        This paper presents the tensile behavior of the stud connection between reinforced concrete(RC) and steel members. Hanger reinforcements are placed around the studs to transfer the tensile loads to the opposite side of the concrete member. Six specimens, of which variables were arrangement details of hanger reinforcements and studs. Tensile test results show that hanger reinforcements are effective to increase tensile strength and ductility. Test tensile strengths of specimens are compared with the nominal strengths by ACI codes(ACI 318-02, ACI 349-99) and PCI 1999.

      • User-expected price-based demand response algorithm for a home-to-grid system

        <P><B>Abstract</B></P> <P>Demand response algorithms can cut peak energy use, driving energy conservation and enabling renewable energy sources, as well as reducing greenhouse-gas emissions. The use of these technologies is becoming increasingly popular, especially in smart-grid scenarios. We describe a home-to-grid demand response algorithm, which introduces a UEP (“user-expected price”) as an indicator of differential pricing in dynamic domestic electricity tariffs, and exploits the modern smart-grid infrastructure to respond to these dynamic pricing structures. By comparing the UEP with real-time utility price data, the algorithm can discriminate high-price hours and low-price hours, and automatically schedule the operation of home appliances, as well as control an energy-storage system to store surplus energy during low-price hours for consumption during high-price hours. The algorithm uses an exponential smoothing model to predict the required energy of appliances, and uses Bayes' theorem to calculate the probability that appliances will demand power at a given time based on historic energy-usage data. Simulation results using pricing structures from the Ameren Illinois power company show that the proposed algorithm can significantly reduce or even eliminate peak-hour energy consumption, leading to a reduction in the overall domestic energy costs of up to 39%.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Introduce a UEP (“user-expected price”) as an indicator of differential pricing in dynamic domestic electricity tariffs. </LI> <LI> Discriminate high-price hours and low-price hours, and automatically schedule the operation of home appliances. </LI> <LI> Control an energy-storage system to store surplus energy during low-price hours for consumption during high-price hours. </LI> <LI> Simulation results show that the proposed algorithm can significantly reduce or even eliminate peak-hour energy consumption. </LI> </UL> </P>

        Improvement of Welan Gum Production and Redistribution of Metabolic Flux under pH Control Process in Alcaligenes sp. CGMCC2428

        Batch fermentations of welan gum from Alcaligenes sp. CGMCC2428 at pH values of 5.5 ~ 7.0were studied. Based on the kinetic analysis, a pH control process for improving welan production was developed. By maintaining the culture pH at 7.0, the process significantly improved the maximal welan concentration and productivity to reach 25.1 ± 0.65 g/L and 0.42 ± 0.003 g/L/h, respectively,compared with those in native pH processes where pH value would decrease from 7.0 to 5.1 (18.5 ± 0.72 g/L and 0.28 ± 0.002 g/L/h). This improvement may be due to the increased metabolic flux of glucose-1-phosphate to welan induced by pH control process. Furthermore, scale-up fermentation under controlled pH was implemented using 300-L fermentor. The highest welan concentration of 27.5± 0.97 g/L was obtained. This simplified process proved effective in industry fermentation for high welan production.

      • Expression of Proteasome Activator REGγ in Human Laryngeal Carcinoma and Associations with Tumor Suppressor Proteins

        The functional significance of the proteasome activator $REG{\gamma}$ in the regulation of cell proliferation and apoptosis has been recognized. However, pathological contributions to tumor development remain to be elucidated. Both oncogenic proteins and tumor suppressors are targeted by $REG{\gamma}$ for proteasomal degradation. It has been proposed that the role of the $REG{\gamma}$ in the pathogenesis of cancer is cell- and context-specific. In this study, we aimed to explore the potential involvement of $REG{\gamma}$ in laryngeal carcinomas, comparing protein expression in tumor and adjacent tissues by immunohistochemical staining and Western blot analysis. We also characterized the correlation between the expression of $REG{\gamma}$ and the previously identified substrates p53 and p21. We showed that $REG{\gamma}$ was abnormally highly expressed in cancer tissues. Statistical analysis revealed that there was a positive relationship between the level of $REG{\gamma}$ and the expression of p53 and p21. Our study suggests that $REG{\gamma}$ overexpression can facilitate the growth of laryngeal cancer cells.

      • MicroRNA-451 Inhibits Growth of Human Colorectal Carcinoma Cells via Downregulation of Pi3k/Akt Pathway

        MicroRNAs (MiRNAs) play important roles in coordinating a variety of cellular processes and abnormal expression has been linked to the occurrence of several cancers. The miRNA miR-451 is downregulated in colorectal carcinoma (CRC) cells, suggested by several research groups including our own. In this study, synthetic miR-451 mimics were transfected into the SW620 human CRC cell line using Lipofectamine 2000 and expression of miR-451 was analyzed by real time PCR, while expression of CAB39, LKB1, AMPK, AKT, PI3K and Bcl2 was analyzed by Western blot, and cell growth was detected by MTT assay. In comparison to the controls, a significant increase in the expression of miR-451 was associated with significantly decreased expression of CAB39, LKB1, AMPK, AKT, PI3K and Bcl2. The capacity of cell proliferation was significantly decreased by miR-451 expression, which also inhibited cell growth. Our study confirmed that miR-451 has a repressive role in CRC cells by inhibiting cell growth through down-regulating the P13K/AKT pathway.

        Theoretical Study of the Elastic and the Thermodynamic Properties of Re$_2$C under High Pressure

        The structural properties of Re$_2$C in anti-MoS$_2$ and anti-ReB$_2$ structures have been investigated by using the pseudopotential plane wave methods based on the density functional theory. The anti-ReB$_2$ structure is found to be more stable than the anti-MoS$_2$ structure. In particular, for the first time, we have studied the elastic properties of Re$_2$C in the anti-ReB$_2$ structure under high pressure. The ductile-brittle behavior and Vickers hardness for the anti-ReB$_2$ structure are also been analyzed. In addition, the Debye temperature, heat capacity and thermal expansion coefficient are discussed by using the quasiharmonic Debye model method.

        Isolation and molecular characterization of two novel HMW-GS genes from Chinese wheat (Triticum aestivum L.) landrace Banjiemang

        High molecular weight glutenin subunits(HMW-GS) play a significant role in determining breadmakingqualities of common wheat flour. The study presentedin the paper identified two novel HMW-GS (designatedas 1Bx14* and 1By15*) at the Glu-B1 locus inChinese wheat (Triticum aestivum L.) landrace Banjiemangby SDS-PAGE. The open reading frames (ORFs) of thegenes were amplified and cloned with designed specificprimers. Both of them had sequences highly similar tothose of other HMW-GS genes, but displayed minormodification. The ORF of 1Bx14* was 2367 bp long andencoded 789 aa with 4 cysteines, which included 25nanopeptides, 65 hexapeptides and 7 tripeptides motifs. The ORF of 1By15* was 2106 bp long and encoded 702 aawith 7 cysteines, which included 23 nanopeptides and 53hexapeptides motifs. The predicted molecular weights ofthe mature proteins encoded by 1Bx14* and 1By15* geneswere 83.1 and 73.2 k Da, respectively. The presence andauthenticity of the two genes in Banjiemang were confirmedby bacterial expression and liquid chromatographyelectrosprayionization mass spectrometry (LC–ESI–MS). By phylogenetic analysis, the 1Bx14* was clustered withx-type subunits but had a high divergence with other 1Bxsubunits, whereas the 1By15* was clustered with y-typesubunits and closely related to subunit 1By15. The studyconcluded that 1Bx14* and 1By15* from Banjiemang werenovel allelic variations of HMW-GS at Glu-B1 locus,which were probably exploitable as new resources forwheat quality improvement.

        Based on the reciprocity theory, a dynamic reciprocal behavior model is verified by sampling the US and French group tourists in Tibet. Result shows that the US group tourists have only negative reciprocity, and French ones have both positive and negative reciprocity, indicating that tourists are more willing to sacrifice their interests to revenge hostile persons.

        Strain Improvement and Metabolic Flux Modeling of Wild-type and Mutant Alcaligenes sp. NX-3 for Synthesis of Exopolysaccharide Welan Gum

        Low-energy nitrogen ion beam implantation technique was used for the strain improvement of Alcaligenes sp. NX-3 for the production of exopolysaccharide welan gum. A high welan gum producing mutant, Alcaligenes sp. NX-3-1, was obtained through 20 keV N+ ion beam irradiation. Starting at a concentration of 50 g/L of glucose, mutant NX-3-1 produced 25.0 g/L of welan gum after 66 h of cultivation in a 7.5 L bioreactor, which was 34.4% higher than that produced by the wild-type strain. The results of metabolic flux analysis showed that the glucose-6-phosphate and acetyl coenzyme A nodes were the principle and flexible nodes, respectively. At the glucose-6-phosphate node, the fraction of carbon measured from glucose-6-phosphate to glucose-1-phosphate was enhanced after mutagenesis, which indicated that more flux was used to synthesize welan gum in the mutant. By analyzing the activities of related enzymes in the biosynthetic pathway of sugar nucleotides essential for welan gum production, we found that the specific activities of phosphoglucomutase, UDP-glucose pyrophosphorylase,UDP-glucose dehydrogenase, and dTDP-glucose pyrophosphorylase in the mutant strain were higher than those in the wild-type strain. These improvements in enzyme activities could be due to the affected of ion beam implantation.

        Structural damage identification based on genetically trained ANNs in beams

        This study develops a two stage procedure to identify the structural damage based on the optimized artificial neural networks. Initially, the modal strain energy index (MSEI) is established to extract the damaged elements and to reduce the computational time. Then the genetic algorithm (GA) and artificial neural networks (ANNs) are combined to detect the damage severity. The input of the network is modal strain energy index and the output is the flexural stiffness of the beam elements. The principal component analysis (PCA) is utilized to reduce the input variants of the neural network. By using the genetic algorithm to optimize the parameters, the ANNs can significantly improve the accuracy and convergence of the damage identification. The influence of noise on damage identification results is also studied. The simulation and experiment on beam structures shows that the adaptive parameter selection neural network can identify the damage location and severity of beam structures with high accuracy.
