Herein, a series of lasiokaurin derivatives were designed and synthesized. All the derivatives together with lasiokaurin and oridonin were tested for their antimicrobial and antiproliferative activity. Compound 16 showed the most promising antimicrobial activity with MICs of 2.0 and 1.0 lg/mL against Gram-Positive bacteria S. aureus and B. subtilis, respectively. All the synthetic lasiokaurin derivatives showed better antiproliferative activity than parent compound lasiokaurin 1. Compound 10 exhibited the strongest cytotoxicity with IC50 values of 0.47 and 0.20 lM against MGC-803 and CaEs-17 cells, accordingly. Moreover, it was shown to have potent antitumor activity in vivo in a murine model of MGC-803 gastric cancer. Preliminary SARs were also concluded based on obtained data. The apoptosis-inducing effects of 10 were further investigated using CaEs-17 cells. The results showed that lasiokaurin derivative 10 could induce apoptosis via mitochondria related pathway and arrest CaEs-17 cell cycle at S phase. Compound 10 could also affect apoptosisrelated proteins that was up-regulation of CDK2 and downregulation of ATM and cyclin A1.
Influence of Semi-Active Suspension on Running Safety of Vehicles
Liu Hong-You,Yu Da-Lian 한국철도학회 2009 International Journal of Railway Vol.2 No.4
Railway vehicles equipped with semi-active suspension system can improve the ride quality of car bodies. Semi-active suspension system is usually applied onto high speed train, and therefore higher running safety requirement is proposed. The influence of semi-active suspension system on safety of vehicles running on straight line and curve line is studied, and the influences of sky hook damping coefficient and system time-delay on operation safety of cars fitted with semiactive suspension system is analyzed. The results show that the vehicles equipped with semi-active suspension system, not only the vibration of car body is decreased, it can also give little influence on running safety of cars, as a result, it will not endanger the running safety of cars.

Railway vehicles equipped with semi-active suspension system can improve the ride quality of car bodies. Semi-active suspension system is usually applied onto high speed train, and therefore higher running safety requirement is desirable. The influence of semi-active suspension system on safety of vehicles running on straight line and curve line is studied, and the influences of sky hook damping coefficient and system time-delay on operational safety of cars fitted with semiactive suspension system is analyzed. The results show that in vehicles equipped with semi-active suspension system, while the vibration of car body is decreased, the running safety of cars is not affected to any significant degree. As a result, the ride quality is much improved with negligible deterioration of the running safety of cars.

Increasing evidence suggests that gut microbiota underpin the development of health and longevity. However, our understanding of what influences the composition of this community of the longevous has not been adequately described. Therefore, illumina sequencing analysis was performed on the gut microbiota of centenarians (aged 100-108 years; RC) and younger elderlies (aged 85-99 years; RE) living in Bama County, Guangxi, China and the elderlies (aged 80-92 years; CE) living in Nanning City, Guangxi, China. In addition, their diet was monitored using a semiquantitative dietary questionary (FFQ 23). The results revealed the abundance of Roseburia and Escherichia was significantly greater, whereas that of Lactobacillus, Faecalibacterium, Parabacteroides, Butyricimonas, Coprococcus, Megamonas, Mitsuokella, Sutterella, and Akkermansia was significantly less in centenarians at the genus level. Both clustering analysis and UniFraq distance analysis showed structural segregation with age and diet among the three populations. Using partial least square discriminate analysis and redundancy analysis, we identified 33 and 34 operational taxonomic units (OTUs) as key OTUs that were significantly associated with age and diet, respectively. Age-related OTUs were characterized as Ruminococcaceae, Clostridiaceae, and Lachnospiraceae, and the former two were increased in the centenarians; diet-related OTUs were classified as Bacteroidales, Lachnospiraceae, and Ruminococcaceae. The former two were deceased, whereas the later one was increased, in the high-fiber diet. The age and high-fiber diet were concomitant with changes in the gut microbiota of centenarians, suggesting that age and high-fiber diet can establish a new structurally balanced architecture of gut microbiota that may benefit the health of centenarians.

Anti-proliferation Effects of Interferon-gamma on Gastric Cancer Cells
Zhao, Ying-Hui,Wang, Tao,Yu, Guang-Fu,Zhuang, Dong-Ming,Zhang, Zhong,Zhang, Hong-Xin,Zhao, Da-Peng,Yu, Ai-Lian Asian Pacific Journal of Cancer Prevention 2013 Asian Pacific journal of cancer prevention Vol.14 No.9
IFN-${\gamma}$ plays an indirect anti-cancer role through the immune system but may have direct negative effects on cancer cells. It regulates the viability of gastric cancer cells, so we examined whether it affects their proliferation and how that might be brought about. We exposed AGS, HGC-27 and GES-1 gastric cancer cell lines to IFN-${\gamma}$ and found significantly reduced colony formation ability. Flow cytometry revealed no effect of IFN-${\gamma}$ on apoptosis of cell lines and no effect on cell aging as assessed by ${\beta}$-gal staining. Microarray assay revealed that IFN-${\gamma}$ changed the mRNA expression of genes related to the cell cycle and cell proliferation and migration, as well as chemokines and chemokine receptors, and immunity-related genes. Finally, flow cytometry revealed that IFN-${\gamma}$ arrested the cells in the G1/S phase. IFN-${\gamma}$ may slow proliferation of some gastric cancer cells by affecting the cell cycle to play a negative role in the development of gastric cancer.
