In inflammatory responses, induction of cytokines and other immune regulator genes in macrophages by pathogen-associated signal such as lipopolysaccharide (LPS) plays a crucial role. In this study, the gene expression profile changes by LPS treatment in the macrophage/monocyte lineage cell line RAW264.7 was investigated. A 60-mer oligonucleotide microarray of which probes target 32381 mouse genes was used. A reverse transcription-in vitro translation labeling protocol and a chemileuminescence detection system were employed. The mRNA expression levels in RAW264.7 cells treated for 6 h with LPS and the control vehicle were compared. 747 genes were up-regulated and 523 genes were down-regulated by more than 2 folds. 320 genes showing more than 4-fold change by LPS treatment were further classified for the biological process, molecular function, and signaling pathway. The biological process categories that showed high number of increased genes include the immunity and defense, the nucleic acid metabolism, the protein metabolism and modification, and the signal transduction process. The chemokine-cytokine signaling, interleukin signaling, Toll receptor signaling, and apoptosis signaling pathways involved high number of genes differentially expressed in response to LPS. These expression profile data provide more comprehensive information on LPS-target genes in RAW264.7 cells, which will be useful in comparing gene expression changes induced by extracts and compounds from anti-inflammatory medicinal herbs.

Ni/Ni-Sn/Ni sandwiched simulated package structures weresuccessfully bonded under low temperature and low pressure byNi-Sn transient liquid-phase sintering bonding. The results showthat, after isothermally holding for 240 min at 300 °C and 180 minat 340 °C, Sn was completely transformed into Ni3Sn4 intermetalliccompounds. When the Ni3Sn4 phases around Ni particles werepressed together, the porosity of the bonding layer increased, whichobviously differed from the normal sintering densification process. With further analysis of this phenomenon, it was found that largevolume shrinkage (14.94% at 340 °C) occurred when Ni reactedwith Sn to form Ni3Sn4, which caused void formation. Amechanistic model of the microstructural evolution in the bondinglayer was proposed. Meanwhile, the resistivity of the bonding layerwas measured and analyzed by using the four-probe method; themicrostructural evolution was well reflected by the resistivity ofthe bonding layer. The relationship between the resistivity andmicrostructure was also discussed in detail.
Study on Thermodynamics of Three Kinds of Benzindocarbocyanine Dyes in Aqueous Methanol Solution

Aggregation behavior of three kinds of benzindocarbocyanine dyes in aqueous methanol solution was studied by UV-Vis absorption spectrum. The results indicated that the three dyes all existed monomer-dimer equilibrium in aqueous methanol solution (concentration range $10^{−5}\;to\;10^{−6}$ M) at 25.0$\sim$41.0 ${^{\circ}C}$ for Dye 1, 28.0$\sim$49.0 ${^{\circ}C}$ for Dye 2 and 26.0$\sim$47.0 ${^{\circ}C}$ for Dye 3. The fundamental property of the three dyes as the dimeric association constant KD, the dimeric free energy ${\Delta$}G_D, the dimeric entropy ${\Delta$}S_D, and the dimeric enthalpy ${\Delta$}H_D were determined. The ${\Delta$}H_D of three dyes: Dye 1, Dye 2 and Dye 3 was -42.5, -15.1 and -18.9 kJ/mol, respectively. The experimental observations were the subject of a theoretical study including the ground-state geometries which were fully optimized using DFT at B3LYP/6-31G level. The effect of dye molecule structure on ${\Delta$}H_D was discussed by theoretical calculations.
A Dual Buck Three-Level PV Grid-Connected Inverter

The use of a PV grid-connected inverter with non-isolated topology and without a transformer is good for improving conversion efficiency; however, this inverter has become increasingly complicated for eliminating leakage current. To simplify the complicated architecture of traditional three-level dual buck inverters, a new dual Buck three-level PV grid-connected inverter topology is proposed. In the proposed topology, the voltage on the grounding stray capacitor is clamped by large input capacitors and is equal to half of the bus voltage; thus, leakage current can be eliminated. Unlike in the traditional topology, the current in the proposed topology passes through few elements and does not flow through the body diodes of MOSFET switches, resulting in increased efficiency. Additionally, a multi-loop control method that includes voltage-balancing control is proposed and analyzed. Both simulation and experimental results are demonstrated to verify the proposed structure and control method.

AMFR, autocrine motility factor receptor, also called gp78, is a cell surface cytokine receptor which has a dual role as an E3 ubiquitin ligase in endoplasmic reticulum-associated degradation. AMFR expression is associated with tumor malignancy. We here investigated the clinical significance of AMFR and its role in metastasis and prognosis in gastric cancer. Expression of AMFR, E-cadherin and N-cadherin in cancer tissues and matched adjacent normal tissues from 122 gastric cancer (GC) patients undergoing surgical resection was assessed by immunohistochemistry. Levels of these molecules in 17 cases selected randomly were also analysed by Western blotting. AMFR expression was significantly increased in gastric cancer tissues, and associated with invasion depth and lymph node metastasis. Kaplan-Meier analysis showed AMFR expression correlated with poor overall survival and an increased risk of recurrence in the GC cases. Cox regression analysis suggested AMFR to be an independent predictor for overall and recurrence-free survival. E-cadherin expression was decreased in gastric cancer tissues; conversely, N-cadherin was increased. Expression of AMFR negatively correlated with E-cadherin expression, whereas N-cadherin expression showed a significant positive correlation with AMFR expression. AMFR might be involved in the regulation of epithelial-mesenchymal transition, with aberrant expression correlating with a poor prognosis and promoting invasion and metastasis in GCs.
A Family of Non-Isolated Photovoltaic Grid Connected Inverters without Leakage Current Issues

Transformerless solar inverters have a higher efficiency than those with an isolation link. However, they suffer from a leakage current issue. This paper proposes a family of single phase six-switch transformerless inverter topologies with an ac bypass circuit to solve the leakage current problem. These circuits embed two unidirectional freewheeling current units into the midpoint of a full bridge inverter, to obtain a freewheeling current path, which separates the solar panel from the grid in the freewheeling state. The freewheeling current path contains significantly fewer devices and poor performance body diodes are not involved, leading to a higher efficiency. Meanwhile, it is not necessary to add a voltage balancing control method when compared with the half bridge inverter. Simulation and experiments are provided to validate the proposed topologies.
Molecular Cloning and Characterization of clyA Genes in Various Serotypes of Salmonella enterica

Cytolysin A (ClyA) is a pore-forming hemolytic protein encoded by the clyA gene. It has been identified in Salmonella enterica serovars Typhi and Paratyphi A. To identify and characterize the clyA genes in various Salmonella enterica strains, 21 different serotypes of strains isolated from clinical specimens were presently examined. Full-length clyA genes were found in S. enterica serovar Brandenburg, Indiana, Panama, and Schwarzengrund strains by polymerase chain reaction amplification. The ClyA proteins from these four strains showed >97% amino acid identity to that of S. enterica serovar Typhi. Although all four serovars expressed detectable levels of ClyA as determined by Western blot analysis, they did not show a strong hemolytic effect on blood agar, indicating that ClyA may not be efficiently expressed or secreted. Escherichia coli transformed with clyA genes from the four serovars enhanced production of ClyA proteins and hemolytic activities to a level similar to S. enterica serovar Typhi ClyA. The present results suggest that ClyA may play a role in the pathogenesis of S. enterica serovar Brandenburg, Indiana, Panama and Schwarzengrund.
