        Photosynthetic Characteristics of Resistance and Susceptible Lines to High Temperature Injury in Panax ginseng Meyer

        In this study, photosynthetic parameters such as the net photosynthesis rate, stomatal conductance, intercellular CO2 concentration, and transpiration rate were examined in selected ginseng varieties and/or lines that are resistant (Yunpoong, HTIR 1, HTIR 2, and HTIR 3) and susceptible (Chunpoong) to high temperature injury (HTI). The net photosynthesis rate increased with the increase in the light intensity in all the HTI-resistant and -susceptible ginseng lines with a light saturation point of 200 μmol m-2s-1, except for Yunpoong that had a light saturation point of 400 μmol m-2s-1. At the light saturation point, the net photosynthesis rate in July was highest in HTIR 3, at 4.2 μmol CO2 m-2s-1, and was lowest in Yunpoong, HTIR 1, Chunpoong, and HTIR 2, in that order, at 1.9 to 3.7 μmol CO2 m-2s-1. The net photosynthesis rate in August was highest in Yunpoong at 5.9 μmol CO2 m-2s-1, and lowest in HTIR 1 and HTIR 3 (4.5 μmol CO2 m-2s-1) and in other lines, in that order, at 2.8 to 2.9 μmol CO2 m-2s-1. The stomatal conductance in July was highest in HTIR 3 (0.055 mol H2O m-2s-1) and Yunpoong, Chunpoong, HTIR 1, and HTIR 2 were 0.038, 0.037, 0.031, and 0.017 in that orders. In August, meanwhile, HTIR 1 showed the highest as 0.075, and followed by HTIR 3, Chungpoong, and HTIR 2 with 0.070, 0.047, and 0.023, respectively. The intercellular CO2 concentration at the light saturation point in July and August was much lower in HTIR 2 at 139 and 185 μmol mol-1 than in the other ginseng lines at 217 to 257 and 274 to 287 μmol mol-1, respectively. The transpiration rate in July and August was higher in the HTI-resistant lines of Yunpoong, HTIR 1, and/or HTIR 3 at 0.83 to 1.03 and 1.67 to 2.10 mol H2O m-2s-1 than in the other ginseng lines at 0.27 to 0.79 mol H2O m-2s- 1 and 0.51-1.65 mol H2O m-2s-1, respectively. Conclusively, all the photosynthetic parameters that were examined in this study were generally higher in the HTI-resistant ginseng lines than in the HTI-susceptible lines, except for HTIR 2, and were much higher in August than in July, especially in the resistant ginseng lines. All these results can be used to provide basic information for the selection of HTI-resistant ginseng lines and the application of cultural practices that are effi cient for ginseng growth, based on the photosynthetic characteristics of the lines.

        In Vitro Suppression of Natural Killer Cell Cytolytic Activities by Prednisolone in Patients with History of Unexplained Recurrent Spontaneous Abortion and Elevated Peripheral Blood NK Cell Fraction

        We aimed to evaluate the effect of prednisolone (PDS) on natural killer cell (NK cell) cytolytic activity in vitro. Methods Blood samples from 74 patients with history of unexplained recurrent spontaneous abortion who elevated peripheral blood NK cell fraction were collected prospectively. Peripheral blood monocytes which containing NK cells were isolated and separated to three different tubes which containing target (K562) cells by the 50:1 effector to target (E:T) ratio. PDS or intravenous immunoglobulin (IVIG) was additionally added to 2 tubes for evaluate their suppressive effect. The percentage killing of target cells was recorded numerically by using flow cytometer and the values between groups were statistically analyzed. Results The mean target cell killing percentage was 40.5% in co-culture tube which was not added PDS or IVIG. In culture experiments which was added IVIG, the killing percentage is reduced to 37.7% which showed no significant differences compared to that of co-cultured tube which was not added PDS or IVIG. But, in experiments with added PDS, the killing percent was reduced to 19.5% and the difference was statistically significant ( P<0.001) compared to that of co-cultured tube which was not added PDS or IVIG. On comparing the reduction in killing percentage of target cells by PDS and IVIG, statistically significant reduction in the PDS coculture was noted ( P<0.005). Conclusion NK cell cytolytic activity is effectively down-regulated by using PDS in vitro. Moreover, the effect of PDS in down-regulation of NK cell cytolytic activity is seems to superior than that of IVIG. But, large scaled in vivo study is needed.

        중증 자궁내막증의 수술적 치료 후 체외수정의 결과

        목적: 중증 자궁내막증 환자에서 수술적 처치가 체외수정 결과에 미치는 영향을 평가하고, 특히 난소 자궁내막증과 복막 자궁내막증 환자의 수술적 처치 후 체외수정시술의 척도들을 비교해 보고자 한다. 연구방법: 1997년에서 2004년까지, 복강경검사로 중증 자궁내막증을 진단 받고 체외수정을 시행한 506주기를 대상으로 후향적 연구를 하였다. 170주기는 수술받지 않았고, 336주기는 수술적 치료(151주기는 난소 자궁내막증, 185 주기는 복막 자궁내막증) 후 체외수정시술을 받았다. 대조군은 동일 기간 내에 난관요인에 의해 체외수정시술을 받은 313주기를 대상으로 하였다. 결과: 자궁내막증의 수술적 처치를 한 그룹과 수술을 받지 않은 그룹에서, 채취된 난자 수(9.6 ± 7.3 vs. 11.2 ± 7.5 vs. 14.8 ± 7.0, P <0.0001) 와 이식된 배아 수(3.5 ± 1.4 vs. 3.5 ± 1.4 vs. 3.9±0.9, P = 0.001)가 유의하게 대조군 보다 낮았다. 하지만, 착상률(17.2% vs. 15.2% vs. 15.9%)과 임신율(36.9% vs. 35.9% vs. 39.6%), 태아생존율(32.1% vs. 28.2% vs. 29.7%)은 유사하였다. 수술받은 복막 자궁내막증 그룹의 체외수정에서는 채취된 난자 수(9.6 ± 7.9 vs. 9.8 ± 6.5), 이식된 배아수(3.4 ± 1.4 vs. 3.7 ± 1.3), 착상률 (15.5% vs. 18.0%), 임신율(34.6% vs. 39.7%), 태아생존율(29.2% vs. 35.8%)이 수술받은 난소 자궁내막증 그룹과 유의한 차이를 보이지 않았다. 결론: 자궁내막증 환자는 과거의 수술적 치료 유무와 상관없이 체외수정시술 척도가 낮았으나, 임신율에는 차이가 없었다. 수술적 치료를 받은 복막 자궁내막증 그룹은 난소 자궁내막증과 유사하게 낮은 체외수정시술 척도를 보였다. 따라서, 난소 자궁내막증 수술 여부와 관계없이 자궁내막증 자체만으로 체외수정시술 척도에 안 좋은 영향을 미치는 것으로 생각된다. Objective: The aim of this study is to evaluate in vitro fertilization (IVF) outcomes in advanced endometriosis with/without surgical treatment and to compare between cyst enucleation and surgically treated peritoneal lesions without endometrioma. Methods: We performed a retrospective analysis of 506 IVF cycles (345 patients) with stage III-IV endometriosis (170 cycles without surgical treatment, 336 cycles with the surgical treatment) between 1997 and 2004. Among the surgical treatment, 151 cycles were performed previously cyst enucleation with endometrioma and 185 cycles were performed only resection of peritoneal lesions without endometrioma enucleation. 313 cycles (246 patients) with the tubal factor infertility were used as controls. Results: In the endometriosis group with/without surgical treatment, the number of retrieved oocytes (9.6±7.3 vs. 11.2 ± 7.5 vs. 14.8 ± 7.0, P < 0.0001) and the transferred embryos (3.5 ± 1.4 vs. 3.5 ± 1.4 vs. 3.9 ± 0.9, P = 0.001) was significantly lower than controls. But, implantation rates (17.2% vs. 15.2% vs. 15.9%), pregnancy rates (36.9% vs. 35.9% vs. 39.6%) and live birth rates (32.1% vs. 28.2% vs. 29.7%) were similar. Surgically treated peritoneal lesions group showed no difference in the number of retrieved oocytes (9.6 ± 7.9 vs. 9.8 ± 6.5), the number of transferred embryos (3.4 ± 1.4 vs. 3.7 ± 1.3), implantation rates (15.5% vs. 18.0%), pregnancy rates (34.6% vs. 39.7%) and live birth rates (29.2% vs. 35.8%) with cyst enucleation group. Conclusion: IVF parameters in the patients with endometriosis are inferior to tubal factor infertility, but IVF outcomes are comparable. Surgically treated peritoneal lesions group without endometrioma enucleation shows IVF parameters as bad as cyst enucleation group. We might be considered endometriosis itself worse IVF parameters regardless previous cyst enucleation.

        영아를 둔 아버지들의 부모 효능감, 놀이 신념, 그리고 영아의 놀이성 간의 관계 연구

        영아를 둔 아버지의 부모 효능감, 놀이 신념, 그리고 영아의 놀이성 간의 관계를 살펴보기 위해 본 연구를 실시하였다. 본 연구의 참여자는 인천 및 경기도에 위치한 보육시설에 다니고 있는 만 2세의 영아를 둔 아버지 152명과 영아를 담당한 교사들 15명이었다. 만 2세의 영아를 둔 아버지의 부모 효능감과 놀이 신념을 측정하기 위해 부모 효능감 척도(Parent Sense of Competence: PSCO)와 부모 놀이 신념 척도 (The Parent Play Beliefs Scale: PPBS)를 사용하였고, 영아를 담당한 교사들이 영아의 놀이성을 측정하기 위해 영아의 놀이성 척도 CPS(Children``s Playfulness Scale)를 사용하였다. 수집된 자료는 연구문제에 따라 독립표본t-test, 일원변량분석(one-way ANOVA), Pearson의 상관관계 분석을 실시하였다. 본 연구의 결과는 다음과 같다. 먼저, 아버지의 효능감은 아버지 연령, 학력, 소득의 종류에 따라 차이가 없었다. 아버지의 놀이 신념은 연령에 따라 차이가 없었지만, 학력에 따라 놀이지지 놀이신념은 차이가 있었고, 소득에 따라 학습 중심의 놀이 신념은 차이가 있었다. 영아의 놀이성은 아버지의 연령에 따라 차이가 없었지만, 아버지의 학력, 소득에 따라 차이가 있었다. 둘째, 아버지의 부모효능감과 놀이지지 놀이신념은 긍정적인 관계가 있었고, 아버지의 부모효능감은 영아 놀이성의 하위 영역인 사회적 자발성에서 긍정적인 관계가 있었다. 그러나 아버지의 놀이 신념과 영아 놀이성은 통계적으로 유의한 상관관계가 존재하지 않았다. 본 연구의 결과는 영아의 놀이성에 영향을 미치는 아버지의 부모효능감과 놀이신념에 대한 올바른 이해와 더 나아가 영아를 둔 아버지의 자녀양육을 어떻게 도움을 줄 수 있는지 살펴보는데 그 의의가 있다. The aims of this study are to investigate the differences between parent efficacy, play beliefs, and toddlers’ playfulness according to the sociodemographic variables of two-year-old toddlers’ fathers and to examine the correlation between parent efficacy, play beliefs, and toddlers’ playfulness toward their fathers. The subjects of this study were 152 two-year-old toddlers enrolled in 30 child care facilities, including public, private, and home child care facilities, in Incheon and Gyeoggido Province. In order to measure the fathers’ play beliefs, parent efficacy and toddlers’ playfulness the Parent Play Beliefs Scale (PPBS), the Parent Sense of Competence (PSCO), and the Children``s Playfulness Scale (CPS) were used respectively. The collected data were analyzed throught-test, one-way ANOVA, and Pearson correlation coefficient test according to the research questions. The major results of this study are as follows. No differences were found between play-centered play beliefs andlearning-centered play beliefs in relation to the variables of the fathers’ ages. However, there was a difference in play-centered play beliefs in relation to the fathers’ educational backgrounds, while a difference in learning-centered play beliefs was also found in regard to the variable of income. Fathers’ efficacy was indifferent to the variables, including age, educational background, and income. There were no differences in toddlers’ playfulness in relation to fathers’ ages, but a difference was noted in toddlers’ playfulness in relation to fathers’ educational backgrounds and incomes. Fathers’ play beliefs were positively correlated with parent efficacy but proved not to have a statistically significant correlation with toddlers’ playfulness. The value of this research lies not only in its help in understanding accurately and better utilizing play by providing basic data that serve as positive factors in fathers’ beliefs affecting toddlers’ play but also in its potential role to assist child care by theorizing about fathers’ roles and the importance of parenting styles.

      • Implication of microRNA regulation in <i>para</i> -phenylenediamine-induced cell death and senescence in normal human hair dermal papilla cells

        <P><I>Para</I>-phenylenediamine (PPD) is a major component of hair coloring and black henna products. Although it has been largely demonstrated that PPD induces allergic reactions and increases the risk of tumors in the kidney, liver, thyroid gland and urinary bladder, the effect on dermal papilla cells remains to be elucidated. Therefore, the current study evaluated the effects of PPD on growth, cell death and senescence using cell-based assays and microRNA (miRNA) microarray in normal human hair dermal papilla cells (nHHDPCs). Cell viability and cell cycle analyses demonstrated that PPD exhibited a significant cytotoxic effect on nHHDPCs through inducing cell death and G<SUB>2</SUB> phase cell cycle arrest in a dose-dependent manner. It was additionally observed that treatment of nHHDPCs with PPD induced cellular senescence by promoting cellular oxidative stress. In addition, the results of the current study indicated that these PPD-mediated effects were involved in the alteration of miRNA expression profiles. Treatment of nHHDPCs with PPD altered the expression levels of 74 miRNAs by ≥2-fold (16 upregulated and 58 downregulated miRNAs). Further bioinformatics analysis determined that these identified miRNA target genes were likely to be involved in cell growth, cell cycle arrest, cell death, senescence and the induction of oxidative stress. In conclusion, the observations of the current study suggested that PPD was able to induce several cytotoxic effects through alteration of miRNA expression levels in nHHDPCs.</P>

      • Thuja orientalis reduces airway inflammation in ovalbumin-induced allergic asthma

        <P>Thuja orientalis (TO) may be used as a herbal remedy for the treatment of numerous inflammatory diseases. In the present study, the effects of TO were evaluated on airway inflammation in ovalbumin (OVA)?induced allergic asthma and RAW264.7 murine macrophage cells. The effects of TO on the production of proinflammatory mediators, were determined in RAW264.7 cells that had been stimulated with lipopolysaccharide (LPS). Furthermore, an in vivo experiment was performed on mice that were sensitized to OVA and then received an OVA airway challenge. TO was administered by daily oral gavage at a dose of 30 mg/kg, 21?23 days after the initial OVA sensitization. TO was shown to reduce nitric oxide production and reduce the relative mRNA expression levels of inducible nitric oxide synthase (iNOS), interleukin (IL)?6, cyclooxygenase?2, matrix metalloproteinase (MMP)?9, and tumor necrosis factor?α in RAW264.7 cells stimulated with LPS. In addition, TO markedly decreased the inflammatory cell counts in bronchial alveolar lavage fluid, reduced the levels of IL?4, IL?5, IL?13, eotaxin and immunoglobulin E, and reduced airway hyperresponsivenes, in the OVA sensitized mice. Furthermore, TO attenuated airway inflammation and mucus hypersecretion, induced by the OVA challenge of the lung tissue. TO also reduced the expression of iNOS and MMP?9 in lung tissue. In conclusion, TO exerted anti?inflammatory effects in an OVA?induced allergic asthma model, and in LPS?stimulated RAW264.7 cells. These results suggest that TO may be a useful therapeutic agent for the treatment of inflammatory diseases, including allergic asthma.</P>

        Illudins C<sub>2</sub> and C<sub>3</sub> Stimulate Lipolysis in 3T3-L1 Adipocytes and Suppress Adipogenesis in 3T3-L1 Preadipocytes

        <P>The secondary metabolites illudins C<SUB>2</SUB> (<B>1</B>) and C<SUB>3</SUB> (<B>2</B>), obtained from the culture broth of <I>Coprinus atramentarius</I>, have been shown to possess antimicrobial activity. In the present study, we discovered novel biological activities of <B>1</B> and <B>2</B> in lipolysis of differentiated 3T3-L1 adipocytes and adipogenesis of 3T3-L1 preadipocytes. Compounds <B>1</B> and <B>2</B> exhibit a dose-dependent increase in glycerol release and thereby reduce intracellular lipid accumulation. The stimulatory effects of <B>1</B> and <B>2</B> on lipolysis are prevented by cAMP-dependent protein kinase (PKA) and extracellular signal-regulated kinase (ERK) inhibitors. Compounds <B>1</B> and <B>2</B> down-regulated perilipin and also affected the mRNA and protein levels of adipose triglyceride lipase (ATGL) and hormone-sensitive lipase (HSL). However, <B>1</B> and <B>2</B> treatment leads to a significant increase in PKA-mediated phosphorylation of HSL at S563 and S660. In addition, <B>1</B> and <B>2</B> treatment in 3T3-L1 preadipocytes induces down-regulation of the critical transcription factors, CCAAT/enhancer binding protein α and β (C/EBPα and C/EBPβ), and peroxisome proliferator activated receptor γ (PPARγ), which are required for adipogenesis, and accordingly inhibits adipogenesis.         Objective:The aim of this study is to compare the frequency of postoperative epilepsies of patients with chronic as opposed to recent onset epilepsy due to glial tumors in the frontal or temporal lobe with the hypothesis that patients with chronic epilepsy do worse. Methods : We compared the clinical and diagnostic characteristics of the patients(n=73) who had seizures preoperatively to those of the patients(n=153) who did not. Among those who have had seizures preoperatively, we compared those(n=32, chronic seizure group) who had seizures a year or more prior to surgery to those(n=41, acute seizure group) who had seizures within a year prior to surgery. Results : Among the various factors, the frequency of benign pathology and favorable neurological state were higher in seizure group than in non-seizure group(p<0.05). Complex partial seizure and low-grade tumors were frequent in chronic seizure group, whereas simple partial seizure and high-grade tumors were frequent in acute seizure group. Seizure-free rate was significantly higher in acute seizure group than in chronic one(p<0.05). Also, the difference of seizure control rate between surgical strategies were statistically significant(p<0.05). Conclusion : This study indicates that preoperative seizure durations and frequencies have a close relationship with the frequency of postoperative epilepsy of glial tumors. A longer lapse may allow the formation of epileptogenic foci, leading to chronic epilepsy, and eventually having a negative effect on the prognosis of the patients. Factors including histopathological characteristics of the tumor, its location, seizure duration/frequency, and semiology should be taken account of deciding on surgical strategies.         Background: This study was aimed to investigate the initial treatment outcomes and prognosis and to analyze factors associated with initial treatment failure and short-term (30-day)mortality of spontaneous bacterial peritonitis (SBP) in cirrhotic patients. Methods: A total 151 cases of SBP diagnosed in patients with liver cirrhosis between January 2003 and December 2011 were included. SBP was diagnosed based on a polymorphonuclear cell count in ascitic fluid of >250 cells/㎟ in the absence of data compatible with secondary peritonitis. Results: The mean age was 57.6 years, 79.5% of the SBP occurred in men. The rate of initial treatment failure was 58/151 (38.4%). In multivariate analysis, hepatocellular carcinoma (Odd ratio 2.770, p=0.021), gastrointestinal hemorrhage (Odd ratio 4.966, p=0.015), and renal impairment (Odd ratio 4.564, p=0.006) were most important factors associated with initial treatment failure. 7-day and 30-day mortality were 15.2% and 27.8%, respectively. Hepatocellular carcinoma (Odd ratio 2.96, p=0.007) and renal impairement (Odd ratio 4.13, p<0.001) were independent prognostic factors of 30-day mortality. Conclusions: Hepatocellular carcinoma and renal impairment is the main factors associated with initial treatment failure and short-term mortality in patients with SBP.
