        Activation of Transient Receptor Potential Melastatin Family Member 8 (TRPM8) Receptors Induces Proinflammatory Cytokine Expressions in Bronchial Epithelial Cells

        Purpose: Cold air is a major environmental factor that exacerbates asthma. Transient receptor potential melastatin family member 8 (TRPM8) is a cold-sensing channel expressed in the airway epithelium. However, its role in airway inflammation remains unknown. We investigated the role of TRPM8 in innate immune responses in bronchial epithelial cells and asthmatic subjects. Methods: The TRPM8 mRNA and protein expression on BEAS2B human bronchial epithelial cells was examined by real-time polymerase chain reaction (PCR), immunofluorescence staining and western blotting. Additionally, interleukin (IL)-4, IL-6, IL-8, IL-13, IL-25 and thymic stromal lymphopoietin (TSLP) levels before and after menthol, dexamethasone and N-(4-tert-butylphenyl)-4-(3-chloropyridin-2-yl) piperazine-1-carboxamide (BCTC) treatments were measured via real-time PCR. TRPM8 protein levels in the supernatants of induced sputum from asthmatic subjects and normal control subjects were measured using enzyme-linked immunosorbent assay, and mRNA levels in sputum cell lysates were measured using real-time PCR. Results: Treatment with up to 2 mM menthol dose-dependently increased TRPM8 mRNA and protein in BEAS2B cells compared to untreated cells (P < 0.001) and concomitantly increased IL-25 and TSLP mRNA (P < 0.05), but not IL-33 mRNA. BCTC (10 μM) significantly abolished menthol-induced up-regulation of TRPM8 mRNA and protein and IL-25 and TSLP mRNA (P < 0.01). TRPM8 protein levels were higher in the supernatants of induced sputum from asthmatic subjects (n = 107) than in those from healthy controls (n = 19) (P < 0.001), and IL-25, TSLP and IL-33 mRNA levels were concomitantly increased (P < 0.001). Additionally, TRPM8 mRNA levels correlated strongly with those of IL-25 and TSLP (P < 0.001), and TRPM8 protein levels were significantly higher in bronchodilator-responsive asthmatic subjects than in nonresponders. Conclusions: TRPM8 may be involved in the airway epithelial cell innate immune response and a molecular target for the treatment of asthma.

        방사선조사와 저칼슘식이가 치아형성시 Interleukin-1의 분포에 미치는 영향에 관한 연구

        Purpose : To elucidate the effects of the irradiation and calcium-deficient diet on expression of interleukin (IL)-1 during tooth formation of rat molar Materials and Methods : The pregnant three-week-old Spague-Dawley rats were used for the study. The control group was non-irradiation/normal diet group, and the experimental groups were irradiation/normal diet group and irradiation/calcium-deficient diet group . The abdomen of the rats on the 9th day of pregnancy were irradiated with single dose of 350 cGy. The rat pups were sacrificed on the 14th day after delivery and the maxillae tooth germs were taken. The specimen were prepared to make sections for light microscopy, and some of tissue sections were stained immunohistochemically with anti-IL-1 antibody. Results : In the irradiation/normal diet group, dental follicle showed fewer blood vessels, mononuclear cells, and fusions of mononuclear cells than in non-irradiation/normal diet group. Alveolar bone showed a few osteoblasts and osteoclasts. Periodontal ligament showed collagen fibers and fibroblasts with irregularity. Weak immunoreactivity for IL-1 was shown in dental follicle, alveolar bone, and periodontal ligament, In the irradiation/calcium-deficient diet group, dental follicle showed sparse cellularity. Alveolar bone showed diminished number of osteoblasts. Periodontal ligament showed irregular collagen fibers and atrophy of cementoblasts and fibroblasts. No immunore-activity for IL-1 was shown in dental follicle, alveolar bone, and periodontal ligament. Conclusion : Irradiation and calcium-deficient diet seems to cause disturbance of the expression of interleukin-1 during tooth formation of rat molar.         <i>In vivo</i> genotoxicity evaluation of lung cells from Fischer 344 rats following 28 days of inhalation exposure to MWCNTs, plus 28 days and 90 days post-exposure

        <P>Despite their useful physico-chemical properties, carbon nanotubes (CNTs) continue to cause concern over occupational and human health due to their structural similarity to asbestos. Thus, to evaluate the toxic and genotoxic effect of multi-wall carbon nanotubes (MWCNTs) on lung cells <I>in vivo</I>, eight-week-old rats were divided into four groups (each group = 25 animals), a fresh air control (0 mg/m<SUP>3</SUP>), low (0.17 mg/m<SUP>3</SUP>), middle (0.49 mg/m<SUP>3</SUP>), and high (0.96 mg/m<SUP>3</SUP>) dose group, and exposed to MWCNTs <I>via</I> nose-only inhalation 6 h per day, 5 days per week for 28 days. The count median length and geometric standard deviation for the MWCNTs determined by TEM were 330.18 and 1.72 nm, respectively, and the MWCNT diameters ranged from 10 to 15 nm. Lung cells were isolated from five male and five female rats in each group on day 0, day 28 (only from males) and day 90 following the 28-day exposure. The total number of animals used was 15 male and 10 female rats for each concentration group. To determine the genotoxicity of the MWCNTs, a single cell gel electrophoresis assay (Comet assay) was conducted on the rat lung cells. As a result of the exposure, the olive tail moments were found to be significantly higher (<I>p</I> < 0.05) in the male and female rats from all the exposed groups when compared with the fresh air control. In addition, the high-dose exposed male and middle and high-dose exposed female rats retained DNA damage, even 90 days post-exposure (<I>p</I> < 0.05). To investigate the mode of genotoxicity, the intracellular reactive oxygen species (ROS) levels and inflammatory cytokine levels (TNF-α, TGF- β, IL-1, IL-2, IL-4, IL-5, IL-10, IL-12 and IFN-γ) were also measured. For the male rats, the H<SUB>2</SUB>O<SUB>2</SUB> levels were significantly higher in the middle (0 days post-exposure) and high- (0 days and 28 days post-exposure) dose groups (<I>p</I> < 0.05). Conversely, the female rats showed no changes in the H<SUB>2</SUB>O<SUB>2</SUB> levels. The inflammatory cytokine levels in the bronchoalveolar lavage (BAL) fluid did not show any statistically significant difference. Interestingly, the short-length MWCNTs deposited in the lung cells were persistent at 90 days post-exposure. Thus, exposing lung cells to MWCNTs with a short tube length may induce genotoxicity.</P>

        2자유도 제어기를 이용한 유도전동기 속도제어

        This paper describes a design of an induction motor control system using a 2 degree-of-freedom PI controller to compensate the effects of disturbance without degrading tracking performance. On the basis of vector control principle, the control system is simulated by using the ACSL and implemented on a DSP system(TMS320C31). In designing the 2 DOF controller, we can tune the performance of either the tracking or disturbance rejection independently without affecting the other. With the experimental results, the 2 DOF controller has shown a better performance in command tracking and disturbance rejection than a conventional PI controller.

        It is the purpose of this study to evaluate the adaptation status to the teaching career of the teachers college graduates and to systematically analyze and rearrange the materials concerned so that some suggestions for the studies related to this field might be concluded. Procedure The teacher's quality for the profession has been measured by an evaluating scale and the attitude for the profession has been measured by standardized tools. And a questionaire of the writer's own composing has been used in examining the general trend of the teacher's quality. This study has been carried on with the following limitations: (1) The area of investigation has been limitted to Busan area. (2) The evaluated teachers have not had any previous training. (3) The actual visiting or interviewing process has not been accompanied. Results The results of the study can be summarized as the following: 1. In the investigation of the teacher's attitude toward the profession, the attitude point of the teachers college graduates was higher by 4.47 points, and the standard of the attitude of the woman teachers was a little higher. 2. In the investigation of the view of value-judgement, "the pride for the teaching profession' marked a higher proportion with the group of teachers college graduates(77.76%). 3. In the item of the degree of enthusiasm for the guidance of the problem child, the answer of "feel keenly the necessity of it" was shown 70%, which was considerably high standard of all sampling items, especialy the zeal of the woman teachers for this guidance was very high. 4. The standard of the teacher's quality of the teacher college graduates of both sexes showed a high percentage. 5. In the investigation of the trend of the teacher's quality through a questionaire, there was no difference among each sampling item. 6. In the self-evaluation for the ability of subject instruction, the teachers, as a whole, admitted the shortage of their ability in the teaching skill. 7. The problems in the teaching of each subject were found in the following fields: (1) "dancing" and "ball games" in the physical education, "instrumental music and concert" in the music subject, "drawing"a and "writing" in the art, (2) "civic course" in the social life, "gathering" in the arithmetic, "geology" in the natural science, and "individual guidance" in the moral life. 8. The degree of application of the educational theories to the professional life was answered in the affirmative in each field. Recommondation Following suggestions have been concluded as a result of this study: 1. The standard the teacher's attitude toward the teaching profession is considered generally low, and it is the writer's belef that the education that the education course should be extended and strengthened in the curriculum of the teachers college and the teachers should be given a chance of continuous in-service training program. 2. The teacher's quality of the woman teachers is lower than that of the man teachers. And it is desirable that more opportunities of professional training for the woman teachers should be considered. 3. In order that the teaching skills studied during the cadet teacher practice period be utilized in the actual class room activities, more practical problems should be treated during the cadet teacher practice course. 4. The educational plan for the charactor-improving in the aspect of the practical moral life, which is tend to be neglected easily in the college life, should be treated more seriously in the teachers college curriculum.

        홍화씨 열수 추출 분말을 이용한 우리 밀 라면의 제조

        The purpose of this study was to make ramyon from Korean cultured wheat by adding with hot water extract powder from safflower seed in order to add the value of it. The cooking quality, instrumental texture and sensory characteristics of ramyon were analyzed. The ramyons with 0.1%, 0.3%, 0.5%, and 0.7% of hot water extract powder(HEP) from safflower seed, control, and those with 3% and 5% of dried powder(DP) from safflower seed were compared. The yield of HEP was 7.8%. Lightness, redness, Max. weight, strength, hardness, adhesiveness, cohesiveness and springiness were measured. As the amounts of HEP and DP was increasing, ramyons smelled stronger and was getting harder and chewier, while became less transparent and had no difference in elasticity and adhesiveness. In overall acceptability, both control and ramyon with HEP had similar points. From three important factors, appearance, color and smell to make ramyon more acceptable, addition of 0.3~0.5% of hot water extract powder from safflower seed was found to be the best. However, further studies on smells are needed to make processed foodstuffs with safflower seed.

      • Clinical Outcome of COVID-19 Patients with Mechanical Ventilator at Intensive Care Unit

        The coronavirus disease 2019 (COVID-19) is a pandemic caused by severe acute respiratory syndrome coronavirus (SARS-CoV2) originated from Wuhan, China. The hospitalization and mortality rate of Intensive care units due to COVID-19 infection have varied from study to study. However, there is not enough information of ICU patients who require mechanical ventilation. The main objective of the study is to describe clinical outcomes and characteristics of critically ill patients with a requirement for mechanical ventilation. Methods This is retrospective cohort study. We reviewed medical charts of patients with COVID-19 admitted to the ICU. COVID-19 was confirmed by using a positive result on a real-time reverse transcriptase-polymerase chain reaction (RT-PCR) assay. Result From February 29, 2020 to August 9, 2021, 71 adult patients with COVID-19 were admitted to the ICU. The median age of admitted patients was 68.5 years old (IQR 63-72.5). Among these 71 patients, 32 (45.1%) received invasive mechanical ventilation. 18 patients (56.3%) were male. 22 patients (68.8%) received CRRT and vasopressor was used on 27 patients (84.4%). 20 patients (62.5%) required extracorporeal membrane oxygenation (ECMO) support. Among 32 patients who have been treated with mechanical ventilation, 21 patients (29.6%) died in the ICU. 11 patients (15.5%) were discharged or transferred to another hospital. Median length of stay at the ICU was 44.5 days (IQR 17-64). The median time between the time of diagnosis to the initiation of invasive mechanical ventilation was 6.5 days (IQR 1-10), and the median time between initiation time to use of mechanical ventilation and ECMO was 2 days (IQR1-4). Conclusion Our study showed ICU mortality rate of patients with invasive mechanical ventilation support was 29.6%. This result may show improvement of survival and clinical outcome in critically ill COVID-19 patients receiving mechanical ventilation support

      • Comparison of the Therapeutic Effect of Peramivir and Oseltamivir in the Treatment of Influenza in the Intensive Care Unit

        Introduction The annual epidemic of influenza is responsible for substantial morbidity and mortality in Korea. About 5 to 10% of hospitalized patients infected with influenza require intensive care. Antiviral drugs including neuraminidase (NA) inhibitors, peramivir, and oseltamivir are used as treatment for influenza. However, there is not much research of the therapeutic outcome of each antiviral drug on intensive care inpatients. This study aims to compare the Results of the use of two antiviral drugs in patients admitted to the intensive care unit(ICU) after being diagnosed with influenza. Methods We retrospectively reviewed medical charts of patients with influenza admitted to the ICU. Influenza infection (A or B) was confirmed by using influenza antigen test or real-time reverse transcriptase-polymerase chain reaction (RT-PCR) assay. Results Among the consecutive patients admitted to the ICUs with influenza (A or B) viral infection from June 2015 to December 2018, 104 patients were confirmed for enrolment. Divided into three groups, 32 were treated with oseltamivir, 61 with peramivir, and 11 with the combination of oseltamivir-peramivir. There were no significant differences between the median durations from onset of symptoms to administration of antiviral drugs (3 days in oseltamivir, 3 day in peramivir, and 4 days in combination), the median duration of treatment (5 days in oseltamivir, 1 day in peramivir, and 5 days in combination) or the median duration of ICU stays (3.5 days in oseltamivir, 5 days in peramivir, and 8 days in combination) between the three groups. There were also no significant differences in hospital mortality (5.8% (n=6) in oseltamivir, 26.9% (n=28) in peramivir, and 6.7% (n=7) in combination), ICU mortality (3.8% (n=4) in oseltamivir, 24.0% (n=25) in peramivir, and 5.8% (n=6) in combination) between the three groups. Conclusion Peramivir also showed similar clinical outcome and efficacy of oseltamivir in influenza patients admitted to ICU.
