      • EFNA1 ligand and its receptor EphA2: potential biomarkers for hepatocellular carcinoma

        <P>Novel biomarkers are needed for early detection and progression evaluation of hepatocellular carcinoma (HCC). The purpose of this study was to identify useful biomolecular markers for HCC. The 26 genes that encode membrane or secretory proteins were identified from cDNA microarray data. We further examined the expression of EFNA1 and its receptor EphA2 and determined their biological implications during the development and progression of HCC. The EFNA1 mRNA was overexpressed in most HCCs as compared with its expression in corresponding nontumor tissues (36 out of 40 cases, 90%), but EphA2 expression was noted in only half of the HCC tissues (20 of 40 cases, 50%). In most of the hepatoma cell lines, the EFNA1 protein expression was positively associated with alpha-fetoprotien (AFP) expression but inversely associated with EphA2 expression. Furthermore, EFNA1 levels were detectable in the supernatant of the cultured hepatoma cells and in the serum of patients with HCC. In contrast, EphA2 expression was prominent in highly invasive hepatoma cells, and its overexpression was significantly correlated with decreased differentiation (r = 0.0248, p < 0.010) and poor survival (p = 0.0453) for HCC patients. EFNA1 and EphA2 may be useful serum markers for the detection of HCC development and progression, respectively.</P>

        문화 부등성에 따른 중국인 학습자의 비판적 수용 연구 - 『82년생 김지영』의 원본 및 중국어 번역본 비교를 중심으로

        한국 독서시장에서의 한국문학을 ‘순문학’이라고 규정지어 살펴보면 중국 내 도서시장에서의 점유율 및 영향력은 그다지 크지 못하다. 즉 유통되고 있는 한국순수문학 작품은 저조한 시장성과 적은 독자군(群)을 가지고 있다. 그 원인은 번역물과 독자수용 사이의 거리이고, 또 하나의 문제는 한류열풍이 크지만 그에 비해 한국현대소설의 독자지형이 너무 협소하다는 것이다. 때문에 한국문학의 순문학 개념에 대한 정초가 우선이다. 그다음 한국문학작품에 대한 감상이 뒤따르는데, 본 연구에서 중국인 학습자를 대상으로 조남주의 장편소설 『82년생 김지영』의 원천 텍스트와 2019년 9월에 출판된 중국어판 『82年生的金智英』을 동시 접근하는 장치를 마련해줬다. 학습자는 텍스트를 읽는 과정에서 차이점을 체감하면서 문화의 충돌을 대리 경험하게 되고 다중언어문화의 소양을 쌓아가는 것이 가능해진다. 본 연구는 중국인 한국어 학습자가 한국문학을 감상함에 있어서, 원천·번역 텍스트를 동시에 살펴보고, 두 문화 사이의 간극을 발견하고; 번역과정에 있어서, 개입된 여러 요인에 대하여 비판적 반응 양상을 산출하는 과정을 거쳐 마지막으로 문화번역의 주체로 거듭나서 양 텍스트 간의 전환과 감상이 자유로워지고 나아가 양 문화를 영위한 개체로 성장하게 될 것이라고 피력하고자 한다. If Korean literature is defined as "pure literature", its share and influence in the book market in China are not very significant. In other words, pure Korean literature works in circulation in China have significantly poor marketability and a small group of readers compared to the literature of other countries. This is because there is distance between translationed works and readers' reception, and another problem is that although Korean Wave has been remarkable in its influence, modern Korean novels still have a narrow reader base. Therefore, it is necessary first to define the concept of pure literature in Korean literature. In this study, we examined the original Korean text of Cho Nam-joo's novel Kim Jiyoung, Born 1982 and the Chinese publication translated in September 2019. It is possible to make Chinese learners build up the knowledge of multilingual culture. In this study, Chinese learners discovered the gap between the two cultures through Korean literature; finally, readers will be born again as a subject of cultural translation, and will be able to freely experience conversion and appreciation between the two texts, and furthermore, will grow into an individuals that drives both cultures.

        中国公司法之司法续造的实证分析:困惑与出路

        基于公司法时滞性、技术性、自治性立法层面的障碍, 《公司法》无法预期涵摄后出现的社会关系是必然的, 这也必将致使法律漏洞的产生, 《公司法》这个无法弥补的先天遗憾, 却促使司法实践中不得不在一定程度上引入法官的主观能动性, 法官在公司法立法层面日趋完善而司法层面实施弱化的大背景之下, 仍然依靠三段论的推理模式解决公司 法律规制阙如的司法困惑显然力不从心。司法不得不针对公司法律规则的不完备性进行续造, 以期弥补公司法理论与公司法实务之间的缝隙。只是司法续造的过程中应当注意防守司法介入的底限, 不仅不能威胁公司自治精神的存在, 而且也不能窒息公司自治的功能。文章在分析司法续造影响性因素的基础之上, 为解决“同案不同判”的冲突, 走出司法能动性与成文法稳定性的两难困境, 为填补公司法律漏洞提供统一的司法解决方案, 提出公司法司法续造的方法论基础。在宏观原则即:信奉统一的司法哲学、审慎理性的司法裁量、实施性效果前瞻性考量的指导下, 对于法律漏洞的克服, 进行类别化识别与填补。 While centered around the theme of judicial development of company law against the macro-background characteristic of perfecting legislation and weakening jurisdiction, this dissertation presents an exploring research on the mechanism ensuring a positive reactive relationship and smooth dialogue between statute company law and jurisdiction thereof.The current research focuses on how jurists who bear the responsibility of forwarding decisions in specific cases by way of syllogism, interpret and develop company law in the absence of statute law adequately applicable to cases in hand ? Based on the legislative obstacles of the Company Law, like time lag, technicality and autonomy, judiciary has to create law for the incompleteness of the Company Law. In this process, it is the bottom of judicial intervention that should be concerned to be defended, which not only cannot threaten the existence of the spirit of company autonomy but also cannot to stifle the functions of company autonomy. Empirical methods are adopted in this research, in combination with the achievements by other scholars worked on related themes, the latest theoretical developments and overseas practices in similar areas as well, to deal with the challenges specified above. The purported purpose of this research is to work out a uniform mechanism for structuring a general theory on the theme so as to solve problems with respect to conflicts between decisions among individual cases, to find a way out of the dilemma between dynamic judicial innovations and the stability of legislation, and also to fill up the gap between company law theories and judicial practices and the leaks in statute company law. Based on the analysis of the influencing factors of judicial renewing, the article promotes the methodology foundation of the judicial renewing in Company Law. The macroscopic principle: under the guidance of believing in uniform judicial philosophy, prudent and rational discretion and foresight examination for implementation effect, it is through the categorized recognition and padding that the legal loopholes can be overcame.

        The Antibacterial Mechanism of Zn(II) Frame Supported on Alginate Membrane

        In order to solve the problem of antibiotic-resistant bacteria caused by excessive use of antibiotics, herein, an antibacterial membrane composed of natural sodium alginate (ALG), zeolite imidazolate skeleton (ZIF-8) and niflumic acid (NIF) was reported. The membrane serves as a versatile platform for local antibacterial. This report carried out in-depth research on the physical properties and antibacterial mechanism of the synthesized sodium alginate composite film. The data shows that the sodium alginate-based antibacterial film has a continuous antibacterial effect, and the release of antibacterial molecules can be controlled according to changes in the external environment. The results show that the complex has stronger mechanical and bacteriostatic properties. Niflumic acid and Zn(II) have synergistic antibacterial effect. The complex promotes bacterial death by hindering bacterial respiratory metabolism and destroying cell membrane structure.

        Oncomodulin/Truncated Protamine-Mediated Nogo-66 Receptor Small Interference RNA Delivery Promotes Axon Regeneration in Retinal Ganglion Cells

        The optic nerve often suffers regenerative failure after injury, leading to serious visual impairment such as glaucoma. The main inhibitory factors, including Nogo-A, oligodendrocyte myelin glycoprotein, and myelin-associated glycoprotein, exert their inhibitory effects on axonal growth through the same receptor, the Nogo-66 receptor (NgR). Oncomodulin (OM), a calcium-binding protein with a molecular weight of an ~12 kDa, which is secreted from activated macrophages, has been demonstrated to have high and specific affinity for retinal ganglion cells (RGC) and promote greater axonal regeneration than other known polypeptide growth factors. Protamine has been reported to effectively deliver small interference RNA (siRNA) into cells. Accordingly, a fusion protein of OM and truncated protamine (tp) may be used as a vehicle for the delivery of NgR siRNA into RGC for gene therapy. To test this hypothesis, we constructed OM and tp fusion protein (OM/tp) expression vectors. Using the indirect immunofluorescence labeling method, OM/tp fusion proteins were found to have a high affinity for RGC. The gel shift assay showed that the OM/tp fusion proteins retained the capacity to bind to DNA. Using OM/tp fusion proteins as a delivery tool, the siRNA of NgR was effectively transfected into cells and significantly down-regulated NgR expression levels. More importantly, OM/tp-NgR siRNA dramatically promoted axonal growth of RGC compared with the application of OM/tp recombinant protein or NgR siRNA alone in vitro. In addition, OM/tp-NgR siRNA highly elevated intracellular cyclic adenosine monophosphate (cAMP) levels and inhibited activation of the Ras homolog gene family, member A (RhoA). Taken together, our data demonstrated that the recombinant OM/tp fusion proteins retained the functions of both OM and tp, and that OM/tp-NgR siRNA might potentially be used for the treatment of optic nerve injury.

        BOTDA based water-filling and preloading test of spiral case structure

        In the water-filling and preloading test, the sensing cables were installed on the surface of steel spiral case and in the surrounding concrete to monitor the strain distribution of several cross-sections by using Brillouin Optical Time Domain Analysis (BOTDA), a kind of distributed optical fiber sensing (DOFS) technology. The average hoop strain of the spiral case was about $330{\mu}{\varepsilon}$ and $590{\mu}{\varepsilon}$ when the water-filling pressure in the spiral case was 2.6 MPa and 4.1 MPa. The difference between the measured and the calculated strain was only about $50{\mu}{\varepsilon}$. It was the first time that the stress adjustment of the spiral case was monitored by the sensing cable when the pressure was increased to 1 MPa and the residual strain of $20{\mu}{\varepsilon}$ was obtained after preloading. Meanwhile, the shrinkage of $70{\sim}100{\mu}{\varepsilon}$ of the surrounding concrete was effectively monitored during the depressurization. It is estimated that the width of the gap between the steel spiral case and the surrounding concrete was 0.51 ~ 0.75 mm. BOTDA based distributed optical fiber sensing technology can obtain continuous strain of the structure and it is more reliable than traditional point sensor. The strain distribution obtained by BOTDA provides strong support for the design and optimization of the spiral case structure.

      • Activation of mammalian target of rapamycin complex 1 (mTORC1) and Raf/Pyk2 by growth factor‐mediated Eph receptor 2 (EphA2) is required for cholangiocarcinoma growth and metastasis

        <P><B>Abstract</B></P><P>Eph receptor 2 (EphA2) overexpression is frequently accompanied by the loss of its cognate ligand during tumor progression. However, the molecular mechanism of this ligand‐independent promotion of tumor by EphA2 remains unclear in highly malignant and fatal cholangiocarcinoma (CC). We examined the biological role of EphA2 in tumor growth and metastasis in CC tissues and cells according to the degree of differentiation and we explored the downstream signaling pathways of EphA2. Growth factor‐mediated EphA2 overexpression itself leads to the activation of the mammalian target of rapamycin complex 1 (mTORC1) and extracellular signal‐regulated kinase (ERK) pathways through ligand‐independent activation of EphA2 (phosphorylation of S897). An <I>in vitro</I> soft agar assay and <I>in vivo</I> orthotopic or subcutaneous tumor model showed that EphA2 enhanced colony formation and accelerated tumor growth, and which seemed to be mainly associated with Akt (T308)/mTORC1 activation. Aberrant expression and activation of EphA2 was also associated with poorer differentiation and higher metastatic ability. Enhanced metastatic ability was also observed in an orthotopic tumor model or lung metastasis model, correlating with Pyk2(Y402)/c‐Src/ERK activation in addition to activation of the canonical Raf/MEK/ERK pathway. The mTORC1 and Raf/Pyk2 pathways also appeared to affect each other. These results suggest that growth factor‐mediated EphA2 might be involved in tumor growth and metastasis through activation of the mTORC1 and Raf/Pyk2 pathways. Therapeutic strategies that target EphA2 and its downstream effectors may be useful to control CC. (H<SMALL>EPATOLOGY</SMALL> 2013;57:2248–2260)</P>
