        The prediction of recovery percent of water-free stage and its application in the correction of theoretical water cut

        Water-free oil production period is an important stage for oil production, prediction and water cut prediction. Because there are many factors affecting the water-free oil production period and the influencing mechanism is complex, it is very difficult to calculate the water-free oil production period. In this paper, the main influencing factors of water-free oil production period are analyzed by the stream tube approach method. The influence law of four influencing factors on water-free oil production period was analyzed and the quantitative relationship between four influencing factors and water-free oil production period were established. The average water saturation of reservoir and water saturation near producers are different which is important for the calculation of theoretical water cut. The quantitative relationship between average water saturation of reservoir and water saturation near producers is established. On this basis, a new method for calculating theoretical water cut is built and is applied to BZ reservoir. The result shows the theoretical water cut calculated by this method is consistent with field data.

        Genome-wide analysis of Gro/Tup1 family corepressors and their responses to hormones and abiotic stresses in maize

        Gro/Tup1 proteins act as negative transcriptional regulators and play crucial roles in many growth and developmental processes in a wide range of organisms. However, our understanding of Gro/Tup1 protein functions in plants is confined to the model plant Arabidopsis. Here, 11 Gro/Tup1 genes, which were characterized by the typical LisH and WD40 repeat domains, were identified in maize through a genome-wide survey. A phylogenetic analysis revealed that maize Gro/Tup1 proteins could be divided into three subfamilies, in which members shared similar protein and gene structures. The predicted maize Gro/Tup1 genes were distributed on seven chromosomes and segmental duplication contributed to their expansion. Many predicted cis-elements associated with hormones, biotic- or abioticstress responses, meristem and seed development, and circadian rhythms, were found in their putative promoter regions. A potential associated protein analysis identified a large number of candidates, including transcription factors, chromatin-modifying enzymes, protein kinases, and ubiquitinconjugating enzymes. An expression profile derived from the RNA-seq data indicated that Gro/Tup1 genes in maize were widely expressed in various organs and tissues. Quantitative real-time PCR revealed that these genes responded to at least one hormone or abiotic stress, either in roots or in shoots. Our study provides useful information on the Gro/Tup1 genes in maize and will facilitate the further functional validation of these genes in growth and development, hormone responses, and biotic- or abiotic-stress resistance.

        Research and practice on oil displacement law of different well pattern in offshore oilfield

        BZ oilfield is the first oilfield successfully developed with large-scale horizontal well pattern in Bohai sea. In order to study the well pattern suitable for offshore fluvial sedimentary oilfield, based on the characteristics of offshore oilfield, such as large well spacing, few wells and complex well pattern, the authors designed a set of heterogeneous reservoir models under the guidance of similarity theory. The different well patterns, different displacement rates and different heterogeneity were simulated. The different displacement effects were studied by measuring the electrical characteristics, injection volume, oil production and water production of the model. It is found that the horizontal well pattern is used for low-speed development, the water breakthrough of oil wells is slow in the relatively homogeneous reservoir. In the heterogeneous high permeability reservoir(Jk = 4), the oilfield adopts low-speed development by using staggered well pattern, the water breakthrough of the oil well is slow, and the EOR is large. In the heterogeneous high permeability reservoir(Jk = 6), the oilfield adopts the combined well pattern for low-speed development, the EOR is large. This study deepens the understanding of the development law of horizontal well pattern and the distribution law of remaining oil, and provides a basis for oilfield development and adjustment.

        Differentially expressed serum proteins associated with calcium regulation and hypocalcemia in dairy cows

        Objective: Hypocalcemia is an important metabolic disease of dairy cows during the transition period, although the effect of hypocalcemia on biological function in dairy cows remains unknown. Methods: In this study, proteomic, mass spectrum, bioinformatics and western blotting were employed to identify differentially expressed proteins related to serum Ca concentration. Serum samples from dairy cows were collected at three time points: 3rd days before calving (day –3), the day of calving (day 0), and 3rd days after calving (day +3). According to the Ca concentration on day 0, a total of 27 dairy cows were assigned to one of three groups (clinical, subclinical, and healthy). Samples collected on day –3 were used for discovery of differentially expressed proteins, which were separated and identified via proteomic analysis and mass spectrometry. Bioinformatics analysis was performed to determine the function of the identified proteins (gene ontology and pathway analysis). The differentially expressed proteins were verified by western blot analysis. Results: There were 57 differential spots separated and eight different proteins were identified. Vitamin D-binding protein precursor (group-specific component, GC), alpha-2-macroglobulin (A2M) protein, and apolipoprotein A-IV were related to hypocalcemia by bioinformatics analysis. Due to its specific expression (up-regulated in clinical hypocalcemia and down-regulated in subclinical hypocalcemia), A2M was selected for validation. The results were consistent with those of proteomic analysis. Conclusion: A2M was as an early detection index for distinguishing clinical and subclinical hypocalcemia. The possible pathogenesis of clinical hypocalcemia caused by GC and apolipoprotein A-IV was speculated. The down-regulated expression of GC was a probable cause of the decrease in calcium concentration.
