        Detection and location of bolt group looseness using ultrasonic guided wave

        Bolted joints are commonly used in civil infrastructure and mechanical assembly structures. Monitoring and identifying the connection status of bolts is the frontier problem of structural research. The existing research is mainly on the looseness of a single bolt. This article presents a study of assessing the loosening/tightening health state and identifying the loose bolt by using ultrasonic guided wave in a bolt group joint. A bolt-tightening index was proposed for evaluating the looseness of a bolt connection based on correlation coefficient. The tightening/loosening state of the bolt was simulated by changing the bolt torque. More than 180 different measurement tests for total of six bolts were conducted. The results showed that with the bolt torque increases, value of the proposed bolt-tightening index increases. The proposed bolt-tightening index trend was very well reproduced by an analytical expression using a function of the torque applied with an overall percentage error lower than 5%. The developed damage index based on the proposed bolt-tightening index can also be applied to locate the loosest bolt in a bolt group joint. To verify the effectiveness of the proposed method, a bolt group joint experiment with different positions of bolt looseness was performed. Experimental results show that the proposed approach is effective to detect and locate bolt looseness and has a good prospect of finding applications in real-time structural monitoring.

        Robust Fault Diagnosis Observer Design for Uncertain Switched Systems

        In order to improve the fault diagnosis theory of switched systems, observer-based method is proposed to deal with the fault diagnosis issue subjected to the parameter uncertainty, unknown process disturbances and unexpected faults. By employing proportional-integral observer (PIO) technique and the average dwell-time (ADT) method, an H∞/H− performance level is considered to ensure the worst case of robustness to the disturbance and sensitivity to the fault simultaneously. Moreover, the obtained fault diagnosis observer can give accurate fault estimation online for the piecewise constant fault. Finally, a two-tank system example is introduced in the end to show the effectiveness of the proposed results.

        Time-Dependent Reliability Analysis of Oil Derrick Structures in Mechanism Failure Mode

        Most research studies on the reliability of oil derrick structures have focused on static reliability analysis. Strength degradation with time of oil derrick structures and mechanism failure should also be considered in reliability analysis. In order to address this issue, we propose a new time-dependent reliability analysis method for oil derrick structures in mechanism failure mode and apply it to a tower derrick structure. We used the Monte Carlo fi nite element method to compute the static failure probability of each derrick component, and steps were employed to determine the main failure mechanisms. The time-dependent failure probabilities of each derrick component in the main failure mechanisms were calculated by the total probability theorem. The time-dependent reliability of a derrick structure system was obtained by considering the system as a series of parallel subsystems. A tower derrick example was used to demonstrate the accuracy and applicability of the proposed method. The results revealed that the proposed method is accurate and reliable.

        The quorum sensing regulator OpaR is a repressor of polar flagellum genes in Vibrio parahaemolyticus

        Vibrio parahaemolyticus possesses two types of flagella: asingle polar flagellum (Pof) for swimming and the peritrichouslateral flagella (Laf) for swarming. Expression of Lafgenes has previously been reported to be regulated by the quorumsensing (QS) regulators AphA and OpaR. In the presentstudy, we showed that OpaR, the QS regulator at high cell density(HCD), acted as a negative regulator of swimming motilityand the transcription of Pof genes in V. parahaemolyticus. OpaR bound to the promoter-proximal DNA regionsof flgAMN, flgMN, and flgBCDEFGHIJ within the Pof geneloci to repress their transcription, whereas it negatively regulatesthe transcription of flgKL-flaC in an indirect manner. Thus, this work investigated how QS regulated the swimmingmotility via direct action of its master regulator OpaR onthe transcription of Pof genes in V. parahaemolyticus.

        The Role of Neutrophil-to-Lymphocyte Ratio in Predicting Pathological Response for Resectable Non–Small Cell Lung Cancer Treated with Neoadjuvant Chemotherapy Combined with PD-1 Checkpoint Inhibitors

        Purpose The aim of our study was to investigate the value of baseline and preoperative neutrophil-to-lymphocyte ratio (NLR) in predicting the pathological response and disease-free survival (DFS) of neoadjuvant chemotherapy alone or combined with programmed cell death-1 (PD-1) checkpoint inhibitors in patients with resectable non–small cell lung cancer (NSCLC). Materials and Methods Resectable NSCLC patients who underwent neoadjuvant chemotherapy alone or combined with PD-1 checkpoint inhibitors between January 2018 and January 2020 were included. Peripheral venous blood samples of the patients were collected within 3 days prior to the first neoadjuvant treatment and within 3 days prior to surgery. Results A total of 79 patients in neoadjuvant chemotherapy combined with PD-1 checkpoint inhibitors group and 89 patients in neoadjuvant chemotherapy alone group were included. Thirty-five point four percent of the patients achieved pathological complete response (pCR) in neoadjuvant chemotherapy combined with PD-1 checkpoint inhibitors group, whereas only 9.0% reached pCR in the group of neoadjuvant chemotherapy. High NLR level were correlated with poor pathological response and DFS in neoadjuvant chemotherapy or combined with PD-1 checkpoint inhibitors group. Multivariate analysis revealed that baseline NLR could independently predict pathological response and DFS in the neoadjuvant chemotherapy combined with PD-1 checkpoint inhibitors group. Conclusion High NLR level were correlated with poor pathological response and shorter DFS in patients with NSCLC undergoing neoadjuvant chemotherapy or combined with PD-1 checkpoint inhibitors. Meanwhile, baseline NLR could independently predict response to pathological response and DFS, revealing its potential as a screening tool in NSCLC patients who received neoadjuvant chemotherapy combined with PD-1 checkpoint inhibitors.
