        Genetic Characterization of Haemagglutinin 1 Domain of Influenza B Viruses Isolated in Korea during $1988{\sim}1999$

        Although Korean influenza virus isolates have been genetically associated with the vaccine strains of the corresponding year, influenza B viruses have prevailed almost every year in Korea during the past decades. We have 'analyzed the genetic characteristics and evolutionary patterns of the haemagglutinin (HA) 1 domains of influenza B viruses isolated during 1988-1999 using direct RT-PCR and sequencing. Phylogenetic analysis of influenza B viruses isolated in Korea indicated that antigenically and genetically distinguishable strains of the lineage II and lineage III variants had been cocirculated. Variants prevailed in early 1990s are represented in 1996/97 and 1998/99 and some different variants have been co circulated geographically and prevailed concurrently in Korea. All HA1s of Korean isolates have amino acid substitutions mainly in the region between position 124 and 310, which was previously proposed an immunodominant region. Insertion-deletion patterns of the HA gene revealed that Korean influenza B viruses were evolved from Lee40 with different manner between lineage II and III viruses. Lineage III viruses were also divided into two groups as conserved group and inserted group, in relation to Lee40. But lineage II viruses had evolved with directional pattern. Antigenic index proposed that influenza B isolates prevailed since 1996/97 seasons might had emerged from the antigenic variants of a Seo1697-like virus and that new variants might appear from the lineage II viruses resulting in persistent prevalence in Korea.

      • Thiocholine ester 기질을 이용한 Acetylcholinesterase 활성부위의 구조특성

        The inhibition pattern of three inhibitors(tacrine, decamethonium and propidium) on the hydrolysis of various thiocholine ester substrates by eel acetylcholinesterase was comparatively examined. When the substrate was acetylthiocholine, it showed a similar competitive inhibition by tacrine inhibitor, and a mixed type inhibition by decamethonium and propidium inhibitors. When the substrate was pentanoylthiocholine, it showed an uncompetitive inhibition by tacrine, and a noncompetitive inhibition by decamethonium. When the substrate was laurylthiocholine, it showed mixed type and uncompetitive inhibition by tacrine, and a competitive inhibition by decamethonium and propidium. Those results suggest that the active of acetylcholinesterase has the existence of hydorphobic site besides the anionic and esteratic site.

      • Metalloporphyrin으로 도핑된 폴리이미드의 광전사적 성질

        본 연구에서는 ZnTPP와 MgTPP를 도우핑한 폴리이미드의 과악적 성질을 UV spectrum과 emission spectrum을 이용하여 조사하였다. Tetraphenylporphyrin-zinc (ZnTPP)와 Tetraphenyl-porphyrin-magnesium (MgTPP)는 전하이동현상을 보였다. 폴리이미드와 폴피린관련 자외흡수피크에서 red-shift를 관찰할 수 있었다. 이는 폴리이미드의 이미드와 폴피린에 포함된 금속 물질과의 전하이동착체의 형성때문으로 설명할 수 있다. In this research, we fabricated metalloporphyrin doped polymides(PI) and characterized them with ultraviolet spectrophotometer and emission spectrophotometer. Tetraphenylporphyrin-zinc-doped and Tetraphenylporphyrin-magnesium-doped PIs showed charge transfer phenomena. Red-shift was obseved in polyimide and porphyrin related bands at the same time. This phenomena can be explained that charge transfer complex(CTC) formed between imide group of PI and metal in the porphyrin.

      • p-Nitrostyrylphenylsulfone 基質에 對한 L-Cysteine의 反應

        The kinetics and mechanism of nucleophilic addition of L-Cysteine to P-nitrostyrlphenyl-sulfone substrate over wide pH range was presented and analyzed in detail. The rate constants were determined by ultra-violet spectrophotometry in 50% methanol water mixed solvent at 25℃ and the following rate equation which can be applied over wide pH range was obtained. i.e kt = ko + kp [OH^(-)] =(1.92×10^(-3)+3.71×10^(3)[OH^(-)]/8.94×10^(-2)+1.03×10^(5)[OH^(-)]) +4.56×10^(2)OH^(-)] From the rate equation, the following mechanism were proposed; above pH 10. 50, the reaction proceed by the attack of L-Cy-S^(-) on the activated carbon-carbon double bind of substrate. Below pH 8.50, the reaction proceed by addition of L-Cysteine to p-nitrostyrl phenylsulfone substrate. And in the range of pH 8.50-10.50, these two reaction occur competitively.

      • 몇가지 金屬이온이 밀 幼植物의 室酸還元 酵素 活量에 미치는 效果

        The initial metabolism of nitrate to nitrite can be readily demonstrated in vivo by incubating plant tissue in the presence of nitrate in the dark under aerobic condition. All wheat seedlings during germination were in a dark incubator, and eight days-old seedlings were used for these experiments. Few other dater on nitrate reductase inhibition by metal ions can be found and no detailed studies of some inhibition types have been reported. For this reason, the authors have carried out some experiments on the inhibition types of nitrate reductase by some metal ions. 1mM CuCl and ZnCl_2 have inhibited nitrate reductase activity to a range of great degree (20-40%), while 1mM MnCl_2 and CoCl_2 have acted as less potent nitrate reductase inhibitors under 10% decreased condition. Relatively, 1mM KCl, MgCl_2 and CaCl_2 have shown the result acting as an activator of nitrate reductase. On the other hand, nitrate reductase levels have shown a phenomenon of fluctuation in response to changes depending upon 0.001-0.1mM concentration of some chloride salts, and shown constant value to a high concentration(1mM) of inhibited chloride salts. Consequently, it is assumed that the results of inhibition are belong to mixed type among various inhibitions, especially with respect to the nitrate reductase activity of wheat seedlings treated with CuCl, ZnCl_2, MnCl_2 and CoCl_2.

        Spectroscopic investigations of spermine effects on the ethidium-DNA binding equilibrium, and on the absorbance-temperature profile of the DNA in the presence of ethidium bromide at various concentrations were performed, as the ethidium binding may be used as the probe of the DNA interactions with environmental ligands. The cooperativity, enthalpy, and the midpoint of the phase transition to the trough region, of the anomalous absorbance-temperature profile, which occurs at the spermine concentration where the DNA collapses into a compact structure, are more sensitive to ethidium than the Tm region phase, and the trough can be eliminated by increased concentration of ethidium.

        We have investigated the effect of hydrophobic environmental reagents (urea and DMSO) on the anomalous absorbance-temperature profile of condensed DNA, induced by spermine. The cooperativity of the phase transition to the trough region in the anomalous absorbance-temperature profile is more sensitive to the hydrophobic environmental reagents than that of the melting region, and the trough peak size can be decreased by increased concentration of the hydrophobic reagents. The present data indicate the importance of hydrophobic interaction in the stabilization of the condensed state of DNA induced by spermine, and that the anomalous absorbance-temperature profile is due to the thermal stabilization of the tertiary structure of the spermine-induced collapsed state of the DNA.

      • Ethylcinnamate에 對한 Iso-propylmercaptan의 親核性 添加反應에 關한 反應速度論的 硏究

        ethylcinnamate에 대한 iso-propylmercaptan의 친핵성 첨가반응 속도상수를 측정하여 넓은 pH 범위에서 잘맞는 반응속도식을 얻었다. 이 식에 의하면 pH 6.5 이하에서는 iso-propylmercaptan과 iso-propylmercaptide ion의 첨가가 경쟁적으로 일어난다. pH 10이상에서는 ethylcinnamate에 iso-propylmercaptide ion의 첨가만이 일어남을 알수 있다. 그러므로 본 실험에서 구한 반응속도식으로부터 실험측정값과 계산 값(이론 값)이 여러 pH를 통하여 잘 맞기 때문에 넓은 pH범위내에서 일어나는 친핵성 첨가반은 mechanism을 잘 설명할 수 있었다. The rate constants of nucleophilic addition reaction of iso-propylmercaptan to ethylcinnamate were determined at various pH and a rate equation which can be applied overwide pH range was obtained; ?? The rate equation reveals that below pH 6.5 the reaction is initiated by addition of iso-propylmercaptan molecule, from pH 6.5 to 10.0 iso-propylmercaptan and iso-propylmercaptide ion are competitively added. Above pH 10.0 the addition reaction of iso-propylmercaptide ion occurs. From these rate equation, we found that observed data accorded satisifactorily with theoretical value over various pH range. Hence, the mechanism of nucleophilic addition reaction over wide pH range can be fully explained.

      • 스페르민-DNA 복합체의 이례적 흡광도-온도 양상에 미치는 DMSO의 영향

        DNA의 응축을 유발시키는 spermine 농도 존재시 downward peak(trough)를 갖는 이례적 흡광도-온도 양상에 hydrophobic environment를 조성하는 DMSO의 영향을 조사하였다. DMSO 농도증가에 따라 downward peak의 깊이가 점차 얕아졌으며 그 phase transition enthalpy와 relative cooperative length(n)가 melting region보다는 downward peak(trough) phase transition에 더 예민하게 영향을 받았다. 따라서 본 data는 spermine에 의하여 유발되는 응축된 DNA 구조에 이르는 hydrophobic interaction에 의한 tertiary structure 형성이 온도상승에 따라 유리해짐으로써 downward peak가 형성된다는 추리를 확인하는 실험결과가 될 것이다. In an effort to improve our knowledge on the mechanism of the collapse of DNA into a compact structure by polyamines, we investigated the effect of dimethyl sulfoxide (DMSO), a hydrophobic environmental reagent on the anomalous absorbance-temperature profile of condensed DNA, induced by spermine and having a trough preceeded by the initiation of the melting phase. The trough peak size can be decreased by increased concentration of DMSO, and the cooperativity of the phase transition to the region in the anomalous absorbance-temperature profile is more sensitive to DMSO than that of the melting regions of the profile. The present data indicate that the anomalous absorbance-temperatuve profile is due to the thermal stabilization of the tertiary structure of the spermine-induced collapsed state of DNA and the importance of hydrophobic interaction in the stabilization of the condensed state of DNA induced by spermine.
