8월 10일까지 자가보고식 설문지를 통해 총 150명의 간호사에게 데이터를 수집하였으며, 그 중 147명의 자료가 연구에 활용되었다. 다중회귀분석에서 자원의 지원과 코로나19 지식, 회복탄력성이 코로나19 감염관리 수행의 의미 있는 요인으로 분석되었다. 지속되는 코로나19 팬데믹 상황에서 간호사의 코로나19 감염관리 수행을 향상시키기 위해 조직 차원의 지원과 개인의 감염관리 역량을 증가시킬 수 있는 전략을 위한 프로그램 개발 및 평가가 필요하다. This study was performed to identify the factors affecting ‘the organizational culture for infection control’, ‘the resource support’, ‘the knowledge about COVID-19’, and ‘the resilience’ among hospital nurses on the performance of COVID-19 infection control. A total of 150 nurses, registered in Korea, participated in the current study. The data were collected through self-report questionnaires. From 27th July to 10th August, 2022, 147 questionnairesout of 150 were collected. In multiple regression analysis, ‘the resource support’, ‘the knowledge about COVID-19’ and ‘the resilience’ were analyzed as significant factors in performing of the infection control for COVID-19 (β = 0.68, p < .001, β = 0.12, p = .023 and β = 0.20, p = .005 respectively). For improving the nurses’ performance of infection control against Coronavirus Disease 2019(COVID-19) during the pandemic, it is necessary to develop and evaluate the programs through the organizational support and strategies to improve individual knowledge.

        Mettl14 mutation restrains liver regeneration by attenuating mitogens derived from non-parenchymal liver cells

        Liver regeneration is a well-known systemic homeostatic phenomenon. The N6-methyladenosine (m6A) modification pathwayhas been associated with liver regeneration and hepatocellularcarcinoma. m6A methyltransferases, such as methyltransferase 3(METTL3) and methyltransferase 14 (METTL14), are involvedin the hepatocyte-specific-regenerative pathway. To illustratethe role of METTL14, secreted from non-parenchymal liver cells,in the initiation phase of liver regeneration, we performed 70%partial hepatectomy (PH) in Mettl14 heterozygous (HET) andwild-type (WT) mice. Next, we analyzed the ratio of liver weightto body weight and the expression of mitogenic stimulatorsderived from non-parenchymal liver cells. Furthermore, weevaluated the expression of cell cycle-related genes and thehepatocyte proliferation rate via MKI67-immunostaining. Duringregeneration after PH, the weight ratio was lower in Mettl14HET mice compared to WT mice. The expressions of hepatocytegrowth factor (HGF) and tumor necrosis factor (TNF)-α,mitogens derived from non-parenchymal liver cells that stimulatethe cell cycle, as well as the expressions of cyclin B1 andD1, which regulate the cell cycle, and the number of MKI67-positive cells, which indicate proliferative hepatocyte in the lateG1-M phase, were significantly reduced in Mettl14 HET mice72 h after PH. Our findings demonstrate that global Mettl14mutation may interrupt the homeostasis of liver regenerationafter an acute injury like PH by restraining certain mitogens,such as HGF and TNF-α, derived from sinusoidal endothelialcells, stellate cells, and Kupffer cells. These results providenew insights into the role of METTL14 in the clinical treatmentstrategies of liver disease.

        Setting School-Level Educational Goal and Objectives with the Modified Delphi Method

        Purpose: The aim of this study was to set the school-level educational goals and objectives, based on the needs analysis, by modified Delphi method. Methods: A needs analysis and workshops were performed to establish educational goals and objectives. The needs analysis comprised 3 rounds of questionnaires and a panel and reference group that compared the results. Various workshops were held to set, outline, develop, and promote the educational goals and objectives and perform a satisfaction survey. Results: In the needs analysis, we identified 8 keywords for ‘ends’ and 12 for ‘means’ with regard to educational goals and 25 keywords for educational objectives, which were summarized in 5 factors (categories). There were significant differences between the panel and reference groups. Through the workshops, we established new educational goal and objectives that met with high satisfaction among members. Conclusion: The developmental process with which the educational goals and objectives were established through a needs analysis and workshops was effective, efficient, and supportive in medical education.

        <P><B>Quasi‐amorphous colloidal structures</B> exhibiting angle‐independent tunable photonic colors in response to the electric stimuli. Moderately polydisperse colloidal Fe<SUB>3</SUB>O<SUB>4</SUB>@SiO<SUB>2</SUB> nanoparticles dispersed in organic solvents exclusively form quasi‐amorphous photonic materials at sufficiently high concentrations, and which reversibly reflect incident light in visible region in response to the relatively low bias voltages. </P>
