        Vegetable consumption is associated with health benefits. Recently, the interest in these food groups has grown among due to the identification of new compound in their chemical composition. Most of the vegetables are cooked by blanching, boiling, steaming, microwaving before consumed. These cooking processes may change not only food physical characteristics but also chemical composition. The present study was undertaken to investigate the effect of different cooking methods on phytochemical content and antioxidant capacity in Ipomoea batatas (L.) Lam stem. The result of study, Total polyphenol of phytochemical in Ipomoea batatas (L.) Lam stem was 6.88-12.65 mg GAE/100 g and flavonoid was 3.16-10.46 mg CE/100 g. Compared with the raw of Ipomoea batatas (L.) Lam stem, all cooking methods were found to significant correlations between the antioxidant activity assessed by ABTS, DPPH and polyphenol(p<0.05). These results can be provide the basic data how to prevent lose of nutrition in Ipomoea batatas (L.) Lam stem when cooking.

        Up-regulation of Heme Oxygenase-1 by Korean Red Ginseng Water Extract as a Cytoprotective Effect in Human Endothelial Cells

        Korean red ginseng (KRG) is used worldwide as a popular traditional herbal medicine. KRG has shown beneficial effects on cardiovascular diseases, such as atherosclerosis, diabetes, and hypertension. Up-regulation of a cytoprotective protein, heme oxygenase (HO)-1, is considered to augment the cellular defense against various agents that may induce cytotoxic injury. In the present study, we demonstrate that KRG water extract induces HO-1 expression in human umbilical vein endothelial cells (HUVECs) and possible involvement of the anti-oxidant transcription factor nuclear factor-eythroid 2-related factor 2 (Nrf2). KRG-induced HO-1 expression was examined by western blots, reverse transcriptase polymerase chain reaction and immunofluorescence staining. Specific silencing of Nrf2 genes with Nrf2-siRNA in HUVECs abolished HO-1 expression. In addition, the HO inhibitor zinc protoporphyrin blunted the preventive effect of KRG on H<sub>2</sub>O<sub>2</sub>-induced cell death, as demonstrated by terminal transferase dUTP nick end labeling assay. Taken together, these results suggest that KRG may exert a vasculoprotective effect through Nrf2-mediated HO-1 induction in human endothelial cell by inhibition of cell death.

        Effect of Korean Red Ginseng treatment on the gene expression profile of diabetic rat retina

        Background: Korean Red Ginseng (KRG) is a herbal medicine used in Asian countries and is very popular for its beneficial biological properties. Diabetes mellitus (DM) and its complications are rapidly becoming a global public health concern. The literature on transcriptional changes induced by KRG in rat models of diabetic retinopathy is limited. Considering these facts, we designed this study to determine whether retinopathy-associated genes are altered in retinas of rats with DM and whether the induced changes are reversed by KRG. Methods: Male SpragueeDawley rats were intravenously injected with streptozotocin (50 mg/kg body weight) to induce DM, following which, KRG powder (200 mg/kg body weight) was orally administered to the KRG-treated DM rat group for 10 wks. The rats were then sacrificed, and their retinas were harvested for total RNA extraction. Microarray gene expression profiling was performed on the extracted RNA samples. Results: From among > 31,000 genes investigated, the expression of 268 genes was observed to be upregulated and that of 58 genes was downregulated, with twofold altered expression levels in the DM group compared with those in the control group. Moreover, 39 genes were upregulated more than twofold and 84 genes were downregulated in the KRG-treated group compared to the DM group. The expression of the genes was significantly reversed by KRG treatment; some of these genes were analyzed further to verify the results of the microarray experiments. Conclusion: Taken together, our data suggest that reversed changes in the gene expression may mediate alleviating activities of KRG in rats with diabetic retinopathy.

      • Hemeoxygenase-1 Mediates an Adaptive Response to Spermidine-Induced Cell Death in Human Endothelial Cells

        <P>Spermidine (SPD) is a ubiquitous polycation that is commonly distributed in living organisms. Intracellular levels of SPD are tightly regulated, and SPD controls cell proliferation and death. However, SPD undergoes oxidation in the presence of serum, producing aldehydes, hydrogen peroxide, and ammonia, which exert cytotoxic effect on cells. Hemeoxygenase-1 (HO-1) is thought to have a protective effect against oxidative stress. Upregulation of HO-1 in endothelial cells is considered to be beneficial in the cardiovascular disease. In the present study, we demonstrate that the ubiquitous polyamine, SPD, induces HO-1 in human umbilical vein endothelial cells (HUVECs). SPD-induced HO-1 expression was examined by Western blot and reverse transcription-polymerase chain reaction (RT-PCR). Involvement of reactive oxygen species, serum amine oxidase, PI3K/Akt signaling pathway, and transcription factor Nrf2 in the induction of HO-1 by SPD was also investigated. Furthermore, small interfering RNA knockdown of Nrf2 or HO-1 and treatment with the specific HO-1 inhibitor ZnPP exhibited a noteworthy increase of death of SPD-stimulated HUVECs. In conclusion, these results suggest that SPD induces PI3K/Akt-Nrf2-mediated HO-1 expression in human endothelial cells, which may have a role in cytoprotection of the cells against oxidative stress-induced death.</P>

        An integrated analysis of microRNA and mRNA expression in salvianolic acid B-treated human umbilical vein endothelial cells

        MicroRNAs (miRNAs) are a class of small noncoding RNAs, producing transcripts of about 22 nucleotides in length. miRNAs usually function as antisense regulators of other RNAs by degrading their targets. Recently, miRNAs have emerged as interesting new drug targets due to their regulatory role in essential biological processes. Salvianolic acid B (SalB) is one of the major pharmacologically active ingredients of Salvia miltiorrhiza, a traditional oriental medicine for treatment of cardiovascular disorders. In this study, we determined whether miRNAs play a role in regulation of various gene expression responses to SalB in human umbilical vein endothelial cells (HUVECs). We used the microarray approach to evaluate levels of both miRNA and mRNA, and found that 171 miRNAs were differentially expressed in SalB-treated HUVECs. We additionally identified 848 messenger RNAs (mRNAs) that are anti-correlated with the miRNAs expression. The Gene Ontology (GO) term enrichment was analyzed for identification of biological processes of target genes affected by differential expression of miRNA. Among 848 genes investigated, cardiovascular disease-related genes were selected in SalB-treated HUVECs. These results suggest that SalB may modulate miRNA and their target gene expression in order to exert vascular protective effects in human endothelial cells.

        덕티드팬 항공기에 적용 가능한 터보샤프트 엔진에 대한 사이클 해석을 Gasturb 12 소프트웨어를 이용하여 수행하였다. 덕티드 팬 항공기의 임무별 동력요구조건에 대한 터보샤프트 엔진의 추력과 비연료소모율(SFC) 특성을 도출하였다. 항공기의 순항조건에서 비연료소모율 향상을 위하여 VGV(Variable Guide Vane)을 도입하여 그 효과를 검토하였고, 탈 설계점에서의 엔진성능향상을 위하여 여러 가지 설계점 변경을 통하여 그 특성을 연구하였다. The cycle analysis of the turboshaft engine for the ducted-fan rotorcraft was performed using Gasturb 12 software. The characteristics of the thrust and specific fuel consumption with mission requirements of the ducted-fan rotorcraft were deducted. To enhance the specific fuel consumption performance in cruise flight condition, variable guide vane system is adopted and the effect was analyzed. Also several design points were studied for improving off design engine performance.

        Conversion of Rutin to Quercetin by Acid Treatment in Relation to Biological Activities

        De-glycosylation could be an important process for enhancing the biological activities of flavonoids. In this study we investigated de-glycosylation of rutin by acid treatment by comparing hydrolysis of rutin to quercetin with two different solvents and acid concentrations. Antioxidant activity was measured using chemical methods and biological activities were examined in cell-based systems. Rutin hydrolysis occurred more rapidly when 80% ethanol was used as the reaction solvent (as compared to water), and the rate of hydrolysis accelerated as acid concentrations increased. In reactions of rutin with 0.5 M HCl in 80% ethanol for 3 h, almost all the rutin was converted into quercetin. 2,2-Diphenyl-1-picrylhydrazyl, 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid radical scavenging activities, and reducing powers were correlated with conversion rate. Protective activity in HepG2 cells, anti-inflammatory activity in RAW264.7 cells, and antiadipogenic activity were increased with increased conversion of rutin to quercetin. This study suggests that de-glycosylation of glycoside flavonoids may increase physiological activity and, therefore, enhance its use in various fields.
