        Cloning and Functional Analysis of the Gβ Gene Mgb1 and the GγGene Mgg1 in Monascus ruber

        The ascomycetous fungus Monascus ruber is one of the most well-known species widely used to produce Monascus-fermentation products for natural food colorants and medicine. Our previous research on the Gα subunit Mga1 and the regulator of G protein signaling MrflbA indicated that heterotrimeric G protein signaling pathways were involved in aspects of growth, sporulation and secondary metabolite production in M. ruber. To better understand the G protein signaling pathways in this fungus, a Gβ subunit gene (Mgb1)and a Gγ subunit gene (Mgg1) were cloned and investigated in the current study. The predicted Mgb1 protein consisted of 353 amino acids and Mgg1 consisted of 94 amino acids,sharing marked similarity with Aspergillus Gβ and Gγ subunits,respectively. Targeted deletion (Δ) of Mgb1 or Mgg1resulted in phenotypic alterations similar to those resulting from ΔMga1, i.e., restricted vegetative growth, lowered asexual sporulation, impaired cleistothecial formation, and enhanced citrinin and pigment production. Moreover, deletion of Mgg1suppressed the defects in asexual development and in biosynthesis of citrinin and pigment caused by the absence of MrflbA function. These results provide evidence that Mgb1and Mgg1 form a functional Gβγ dimer and the dimer interacts with Mga1 to mediate signaling pathways, which are negatively controlled by MrflbA, for growth, reproduction and citrinin and pigment biosynthesis in M. ruber.

        Identification and Characterization of a Novel Angiotensin Iconverting Enzyme Inhibitory Peptide (ACEIP) from Silkworm Pupa

        The angiotensin I-converting enzyme inhibitorypeptide (ACEIP) was isolated and characterized fromsilkworm pupae and purified using Sephadex G-25 gelfiltration. The structure and physicochemical properties ofpupa ACEIP were analyzed. The α-P3 fraction exhibitedthe most potent ACE inhibitory activity. After purificationvia semi-preparative reverse-phase HPLC (RP-HPLC) andHPLC, the α-P3-6-b component was revealed to have thehighest ACE inhibitory activity (IC50=28.3 μg/mL). Edmandegradation revealed a Val-Glu-Ile-Ser amino acid sequencein which novel active sequences were identified. Physicochemicalproperty testing showed that purified pupaACEIP exhibits good solubility, heat resistance, and acidresistance that all indicate ACEIP derived from silkwormpupa is an excellent food-derived ACEIP.

      • EFFECTIVENESS OF WATER SURFACE WASHING IN CLEANSING RAPID SAND FILTERS

        Experiments in water surface washing with and without water backwashing were conducted to investigate the mechanism and the effect of water surface washing in the cleansing of rapid sand filters. The results showed that the effect of water surface washing alone was limited only to the upper portion of the filter bed with a depth of less than 8 cm from the sand surface. However, through the combined use with water backwashing, its influence could reach the lower portion of the filter bed, leading to a much higher discharge rate of the turbid matter deposited throughout the whole filter. Moreover, Introducing the surface washing during the middle period, compared to carrying it out during the initial or the final period of water backwashing, proved to be much more effective.

        A Facile Green Synthesis of Sm2O3 Nanoparticles via Microwave-Assisted Urea Precipitation Route and Their Optical Properties

        Samarium oxide (Sm2O3) nanoparticles with a narrow sizedistribution were successfully synthesized by microwaveassistedusing urea as precipitant without surfactant ortemplate. The Sm2O3 particles were characterized using X-raydiffraction analysis, field-emission scanning electronmicroscopy, field-emission transmission electron microscopyand ultraviolet-visible-near-infrared spectrophotometer. Theresults showed that the samples prepared with differentconcentration of urea had different particle sizes. When theconcentration of urea was 1.2 mol/L, the sample had thesmallest particle size. A possible mechanism for the formationof the nanoparticles was proposed. Optical properties ofSm2O3 nanoparticles showed that the nanoparticles had astrong absorption property in the deep ultraviolet regionbetween 200 nm and 270 nm.

        The Long Noncoding RNA NEAT1 Targets miR-34a-5p and Drives Nasopharyngeal Carcinoma Progression via Wnt/β-Catenin Signaling

        Purpose: Long noncoding RNA nuclear paraspeckle assembly transcript 1 (NEAT1) has been deemed an oncogene in many humancancers. However, the underlying mechanism of NEAT1 in nasopharyngeal carcinoma (NPC) progression remains largelyunclear. Materials and Methods: Quantitative real-time PCR assay was performed to assess the expression of NEAT1 and miR-34a-5p inNPC tissues and cells. Western blot analysis was used to observe cell epithelial to mesenchymal transition (EMT) and the activationof Wnt/β-catenin signaling in 5-8F cells. MiRNA directly interacting with NEAT1 were verified by dual-luciferase reporter assayand RNA immunoprecipitation. Cell proliferation ability was determined by CCK-8 assay, and cell migration and invasion capacitieswere assessed by transwell assays. An animal model was used to investigate the regulatory effect of NEAT1 on tumorgrowth in vivo. Results: Our data revealed that NEAT1 is upregulated, while miR-34a-5p is downregulated in NPC tissues and cell lines. NEAT1knockdown repressed tumor growth in vitro and in vivo. Additionally, we discovered that NEAT1 directly binds to miR-34a-5pand suppresses miR-34a-5p expression. Moreover, NEAT1 knockdown exerted suppression effects on cell proliferation, migration,invasion, and EMT by miR-34a-5p. NEAT1 knockdown blocked Wnt/β-catenin signaling via miR-34a-5p. Conclusion: Our study demonstrated that NEAT1 targets miR-34a-5p at least partly to drive NPC progression by regulating Wnt/β-catenin signaling, suggesting a potential therapeutic target for NPC.

        Gamma spectrum denoising method based on improved wavelet threshold

        Adverse effects in the measured gamma spectrum caused by radioactive statistical fluctuations, gamma ray scattering, and electronic noise can be reduced by energy spectrum denoising. Wavelet threshold denoising can be used to perform multi-scale and multi-resolution analysis on noisy signals with small root mean square errors and high signal-to-noise ratios. However, in traditional wavelet threshold denoising methods, there are signal oscillations in hard threshold denoising and constant deviations in soft threshold denoising. An improved wavelet threshold calculation method and threshold processing function are proposed in this paper. The improved threshold calculation method takes into account the influence of the number of wavelet decomposition layers and reduces the deviation caused by the inaccuracy of the threshold. The improved threshold processing function can be continuously guided, which solves the discontinuity of the traditional hard threshold function, avoids the constant deviation caused by the traditional soft threshold method. The examples show that the proposed method can accurately denoise and preserves the characteristic signals well in the gamma energy spectrum

      • DC component Mitigation Utilizing BP-PI and Repetitive Control in Transformless Grid-Connected Inverters

        Transformerless grid-connected inverters, due to their advantages in high efficiency, small volume and light weight, gain more research and interest in recent years. Due to the asymmetrical driving signal in pulse-width-modulation (PWM) caused by time-delay, zero-drift of the current sensors and imparities of the power transistors, etc. Output of the grid current contains dc component. As a result, power quality of the grid is degraded. In this paper, a dc component suppression scheme with recursive integral PI controller are introduced, according to the change tendency of tracking error, the coefficients of PI controller are regulated by BP neural network in real time, and the global optimization requirements are met. Sliding-Window-Double-Iteration-Method (SWDIM) is utilized for fast dc current extraction. Simulation results from a 2-kW three-phase grid-connected inverter confirm the effectiveness of the proposed scheme.

      • Light Load Efficiency Improvement Method for GaN-Based DAB Converter with Hybrid Discontinuous Current Mode

        Conventional phase-shift controlled dual active bridge (DAB) converters which operate in discontinuous current mode (DCM) are hard to realize the zero-voltage switching (ZVS) due to the light load. This paper proposed an efficiency improvement method for the DAB converter with fix-frequency hybrid discontinue current mode (HDCM). By adopting the HDCM control method, synchronous rectification and wide load range ZVS of switching devices are realized simultaneously without changing the components of the conventional DAB power stage in DCM. Besides, to further improve the conversion ratio and efficiency, the voltage-doubled rectification (VDR) is adopted. The HDCM implementation method with a digital signal processor (DSP) is proposed. A 200W laboratory prototype is designed to validate the correctness of HDCM. Compared to the conventional DAB converter which operates in DCM, experimental results demonstrate that the efficiency was increased 1.2% at 10% rated load.

      • Numerator-Denominator Model Based H<SUB>∞</SUB> Robust Control for LCL Filter-Based Grid-Connected Inverters under Weak Grid Conditions

        In the grid-connected inverter controller system, conventional PI or PR controller decrease their lowfrequency gain and bandwidth to maintain the stability of the system. However, this degrades the disturbance rejection and current tracking capability. To solve this problem, this paper gives an H∞ robust control scheme based on Numerator Denominator Model method, which has the advantages of simplified current controller design procedure, can place the poles of closed-loop system at the desired place. By properly selecting the weight function, the proposed control scheme can not only achieve good robustness in disturbance rejection capability, but also gives a straight forward to design the dynamic performance. Simulation and experimental comparison results between PR and H∞ controller is demonstrated. The results prove that the proposed control scheme shows obviously improvements on the robustness of inverter and meanwhile does not lose advantages on the dynamic performance.
