Background/Aims: Data on treatment efficacy and safety of glecaprevir/pibrentasvir (GLE/PIB) for chronic hepatitis C virus (HCV) infection in Asian patients with severe renal impairment are limited. This study aimed to study the treatment and side effects of GLE/PIB in these patients infected with non-1 genotype (GT) HCV. Methods: We prospectively recruited patients with Child’s A cirrhosis and eGFR <30 mL/min/1.73 m2 in Hong Kong and Taiwan during 2017–2018 to receive GLE/PIB treatment. Results: Twenty-one patients (GT2, n=7; GT3, n=6; and GT6, n=8) received GLE/PIB for 11.2±1.8 weeks. All except one were treatment-naïve. GLE/PIB was initiated in 16 patients while on dialysis (seven on peritoneal dialysis [PD] and nine on hemodialysis) and in five patients before dialysis. One patient died of PD-related peritonitis during treatment and two were lost to follow up. The SVR12 rate in the remaining 18 patients was 100%. All patients achieved undetectable levels at 4-, 12-, 24- and 48-week after treatment. Patients with deranged alanine aminotransferase showed normalization after 4 weeks and the response was sustained for 48 weeks. No significant adverse event was observed. Conclusions: GLE/PIB treatment was associated with high efficacy and tolerability in HCV-infected patients with severe renal impairment.

The use of laboratory procedures is often inefficient for materialisation of recombinant therapeutic proteins in Escherichia coli (E. coli) for pre-clinical evaluation. Approaches such as scaling out shake flask cultivation can be laborious, inefficient and expensive. These inefficiencies can be compounded if the protein requires post-translational modification such as multimerisation. We previously used laboratory methods to produce the < 60 kDa, recombinant biotherapeutic, RB1. We were aware, a priori, that dimerisation of RB1 could double the molecular weight of the protein and increase its systemic retention in the human body by avoiding renal filtration. Here we modified RB1 by substituting a native residue for an unpaired cysteine, generating eRB1, in order to favour its dimerisation. Laboratory methods failed to achieve > 20% disulphide-bridged homodimerisation or monomer of sufficient purity to enable chemi-dimerisation. As such we established a set of high performance, bench-scale, unit operations for cultivation of E. coli cells expressing eRB1, the isolation of eRB1 inclusion bodies, refolding and disulphide-based dimerisation of ≥ 40% of total eRB1 and finally successful chemi-dimerisation of remaining monomeric eRB1. The establishment of scalable procedures can now enable future investigations of eRB1 and other < 60 kDa biologics for which significant bench-scale production is required for pre-clinical evaluation.

Vivax malaria is a significant military and civilian health threat in the north of the Republic of Korea (ROK). The island of Baengnyeong-do is the westernmost point of the ROK and is located close to the southwestern coast of the Democratic People’s Republic of Korea (DPRK). Mosquitoes were collected using a black light trap on Baengnyeong-do, and Anopheles spp. were assayed by PCR, to identify the species, and screened for sporozoites of Plasmodium vivax. Of a subsample of 257 mosquitoes, Anopheles lesteri was the most frequently collected (49.8%), followed by Anopheles sinensis (22.6%), Anopheles pullus (18.7%), Anopheles kleini (7.8%), and Anopheles belenrae (1.2%). The overall sporozoite rate was 3.1%, with the highest rates observed in An. kleini (15.0%), An. sinensis (5.2%), and An. lesteri (1.6%). No sporozoite positive An. pullus or An. belenrae were observed. The results extend our knowledge of the distribution and potential role in malaria transmission of An. kleini, An. lesteri, and An. sinensis, for an area previously considered to be at a low risk for contracting vivax malaria.
The Asenze Cohort Study in KwaZulu-Natal, South Africa: Protocol and Cohort Profile

The Asenze cohort is set in South Africa, a middle-income country impacted by one of the highest global rates of people living with HIV/AIDS and high levels of socioeconomic inequality. This longitudinal population-based cohort of children and their primary caregivers assesses household and caregiver functioning, child health, social well-being, and neuro-development from childhood through adolescence. Almost 1,600 children born at the peak of the human immunodeficiency virus epidemic (2003-2005) were followed (with their primary caregivers) in 3 waves, between 2008 and 2021, at average ages of 5, 7, and 16. Wave 3 is currently underway, having assessed over 1,100 of the original wave 1 children. Wave 4 begins in 2022. The study, with a dyadic structure, uses a broad range of measures, validated in South Africa or recommended for global use, that address physical, social and neuro-development in childhood and adolescence, and the social, health, and psychological status of children’s primary caregivers. The Asenze study deepens our understanding of childhood physical, cognitive, and social abilities and/or disabilities, including risk-taking behaviors, and biological, environmental, and social determinants of health. We anticipate the findings will contribute to the development of community-informed interventions to promote well-being in this South African population and elsewhere.
Ivy Halls and Ivy Walls: The Continuing Legacy of the Ivy League

This article analyses the changes in access to U.S. Ivy League universities over the last forty years in terms of gender, racial and ethnic diversity, to argue that despite greater access for women and racialized populations, these elite institutions still largely reproduce social class, with only a small percentage of their students drawn from the lower middle and working classes. The continuing lure of the Ivy League is examined, including specifically the lure for students in South Korea, and the article concludes with a discussion of which aspects of elite education should be widely available and might be transferable to other domains like public education.
Biodiversity of harvestmen in Hong Kong and their relationship with amphibians

Despite being a common animal found in the wild, there are very few studies on global biodiversity of harvestmen, and even less so in Hong Kong, China. This study will uncover the number of species in Hong Kong, as well as describe and classify new species using updated morphological techniques and phylogenetic analysis. We will also describe their role in the ecosystem as prey items of amphibians, as documented both in/ex-situ. The project will introduce these mysterious creatures, and hope will stimulate further harvestmen research in Hong Kong and Korea.
