Hyperactivated ${\alpha}2$-6-sialylation on N-glycans due to overexpression of the Golgi enzyme ${\beta}$-galactoside: ${\alpha}2$-6-sialyltransferase (ST6Gal-I) often correlates with cancer progression, metastasis, and poor prognosis. This study was aimed to determine the association between ST6Gal-I expression and the risk of recurrence and survival of patients with localized clear-cell renal cell carcinoma (ccRCC) following surgery. We retrospectively enrolled 391 patients (265 in training cohort and 126 in validation cohort) with localized ccRCC underwent nephrectomy at a single center. Tissue microarrays were constructed for immunostaining of ST6Gal-I. Prognostic value and clinical outcomes were evaluated. High ST6Gal-I expression was associated with Fuhrman grade (p<0.001 and p=0.016, respectively) and the University of California Los-Angeles Integrated Staging System (UISS) score (p=0.004 and p=0.017, respectively) in both cohorts. Patients with high ST6Gal-I expression had significantly worse overall survival (OS) (p<0.001 and p<0.001, respectively) and recurrence free survival (RFS) (p<0.001 and p=0.002, respectively) than those with low expression in both cohorts. On multivariate analysis, ST6Gal-I expression remained associated with OS and RFS even after adjusting for the UISS score. Stratified analysis suggested that the association is more pronounced among patients with low and intermediate-risk disease defined by the UISS score. High ST6Gal-I expression is a potential independent adverse predictor of survival and recurrence in ccRCC patients, and the prognostic value is most prominent in those with low and intermediate-risk disease defined by the UISS score.
Estimation of the net energy requirement for maintenance in broilers

Objective: The net energy requirement for the maintenance (NEm) of broilers was determined using regression models by the indirect calorimetry method (ICM) or the comparative slaughter method (CSM). Methods: A 2×4 factorial arrangement of treatments including the evaluation method (ICM or CSM) and feed intake (25%, 50%, 75%, or 100% of ad libitum recommended) was employed in this experiment. In the ICM, 96 male Arbor Acres (AA) birds aged d 15 were used with 4 birds per replicate and 6 replicates in each treatment. In the CSM, 116 male AA birds aged d 15 were used. Among these 116 birds, 20 were selected as for initial data and 96 were assigned to 4 treatments with 6 replicate cages and 4 birds each. The linear regression between retained energy (RE) and metabolizable energy intake (MEI) or the logarithmic regression between heat production (HP) and MEI were used to calculate the metabolizable or net energy requirement for maintenance (MEm) or NEm, respectively. Results: The evaluation method did not detect any differences in the metabolizable energy (ME), net energy (NE), and NE:ME of diet, and in the MEI, HP, and RE of broilers. The MEI, HP, and RE of broilers decreased (p<0.01) as the feed intake decreased. No evaluation method× feed intake interaction was observed on these parameters. The MEm and NEm estimated from the linear relationship were 594 and 386 kJ/kg of body weight (BW)0.75/d in the ICM, and 618 and 404 kJ/kg of BW0.75/d in the CSM, respectively. The MEm and NEm estimated by logarithmic regression were 607 and 448 kJ/kg of BW0.75/d in the ICM, and were 619 and 462 kJ/kg of BW0.75/d in the CSM, respectively. Conclusion: The NEm values obtained in this study provide references for estimating the NE values of broiler diets.

Objective: The objective of current study was to investigate the lactation performance and rumen fermentation characteristics of dairy cows fed a diet with alfalfa hay replaced by corn stover but supplemented with molasses. Methods: Sixteen Holstein cows in mid-lactation were randomly assigned to 1 of 2 dietary treatments: i) alfalfa based diet (AH), and ii) corn stover based diet supplemented with molasses (CSM). The experiment was conducted according to a 2×2 crossover design with 22-d each period, consisting of 17 d for adaptation and 5 d for data and samples collection. Results: Dry matter intake and milk yield were higher for cows fed AH than CSM (p<0.01). Milk protein content and nitrogen conversion were higher (p<0.05), while milk urea nitrogen was lower (p<0.01) for cows fed AH than CSM-fed cows. Contents of milk total solids, fat and lactose were not different between two groups (p>0.10). Total rumen volatile fatty acid concentration tended to be higher (p = 0.06) for cows fed AH than CSM-fed cows. Molar proportion of acetate was lower (p = 0.04), but valerate was higher (p = 0.02) in cows fed AH than CSM-fed cows. Rumen concentration of propionate, and isobutyrate, and ratio of acetate to propionate tended to be different (p<0.10) between two groups. The feed cost per kilogram of milk was lower in CSM than AH (p<0.01). No differences were found in feed efficiency and most plasma parameters tested (p>0.10). Conclusion: In comparison with AH diet, CSM diet could be fed to dairy cows without negative effect on feed efficiency, ruminal fermentation, but economically beneficial, indicating that CSM could be an alternative choice for dairy farms instead of AH to feed mid-lactation dairy cows.
Optical Properties of K9 Glass Waveguides Fabricated by Using Carbon-Ion Implantation

K9 glass is a material with promising properties that make it attractive for optical devices. Ion implantation is a powerful technique to form waveguides with controllable depth and refractive index profile. In this work, optical planar waveguide structures were fabricated in K9 glasses by using 6.0-MeV C3+-ion implantation with a fluence of 1.0 × 1015 ions/cm2. The effective refractive indices of the guided modes were measured by using a prism-coupling system. The refractive index change in the ion-irradiated region was simulated by using the intensity calculation method. The modal intensity profile of the waveguide was calculated and measured by using the finite difference beam propagation method and the end-face coupling technique, respectively. The transmission spectra before and after the implantation showed that the main absorption band was not influenced by the low fluence dopants. The optical properties of the carbon-implanted K9 glass waveguides show promise for use as integrated photonic devices.
Balloon dilation of epidermolysis bullosa-related esophageal strictures: A report of two cases

Epidermolysis bullosa (EB) is a very rare inherited disease featured with skin blistering resulting from minor trauma. Sometimes the esophageal mucosa could also be involved, which leads to esophageal strictures. Here we report two cases of EB-related esophageal strictures who were successfully treated with esophageal balloon dilations. The two cases with EB had severe dysphagia. Clinical examination showed signs of malnutrition, skin blisters and loss of toenails due to EB. They underwent careful fluoroscopic balloon dilation with 10- and 16-mm-sized balloon catheters, respectively. They could ingest soft and some solid foods after the procedure and maintained during the 20 months and 16 months follow-up periods.
Improvement of dynamic responses of a pedestrian bridge by utilizing decorative wind chimes

A novel approach is presented to improve dynamic responses of a pedestrian bridge by utilizing decorative wind chimes. Through wind tunnel tests, it was verified that wind chimes can provide stabilization effects against flutter instability, especially at positive or negative wind angles of attack. At zero degrees of angle of attack, the wind chimes can change the flutter pattern from rapid divergence to gradual divergence. The decorative wind chimes can also provide damping effects to suppress the lateral sway motion of the bridge caused by pedestrian footfalls and wind forces. For this purpose, the swing frequency of the wind chimes should be about the same as the structural frequency, which can be achieved by adjusting the swing length of the wind chimes. The mass and the swing damping level are other two important and mutually interactive parameters in addition to the swing length. In general, 3% to 5% swing damping is necessary to achieve favorite results. In the study case, the equivalent damping level of the entire system can be increased from originally assumed 1% up to 5% by using optimized wind chimes.

<P>We explored a very interesting gold nanoparticle system—pegylated gold in colloidal solution—and analyzed its uptake by mice colorectal adenocarcinoma CT26 tumor cells and the impact on the cell’s response to x-ray irradiation. We found that exposure to polyethylene glycol (PEG) modified (‘pegylated’) 4.7 ± 2.6 nm gold nanoparticles synthesized by a novel synchrotron-based method enhances the response of CT26 cells to x-ray irradiation. Transmission electron microscopy (TEM) and confocal microscopy revealed that substantial amounts of such nanoparticles are taken up and absorbed by the cells and this conclusion is supported by quantitative induced coupled plasma (ICP) results. Standard tests indicated that the internalized particles are highly biocompatible but strongly enhance the cell damage induced by x-ray irradiation. Synchrotron radiation Fourier transform infrared (SR-FTIR) spectromicroscopy analyzed the chemical aspects of this phenomenon: the appearance of C = O stretching bond spectral features could be used as a marker for cell damage and confirmed the enhancement of the radiation-induced toxicity for cells.</P>
Kinetics and computational docking studies on the inhibition of tyrosinase induced by oxymatrine.

<P>A combination of enzymatic inhibition kinetics and computational prediction was employed to search for an effective inhibitor of tyrosinase. We found that oxymatrine significantly inhibited tyrosinase, and that this reaction was not accompanied by detectable conformational changes. Kinetic analysis showed that oxymatrine reversibly inhibited tyrosinase in a mixed-type manner. Measurements of intrinsic and ANS-binding fluorescences showed that oxymatrine did not induce any conspicuous changes in the tertiary structure. We also conducted a docking simulation between tyrosinase and oxymatrine using two docking programs, Dock6.3 and AutoDock4.2 (binding energy was -118.81 kcal/mol for Dock6 and -8.04 kcal/mol for AutoDock4). The results also suggested that oxymatrine interacts mostly with the residues of CYS83 and HIS263 in the active site of tyrosinase. This strategy of predicting tyrosinase inhibition by simulation of docking coupling with kinetics may prove useful in screening for potential tyrosinase inhibitors. Knowledge of tyrosinase inhibition can provide medical, cosmetic, and agricultural applications. Our study suggests that oxymatrine is an important agent for various applications related to pigment formation.</P>
