        Utility of DNA barcoding for identification of common Vespa species (Hymenoptera: Vespidae) from Yunnan, China

        As traditional Chinese medicine and edible insects, Ve s p a hornets have high economic value and huge market demand. Hornet breeding has become an important way to increase the income of mountain farmers in China, but the limitation of traditional identification methods has become the bottleneck of the modern development of hornet resources. In this study, a simple and reliable method for the molecular identification of typical hornets from Yunnan was successfully established. In the NCBI database, DNA sequences of hornet samples were aligned as Vespa analis, Ve s p a b a s a l i s , Vespa velutina, Vespa mandarinia, Ve s p a m a g n i f i c a , Ve s p a b i n g h a m i , Ve s p a s o r o r by BLAST search. The efficiency of the PCR amplification of mitochondrial cytochrome C oxidase I (COI) gene sequences in all samples was 100% by using the optimized program. Moreover, a reference database for DNA barcodes of Vespa materials in Yunnan was constructed. This study provided a successful example for the identification of local insects of Yunnan and will be helpful to the development of wasp resources.

        Active Disturbance Rejection Control-based Robust Model Predictive Current Control for Induction Motor

        Induction motor (IM) is widely used in numerous industrial applications. However, achieving high-performance control of IM by using conventional control algorithms is diffi cult because of the mathematical model of high-order, strong coupling, and nonlinearity. Although fi nite control set model predictive current control (FCS-MPCC) has attracted considerable attention because of its simple structure, fast dynamic response, and excellent performance, its performance is degraded substantially if a mismatch exists between model parameters and actual machine parameters. To overcome these problems, active disturbance rejection controller based MPCC (ADRC-MPCC) was proposed for improving the dynamic property and robustness of IM in this paper. The model errors on the IM caused by the load torque and parameter changes were observed and compensated using the extended state observer and nonlinear state error feedback. Two outer loops, namely the ADRC speed loop controller and ADRC fl ux loop controller were designed to improve the rejection disturbance performance of IM speed control system. Furthermore, the calculation process of MPCC of the inner loop is also simplifi ed by ADRC-MPCC. The proposed control strategy was compared and verifi ed with PI control and FCS-MPCC by using MATLAB/Simulink. Simulation and experimental results confi rmed the robustness and dynamic performance of the proposed control strategy

        Development of AFLP and SCAR Markers Linked to a Recessive Genic Male Sterile Gene (Bnms3) in Rapeseed for Marker-assisted Selection

        9012AB is a recessive genic male sterility (RGMS) line in rapeseed, of which the male sterility is controlled by two pairs of recessive duplicate male sterile genes (Bnms3 and Bnms4) interacting with a recessive epistatic suppressor gene (esp). The recessive homozygosity at the esp locus (espesp) can suppress the expression of the recessive male sterility trait in homozygous plants (Bnms3Bnms3Bnms4Bnms4) and result in fertility restoration. A F2 population of 188 plants, derived from self-pollinated progenies of a 9012 AB fertile plants (BnMs3Bnms3Bnms4Bnms4EspEsp), was conducted to identify molecular markers linked to the recessive male sterility gene (Bnms3). By amplified fragment length polymorphism (AFLP) assay combining with bulked segregant analysis (BSA), 13 markers linked to Bnms3 were identified. Linkage analysis indicated that 13 AFLP markers were tightly linked to the Bnms3 gene with a genetic distance varying from 1.3 cM to 7.1 cM. Among them, one marker was co-dominant marker, 6 markers were in coupling phase with Bnms3, and the others were in repulsion phase with Bnms3 gene, One AFLP marker with a genetic distance of 1.4cM was further converted into a SCAR marker successfully, which have been applied in marker-assisted selection of RGMS lines and their temporary maintainers effectively.

        United Electromagnetic Characteristics and Online Monitoring Method of Static Air-gap Eccentricity of Turbo-Generator

        The purpose of this paper is to investigate the united Electromagnetic characteristics for the effective monitoring on the static air-gap eccentricity (SAGE) of turbo-generator. Different from other studies, this paper not only studies on the unbalanced magnetic pull (UMP) and the vibration characteristics of the stator and the rotor, but also investigates the harmonic features of the magnetic flux density and the circulating current inside the parallel branches (CCPB). The theoretical calculation, together with the finite-element-method (FEM) simulation and the experiment verification, is taken for a SDF-9 type non-salient generator. It is shown that, when SAGE occurs, apparent doublefrequency UMP and vibrations will be produced both on the stator and the rotor, while the CCPB will have an obvious increment at the 1<SUP>st</SUP> harmonic component. In addition, the amplitude of the magnetic flux density will be of cosine distribution in the circumferential position of the air-gap, while in normal condition it is a constant. Moreover, the pass-band amplitude, together with the 1<SUP>st</SUP> harmonic of the magnetic flux density, will be enlarged as well. These united electromagnetic characteristics can be used as the diagnosis and monitoring criterion for SAGE.

        Effect of male inflorescence of Castanea mollissima on the reproductive development and lifetime of Bactrocera minax

        Previous work reveals that newly emerged adult Chinese citrus flies, Bactrocera minax (Enderlein), emerges from the ground in the mosaic-type citrus orchard of the hilly terrain landform dispersed into adjacent forest, and the newly emerged adult flies fed on male inflorescence of the Chinese chestnut, Castanea mollissima Blume (Fagaceae). The potential impact of male inflorescence of C. mollissima on the longevity, and survival of adult females and males, the ovarian development of adult females, and the abdominal index, ovarian index, and egg length of the fourth ovarian grade of living females, as well as the flowering process of the male inflorescence of the Chinese chestnut, were studied with the three methods of rearing (normal rearing, hungry rearing, and bagging rearing). For all three rearing treatments, the longevity of all adults could live up to 38 days, there were similar time dynamics for the average survival of female and male adult flies. Of the total females, 32.1%–100% females with bagging rearing could attain the fourth ovarian grade like those in rearing and hunger rearing. The abdominal index, ovarian index, and egg length of the fourth ovarian grade of 27–39 days’ old living females with bagging rearing (it corresponds to the late flower withering period), were 75.1%, 49.8% and 89.6% of those with normal rearing, and 73.2%, 60.6% and 90.8% of those with hungry rearing, respectively. The male inflorescence of C. mollissima can satisfy the nutritional requirements of adult B. minax, enabling their survival and normal reproduction. This study can provide references for development and application of artificial feed and food attractants of adult B. minax.

      • Genetic Variations in the HIF1A Gene Modulate Response to Adjuvant Chemotherapy after Surgery in Patients with Colorectal Cancer

        Background: Hypoxia-inducible factor $1{\alpha}$ (HIF-$1{\alpha}$) plays an important role in regulating cell survival and angiogenesis, which are critical for tumor growth and metastasis. Genetic variations of HIF1A have been shown to influence the susceptibility to many kinds of human tumors. Increased expression of HIF-$1{\alpha}$ has also been demonstrated to be involved in tumor progression. However, the prognostic value of single nucleotide polymorphisms (SNPs) inthe HIF1A gene remains to be determined in most cancer types, including colorectal cancer (CRC). In this study, we sought to investigate the predictive role of HIF1A SNPs in prognosis of CRC patients and efficacy of chemotherapy. Materials and Methods: We genotyped two functional SNPs in HIF1A gene using the Sequenom iPLEX genotyping system and then assessed their associations with clinicopathological parameters and clinical outcomes of 697 CRC patients receiving radical surgery using Cox logistic regression model and Kaplan Meier curves. Results: Generally, no significant association was found between these 2 SNPs and clinical outcomes of CRC. In stratified analysis of subgroup without adjuvant chemotherapy, patients carrying CT/TT genotypes of rs2057482 exhibited a borderline significant association with better overall survival when compared with those carrying CC genotype [Hazard ratio (HR), 0.47; 95% confidence interval (95% CI): 0.29-0.76; P < 0.01]. Moreover, significant protective effects on CRC outcomes conferred by adjuvant chemotherapy were exclusively observed in patients carrying CC genotype of rs2057482 and in those carrying AC/CC genotype of rs2301113. Conclusions: Genetic variations in HIF1A gene may modulate the efficacy of adjuvant chemotherapy after surgery in CRC patients.

        Endothelial Aquaporin-1 (AQP1) Expression Is Regulated by Transcription Factor Mef2c

        Aquaporin 1 (AQP1) is expressed in most microvascula-ture endothelial cells and forms water channels that play major roles in a variety of physiologic processes. This study aimed to delineate the transcriptional regulation of AQP1 by Mef2c in endothelial cells. Mef2c cooperated with Sp1 to activate human AQP1 transcription by binding to its proximal promoter in human umbilical cord vein endothelial cells (HUVEC). Over-expression of Mef2c, Sp1, or Mef2c/Sp1 increased HUVEC migration and tube-forming ability, which can be abolished AQP1 knockdown. These data indicate that AQP1 is a direct target of Mef2c in regulating angiogenesis and vasculogenesis of endothelial cells.

        Tracking the movement trajectory of newly emerged adult Chinese citrus flies with insect harmonic radar

        Previous work reveals that the adult Chinese citrus fly, Bactrocera minax (Enderlein), emerges from the ground in citrus orchards and presumably disperses to alternate food sources in neighboring shrubs and trees, where it stays and becomes reproductively mature, and then later returns to the orchard to lay eggs. We investigated the trajectories of early emerged adult Chinese citrus flies, which were tracked with portable harmonic radar in 2013, 2014, 2015, and 2016 in four different habitats in Hubei province of China that presumably provided food sources for adults. This study we investigated whether early emerged adult Chinese citrus flies migrate into forests. A total of 47 adults were tracked following release in a citrus orchard. Our results showed that a total of 33 adults released in a citrus orchard flew into the forest, and 14 adults that disappeared during the tracking process in the citrus orchard. A total of 10 adults (in 2014) that were tracked and released in the forest flew to the forest. Of the adult flies investigated, 54.5% flew directly into the forest, while the remaining 45.5% finally arrived in the forest after repeatedly adjusting their trajectories. When there were Castanea mollissima Blume (Fagaceae) in the surrounding forest, the proportion of frequency of adult landing on the plant represented 58.3% and 88.5% of landing on all species in 2014 and in 2015, respectively. We conclude that early emerged adult Chinese citrus flies in the mosaic-type citrus orchard of the hilly terrain landform dispersed into adjacent forest.
