      • 12-O-Tetradecanoylphorbol-13-Acetate에 의한 HL-60 세포 분화유도중 Vimentin 유전자 전사조절에 대한 AP-1의 역할

        Purpose: To gain insight on the role of AP-1 in transcriptional regulation of vimentin gene during differentiation of HL-60 cells by 12-O-tetradecanoylphorbol-13-acetate (TPA), the levels of vimentin mRNA and AP-1 have been investigated with Northern blot hybridization and DNA mobility shift assay. Materials and Methods: HL-60 cells were grown in RPMI 1640 medium supplemented with 10% heat-inactivated fetal calf serum and antibiotics in a humidified 5% CO_(2) at 37°C. Total RNA was prepared by a modification of the method of Karlinsey et al. Northern blot hybridization was performed by the method of Virca et al. EcoRI fragment of pVIM-GEM was used as probe for vimentin mRNA. DNA mobility shift assay was performed by the method of Lim et al. End labeled DNA probe(Upper strand, 5'-CGCTTGATGAGTCAGCCG- 3') for AP-1 binding activity was mixed with nuclear extracts in a 20 μl reaction volume containing 300 mM KC1, 60 mM HEPES, pH 7.9, 25 mM MgCl_(2), 1 mM EDTA, 1 mM DTT, 60% glycerol, and 2μg of poly[dI-dC]. Results: TPA increased vimentin mRNA levels, with maximal stimulation reached at 24 hr. The level of vimentin mRNA was induced in proportion to the concentration of TPA. TPA-induced vimentin mRNA was almost reduced by actinomycin-D pretreatment. TPA-induced stimulation of vimentin gene was completely reduced by staurosporin pretreatment. In DNA mobility shift assay, AP-1 newly appeared at 24 hr during TPA-induced differentiation and was almost not detected after the pretreatment of staurosporin. Conclusions: These results suggest that the induction of vimentin mRNA during TPA-dependent differentiation in HL-60 cells may be mediated by protein kinases C signal transduction and AP-1 is important to transcriptional regulation.

        Enhancements of encoding and decoding strategy of a discrete bidirectional associative memory (BAM) proposed B. Kosko are presented. This paper describes multiple training, which allow to achieve recall of single trained pair under suitable initial conditions of data (We fixed Hamming distance). Computer simulations are shown to lead to an improvement in some instances over the original Kosko strategy for recall of multiple pairs.

        위암수술 환자에서의 cp 개발과 cp적용에 따른 질 향상 활동에 관한 연구

        Background : In an era of increasing medical costs, safe reduction in postoperative stay has become a major focus to optimize utilization of healthcare resources. The authors aimed to evaluate the clinical significance of the critical pathway(CP) for gastrectomy patients by implementing standardized postoperative management and electronic medical records. Method : From August 2006 to April 2007, critical pathways were introduced to inpatients and outpatients with gastric cancer. 60 consecutive patients undergoing distal gastrectomy were randomly divided into two groups; 30 CP group(A) and 30 non-CP group(B). Simultaneously, we also retrospectively reviewed the records of 438 patients(C:control group) who were able to be enrolled in CP program. We compared group(A) with (B), (C) in terms of hospital stay, complication rate, use of antibiotics and hospital costs. Patient satisfaction was surveyed by questionnaires. Result : There was no significant demographic difference between group(A) and (B). Of 30 patients in group(A), 5 drop-outs and 14 variances occurred. The mean postoperative hospital stays(Laparoscopy-assisted distal gastrectomy: LADG/Open distal gastrectomy: ODG) were 7.6/10.5 days for the group(A), 8.9/12.7 days for the group(B) and 12.6/14.5 days for the group(C) (p=0.02 in LADG). The total hospital cost of group(A) had a tendency to be less than that of group(B) and the amount of hospital cost per day in group(A) was significantly higher than that in group(B). The usage rates of the first generation antibiotics were 73.33%, 63.33%, and 59.2% in group(A), (B), and (C), respectively. Patients’ satisfaction had a tendency to be higher in the CP group than non-CP group. Conclusion : The mean hospital stay in LADG patients was shortened meaningfully and hospital cost per day was enlarged after introduction of clinical pathway. It is necessary to apply a number of patients to the critical pathway and to make an effort to reduce the variation and drop-out rates.

      • 한국의 전자무역 활성화 방안에 관한 연구 : 한국과 주요국의 전자무역 비교 · 분석

        The activation of e-Trade emerge from a major policy issue in many counties. Especially, our country will be adapted and used new trade method quickly then to be application is an important issue of trade policy because our country highly depend on trade rather than other country. Taking a comparison of the progress of e-trade with the major competitor-countries targeting the leading market presence and strategic gateways in e-trade, Korea can be diagnosed as more government-led than the USA featured by the distinctive private sector initiatives, and as more independent than the EU. Compared with Singapore, which is a main competitor in terms of the strategic gateway for e-trade in East Asis region, Korea lacks in its efforts to attract the business. Thus, this dissertation aims to present several methods for more active e-trade in Korea by examining the implications and comparative insights drawn from the e-trade states in major countries. Considering general pump priming policies of the government and the private sectors, the first step to take is to keep up investment in long-term rather than for short-term returns, and to make Korean voices heard and reflected in the decision-making processes for international standards by actively participating in the activities of international organizations and conventions. To sum up, this dissertation is an attempt to build a theoretical model drawn from a comparative investigation of states and progresses of many countries upon undertaking e-trade promotion. However, since the research materials for this dissertation are limited to the existing research papers and press releases of related organizations without analyzing actual conditions, this dissertation has some restrictions in it.

        A strengthening technique for reinforced concrete beams using external unbonded reinforcement offers advantages in speed and simplicity of installation over other, established, strengthening techniques. The purpose of this paper is to investigate the capabilities of a new retrofitting technique, namely external prestressing(Bow), for flexural strengthening of beams. The paper provides a general description of structural behavior of beams strengthened using the technique. Results of 2 physical tests on strengthened reinforced concrete continuous beams are reported and compared. It is shown that the technique can provide greater strength enhancement to lightly reinforced sections and that provision of deflectors enhances efficiency.

        인공 석회화 기질의 개발 및 파골세포에 의한 기질흡수에 관한 연구

        To develop artificial matrix resorbable by osteoclasts can be used as a model system that biological function of osteoclasts would be assayed in vitro in a standardized condition instead of using heterogenous bone or dentin slices. The present study developed artificial substrates consisted of apatite crystals. collagen gen. agar, apatite crystals under collagen film, mixture of apatite crystals and collagen, and mixture of agar and apatite crystals. And osteoclastic attachement and resorption activity on the each substrate was assayed in vitro. Osteoclasts attached to apatite crystals, collagen, mixture of apatite crystals and collagen, and mixture of apatite crystals and agar, however, not to agar. This suggested that apatite crystals and collagen should be exposed on the surface of artificial substrate where osteoclasts can attach. Osteoclasts attached to the substrate resorbed the substrate consisted of apatite crystals, mixture of apatite crysals and collagen, and apatite crystals under thin film of collagen, however, not of collagen, mixture of apatite crystals and agar. This suggested that the first phase of osteoclasts in resorbing matrix is to secrete acid and the end result of this phase, whatever it may be, may activate secretion of hydrolytic enzymes specific to organic components.

        저칼슘식이로 사육한 생쥐의 골수세포 배양에 의한 파골세포양세포의 형성

        Although it is now well established that osteoclasts are derived from mononuclear hematopoietic stem cells, most likely within the granulocyte-macrophage lineage, the exact differentiation process of osteoclast precusors has only been partially discovered. This study was carried out to investigate the following topics by examining the osteoclast-like cell formation from the calcium-deficient mouse bone marrow cells; 1) the effect of calcium deficiency on the formation of the mononuclear phagocytes and the mononuclear precusors of the osteoclast in the mouse bone marrow; 2) the effect of TNF (tumor necrosis factor)-α on the differentiation of osteoclast-like cells from the calcium-deficient mouse bone marrow culture; 3) the formation of the osteoclast-like cell from the calcium-deficient mouse bone marrow mononuclear cells and the relationship between the osteoblast cell line(MC3T3E1) and osteoclast-like cell formation; 4) formation of the osteoclast-like cells from the mononuclear cells which were col1ected from PTH-pretreated mouse bone marrow cells to investigate the relationship between PTH and calcium deficiency in osteoclast-like cell formation. Bone marrow cells were isolated from marrow of mouse feeding normal diet or calcium deficient diet for 7 and 14 days. Bone marrow mononuclear cells were fractionated by cetrifugation on Hypaque-Ficoli density gradients. PTH-pretreated mouse bone marrow mononuclear cells were obtained by collecting the nonadherent cells after the culture of normal mouse marrow cells with PTH for 6 days. The prepared cells were cultured on dentin disc at the concentration of 1.5-2 X 10^(6) cells per 0.5ml of a-MEM containing 10% FCS for 3, 5 and 7 days. Thereafter, TRAP staining, NSE staining, effect of calcitonin on the osteoclast-like cell and scanning electron microscope analysis of resorption lacunae on the dentin disc were performed. The results were as follows; 1. Number of the osteoclast precusors and mononuclear phagocytes in bone marrow was increased by calcium-deficient diet. 2. More osteoclast-like cells were formed from the calcium-deficient mouse bone marrow cells than from normal cells. 3. Formation of osteoclast-like cells was significantly stimulated by TNF-α in normal mouse bone marrow cells and calcium-deficient mouse bone marrow cells at concentration of l0ng/ml and 100ng/ml. 4. Osteoblast cell line(MC3T3E1) did not play an important role in osteoclast-like cell formation. 5. Formation of the osteoclast-like cells from the mononuclear cells which were collected from PTH-pretreated mouse bone marrow cells was similar to that of calcium-deficient mouse bone marrow mononuclear cells, which suggests that the effect of calcium deficiency in osteoclast formation may be mediated by PTH.

      • Experimental excitation and characterization of cladding modes in photonic crystal fiber

        <P>We extend the previous theoretical study on the effect of outer silica cladding to the analysis on real field profiles in a practical PCF. Clear field profiles for the first higher-order cladding modes are presented and discussed. The observed mode fields are reproduced by numerical calculations, and it turns out that they correspond to LP(16) mode groups. Optical properties of the observed modes such as lobe direction and polarization are also investigated. The results of this study will be useful in the design of the PCF-based optical devices utilizing cladding mode coupling.</P>
