      • Interpretation of cryogenic-temperature Charpy fracture initiation and propagation energies by microstructural evolution occurring during dynamic compressive test of austenitic Fe-(0.4,1.0)C-18Mn steels

        In the present study, Charpy impact energy (E<SUB>T</SUB>) composed of fracture initiation energy (E<SUB>I</SUB>) and propagation energy (E<SUB>P</SUB>) of austenitic Fe-(0.4,1.0)C-18Mn steels was evaluated in the temperature range from room to cryogenic temperatures by an instrumented Charpy impact tester, and was interpreted by microstructural evolution of dynamically compressed specimens. In the 1.0C-18Mn steel, the E<SUB>I</SUB> and E<SUB>P</SUB> decreased slightly with decreasing temperature, but the E<SUB>P</SUB>/E<SUB>T</SUB> ratio was kept to be about 0.5. In the 0.4C-18Mn steel, the E<SUB>I</SUB> remained almost constant or slightly decreased with decreasing temperature, while the E<SUB>P</SUB>/E<SUB>T</SUB> ratio steadily decreased, thereby leading to the lower (about 30%) cryogenic-temperature E<SUB>T</SUB> than that of the 1.0C-18Mn steel. Under the dynamic compressive loading, a considerable number of ε-martensites were formed in the 0.4C-18Mn steel, whereas they were not found in the 1.0C-18Mn steel, and their volume fractions increased steadily with decreasing temperature. This γ→ε-martensite transformation was attributed to the decrease in stacking fault energy, and resulted in the very low E<SUB>P</SUB> and resultant E<SUB>T</SUB>.

        The saponins of two- and four-year-old American ginseng plants (Panax quinquefolium L.) (Araliaceae) collected in July and September were studied. American ginseng saponins (panaquilins) differ from Korean ginseng (Panax ginseng C.A. MEYER) saponins (ginsenosides). The American ginseng saponins separated and named were panaquilins A, B, C, D, E-1, E-2, E-3, G-1, G-2, (c) and (d). One-dimensional thin-layer chromatography did not completely separate panaquilin mixture and was subject to misinterpretation. The panaquilins were more accurately separated and identified by the two-dimensional thin-layer method established. Some differences in American ginseng saponins were dependent upon the plant age, time of collection, and part extracted. The American ginseng sapogenin components are panaxadiol (panaquilins B and C), oleanolic acid (panaquilin D) and panaxatriol (panaquilin G-1). The panaquilins E-1, E-2 and E-3 mixture contained both panaxadiol and panaxatriol. The genins of panaquilins A, (c), (d) and G-2 were not identified. In addition, β-sitosterol and stigmasterol were identified from the root ether extracts.

        Differential Ubiquitin Binding by the Acidic Loops of Ube2g1 and Ube2r1 Enzymes Distinguishes Their Lys-48-ubiquitylation Activities

        <P>The ubiquitin E2 enzymes, Ube2g1 and Ube2r1, are able to synthesize Lys-48-linked polyubiquitins without an E3 ligase but how that is accomplished has been unclear. Although both E2s contain essential acidic loops, only Ube2r1 requires an additional C-terminal extension (184–196) for efficient Lys-48-ubiquitylation activity. The presence of Tyr-102 and Tyr-104 in the Ube2g1 acidic loop enhanced both ubiquitin binding and Lys-48-ubiquitylation and distinguished Ube2g1 from the otherwise similar truncated Ube2r1<SUP>1–183</SUP> (Ube2r1C). Replacement of Gln-105–Ser-106–Gly-107 in the acidic loop of Ube2r1C (Ube2r1C<SUP>YGY</SUP>) by the corresponding residues from Ube2g1 (Tyr-102–Gly-103–Tyr-104) increased Lys-48-ubiquitylation activity and ubiquitin binding. Two E2∼UB thioester mimics (oxyester and disulfide) were prepared to characterize the ubiquitin binding activity of the acidic loop. The oxyester but not the disulfide derivative was found to be a functional equivalent of the E2∼UB thioester. The ubiquitin moiety of the Ube2r1C<SUP>C93S</SUP>-[<SUP>15</SUP>N]UB<SUP>K48R</SUP> oxyester displayed two-state conformational exchange, whereas the Ube2r1C<SUP>C93S/YGY</SUP>-[<SUP>15</SUP>N]UB<SUP>K48R</SUP> oxyester showed predominantly one state. Together with NMR studies that compared UB<SUP>K48R</SUP> oxyesters of the wild-type and the acidic loop mutant (Y102G/Y104G) forms of Ube2g1, <I>in vitro</I> ubiquitylation assays with various mutation forms of the E2s revealed how the intramolecular interaction between the acidic loop and the attached donor ubiquitin regulates Lys-48-ubiquitylation activity.</P>

        Cooperativity of E-cadherin and Smad4 Loss to Promote Diffuse-Type Gastric Adenocarcinoma and Metastasis

        <P>Loss of E-cadherin (<I>CDH1</I>), Smad4, and p53 has been shown to play an integral role in gastric, intestinal, and breast cancer formation. Compound conditional knockout mice for Smad4, p53, and E-cadherin were generated to define and compare the roles of these genes in gastric, intestinal, and breast cancer development by crossing with <I>Pdx-1-Cre</I>, <I>Villin-Cre</I>, and <I>MMTV-Cre</I> transgenic mice. Interestingly, gastric adenocarcinoma was significantly more frequent in <I>Pdx-1-Cre;Smad4<SUP>F/F</SUP>;Trp53<SUP>F/F</SUP>;Cdh1<SUP>F</SUP></I><SUP>/+</SUP> mice than in <I>Pdx-1-Cre;Smad4<SUP>F/F</SUP>;Trp53<SUP>F/F</SUP>;Cdh1</I><SUP>+/+</SUP> mice, demonstrating that <I>Cdh1</I> heterozygosity accelerates the development and progression of gastric adenocarcinoma, in combination with loss of Smad4 and p53. <I>Pdx-1-Cre;Smad4<SUP>F/F</SUP>;Trp53<SUP>F/F</SUP>;Cdh1<SUP>F</SUP></I><SUP>/+</SUP> mice developed gastric adenocarcinomas without E-cadherin expression. However, intestinal and mammary adenocarcinomas with the same genetic background retained E-cadherin expression and were phenotypically similar to mice with both wild-type <I>Cdh1</I> alleles. Lung metastases were identified in <I>Pdx-1-Cre;Smad4<SUP>F/F</SUP>;Trp53<SUP>F/F</SUP>;Cdh1<SUP>F</SUP></I><SUP>/+</SUP> mice, but not in the other genotypes. Nuclear β-catenin accumulation was identified at the invasive tumor front of gastric adenocarcinomas arising in <I>Pdx-1-Cre;Smad4<SUP>F/F</SUP>;Trp53<SUP>F/F</SUP>;Cdh1<SUP>F</SUP></I><SUP>/+</SUP> mice. This phenotype was less prominent in mice with intact E-cadherin or Smad4, indicating that the inhibition of β-catenin signaling by E-cadherin or Smad4 downregulates signaling pathways involved in metastases in <I>Pdx-1-Cre;Smad4<SUP>F/F</SUP>;Trp53<SUP>F/F</SUP>;Cdh1<SUP>F</SUP></I><SUP>/+</SUP> mice. Knockdown of β-catenin significantly inhibited the migratory activity of <I>Pdx-1-Cre;Smad4<SUP>F/F</SUP>;Trp53<SUP>F/F</SUP>;Cdh1<SUP>F</SUP></I><SUP>/+</SUP> cell lines. Thus, loss of E-cadherin and Smad4 cooperates with p53 loss to promote the development and metastatic progression of gastric adenocarcinomas, with similarities to human gastric adenocarcinoma.</P><P><B>Implications:</B> This study demonstrates that inhibition of β-catenin is a converging node for the antimetastatic signaling pathways driven by E-cadherin and Smad4 in <I>Pdx-1-Cre;Smad4<SUP>F/F</SUP>;Trp53<SUP>F/F</SUP>;Cdh1<SUP>F</SUP></I><SUP>/+</SUP> mice, providing novel insights into mechanisms for gastric cancer metastasis. <I>Mol Cancer Res; 12(8); 1088–99. ©2014 AACR</I>.</P>

        Purposes :The purposes of this study was to investigate the self-managing status according to the life styles and the diabetic knowledge of early adulthood. The subjects in this study were 240people, the early adult aged twenties to thirties living in the metropolitan area. Data were collected by using self-administered questionnaires that measure about the knowledge of diabetes; the dietetic behavior; the knowledge of diet; the exercise behavior; the knowledge of exercise. Descriptive statistics, t-test, ANOVA, and correlation were used for data analysis. Result :1. At the knowledge of diabetes degree based on the general character, the subjects who had experiences of diabetic education have higher degree of the diabetic knowledge. 2. At the knowledge of diabetes degree according to the life style, the subjects who have regular diet are appeared highly. Also the subjects who don't take much food of animal origin have higher degree. 3. At the knowledge of diet based on the life style, the subjects who don't keep three times meals a day are appeared highly. Also the subjects who have regular diet have higher degree. 4. At the knowledge of exercise according to the life style, the subjects who take extra nutritions 3-4 times a week are appeared highly. Although the subjects tend to know about dietary, exercise and health management, it showed that they actually don't perform what they know in their everyday of life. The reason for this could be the lack of perceived seriousness about diabetes mellitus. Thus as nursing professionals, we should provide them proper health education and support. Moreover, advanced research on the empowerment is warranted.

      • Effects of estrogen and estrogenic compounds, 4-tert-octylphenol, and bisphenol A on the uterine contraction and contraction-associated proteins in rats

        We examined the effects of estradiol (E2), 4-tert-octylphenol (OP), and bisphenol A (BPA) on uterine contractions in immature rats. The expression and localization of contraction-associated proteins (CAPs), and contractility of rat uterus with a collagen gel contraction assay were analyzed. E2, OP, and BPA all increased oxytocin (OT)-related pathway, while the prostaglandin-related signaling was reduced. Interestingly, E2 and estrogenic compounds showed distinct effects on the contractile activity of uterine cells. E2 enhanced the contractility, while OP and BPA significantly decreased it. Immunohistochemical analysis of CAPs showed distinct regulation of prostaglandin F receptor localization by E2 and estrogenic compounds, which may explain the different contractile activities of those reagents. In summary, we demonstrate that E2, OP, and BPA regulate CAP expression in a similar manner in the immature rat uterus, however, the effects on contractile activity were modulated differently. These findings suggest that OP and BPA interfere with uterine contractility.

        Rotational viscosity calculation method for liquid crystal mixture using molecular dynamics

        This paper presents the directly obtained rotational viscosity values of E7, which includes pentylcyanobiphenol, heptylcyanobiphenol, 4-cyano-4'-n-octyloxy-1,1'-biphenyl, and 4-cyano-4"-n-pentyl-1,1',1"-terphenyl, at various tempe using molecular dynamics computer simulation. The director mean squared displacement was achieved from the squared displacement of the mean director using the concept of the mean director of various nematic liquid crystals. The calculated values were compared with the experiment results that predicted a good agreement. Additional points that must be considered for further study are also discussed.

        Conjugated Linoleic Acid Reduction of Vascular Endothelial Growth Factor Expression in Murine Mammary Tumor Cells through Alteration of Prostaglandin E₂

        Conjugated linoleic acid (CLA) is a group of positional and geometric isomers of linoleic acid that have been used to reduce the incidence, growth and metastasis of breast, colon, prostate and gastric cancer in animals. CLA could reduce tumor growth by altering angiogenesis; a process requiring associated angiogenic factors such as vascular endothelial growth factor (VEGF). In this study, we determined whether CLA could modulate the expression of VEGF in murine mammary tumor cells and adipocytes. The c9, t11-CLA isomer reduced VEGF transcripts and protein when mammary tumor cells were stimulated with PMA. That isomer also reduced VEGF expression in unstimulated mouse 3T3-L1 adipocytes. Since VEGF can be regulated by cyclooxygenase-2 (COX-2), we determined whether CLA could alter COX-2 enzyme expression and PGE₂ production. The c9, t11-CLA isomer reduced not only COX-2 enzyme expression but also PGE₂ production. Thus, c9, t11-CLA could modulate neovascularization by alteration of VEGF expression from mammary tumor cells and adipocytes by reducing COX-2 metabolites.

      • Characterization of Stearoyl-CoA Desaturases from a Psychrophilic Antarctic Copepod, Tigriopus kingsejongensis

        <P>Stearoyl-CoA desaturase is a key regulator in fatty acid metabolism that catalyzes the desaturation of stearic acid to oleic acid and controls the intracellular levels of monounsaturated fatty acids (MUFAs). Two stearoyl-CoA desaturases (SCD, Delta 9 desaturases) genes were identified in an Antarctic copepod, Tigriopus kingsejongensis, that was collected in a tidal pool near the King Sejong Station, King George Island, Antarctica. Full-length complementary DNA (cDNA) sequences of two T. kingsejongensis SCDs (TkSCDs) were obtained from next-generation sequencing and isolated by reverse transcription PCR. DNA sequence lengths of the open reading frames of TkSCD-1 and TkSCD-2 were determined to be 1110 and 681 bp, respectively. The molecular weights deduced from the corresponding genes were estimated to be 43.1 kDa (TkSCD-1) and 26.1 kDa (TkSCD-2). The amino acid sequences were compared with those of fatty acid desaturases and sterol desaturases from various organisms and used to analyze the relationships among TkSCDs. As assessed by heterologous expression of recombinant proteins in Escherichia coli, the enzymatic functions of both stearoyl-CoA desaturases revealed that the amount of C16:1 and C18:1 fatty acids increased by greater than 3-fold after induction with isopropyl beta-d-thiogalactopyranoside. In particular, C18:1 fatty acid production increased greater than 10-fold in E. coli expressing TkSCD-1 and TkSCD-2. The results of this study suggest that both SCD genes from an Antarctic marine copepod encode a functional desaturase that is capable of increasing the amounts of palmitoleic acid and oleic acid in a prokaryotic expression system.</P>
