Background: Panax ginseng Meyer (P. ginseng), a herb distributed in Korea, China and Japan, exerts benefits on diverse inflammatory conditions. However, the underlying mechanism and active ingredients remains largely unclear. Herein, we aimed to explore the active ingredients of P. ginseng against inflammation and elucidate underlying mechanisms. Methods: Inflammation model was constructed by lipopolysaccharide (LPS) in C57BL/6 mice and RAW264.7 macrophages. Molecular docking, molecular dynamics, surface plasmon resonance imaging (SPRi) and immunofluorescence were utilized to predict active component. Results: P. ginseng significantly inhibited LPS-induced lung injury and the expression of proinflammatory factors, including TNF-a, IL-6 and IL-1b. Additionally, P. ginseng blocked fluorescence-labeled LPS (LPS488) binding to the membranes of RAW264.7 macrophages, the phosphorylation of nuclear factor-kB (NF-kB) and mitogen-activated protein kinases (MAPKs). Furthermore, molecular docking demonstrated that ginsenoside Ro (GRo) docked into the LPS binding site of toll like receptor 4 (TLR4)/myeloid differentiation factor 2 (MD2) complex. Molecular dynamic simulations showed that the MD2-GRo binding conformation was stable. SPRi demonstrated an excellent interaction between TLR4/MD2 complex and GRo (KD value of 1.16 × 10-9 M). GRo significantly inhibited LPS488 binding to cell membranes. Further studies showed that GRo markedly suppressed LPS-triggered lung injury, the transcription and secretion levels of TNF-α, IL-6 and IL-1β. Moreover, the phosphorylation of NF-kB and MAPKs as well as the p65 subunit nuclear translocation were inhibited by GRo dose-dependently. Conclusion: Our results suggest that GRo exerts anti-inflammation actions by direct inhibition of TLR4 signaling pathway.

This study combines the feedback linearization method and our recent researches of Chen electrical unifying approach to propose a global tracking controller design for nonlinear uncertain diff erential-diff erence systems with time delay. One example, which cannot be solved by the fi rst paper on the almost disturbance decoupling problem, is presented in this study to show the essential points that the tracking performance is easily solved by the proposed approach. The study applies Chen electrical unifying approach to take the place of using the disorganized virtual ground technique and the Kirchhoff ’s law for those traditional approaches. In order to show its signifi cant practicability, the study has fi rstly designed an easy-to-implement adder and a time-delay circuit, and an unstable time-delay operational amplifi er control system based on the Chen electrical unifying approach, and then has proposed a stable controller.

The surface modification of poly (vinylidene fluoride) (PVDF) membrane was performed via a two-step polymerization reactions. Poly (acrylic acid) (PAAc) was first grafted onto the membrane surface for the preparation of PVDF-g-PAAc membrane, and then poly (ethylene glycol) 200 (PEG 200) was immobilized on the membrane surface by the esterification reaction for the fabrication of PVDF-g-PEGA membrane. Attenuated total reflectance (ATR) FTIR, X-ray photoelectron spectroscopy (XPS), scanning electron microscope (SEM), and protein adsorption, water flux, water content and dynamic contact angle were conducted to characterize the structures and performance of the resultant PVDF membranes. The experimental results showed that the adsorption of bovine serum albumin (BSA) on the PVDF-g-PEGA membrane decreased about 80% when the grafting ratio reached to 15 wt%, compared with the pristine PVDF membrane. Moreover, the water contact angle of the membrane dropped to 60.5o, while the membrane pore sizes remained little changed.

Osmanthus fragrans is a woody, evergreenspecies of shrubs and small trees that is extensively plantedin sub-tropical and temperate climates as an ornamentalplant in gardens and for its health benefits. The flower colorranges from ivory to orange to pink among different varietiesand even color difference during the whole blossom inthe sweet osmanthus. Sweet osmanthus is widely cultivatedthroughout China and other countries due to its prominentfragrance, colorful flowers, and medicinal properties. However, the scanty genomic resources in the Olea familyhave greatly hindered further exploration of its geneticmechanism on these economically important traits. In thisstudy, transcriptome sequencing of O. fragrans was performedusing the Illumina HighSeqTM2000 sequencingplatform. Next generation sequencing (NGS) of the transcriptomeof O. fragrans produced 31.7G of clean bases(211,266,818 clean reads) that were assembled into256,774 transcripts and 117,595 unigenes. Of them, 197and 237 candidate genes involved in fragrance and pigmentbiosynthesis respectively were identified based on functionannotation. Meanwhile, 1 unnamed protein and 468 functionalunknown genes were also identified. Furthermore,mRNA sequencing expression profiling of O. fragranswere compared to previous genes’. In summary, thiscomprehensive transcriptome dataset allows the identificationof genes associated with several major metabolicpathways and provides a useful public information platformfor further functional genomic studiesin O. fragransLour.

This meta-analysis was performed to evaluate and compare the outcomes of robotic gastrectomy (RG) and laparoscopic gastrectomy (LG) for treating gastric cancer. A systematic literature search was carried out using the PubMed database, Web of Knowledge, and the Cochrane Library database to obtain comparative studies assessing the safety and efficiency between RG and LG in May, 2013. Data of interest were analyzed by using of Review Manager version 5.2 software (Cochrane Collaboration). A fixed effects model or random effects model was applied according to heterogeneity. Seven papers reporting results that compared robotic gastrectomy with laparoscopic gastrectomy for gastric cancer were selected for this meta-analysis. Our metaanalysis included 2,235 patients with gastric cancer, of which 1,473 had undergone laparoscopic gastrectomy, and 762 had received robotic gastrectomy. Compared with laparoscopic gastrectomy, robotic gastrectomy was associated with longer operative time but less blood loss. There were no significant difference in terms of hospital stay, total postoperative complication rate, proximal margin, distal margin, numbers of harvested lymph nodes and mortality rate between robotic gastrectomy and laparoscopic gastrectomy. Our meta-analysis showed that robotic gastrectomy is a safe technique for treating gastric cancer that compares favorably with laparoscopic gastrectomy in short term outcomes. However, the long term outcomes between the two techniques need to be further examined.

Insect neuropeptides are involved in many important developmental processes. Here, we cloned three neuro peptide genes (SfEH, SfETH, and SfCCAP) from Sogatella furcifera (Horváth) and analyzed the relative expression levels of these genes through Real-time Quantitative polymerase chain reaction (RT-qPCR) during different developmental stages and in different wing types of S. furcifera following exposure to sublethal concentrations (LC 10 and LC 25 ) of methylaminoavermectin benzoate. The results revealed that the three neuropeptide genes (SfEH, SfETH, and SfCCAP) of S. furcifera contained open reading frame sequences of 222, 636, and 408 bp, encoding 73, 211, and 135 amino acids, respectively. Moreover, the expression of SfEH and SfETH reached its peak on day 3 of the 5 th instar nymph stage, while the SfCCAP expressed pinnacle on 40 min after eclosion. Furthermore, the relative expression levels of the three genes were in the following order: SfEH, short-winged female > long-winged female > long-winged male; SfETH, long-winged female > short-winged female > longwinged male; and SfCCAP, short-winged female > long-winged male > short-winged female. Upon calculating the relative expression of the three peptide genes after 24 and 48 h of treatment with sublethal concentrations of methylaminoavermectin benzoate, it was found that the relative mRNA expression levels of SfEH and SfETH declined upon LC 10 treatment but increased upon LC 25 treatment. Meanwhile, the relative mRNA expression level of SfCCAP declined upon both LC 10 and LC 25 treatments. This result will help us understand the effects of pesticides on insect growth and development at the molecular level.

Introduction: The esophagus squamous cell carcinoma (ESCC) is one of the most deadly malignances, and a current challenge is the development of effective therapeutic agents. Our present work addressed the effect of HIF-$1{\alpha}$ siRNA alone or in combination with cisplatin on the growth of ESCC in nude mice. Materials and Methods: Xenografts were established by inoculating ESCC TE-1 cells in nude mice, and transplanted tumors were treated with HIF-$1{\alpha}$ siRNA, cisplatin alone or together. Growth was assessed by measuring tumor volume. HIF-$1{\alpha}$ mRNA and protein expression were detected using RT-PCR and immunohistochemistry, respectively. Apoptosis of ESCC TE-1 cells was analyzed by flow cytometry. Results: In our nude mice model, HIF-$1{\alpha}$ siRNA effectively inhibited the growth of transplanted ESCC, downregulating HIF-$1{\alpha}$ mRNA and protein expression, and inducing ESCC TE-1 cell apoptosis. Notably when combinated with cisplatin, HIF-$1{\alpha}$ siRNA showed synergistic interaction in suppressing tumor growth. Furthermore, the proportion of apoptotic cells in HIF-$1{\alpha}$ siRNA plus cisplatin group was significantly higher than that in cisplatin or HIF-$1{\alpha}$ siRNA-treated groups (P<0.05). Conclusions: Down-regulated HIF-$1{\alpha}$ expression induced by siRNA could effectively suppress the growth of transplanted ESCC $in$ $vivo$. HIF-$1{\alpha}$ siRNA could enhance the cytotoxicity of cisplatin, which suggests that a combination of these two agents may have potential for therapy of advanced ESCC.
