      • Integration for Heterogeneous Manufacturing Information Systems Based on Semantic Entity

        To support semantic integration for heterogeneous information systems in manufacturing enterprises, based on semantic gateway, the concept of semantic entity is defined, and the semantic entity-based integration method is studied. Semantic entity sets up conceptual structure by aggregating semantic concept from semantic gateway, and builds dynamic conceptual structure through adding or removing its general concept. By means of key business data, semantic entity identifies business object and obtains business data from its application system, and then instantiates concept structure. Based on instantiation, semantic service can obtain parameters data from business object instant, and satisfies the interface contract. Semantic entity captures data changes of business object, and then establishes status-based integration rules, defines algorithm logic of rules. By instantiating concept structure aimed to specific application system and invoking semantic service, the integration activities among information systems come true. This work has been verified by prototype system, and attained expected integration effect.

        Investigation of the SHM-oriented model and dynamic characteristics of a super-tall building

        Shanghai Tower is a 632-meter super high-rise building located in an area with wind and active earthquake. A sophisticated structural health monitoring (SHM) system consisting of more than 400 sensors has been built to carry out a long-term monitoring for its operational safety. In this paper, a reduced-order model including 31 elements was generated from a full model of this super tall building. An iterative regularized matrix method was proposed to tune the system parameters, making the dynamic characteristic of the reduced-order model be consistent with those in the full model. The updating reduced-order model can be regarded as a benchmark model for further analysis. A long-term monitoring for structural dynamic characteristics of Shanghai Tower under different construction stages was also investigated. The identified results, including natural frequency and damping ratio, were discussed. Based on the data collected from the SHM system, the dynamic characteristics of the whole structure was investigated. Compared with the result of the finite element model, a good agreement can be observed. The result provides a valuable reference for examining the evolution of future dynamic characteristics of this super tall building.

      • IL-12 Regulates B7-H1 Expression in Ovarian Cancer-associated Macrophages by Effects on NF-κB Signalling

        Background and Aim: B7-H1, a co-inhibitory molecule of the B7 family, is found aberrantly expressed in ovarian cancer cells and infiltrating macrophage/dendritic-like cells, and plays a critical role in immune evasion by ovarian cancer. IL-12, an inducer of Th1 cell development, exerts immunomodulatory effects on ovarian cancer. However, whether IL-12 regulates B7-H1 expression in human ovarian cancer associated-macrophages has not been clarified. Therefore, we investigated the effects of IL-12 on the expression of B7-H1 in ovarian cancer-associated macrophages and possible mechanisms. Methods: PMA induced THP-1-derived macrophages or human monocyte-derived macrophages were treated with recombinant IL-12 (rIL-12) or infected with adenovirus carrying human IL-12 gene (Ad-IL-12-GFP) for 24 h, then cocultured with the SKOV3 ovarian cancer cell line for another 24 h. Macrophages were collected for real-time PCR and Western blot to detect the expression of B7-H1, and activation of the NF-${\kappa}B$ signaling pathway. Moreover, supernatants were collected to assay for IL-12, IFN-${\gamma}$ and IL-10 by ELISA. In addition, monocyte-derived macrophages treated with IFN-${\gamma}$ were cocultured with SKOV3 and determined for the expression of B7-H1. Furthermore, the expression of B7-H1 in monocyte-derived macrophages was also evaluated after blocking NF-${\kappa}B$ signaling. Results: The expression of B7-H1 was significantly upregulated in monocyte-derived macrophages treated with rIL-12 or Ad-IL-12-GFP compared with the control groups (p<0.05), accompanied by a remarkable upregulation of IFN-${\gamma}$ (p<0.05), a marked downregulation of IL-10 (p<0.05) and activation of NF-${\kappa}B$ signaling. However, the upregulation of B7-H1 was inhibited by blocking the NF-${\kappa}B$ signaling pathway (p<0.05). Expression of B7-H1 was also increased (p<0.05) in monocyte-derived macrophages treated with IFN-${\gamma}$ and cocultured with SKOV3. By contrast, the expression of B7-H1 in THP-1-derived macrophages was significantly decreased when treated in the same way as monocyte-derived macrophages (p<0.05), and IL-10 was also significantly decreased but IFN-${\gamma}$ was almost absent. Conclusions: IL-12 upregulates the expression of B7-H1 in monocyte-derived macrophages, which is possible though inducing the secretion of IFN-${\gamma}$ and further activating the NF-${\kappa}B$ signal pathway. However, IL-12 downregulates the expression of B7-H1 in THP-1-derived macrophages, associated with a lack of IFN-${\gamma}$ and inhibition of expression of IL-10.

      • Inclinometer-based method to monitor displacement of high-rise buildings

        Horizontal displacement of high-rise building is an essential index for assessing the structural performance and safety. In this paper, a novel inclinometer-based method is proposed to address this issue and an algorithm based on three spline interpolation principle is presented to estimate the horizontal displacement of high-rise buildings. In this method, the whole structure is divided into different elements by different measured points. The story drift angle curve of each element is modeled as a three spline curve. The horizontal displacement can be estimated after integration of the story drift angle curve. A numerical example is designed to verify the proposed method and the result shows this method can effectively estimate the horizontal displacement with high accuracy. After that, this method is applied to a practical slender structure - Shanghai Tower. Nature frequencies identification and deformation monitoring are conducted from the signal of inclinometers. It is concluded that inclinometer-based technology can not only be used for spectrum analysis and modal identification, but also for monitoring deformation of the whole structure. This inclinometer-based technology provides a novel method for future structural health monitoring.

      • Aberrant DNA Methylation of P16, MGMT, hMLH1 and hMSH2 Genes in Combination with the MTHFR C677T Genetic Polymorphism in Gastric Cancer

        Associations of P16, MGMT, hMLH1 and hMLH2 with gastric cancer and their relation with MTHFR status in gastric patients who were confirmed with pathological diagnosis were assessed. Aberrant DNA methylation of P16, MGMT, hMLH1 and hMLH2 and polymorphisms of MTHFR C677T were assayed. The proportional DNA hypermethylation in P16, MGMT, hMLH1 and hMLH2 in cancer tissues was significantly higher than in remote normal-appearing tissues. DNA hypermethylation of P16 and MGMT was correlated with the T and N stages. Individuals with homozygotes (TT) of MTHFR C677T had significant risk of hypermethylation of MGMT in cancer tissues [OR (95% CI)= 3.47(1.41-7.93)]. However, we did not find association between polymorphism in MTHFR C677T and risk of hypermethylation in P16, MGMT, hMLH1 and hMLH2 genes either in cancer or remote normal-appearing tissues. Aberrant hypermethylation of P16, MGMT, hMLH1 and hMLH2 could be predictive of gastric cancer.

        Numerical calculation of irregular tire wear caused by tread self-excited vibration and sensitivity analysis

        Tire wear negatively affects vehicle safety and riding comfort. Abnormal wear is more dangerous and wears tires out more quickly. In this paper, numerical and sensitivity analyses of polygonal wear caused by unstable vibration are presented. The model used for this study was based on the works of Sueoka. Tread self-excited vibration was analyzed in a quantitative sense, which was qualitatively different from the work of Sueoka. Wear was plotted on tire circumference visually. The mechanism governing polygonal tire wear was investigated as that both the polygonal wear and the standing wave are caused by two types of tread vibrations that only differ in the extent of the tread vibration. Sensitivity analysis shows that decreases in tread mass and stiffness and increases in tread damping lead to noticeable reductions in tire wear. This information could help tire manufacturers produce tires that exhibit less wear caused by tread vibration.

        Design and Analysis of an Active Balancing Mechanism for a Vertical Axis Washing Machine

        In recent years, active balancing of washing machines has drawn much attention from manufactures. This paper proposes a novel active balancing mechanism for vibration suppression of a vertical axis washing machine which employs water for counteracting the unbalanced laundry. At first, a novel structure with two balancing planes sharing only one set of nozzles for injecting water is designed and the balancing capability of the structure is analyzed. Then, a dynamic model of the active balancing system is constructed. Vibration characteristics of the system including changes of the vibration amplitude and phase angle under different influences are studied. After that, a strategy for identifying the imbalance is proposed and an active control method is designed. At last, a washing machine armed with the active mechanism is manufactured. Both a fixed eccentric block and a pile of randomly distributed laundry are used to test the balancing effect. The results show that lateral vibrations of the washer can be greatly reduced by the active mechanism. Due to the simplicity of the structure and the satisfactory balancing effect, the mechanism proposed has a big potential to be employed in future washing machines.

        IL-33 promotes IL-10 production in macrophages: a role for IL-33 in macrophage foam cell formation

        We evaluated the role of IL-10- in IL-33-mediated cholesterol reduction in macrophage-derived foam cells (MFCs) and the mechanism by which IL-33 upregulates IL-10. Serum IL-33 and IL-10 levels in coronary artery disease patients were measured. The effects of IL-33 on intra-MFC cholesterol level, IL-10, ABCA1 and CD36 expression, ERK 1/2, Sp1, STAT3 and STAT4 activation, and IL-10 promoter activity were determined. Core sequences were identified using bioinformatic analysis and sitespecific mutagenesis. The serum IL-33 levels positively correlated with those of IL-10. IL-33 decreased cellular cholesterol level and upregulated IL-10 and ABCA1 but had no effect on CD36 expression. siRNA-IL-10 partially abolished cellular cholesterol reduction and ABCA1 elevation by IL-33 but did not reverse the decreased CD36 levels. IL-33 increased IL-10 mRNA production but had little effect on its stability. IL-33 induced ERK 1/2 phosphorylation and increased the luciferase expression driven by the IL-10 promoter, with the highest extent within the − 2000 to − 1752 bp segment of the 5′-flank of the transcription start site; these effects were counteracted by U0126. IL-33 activated Sp1, STAT3 and STAT4, but only the STAT3 binding site was predicted in the above segment. Site-directed mutagenesis of the predicted STAT3-binding sites (CTGCTTCCTGGCAGCAGAA→CTGCCTGGCAGCAGAA) reduced luciferase activity, and a STAT3 inhibitor blocked the regulatory effects of IL-33 on IL-10 expression. Chromatin immunoprecipitation (CHIP) confirmed the STAT3-binding sequences within the − 1997 to − 1700 and − 1091 to − 811 bp locus regions. IL-33 increased IL-10 expression in MFCs via activating ERK 1/2 and STAT3, which subsequently promoted IL-10 transcription and thus contributed to the beneficial effects of IL-33 on MFCs.

      • A preliminary study on dynamic balance performances during normal walking with high-heeled shoes

        A preliminary study on investigating the effects of high-heeled shoes on wearers’ dynamic balance performances during normal walking was conducted with 5 healthy female adults. Motion capture and F-scan systems were used to record the biomechanical data and several parameters including vertical position of center of body mass (COM), ankle joint angles and knee joint angles were calculated afterwards. The experimental data at flat shoes and high-heeled shoes were compared and the results showed that wearing high-heeled shoes would result in significantly smaller RMS of time series COM height and restrict the flexibility of lower limbs. This suggested human dynamic balance performance became worse with high-heeled shoes.

      • Benzochloroporphyrin Derivative Induced Cytotoxicity and Inhibition of Tumor Recurrence During Photodynamic Therapy for Osteosarcoma

        Photodynamic therapy (PDT) is a promising cancer treatment modality that uses dye-sensitized photooxidation of biologic matter in target tissue. This study explored effects of the photosensitizer BCPD-17 during PDT for osteosarcoma. LM-8 osteosarcoma cells were treated with BCPD-17 and cell viability after laser irradiation was assessed in vitro with the 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide assay. The effects of BCPD-17 during PDT recurrence were then examined on tumor-bearing mice in vivo. BCPD-17 had dosedependent cytotoxic effects on LM-8 osteosarcoma cells after laser irradiation which also had energy-dependent effects on the cells. The rate of local recurrence was reduced when marginal resection of mice tumors was followed by BCPD-17-mediated PDT. Our results indicated BCPD-17-mediated PDT in combination with marginal resection of tumors is a potentially new effective treatment for osteosarcoma.
