      • 프로테오믹스 방법을 이용한 인간 SOD1 단백질과 상호작용하는 효모 단백질들의 탐색

        Amyotrophic lateral sclerosis (ALS) is a neurologic disease that rapidly progresses and causes death within 5 years of the onset of symptoms. Mutations of SOD1 gene have been linked to familial cases of ALS (fALS). Although many hypotheses have been suggested, the role of SOD1 in ALS pathogenesis still remains unclear. A hallmark for ALS is the presence of intracellular inclusions containing mutant SOD1 and ubiquitin, suggesting that cells attempt to eliminate SOD1 proteins by ubiquitin-proteasome system. Possibly the neuronal cells' failure to promptly eliminate unfolded SOD1 proteins leads to the protein accumulation and the eventual neuronal death. To study the role of ubiquitin-proteasome system in the degradation of mutant SOD1 proteins, we used yeast Saccharomyces cerevisiae as a model system. When being expressed in yeast, G93A mutant of hSOD1 was degraded rapidly, while wild-type SOD1 was stable. Most of SOD1 expressed in yeast, unlike in mammalian cells, remained soluble. To identify proteins interacting with SOD1 and investigate their roles in the SOD1 degradation, we immunoprecipitated SOD1 and analyzed the binding partners by 2-DE followed by MALDI-TOF mass spectrometry.         재조합 균주 Bacillus subtilis LKS88에 의한 Streptomyces albus KSM-35 Amylase의 생산조건

        Streptomyces albus KSM-35의 amylase의 유전자를 Bacillus subtilis LKS88에서 발현시킨 균주를 사용하여 amylase의 생산에 미치는 몇가지 요인들을 조사하였다. 본 실험에 사용한 탄소원들중에서 sodium citrate와 왕겨의 효소 생산량이 가장 높았고, 이들을 1:1의 비율로 혼합하여 첨가하였을 때 효소의 생산량은 대조군으로 사용한 가용성 전분에 비하여 20배 이상 증가하였다. 질소원으로 대두박을 사용하였을 경우 yeast extract의 첨가량을 줄이면서 효소의 생산량을 증가시켰다. 배양을 위한 초기 pH를 60으로 조절하고 SDS를 0.01%첨가하였을 때 대조구조보다 높은 효소 생산량을 나타내었다. Bacillus subtilis LKS88 (pASA240)을 sodium citrate와 왕겨 각각 1.5%, K_2HPO_4 0.66%, yeast extract 0.3%, 대두박 0.7%, SDS 0.01% 및 초기 pH를 6.0으로 조절한 배지에서 37℃에서 배양시 최대 효소 활성은 배양 후 36시간에서 나타났으며 이때 효소 활성은 56.6 U/ml이었다. The effects of culture conditions on the production of amylase expressed by Bacillus subtilis LKS88 with a cloned gene from Streptomyces albus KSM-35 were investigated The production of amylase was increased significantly by using sodium citrate and rice hull as a carbon source. In addition, the use of a mixture of sodium citrate and rice hull(1:1) resulted in increase of enzyme production by 20-fold when compared to that of soluble starch. The soybean meal as the nitrogen source could be partially replaced with yeast extract without changing the enzyme production yield. The amylase production was also increased by adjusting initial pH to 6.0 or by adding 0.01% SDS. Maximum amylase production was observed in the medium containing 1.5% sodium citrate, 1.5% rice hull, 0.7% soybean meal, 0.3% yeast extract, 0.66% K_2HPO_4, 0.05% MgSO_4·7H_2O, 0.08% CaCl_2·2H_2O, 0.01% SDS with initial pH of 6.0. The maximum yield of amylase reached 56.6 U/ml when B. subtilis LKS88(pASA 240) was cultured at 37℃ for 36 hr.

      • 도토리澱粉이 흰쥐의 脂質代謝에 미치는 影響

        In an attempt to observe the lipid metabolism of acorn starch, five different diets were made: one rice diet (control group) and four diets in which acorn starch was mixed with rice at the rate of 10%, 20%, 30% and 40% respectively. These five diets controlled in such ways are the same level in calorie and protein content. The acorn starch used was cleared from tannin by being heated 20 hours. These diets were each fed to different groups of male albino rats for 6 weeks, during which various items were measured on these rats such as total lipid, total cholesterol, triglyceride, phospholipid and HDL-cholesterol contents. The results obtained through this study are summarized as follow: The content of total lipid in liver of each group was lower in the 2nd week, and that of 30% acorn starch group was lower in the 4th week, and that of 20% acorn starch group was lower in the 6th week, compared with control group. The content of total cholesterol in liver of each group except 10% acorn starch group was lower than that of the control group in the 2nd week, but in the 4th and the 6th week, there was no particular difference in content between each group and the control group. The centents of total lipid, triglyceride, phospholipid, total cholesterol and HDL-cholesterol in serum are almost the same between each group and the control group during each period of feeding. Therefore acorn starch diets mixed with rice at the rate of 10%, 20%, 30% and 40% had little effect on the serum lipid contents.

        사용자 수술명과 수술분류 code(ICD-9-CM) 일치율 향상에 관한 연구 : 수술실 OCS program 사용 활성화를 통하여

        Background : The necessity of unify and standardize codes used at hospital has been emphasized since OCS (Order Communicating System) was adopted. Therefore, the purpose of this study were to standardize operation code by continuous training of the ICD-9-CM code that is used as standard code in OCS program at operating room. Method : In 400 operation data, operation code entered in OCS program at operating room was compared to operation name recorded in medical record. In addition, a matching rate between input data of operation code by medical record department and computing input data of operation code in 3,710 cases was compared for each department. User operation name and operation code were matched and major diagnosis by operation department and operation name were also matched. Results : User operation name was reflected in operation classification code in detail, and operation code entered on user was registered. Input rate and matching rate of operation code were gradually improved after improvement activity. In particular, a matching rate was high at ophthalmology where operation name is segmented. Plastic surgery and orthopedics with a lot of emergency operation and comprehensive operation name show low input rates. Conclusions : As the medical field makes progress in computerlization, awareness of information exchange and sharing becomes higher. Among codes to classified medical institution, codes related to surgical operation are all different by user of hospital and department. Computerlization and standardization is essential. And when efforts of standardization continue in alliance with individual hospital and institution, initiative of preparing medical policy data at a national level will be accelerated.

      • 근위축성 측삭경화증과 연관된 돌연변이 SOD1과 HAX-1과의 상호작용

        Mutations of SOD1 gene have been linked to certain cases of familial amyotrophic lateral sclerosis (ALS). Despite numerous attempts to elucidate its role in the molecular mechanism, how the mutant SOD1 causes death of motor neurons still remains elusive. A hallmark for ALS is the presence of intracellular inclusions containing mutant SOD1 and ubiquitin suggesting that cells' failure to eliminate aberrant proteins by ubiquitin- proteasome system may be a cause of cytotoxicity. Additional proteins also associate with mutant SOD1, some of which are likely involved in the protein aggregation. Previously we have demonstrated that human SOD1 proteins preferentially interact with molecular chaperones and proteasome subunit in yeast Saccharomyces cerevisiae. To identify human proteins interacting with SOD1, we isolated G93A mutant SOD1 proteins expressed in cultured human cells and analyzed the associating proteins by 2-DE followed by MALDI-TOF mass analysis. As with yeast cells, in human cells mutant SOD1 strongly bound to chaperone proteins, mostly hsp70 and hsc71 proteins. Unexpectedly, we also found that HS-1-associated protein-X-1 (HAX-1), a member of apoptotic regulators, interacts with G93A SOD1. These results suggest that the components of both cellular chaperone and apoptotic machinery are sequestered together with misfolded SOD1 which may eventually lead to the neuronal death.       • 도토리澱粉 添加給食時 消化吸收에 關한 硏究

        The study to observe the digestion and absorption rates of acorn starch. The diets used for this study are four: one rice diet and three other diets which were mixed at the rate of 20% with each of the three acorn starches uniquely processed, i.e., the acorn starch was cleared from tannin by being heated, by being processed in the solution of NaCl;NaHCO₃(5:1 v/v), and by being sunk in the water respectively. These diets were fed to different groups of male and female albino rats for 4 weeks, during which growth rate and digestion and absorption rates were measured. The main results obtained are as follows: 1. Growth rate was highest in Group A in case of the male rats(p<0.05). In case of the female rats Group A and Group C showed similar rates compared with Group S, but Group B showed the somewhat lower rate. 2. The rate of protein absorption was somewhat low in all the groups compared with Group S. 3. As for nitrogen retention rates, Group A and Group B were higher in the 1st and 2nd weeks(p<0.01) and Group C was higher in the 2nd week(p<0.05) compared with Group S in case of the male rats. In case of the female rats Group C and Group A were lower than Group S in the 1st week and 3rd week respectively (p<0.01). In the 4th week Group A and Group B showed the higher amount(p<0.01). 4. The rate of carbohydrate absorption was somewhat lower in all the groups including Group S. 5. The rate of carbohydrate absorption was somewhat lower in all the groups compared with Group S throughout the period of feeding. 6. Calory utilization rates was shown to similar in all the groups throughout the period of feeding. These results indicate that the digestion and absorption rates of all the diets turned out be not so different. Thus it could be concluded that the addition of acorn starch at the rate of 20% does not influence the digestion and absorption of food.

        The effect of varying dietary levels of protein on magnesium, sodium and potassium utilization was examined in 18-month-old male rats. In the experiment, the rats were fed on diets of various protein levels for 4 weeks:Group Ⅰ,Ⅱ and Ⅲ received diets of 8%, 16% and 24% protein respectively throughout the experiment: Group Ⅳ received 3-day diet of 8% protein and 1-day diet of 24% protein by turns throughout the experiment:Group Ⅴ received 3-day diet of 24% protein and 1-day diet of 8% protein by turns throughout the experiment. The concentrations of magnesium, sodium and potassium in each organ were not related to dietary protein. The apparent aborption rates and balance of magnesium, sodium and potassium were not affected by dietary protein levels. Sodium intake per urinary sodium excretion ratio were significantly increased in Group Ⅲ and Ⅴ than those in the other groups. Potassium intake per urinary potassium excretion ratio of Group Ⅰ was increased when compared with the other groups. The urinary magnesium excretion levels were similar to each groups.

        우리나라 일부 석면사업장의 석면폐 유병률

        Asbestos industry has been in operation over 60 years in Korea. However, the prevalence of asbestosis has not been yet reported. With rapid turn-over of workers, previous cross-sectional studies of current workers on the job could not find cases with exposures long enough for the development of asbestosis. This study was done to evaluate asbestosis prevalence of those worksites with operation history of more than 20 years. In total, 139 workers from 5 worksites were examined. Asbestos industries covered in the study include 2 asbestos textile, 1 brake lining, and 2 ship repairing worksites. Chest x-ray was taken from all workers and read by two experts familiar with pneumoconiosis classification according to 1980 ILO guidelines. Those with findings compatible with asbestosis were further ckecked with high resolution computerized tomography (HRCT). Pulmonary function tests were done according to ATS guidelines, and occupational and previous medical history was taken through a standardized interview. Air-borne asbestos was measured according to NIOSH method 7400. The air-borne asbestos concentrations ranged from 0.2-1.3 f/cc for asbestos textile, from 0.7-1.0 f/cc for brake lining, and from 6.3-7.8 f/cc during asbestos removal at ship repairing worksite. Of the 139 workers 25 had abnormal chest radiographic findings, and 10 of them had findings compatible with pneumoconiosis. When work history and current asbestos measurements were accounted, 9 workers who had more than 10 years of asbestos exposure history showed chest radiographic findings of pneumoconiosis with Finally, 4 workers showed finding of pulmonary fibrosis and/or pleural thickening at HRCT, and 2 of them had restrictive lung function changes. The study results showed that, among 139 subjects, there were 4(3%) definite asbestosis cases confirmed with HRCT. The prevalence of probable asbestosis was 7% for 10-14 years of exposure, 13% for 15-19 years of exposure, and 23% for 20 or more years of exposure. The prevalence of compensable asbestosis with abnormal lung function was 4-6% for those with 15 or more years of exposure.

        하의용 시판 신축성 소재의 물리적 특성과 맞음새에 관한 연구 : 스커트를 중심으로 Focused on the Tight Skirt

        This study was investigated physical properties of stretch fabrics by KES-FB system to show suitable basic data to making skirts of excellent capacity and develop more organized basic skirt pattern by fitness evaluation. ₁. The results of T.H.V.(Total Hand Value) were as follows: In the kind of blending fiber, cotton/spandex was more excellent than nylon/spandex and polyester/spandex, in the direction of stretch, one-way(weft inserted polyurethan yarn) polyester/spandex and cotton/spandex were higher than two-way(warp and weft inserted polyurethan yarn) polyester/spandex and cotton/spandex, two-way nylon/spandex was higher than one-way nylon/spandex. ₂. The results of calculating the variance between sample stretch fabrics and Japanese s/s women's suit fabrics after standardizing were as follows: Stretch fabrics has ₂range of tensile, bending, shearing, compression, surface, thickness and weight as compared with Japanese s/s women's suit fabrics. In the tensile property, one-way stretch fabrics were almost the same with Japanese s/s women's suit fabrics, only tow-way polyester/spandex had +₁~+₂range. In t도 bending, shear property, there was no difference between sample fabrics and Japanese s/s women's suit fabrics ₃. In the total fitness of the skirt, nylon/spandex is the best in the fabrics and one-way stretch fabric is better than two-way stretch fabric.

        This paper researched the scale of subjective speech quality measurement over VoIP(Voice over IP) service which is a component of broadband networks. Though objective parameters of multimedia services like PSNR or Jitter can be easily measured and defined. those factors are not easily met the users' perceptual recognition. It is suggested that the scale of speech quality measurement through the subjective measurement is more adequate for end-to-end speech quality which is composed of sender-side quality. transmission quality. receiver-side quality. which provides the degree of correctness in speakers' representation. the degree of impairment caused by various factors. and the degree of recognition processed speech. respectively. It's also examined by the proposed method and verified by it's availability.
