This study manufactured model slope in soil. When it is rainfall. It experimented weathered granite soil to be distributed in korea mostly as dividing into two soil sample SP, SM, Moreover, inclination of slope(1:1~1:2) and rainfall strength (50~150mm/hr) divided into five step and it observed seepage behavior of slope according to inclination of slope and rainfall strength. It has manufactured six pore water pressure cells in the lower end of model soil box for measuring pore water pressure to be most important factor of slope failure and analyzed change of pore water pressure according to location of slope when rainfall is continued. Change of saturation depth according to experiment condition is measured through the whole soil box by eyes. In result, in case of being in different rainfall strength in sample inclination. The higher rainfall strength rises, The faster seepage of rain progresses. If angle become gentle from 1:1 to 1:2, The seepage velocity increase, too. If compared theoretical number to calculate by ◁수식 삽입▷ (원문을 참조하세요), that Lumb propose with seepage depth to be measured through indoor model test. As a result, Seepage velocity to measure actually is faster than seepage velocity by formula for Lumb to propose If angle become steep, If shows a tendency for pore water pressure to increase in slope angle from 1:22 to 1:1.5. But in angle from 1:1.5 to 1:1, If angle become steep, It shows a tendency for pore water pressure to decrease gradually as standardizing slope angle 1:1.5, According to rainfall strength increases from 50mm/hr to 150mm/hr. Pore water pressure increases in proportion to rainfall strength.es with the much increase of the pore water pressure.

The purpose of this paper is to study the optimal job assignment in which both the man service time and machine processing time are assumed to be random variables. These machines do not have to be of same type, and the above procedures are programmed by BASIC language to use easily and rapidly in the personal computer. The result of this study can contribute to productivity by increasing the efficencies of both operators and machines.

본 연구에서는 실제 기업을 대상으로 생산과 판매, 물류 각 단계의 동기화를 통하여 단계별로 발생하는 데이터를 실시간으로 공유하며, 적정재고 수준 설정으로 재고 유지 비용의 절감을 도모하여 궁극적으로는 생산 계획의 수립 시에서부터 물류 비용을 고려하여 전체 비용의 절감을 기대할 수 있는 시스템을 제안하였다. 과거 우리나라의 기업은 생산 부문의 합리화와 판매 부문에서의 이익 극대화에 힘입어 고도의 성장을 이루어왔으나 시장 환경의 변화와 시장 상황의 악화로 기업의 생존을 위해서는 원가 절감의 시대적 요구를 반영하지 않을 수 없게 되었다. 본 연구의 대상이 되는 기업에서는 현재 부분적인 전산화가 업무별로 단편적으로 이루어지고 있었다. 이에 본 연구에서는 제품의 대외 경쟁력을 좌우하는 많은 요인들 중에서 원가 절감을 위한 지능형 통합 생산 물류 시스템을 제안하고, 제안된 시스템 모듈을 효과적으로 동기화하여 통합시키는 방법을 제시한다. 본 연구의 제안 시스템은 실제 기업의 실정을 반영한 것으로 보다 현실적인 운용이 가능할 것이다.

<P>The purpose of this study was to develop <SUP>64</SUP>Cu-labeled trastuzumab with improved pharmacokinetics for human epidermal growth factor receptor 2 (HER2). <B>Methods:</B> Trastuzumab was conjugated with SCN-Bn-NOTA and radiolabeled with <SUP>64</SUP>Cu. Serum stability and immunoreactivity of <SUP>64</SUP>Cu-NOTA-trastuzumab were tested. Small-animal PET imaging and biodistribution studies were performed in a HER2-positive breast cancer xenograft model (BT-474). The internal dosimetry for experimental animals was determined using the image-based approach with the Monte Carlo N-particle code. <B>Results:</B> <SUP>64</SUP>Cu-NOTA-trastuzumab was prepared with high radiolabeling yield and radiochemical purity (>98%) and showed high stability in serum and good immunoreactivity. Uptake of <SUP>64</SUP>Cu-NOTA-trastuzumab was highest at 48 h after injection as determined by PET imaging and biodistribution results in BT-474 tumors. The blood radioactivity concentrations of <SUP>64</SUP>Cu-NOTA-trastuzumab decreased biexponentially with time in both mice with and mice without BT-474 tumor xenografts. The calculated absorbed dose of <SUP>64</SUP>Cu-NOTA-trastuzumab was 0.048 mGy/MBq for the heart, 0.079 mGy/MBq for the liver, and 0.047 mGy/MBq for the spleen. <B>Conclusion:</B> <SUP>64</SUP>Cu-NOTA-trastuzumab was effectively targeted to the HER2-expressing tumor in vitro and in vivo, and it exhibited a relatively low absorbed dose due to a short residence time. Therefore, <SUP>64</SUP>Cu-NOTA-trastuzumab could be applied to select the right patients and right timing for HER2 therapy, to monitor the treatment response after HER2-targeted therapy, and to detect distal or metastatic spread.</P>

<P>Gastrin-releasing peptide receptor (GRPR) is overexpressed by a variety of human tumors and in particular, identified to be upregulated in prostate cancers. The current study aimed to develop clinically translatable BBN analogue-based radioligands for positron emission tomography (PET) of GRPR-positive tumors. We developed radiolabeled BBN analogues and modified radiolabeled galacto-BBN analogues and then investigated their tumor-targeting efficacy in vivo. The chelator 1,4,7-triazacyclononane, 1-glutaric acid-4,7 acetic acid (NODAGA) was used to radiolabel the peptides with Cu-64. The peptides were evaluated by measuring cell-based receptor-binding affinities. Biodistribution experiments and small animal imaging using PET were performed in nude mice bearing subcutaneous PC3 human prostate cancer xenografts. The conjugates were radiolabeled with yields >99%. The stability assay showed that [Cu-64] NODAGA-BBN and [Cu-64]NODAGA-galacto-BBN remained stable in both human and mouse serum for 1 h at 37 degrees C. PET images of PC3 tumor-bearing nude mice were acquired at 1, 3, 24, 48 and 72 h after injection. [64Cu]NODAGA-galacto-BBN showed retention in tumors for 72 h, low liver uptake, and rapid renal clearance. PET imaging results were also confirmed by biodistrubution 1 and 3 h after injection. [Cu-64]NODAGA-BBN and [Cu-64]NODAGA-galacto-BBN are promising new PET probes for GRPR-positive prostate cancer.</P>

<P>When the coenocytic green alga <I>Bryopsis plumosa</I> (Huds.) Ag. was cut open and the cell contents were expelled, the cell organelles agglutinated rapidly in seawater to form protoplasts. Aggregation of cell organelles in seawater was mediated by a lectin–carbohydrate complementary system. Two sugars, <I>N</I>-acetyl-<SMALL>D</SMALL>-glucosamine and <I>N</I>-acetyl-<SMALL>D</SMALL>-galactosamine inhibited aggregation of cell organelles. The presence of these sugars on the surface of chloroplasts was verified with their complementary fluorescein isothiacyanate-labeled lectins. An agglutination assay using human erythrocytes showed the presence of lectins specific for <I>N</I>-acetyl-<SMALL>D</SMALL>-galactosamine and <I>N</I>-acetyl-<SMALL>D</SMALL>-glucosamine in the crude extract. One-step column purification using <I>N</I>-acetyl-<SMALL>D</SMALL>-glucosamine-agarose affinity chromatography yielded a homogeneous protein. The protein agglutinated the cell organelles of <I>B. plumosa</I>, and its agglutinating activity was inhibited by the above sugars. Sodium dodecyl sulfate polyacrylamide gel electrophoresis results showed that this protein might be composed of two identical subunits cross-linked by two disulfide bridges. Enzyme and chemical deglycosylation experiments showed that this protein is deficient in glycosylation. The molecular weight was determined as 53.8 kDa by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. The N-terminal 15 amino acid sequence of the lectin was Ser–Asp–Leu–Pro–Thr–X–Asp–Phe–Phe–His–Ile–Pro–Glu–Arg–Tyr, and showed no sequence homology to those of other reported proteins. These results suggest that this lectin belongs to a new class of lectins. We named this novel lectin from <I>B. plumosa</I>“bryohealin.”</P>

$^{64}Cu$-labeled diacetyl-bis($N^4$-methylthiosemicarbazone) is a promising agent for internal radiation therapy and imaging of hypoxic tissues. In the study, we confirmed hypoxia regions in VX2 tumor implanted rabbits with injection $^{64}Cu$-ATSM and $^{18}F$-FDG using positron emission tomography (PET)/computed tomography (CT). PET images with $^{18}F$-FDG and $^{64}Cu$-ATSM were obtained for 40 min by dynamic scan and additional delayed PET images of $^{64}Cu$-ATSM the acquired up to 48 hours. Correlation between intratumoral $O_2$ level and $^{64}Cu$-ATSM PET image was analyzed. $^{64}Cu$-ATSM and $^{18}F$-FDG were intravenously co-injected and the tumor was dissected and cut into slices for a dual-tracer autoradiographic analysis. In the PET imaging, $^{64}Cu$-ATSM in VX2 tumors displayed a specific uptake in hypoxic region for48 h. The uptake pattern of $^{64}Cu$-ATSM in VX2 tumor at 24 and 48 h did not match to the $^{18}F$-FDG. Through ROI analysis, in the early phase (dynamic scan), $^{18}F$-FDG has positive correlation with $^{64}Cu$-ATSM but late phase (24 and 48 h) of the $^{64}Cu$-ATSM showed negative correlation with $^{18}F$-FDG. High uptake of $^{64}Cu$-ATSM in hypoxic region was responded with significant decrease of oxygen pressure, which confirmed by $^{64}Cu$-ATSM PET imaging and autoradiographic analysis. In conclusion, $^{64}Cu$-ATSM can utilize for specific targeting of hypoxic region in tumor, and discrimination between necrotic- and viable hypoxic tissue.
