        두개골의 3차원 영상 분석을 위한 전산화단층촬영 방법의 비교 : 상층 두께가 3차원 영상의 계측에 미치는 영향

        Purpose : To evaluate the quantitative accuracy of three-dimensional (3D) images by means of comparing distance measurements on the 3D images with direct measurements of dry human skull according to slice thickness and scanning modes. Materials and Mathods : An observer directly measured the distance of 21 line items between 12 orthodontic landmarks on the skull surface using a digital vernier caliper and each was repeated five times. The dry human skull was scanned with a Helical CT with various slice thickness (3, 5, 7 mm) and acquisition modes (Conventional and Helical). The same observer measured corresponding distance of the same items on reconstructed 3D images with the internal program of V-works 4.0 (Cybermed Inc., Seoul, Korea). The quantitative accuracy of distance measurements were statistically evaluated with Wilcoxons’ two-sample test. Results : 11 line items in Conventional 3 mm, 8 in Helical 3mm, 11 in Conventional 5 mm, 10 in Helical 5 mm, 5 in Conventional 7 mm and 9 in Helical 7 mm showed no statistically significant difference. Average difference between direct measurements and measurements on 3D CT images was within 2 mm in 19 line items of Conventional 3 mm, 20 of Helical 3 mm, 15 of Conventional 5 mm, 18 of Helical 5 mm, 11 of Conventional 7mm and 16 of Helical 7 mm. Conclusion : Considering image quality and patient’s exposure time, scanning protocol of Helical 5 mm is recommended for 3D image analysis of the skull in CT.

        3차원 전산화단층촬영 영상을 이용한 얼굴 연조직 두께 계측

        Purpose : To evaluate accuracy and reliability of program to measure facial soft tissue thickness using 3D computed tomographic images by comparing with direct measurement. Materials and Methods : One cadaver was scanned with a Helical CT with 3 mm slice thickness and 3 mm/sec table speed. The acquired data was reconstructed with 1.5 mm reconstruction interval and the images were transferred to a personal computer. The facial soft tissue thickness were measured using a program developed newly in 3D image. For direct measurement, the cadaver was cut with a bone cutter and then a ruler was placed above the cut side, The procedure was followed by taking pictures of the facial soft tissues with a high-resolution digital camera. Then the measurements were done in the photographic images and repeated for ten times. A repeated measure analysis of variance was adopted to compare and analyze the measurements resulting from the two different methods. Comparison according to the areas was analyzed by Mann-Whitney test. Results : There were no statistically significant differences between the direct measurements and those using the 3D images (p>0.05). There were statistical differences in the measurements on 17 points but all the points except 2 points showed a mean difference of 0.5 mm or less. Conclusion : The developed software program to measure the facial soft tissue thickness using 3D images was so accurate that it allows to measure facial soft tissues thickness more easily in forensic science and anthropology.       • 고속핵에서 GABA가 동맥압에 미치는 영향

        목적 : NTS (nucleus tractus solitarius)는 압감수성 반사 구심성 신경이 처음 시냅스를 형성하는 곳으로 동맥압 조절에 중요한 역할을 한다. NTS에서 어떤 특정 물질이 동맥압 조절에 관여하는지는 정확히 밝혀지지는 않았지만, NTS에 존재하는 많은 물질들이 이들 과정에 관여하고 있다. 그중 하나인 GABA성 약물이 NTS에서 순환계에 미치는 영향을 구명하고자 본 실험을 시행하였으며 아울러 반대편 NTS를 lidocaine으로 억제하였을 경우의 변화를 관찰하여 NTS에서 신경회로 일단을 밝히고자 하였다. 대상 및 방법 : 실험동물은 체중 300-500g 되는 숫컷 Wistar쥐를 사용하였다. 동맥압 측정과 약물 주입을 위하여 대퇴 동정맥에 삽관하였다. 이어서 기관지절개술을 시행하였으며 체온을 36-37℃ 되게 열판을 이용하여 조절하였다. 쥐를 뇌 정위 고정 장치에 두고 머리를 45°숙여 NTS를 노출시킨 다음 약물을 10초간에 걸쳐 80nl주입하였다. 결과 : 인공 뇌척수액을 주입하면 동맥압의 변화가 거의 없었으나 동량의 GABA를 NTS에 주입하면 동맥압이 증가하였다. GABA에 의해 동맥압이 증가된 상태에서 반대측 NTS에 lidocaine을 주입하여 한쪽 기능을 억제시키면 동맥압이 더욱 증가하였다. 또한 lidocaine주입 후 반대쪽의 건전한 NTS에 GABA를 재차 주입하면 동맥압 증가가 적었다. 그리고 GABA를 NTS에 주입 후 정맥으로 nitroprusside를 주입하면 GABA 주입이 없는 대조군에 비해 동맥압 감소가 더 컸으며, lidocaine으로 마취하여 한쪽 NTS의 기능을 차단한 후 반대쪽 건전한 NTS에 GABA를 주입한 경우는 동맥압 감소가 적었다. 동일한 방법으로 lidocaine을 주입한 후 phenylephrine으로 승압을 시키면 동맥압 증가가 적었다. 압감수성 반사는 NTS에 GABA를 주입하거나 또는 반대편 NTS를 lidocaine으로 마취하여 기능을 억제시킨 경우에 둔화되었다. 결론: NTS에서 GABA가 동맥압의 조절 인자로 작용하며, 양쪽 NTS는 상호작용하고 있는 것으로 보인다. The nucleus tractus solitarius (NTS), the site of termination within the brain of baroreceptor afferent fibers, has been shown to be involved in the regulation of arterial pressure (AP). Many neuroactive substances present within the NTS have been shown to be involved in this process, although the manner in which any specific neurotransmitter acts physiologically in the regulation of AP has not been completely elucidated. The present study was intended to clarify the mechanism of action of GABA in the NTS and also to examine the cardiovascular response to inhibition of the contralateral NTS by lidocaine. Male Wistar rats weighing 300-500 g were employed for the experiment. The femoral artery and vein were cannulated to record the arterial pressure (AP) and to inject nitroprusside or phenylephrine, respectively. The tracheotomy was performed and body temperature was kept to 36-37℃. Rats were mounted in a stereotaxic frame with the head flexed downward to an angle of 45°. After the NTS was exposed. GABA or lidocaine was microinjected into the NTS in a volume of 80 nl over 10 seconds through a glass micropipette (outer diameter of the tip 50 ㎛) connected to a 1 ㎕ Hamilton microsyringe. The results obtained are summarized as follows. Microinjection of artificial CSF into the ipsilateral NTS did not alter basal AP. In contrast, microinjection of the same volume of GABA into the NTS produced a significant increase in the AP. The results presented thus far indicated that the volume injected did not affect AP. In animals with elevated AP by GABA injection, injection-of lidocaine into the contralateral NTS resulted in a further increase in AP. After the contralateral NTS was inhibited with lidocaine. injection of GABA into the NTS produced a less increase in AP. After microinjection of GABA into the NTS, the nitroprusside administered i.v. accentuated the increase of AP but not after microinjection of lidocaine into the contralateral NTS. Similar results were obtained with i.v. injection of phenylephrine after pretreatment of lidocaine into the NTS. The baroreflex sensitivity was reduced by microinjection of GABA into the NTS or microinjection of lidocaine into the contralateral NTS. These results suggest that alterations in GABA-mediated neural transmission in the NTS contribute to the regulation of AP and the NTS of both sides have a mutual interaction.

      • 회전원판 반응조를 이용한 Pseudomonas sp. KS-96에 의한 gallic acid로부터 pyrogallol의 전환

        In previous paper Pseudomonas sp. KS-96 isolated from the soil to bioconversion into pyrogallol from gallic acid. Continuous bioconversion of pyrogallol was carried out using rotatory disc contactor immobilized Pseudomonas sp. KS-96. Enzyme activity of gallate decarboxylase released from Pseudomonas sp. KS-96 were shown at the highest activity on 24h incubation. Culture media containing gallic acid supplied on the flow rate of 20㎖/h until thickness of cells wall reached steady state. Bioconversion rate of pyrogallol from gallic acid showed at highest level ranging from 18hr to 36h according to time courses. Continuous bioconversion of pyrogallol using rotating disc contactor was about 81% and 80% between 6 and 8 days at the feeding rate of 30㎖ per hour in the medium containing 15g/ℓ gallic acid.

        너도밤나무 크레오소트가 흰 쥐의 장내 미생물 변화에 미치는 영향

        설사는 경제동물의 이유시기 폐사율을 일으키는 가장 높은 요인이다. 크레오소트는 전통 의약품으로 오랜 세월동안 지사제로 사용되어 왔다. 본 연구는 흰 쥐 모델에서 크레오소트 급여가 동물의 성장 효율 및 장내 미생물에 미치는 영향을 구명할 목적으로 수행되었다. 4주령의 수컷 흰쥐 24마리를 임의로 대조구, 항생제 그룹, 크레오소트 0.4% 그룹, 크레오소트 0.8% 그룹으로 배치하였다. 대조구는 기초사료, 항생제 그룹은 apramycin 0.5%, 크레오소트 그룹은 크레오소트 0.4%와 0.8% 수준으로 하여 예비시험 기간 1주일, 본시험 기간 4주일 동안 급여하였다. 일당증체량은 실험구간 차이가 없었으나, 사료 섭취량은 Creo 0.8 그룹에서 유의적으로 증가하였다(p<0.05). 장내 미생물에 대한 문(phylum) 수준 분석 결과 Creo 0.8 그룹에서 Firmicutes가 감소하고 Bacteroidetes가 증가하여 F/B 비율을 감소시키는 것으로 나타났다(p<0.05). 과(family) 수준에서는 Lachnospiraceae가 크레오소트 0.8% 수준에서 증가되었으며(p<0.01), 속(genus) 수준에서는 Turicibacter가 감소되었다(p<0.01). 종(species) 수준에서는 Clostridium disporicum이 감소되었다(p<0.01). 이상의 결과는 쥐에 크레오소트 급여는 사료 섭취량을 증가시키고 장내 미생물의 변화에 영향을 미칠 수 있다는 것을 시사한다. Diarrhea is one of the main disorders which cause the highest level mortality of the post-weaning economic animal. Beech-wood creosote has been used as a traditional anti-diarrheic medicament for a long time. The present study was conducted to investigate the effects of dietary supplementation of Beech-wood on growth performance and intestinal microbiota in rats. Twelve 4-week-old rats were randomly assigned to one of four dietary groups and fed a basal diet supplemented with none (CON), 0.5% apramycin (ANTI), 0.4% creosote (Creo 0.4), or 0.8% creosote (Creo 0.8) for 4 weeks following 1 week of adaptation period to the respective diet. Average daily gain was not influenced by the dietary treatment whereas average daily feed intake was greatest for the Creo 0.8 group. In the intestinal microbiota at the level of the phylum, the percentage of Firmicutes bacteria decreased but Bacteroidetes increased in the Creo 0.8 group vs. Control, which resulted in a decreased F/B ratio for the former (p<0.05). Moreover, the percentage of Lachnospiraceae was greater at the level of the family for the Creo 0.8 group than for Control, but the percentages of Turicibacter and Clostridium disporicum were less in the former (p<0.01) at the genus and species levels, respectively. Collectively, the present results indicate that dietary supplementation of creosote increases the feed intake and also influence the intestinal microbiota in rats.

      • Interdisciplinary research: Can plasma technology be applied in the animal developmental biotechnology?

        This study firstly investigated whether plasma treatment of fertilized eggs before hatching could affect the growth and reproduction of chickens. Four-day-incubated fertilized eggs exposed to non-thermal dielectric barrier discharge plasma at 11.7 kV for 2 min resulted in the highest growth in chickens. Plasma growth-promoting effect in chickens was regulated by the reactive oxygen species homeostasis and the improvement of energy metabolism via increasing serum hormones and adenosine triphosphate levels which were resulted from the regulation of the demethylation levels of growth and hormone biosynthesis-related genes. Interestingly, plasma-treated male chickens showed more conspicuous growth characteristics than females. Further, aspects of the male reproductive system were improved by the plasma treatment: testosterone levels, sperm quality and fertility increased via the double regulation of DNA methylation and miRNA in the testis. In summary, optimized plasma treatment of fertilized eggs can improve chicken growth and male reproductive capacity; this finding can improve profitability and next-generation potential production in poultry breeding.

        Characterization and In Vitro Differentiation of Korean Ring-Necked Pheasant (Phasianus colchicus) Male Germ Cells

        Phasianus colchicus is not only a beautiful bird but also a great value in science and under the threat of endanger. Hence, the aim of this study was to isolate the pheasant male germ cells (mGCs) and then induce them into elongated sperm-like cells in vitro. The mGCs were purified and enriched by a two-step plating method based on the different adherence velocities of mGCs and somatic cells. The percentage of the c-kit positive cells and c-kit negative cells examined by flow cytometry analysis (FCA) was 92.87% and 2.57%, respectively. Subsequently, the mGCs were induced for 48h in DMEM/F12 medium supplemented factors such as retinol acid, testosterone and bovine FSH, followed by 5 weeks in culture. We found that some elongated sperm-like cells appeared initially in vitro under inducement of stimulated factors. The elongated sperm-like cells showed in the expression of changed morphology and post-transcriptional marker such as spermatid associated (SPERT), spermatid perinuclear RNA binding protein (STRBP), round spermatid basic protein 1 (RSBN1) and SPER1L. Moreover, in DNA content identified assay, induced cells showed that the 1C DNA population markedly increased in differentiated group but it was not change in undifferentiated group. Successful in vitro differentiation of pheasant testicular germline cells into spermatids appears to offer extremely attractive potential for the conservation of endangered birds and treatment of male infertility.

      • Optimization of Techniques for Isolation and Identification of Bovine Hematopoietic Stem Cells from Peripheral Blood

        Hematopoietic stem cells (HSCs) are the self-renewing, multipotent progenitors that give rise to all types of mature blood cells. The hallmark properties of HSCs are the ability to balance self-renewal versus differentiation cell fate decisions to provide sufficient primitive cells to sustain haematopoiesis, while generating more mature cells with specialized capacities. In the present experiment, we optimized the techniques for isolation and identification of hematopoietic stem cells from cow peripheral blood. The objective of this study was to optimize the more accurate methodology for separation of mononuclear cells (MNCs) from peripheral blood and identification of HSCs by using a specific cell surface marker i.e. CD34. A total 10 peripheral blood samples were collected from Holstein dairy cows from jugular vein. We used Ficoll 400 in different concentrations from 1 to 12% and Ficollpaque Plus (1.077 g/ml) at different centrifugation speed and time. After Giemsa staining, we found more than 98% recovery of monocytes with Ficollpaque Plus (1.077 g/ml). It was demonstrated that Ficollpaque Plus (1.077 g/ml) and centrifugation at 400xg for 30 min is the best method for separation of MNCs from bovine peripheral blood. Separated MNCs were immediately subjected for magnetic activated cell sorting (MACS) by using CD34 microbead kit. HSCs (CD34+ cells) recovery was 0.307% of peripheral blood. Peripheral blood MNCs and CD34+ cells were morphologically characterized by Giemsa staining. CD34+ cells were also confirmed by immunochemistry using FITC conjugated CD34 antibodies. HSCs were also confirmed by progenitor assay including burst forming unit-erythroid (BFU-E), colony forming cells- granulocyte (CFC-G), colony forming cells- macrophage (CFC-M), colony forming cells- granulocyte macrophage (CFU-GM) and colony forming cells- granulocyte erythroid macrophage monocyte (CFCGEMM) on Methocult 4435.
