To report retinal image of in a 6-year-old male castrated poodle dog with a 2-month history of nyctalopia using optical coherence tomography (OCT). Ocular reflexes were present in both eyes and slit lamp examination showed anterior subscapular cataract in the right eye. There were fundus abnormalities in both eyes similar to retinal degeneration. Scotopic electroretinograms (ERGs) revealed significantly subabnormal amplitudes and prolonged implicit time, whereas photopic ERGs were better maintained, although far from normal. OCT of affected dogs revealed generalized retinal thinning much more than a normal age-matched dog. Therefore, OCT scanning is considered to be a useful method for retinal evaluation in dogs with retinal degeneration.

<P><B>Background and Purpose—</B></P><P>We investigated the relationship between the degree of thrombus resolution and the time from stroke onset or thrombus formation to intravenous tissue-type plasminogen activator (tPA) treatment.</P><P><B>Methods—</B></P><P>In patients with stroke, we measured thrombus volume on thin-section noncontrast brain computed tomographic scans taken at baseline and 1 hour after tPA administration. We determined the association between the time from symptom onset to tPA treatment and the degree of thrombus resolution. In a C57/BL6 mouse model of FeCl<SUB>3</SUB>-induced carotid artery thrombosis, we investigated the effect of tPA administered at different time intervals after thrombus formation, using Doppler-based blood flow measurement.</P><P><B>Results—</B></P><P>Of 249 patients enrolled, 171 showed thrombus on baseline computed tomography. Thrombus was resolved by ≥50% in 43 patients (25.1%, good volume reduction) and by <50% in 94 patients (55.0%, moderate volume reduction) 1 hour after tPA treatment. In 34 patients (19.9%, nonvolume reduction; either no change or thrombus volume increased), overall thrombus volume increased. The probability of thrombus resolution decreased as the time interval from symptom onset to treatment increased. On multivariate analysis, good volume reduction was independently related with shorter time intervals from symptom onset to tPA treatment (odds ratio, 0.986 per minute saved; 95% confidence interval, 0.974–0.999). In the mouse model, as the interval between thrombus formation and tPA treatment increased, the initiation of recanalization was delayed (<I>P</I>=0.006) and the frequency of final recanalization decreased (<I>P</I> for trends=0.006).</P><P><B>Conclusions—</B></P><P>Early administration of tPA after stroke onset is associated with better thrombus resolution.</P>

Karyopherin-α3 (KPNA3), a karyopherin- α isoform, is intimatelyassociated with metastatic progression via epithelial-mesenchymaltransition (EMT). However, the molecular mechanismunderlying how KPNA3 acts as an EMT inducer remains to beelucidated. In this report, we identified that KPNA3 was significantlyupregulated in cancer cells, particularly in triple-negativebreast cancer, and its knockdown resulted in the suppressionof cell proliferation and metastasis. The comprehensivetranscriptome analysis from KPNA3 knockdown cells indicatedthat KPNA3 is involved in the regulation of numerous EMTrelatedgenes, including the downregulation of GATA3 andE-cadherin and the up-regulation of HAS2. Moreover, it wasfound that KPNA3 EMT-mediated metastasis can be achievedby TGF-β or AKT signaling pathways; this suggests that thenovel independent signaling pathways KPNA3-TGF-β-GATA3-HAS2/E-cadherin and KPNA3-AKT-HAS2/E-cadherin are involvedin the EMT-mediated progress of TNBC MDA-MB-231 cells. These findings provide new insights into the divergent EMTinducibility of KPNA3 according to cell and cancer type.

Bedlington terrier의 다발성 안기형 1례

To report multiple congenital ocular defects in a Bedlington terrier dog aged 2.5 months with blindness. Routine ophthalmic examinations were performed for the clinical signs. Menace responses and cotton ball test were absent in both eyes (OU), but pupillary light reflexes were normal in OU. Slit lamp biomicroscopy reveled corneal dystrophy, posterior subcapsular cataract, microphthalmia in OU and lenticular coloboma in the right eye. In indirect ophthalmoscopy and ultrasonography, retinal detachment and posterior lenticonus were shown in OU. It is the first report of lenticular coloboma and posterior lenticonus in a Bedlington terrier dog.
