      • Clinical features of vascular events in patients with al amyloidosis

        Background and Aims: AL amyloidosis can cause various vascular events, which are mainly related to vascular amyloid deposition or thromboembolism. Despite their clinical importance, only few reports have been published on the manifestations of vascular events in these patients. Methods: We retrospective reviewed medical records of patients including various imaging study results such as angiography, Doppler sonography, computed tomography, and magnetic resonance imaging to find out clinically significant vascular stenosis and thromboembolism. Results: Between January 2001 and March 2014, 106 patients (male/female=60/46) with biopsy-proven AL amyloidosis were included from 3 hospitals. The median age was 58 years (range, 23-79 years). During a median follow-up of 12.6 months (range, 0.1-156.8 months), 13 events of thromboembolism developed in 13 patients: 10 patients (9.4%) experienced cerebral infarction, 2 patients (1.9%) pulmonary embolism and 1 patient (0.9%) deep vein thrombosis. Patients with higher free light chain difference suffered significantly more from vascular events than the others in multivariable analysis. (20.2% vs. 8.9%, p=0.022). The median overall survival (OS) from pathologic diagnosis was 20.9 months (95% CI, 5.7-36.2). In multivariable analysis, higher BM plasma cell percentage and higher serum B2 microglobulin levels were poor prognostic factors. (p<0.05) Conclusions: The incidence of vascular events was substantial in patients with AL amyloidosis. Vascular events significantly occurred in patients with higher free light chain difference. Poor prognostic factor for overall survival was higher serum beta-2 microglobulin.

      • KDM3A histone demethylase functions as an essential factor for activation of JAK2−STAT3 signaling pathway

        <P><B>Significance</B></P><P>An aberrant covalent histone modification which drives dysregulation of transcriptional program is related to human diseases such as cancer. Thus, identifying signaling pathways modulating transcription factors and epigenetic enzymes are coming into view as attractive therapeutic targets. In this study, we provide evidence that KDM3A is tyrosine-phosphorylated by JAK2, and tyrosine-phosphorylated KDM3A acts as a coactivator for STAT3, thereby exerting increased cancer cell growth and motility. We propose that JAK2-dependent tyrosine phosphorylation of KDM3A could be a potential therapeutic target for epigenetic control of oncogenic effect governed by JAK2−STAT3 signaling pathway.</P><P>Janus tyrosine kinase 2 (JAK2)−signal transducer and activator of transcription 3 (STAT3) signaling pathway is essential for modulating cellular development, differentiation, and homeostasis. Thus, dysregulation of JAK2−STAT3 signaling pathway is frequently associated with human malignancies. Here, we provide evidence that lysine-specific demethylase 3A (KDM3A) functions as an essential epigenetic enzyme for the activation of JAK2−STAT3 signaling pathway. KDM3A is tyrosine-phosphorylated by JAK2 in the nucleus and functions as a STAT3-dependent transcriptional coactivator. JAK2−KDM3A signaling cascade induced by IL-6 leads to alteration of histone H3K9 methylation as a predominant epigenetic event, thereby providing the functional and mechanistic link between activation of JAK2−STAT3 signaling pathway and its epigenetic control. Together, our findings demonstrate that inhibition of KDM3A phosphorylation could be a potent therapeutic strategy to control oncogenic effect of JAK2−STAT3 signaling pathway.</P>

        ORIGINAL ARTICLE: A Clinicopathologic Study of Thirty Cases of Acquired Perforating Dermatosis in Korea

        Background: Acquired perforating dermatosis (APD) is histopathologically characterized by transepidermal elimination of materials from the upper dermis. APD can be divided into four diseases: Kyrle`s disease, perforating folliculitis, elastosis perforans serpiginosa, and reactive perforating collagenosis. APD is usually associated with systemic diseases, especially diabetes mellitus or chronic renal failure. So far, there have only been a few Korean studies of APD, which have a limited number of patients. Objective: The aim of this study is to evaluate the clinical and histopathologic characteristics of 30 cases of APD and to examine the association with systemic diseases. Methods: We retrospectively reviewed the medical records and biopsy specimens of 30 patients who were diagnosed with APD. Results: The mean age was 55.5 years, and the average duration of the lesion was 7.8 months. The lower extremities (73.3%) were the most frequently occurring sites of the lesion. Twenty-five patients (83.3%) had pruritus, and Koebner`s phenomenon was present in 11 patients. Patients of 63.3% had at least one systemic disease. Diabetes mellitus (n=17, 56.7%) and chronic renal failure (n=10, 33.3%) were the most commonly associated conditions. Most patients received topical steroids (93.3%) and antihistamines (80.0%). The most common histopathologic type was reac-tive perforating collagenosis (n=23, 73.3%). Conclusion: In this study, most patients had a systemic association to the diseases. Therefore, we suggest that further evaluation is necessary for patients who present with APD. This includes reviewing patient`s comprehensive past medical history, clinical exam, and additional diagnostic testing to check for the possibility of associated systemic diseases.

        Foodborne Viruses Detected Sporadically in the Fresh Produce and Its Production Environment in South Korea

        <P> Contamination of fresh vegetables and berries with human enteric viruses is a major cause of food poisoning. The aim of this study was to investigate the prevalence of norovirus GI, norovirus GII, hepatitis A virus (HAV), adenovirus, astrovirus, rotavirus, and male-specific coliphage systematically in fresh fruit and vegetables and associated agricultural environmental samples, including irrigation water, soil, and worker's gloves. Enteric viruses were detected by international standard methods (ISO/TS 15216), and male-specific coliphages were isolated using US EPA Method 1601. For the study, 773 samples were collected from June 2016 to April 2017, including Chinese cabbage (n=244), cucumber (n=98), lettuce (n=73), strawberry (n=120), soil (n=191), irrigation water (n=14), and gloves (n=27). Two cucumber and two irrigation water samples were positive for norovirus GI, and one cucumber and two irrigation water samples were positive for norovirus GII. HAV was detected in one strawberry sample and one glove sample. The other tested foodborne viruses were not detected in any of the samples. Sixteen male-specific coliphages were isolated from Chinese cabbage, cucumber, lettuce, cherry tomato, soil, and irrigation water. The isolation of male-specific coliphage would be more practical to investigate the fecal contamination in produce rather than pathogenic viruses. </P>

        Development of high durability plasma filter for air circulating disinfection system

        Deadly diseases are caused by pathogenic bacteria and viruses that spread, among other means, through air circulating systems; hence, it is important to focus on pathogen removal from the air before circulating air through the system. Our paper introduces a novel plasma-based filter that, when used in an air circulating system with particulate air filter, disinfects the air flow. This device, based on dielectric filter discharge (DFD) structure with low pressure drops, indicates easy installation into existing air circulating system. Its performance was evaluated in accordance with the specifications of duct used in hospitals, with consistent O3 generation during 200 h showing high durability. Escherichia coli and Micrococcus luteus were used as the target airborne bacteria; the system exhibited a removal efficiency of approximately 99.99% on bacterial aerosols and continuous bactericidal action, demonstrating that the DFD system can be directly applied to existing air circulating systems.

        지역축제의 서비스품질이 만족과 재방문의도에 미치는 영향 연구

        지역축제는 지역의 문화와 특성을 알릴 수 있는 대표적인 행사이다. 부안의 마실축제는 과거 원전수거물 처리센터 건립을 둘러싼 갈등을 해소하고 지역민의 화합을 개최목적으로 시작하여 군민과 관광객이 함께 만드는 거리·화합형 대표축제로 브랜딩하고 있다. 축제가 제공하는 다양한 프로그램과 서비스는 축제에 참가하는 지역주민 및 외지인에게 중요한 사안이고 끊임없이 모니터링 해야 하는 사항이다. 이러한 서비스 평가를 통하여 축제개발과 기획에 보완사항과 프로그램 개발 및 서비스 증진에 반영하는 것은 기존의 다른 많은 연구에서도 제시되고 있다. 부안 마실축제의 발전과 성장을 위하여 부안 마실축제의 서비스 품질을 평가하는 것은 이러한 축제의 성공과 발전을 위하여 필수적인 사항이라고 할 수 있다. 이에 본 연구는 부안 마실축제 서비스 품질을 축제안내, 프로그램, 기념품, 음식, 부대시설 등의 내용을 포함한 측정변수를 활용하여 확인적 요인분석을 실시하고, 축제만족도 및 충성도에 대한 구조모형분석을 실시하였다. 설문조사는 축제 현장에서 축제행사경험을 마친 방문객을 대상으로 실시하였다. 분석결과, 축제행사내용, 편의시설, 기념품 등이 축제만족도에 구조적 영향관계를 보이고 있었다. 이는 축제의 기본적인 프로그램 내용이 중요하며, 쉴 수 있는 공간, 화장실 등의 편의시설 등이 중요한 사안으로 받아들여지고 있다는 것으로 이해될 수 있다. 특히 부안 마실축제의 기념품 등이 유의적인 관계를 보이는 것은 다른 기존 축제와 다른 특성을 반영한 결과로 이해할 수 있다. 결과적으로 부안은 축제의 프로그램과 내용, 그리고 편의시설강화를 더욱 필요로 하며, 지역 특산 기념품 등에 대한 만족도 향상을 더욱 검토해야 할 것이다. 또한 음식과 안내 등에 대한 보안과 검토를 통하여 이에 대한 만족도가 향상될 수 있도록 노력해야 할 것이다. 부안 마실축제는 거리형 주민화합축제로서 지역주민의 협력과 협조가 더욱 필요한 축제임으로 향후 지역주민의 협력과 관련한 속성 및 협력요인에 대한 연구가 필요하다고 본다. This paper is focused on the service quality of Buan Masil Festival. The objectives of the research were to explore whether or not the service quality of local festival affects overall satisfaction and revisit intention Data were collected using self-administered questionnaires at the 4th Buan Masil Festival on May 6-8, 2015. A total of 373 valid samples were used for the analysis. The results of the study indicate that service quality of program, souvenir, and facility lead to visitors" satisfaction, and visitors" satisfaction influenced revisit intention. The results concludes with a discussion of implication for future research.

        Regulation of Notch1/NICD and Hes1 expressions by GSK-3α/β

        <P>Notch signaling is controlled at multiple levels. In particular, stabilized Notch receptor activation directly affects the transcriptional activations of Notch target genes. Although some progress has been made in terms of defining the regulatory mechanism that alters Notch stability, it has not been determined whether Notch1/NICD stability is regulated by GSK-3alpha. Here, we show that Notch1/NICD levels are significantly regulated by GSK-3beta and by GSK-3alpha. Treatment with LiCl (a specific GSK-3 inhibitor) or the overexpression of the kinase-inactive forms of GSK-3alpha/beta significantly increased Notch1/NICD levels. Endogenous NICD levels were also increased by either GSK-3alpha/beta- or GSK-3alpha-specific siRNA. Furthermore, it was found that GSK-3alpha binds to Notch1. Deletion analysis showed that at least three Thr residues in Notch1 (Thr-1851, 2123, and 2125) are critical for its response to LiCl, which increased not only the transcriptional activity of endogenous NICD but also Hes1 mRNA levels. Taken together, our results indicate that GSK-3alpha is a negative regulator of Notch1/NICD.</P>
