Preparation and Characterization of Antioxidant Peptides from Fermented Goat Placenta

        Hou, Yinchen,Zhou, Jiejing,Liu, Wangwang,Cheng, Yongxia,Wu, Li,Yang, Gongming Korean Society for Food Science of Animal Resource 2014 한국축산식품학회지 Vol.34 No.6

The goat placenta was fermented by Bacillus subtilis and the optimal fermentation parameters of strongest antioxidant capacity of peptides were obtained using response surface methodology (RSM). The effects of fermentation time, initial pH value and glucose content on the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging capacity of the goat peptides were well fitted to a quadric equation with high determination coefficients. According to the data analysis of design expert, the strongest DPPH radical scavenging capacity value was obtained with the following conditions: content of glucose was 2.23%, initial pH value was 7.00 and fermentation time was 32.15 h. The DPPH radical scavenging capacity commonly referring antioxidant activity showed a concentration dependency and increased with increasing peptide concentration. The effects of temperature and pH were assessed to determine the stability of antioxidant peptides prepared from goat placenta. Antioxidant peptides showed good stabilities when temperature was lower than $70^{\circ}C$. However, the antioxidant peptides lost antioxidant activities rapidly under alkaline and excessive acid condition. Ultrafiltration technique was performed to separate fermentation broth with different Mw (molecular weight). It was found that peptides in the range of < 3 KDa mainly accounted for the antioxidant activities.

Distinct Transcriptional and Functional Differences of Lung Resident and Monocyte-Derived Alveolar Macrophages During the Recovery Period of Acute Lung Injury

        Hou Fei,Wang Huan,Zheng Kun,Yang Wenting,Xiao Kun,Rong Zihan,Xiao Junjie,Li Jing,Cheng Baihe,Tang Li,Xie Lixin 대한면역학회 2023 Immune Network Vol.23 No.3

In acute lung injury, two subsets of lung macrophages exist in the alveoli: tissue-resident alveolar macrophages (AMs) and monocyte-derived alveolar macrophages (MDMs). However, it is unclear whether these 2 subsets of macrophages have different functions and characteristics during the recovery phase. RNA-sequencing of AMs and MDMs from the recovery period of LPS-induced lung injury mice revealed their differences in proliferation, cell death, phagocytosis, inflammation and tissue repair. Using flow cytometry, we found that AMs showed a higher ability to proliferate, whereas MDMs expressed a larger amount of cell death. We also compared the ability of phagocytosing apoptotic cells and activating adaptive immunity and found that AMs have a stronger ability to phagocytose, while MDMs are the cells that activate lymphocytes during the resolving phase. By testing surface markers, we found that MDMs were more prone to the M1 phenotype, but expressed a higher level of pro-repairing genes. Finally, analysis of a publicly available set of single-cell RNA-sequencing data on bronchoalveolar lavage cells from patients with SARS-CoV-2 infection validated the double-sided role of MDMs. Blockade of inflammatory MDM recruitment using CCR2−/− mice effectively attenuates lung injury. Therefore, AMs and MDMs exhibited large differences during recovery. AMs are long-lived M2-like tissue-resident macrophages that have a strong ability to proliferate and phagocytose. MDMs are a paradoxical group of macrophages that promote the repair of tissue damage despite being strongly pro-inflammatory early in infection, and they may undergo cell death as inflammation fades. Preventing the massive recruitment of inflammatory MDMs or promoting their transition to pro-repairing phenotype may be a new direction for the treatment of acute lung injury.

      • Controlled Growth of Well-Defined Conjugated Polymers from the Surfaces of Multiwalled Carbon Nanotubes: Photoresponse Enhancement via Charge Separation

        Hou, Wenpeng,Zhao, Ning-Jiu,Meng, Dongli,Tang, Jing,Zeng, Yi,Wu, Yu,Weng, Yangziwan,Cheng, Chungui,Xu, Xiulai,Li, Yi,Zhang, Jian-Ping,Huang, Yong,Bielawski, Christopher W.,Geng, Jianxin American Chemical Society 2016 ACS NANO Vol.10 No.5

The installation of heterojunctions on the surfaces of carbon nanotubes (CNTs) is an effective method for promoting the charge separation processes needed for CNT-based electronics and optoelectronics applications. Conjugated polymers are proven state-of-the-art candidates for modifying the surfaces of CNTs. However, all previous attempts to incorporate conjugated polymers to CNTs resulted in unordered interfaces. Herein we show that well-defined chains of regioregular poly(3-hexylthiophene) (P3HT) were successfully grown from the surfaces of multiwalled CNTs (MWNTs) using surface-initiated Kumada catalyst-transfer polycondensation. The polymerization was found to proceed in a controlled manner as chains of tunable lengths were prepared through variation of the initial monomer-to-initiator ratio. Moreover, it was determined that large-diameter MWNTs afforded highly ordered P3HT aggregates, which exhibited a markedly bathochromically shifted optical absorption due to a high grafting density induced planarization of the polymer chains. Using ultrafast spectroscopy, the heterojunctions formed between the MWNTs and P3HT were shown to effectively overcome the binding energy of excitons, leading to photoinduced electron transfer from P3HT to MWNTs. Finally, when used as prototype devices, the individual MWNT-g-P3HT core-shell structures exhibited excellent photoresponses under a low illumination density.

Comprehensive profiles and diagnostic value of menopausal-specific gut microbiota in premenopausal breast cancer

        Hou Ming-Feng,Ou-Yang Fu,Li Chung-Liang,Chen Fang-Ming,Chuang Chieh-Han,Kan Jung-Yu,Wu Cheng-Che,Shih Shen-Liang,Shiau Jun-Ping,Kao Li-Chun,Kao Chieh-Ni,Lee Yi-Chen,Moi Sin-Hua,Yeh Yao-Tsung,Cheng Chi 생화학분자생물학회 2021 Experimental and molecular medicine Vol.53 No.-

In Western countries, breast cancer tends to occur in older postmenopausal women. However, in Asian countries, the proportion of younger premenopausal breast cancer patients is increasing. Increasing evidence suggests that the gut microbiota plays a critical role in breast cancer. However, studies on the gut microbiota in the context of breast cancer have mainly focused on postmenopausal breast cancer. Little is known about the gut microbiota in the context of premenopausal breast cancer. This study aimed to comprehensively explore the gut microbial profiles, diagnostic value, and functional pathways in premenopausal breast cancer patients. Here, we analyzed 267 breast cancer patients with different menopausal statuses and age-matched female controls. The α-diversity was significantly reduced in premenopausal breast cancer patients, and the β-diversity differed significantly between breast cancer patients and controls. By performing multiple analyses and classification, 14 microbial markers were identified in the different menopausal statuses of breast cancer. Bacteroides fragilis was specifically found in young women of premenopausal statuses and Klebsiella pneumoniae in older women of postmenopausal statuses. In addition, menopausal-specific microbial markers could exhibit excellent discriminatory ability in distinguishing breast cancer patients from controls. Finally, the functional pathways differed between breast cancer patients and controls. Our findings provide the first evidence that the gut microbiota in premenopausal breast cancer patients differs from that in postmenopausal breast cancer patients and shed light on menopausal-specific microbial markers for diagnosis and investigation, ultimately providing a noninvasive approach for breast cancer detection and a novel strategy for preventing premenopausal breast cancer.

        K-doping effect of the superconductivity in K2xFeTe1-xSx (0.07 ≤ x ≤ 0.3)

        Cheng Cheng,Zhenjie Feng,Qing Li,Tao Li,Qiang Hou,Fei Chen,Zhongmin Ou,Jun-Yi Ge,Shixun Cao,Jincang Zhang 한국물리학회 2019 Current Applied Physics Vol.19 No.4

Bulk samples of K doping K2xFeTe1-xSx with x=0.07, 0.1, 0.2, 0.3 are successfully prepared by using easy-to-use stable compound K2S as the reactant. The lattice constant calculated from X-ray diffraction patterns indicate that K ions enter the Fe-Te-S layers. K doping is beneficial enhance the superconductivity transition temperature from the R-T curves. The apparent diamagnetic signal is observed in M-T curves when the content of K is smaller than 0.1. However, differential curves (dM/dT) in K-rich samples appear sharp slope mutations, which means that the Meissner effect signal is covered by the increased excess ferromagnetic ions. The number of excess Fe magnetic ions is proportional to K content, which may play an important role in determining the superconductivity.

      • The Evaluation of Drying Qualities of Tea Dryers in Taiwan

        ( Cheng-hou Chang ),( Suming Chen ),( Chao-yin Tsai ),( Kuo-chih Tung ) 한국농업기계학회 2018 한국농업기계학회 학술발표논문집 Vol.23 No.1

Tea drying is a necessary process of tea manufacturing to prevent deterioration, stabilize the tea quality, and to facilitate packaging and storage. Therefore, the tea dryer is an essential equipment for the tea industry; and the tea drying quality is an index to evaluate tea dryers. In this study, four types of tea dryers were used to conduct tea leaves drying experiments. Four tests of sensory evaluation, color analysis, soluble ingredients analysis and chemical composition analysis were used to evaluate the drying quality of the tea dryers. The experiment results showed that the vacuum roller dryer (VRD) in the overall tea drying qualities had a better evaluation, the next were low-humidity dryer (LHD) and multiple-layer conveyer dryer (MLD), the electric hot air dryer (HAD) was worst among all tea dryers. The comprehensive evaluation of tea drying qualities were investigated using area analysis of radar chart which were VRD (2.38) > LHD (2.15) > MLD(2.12) > HAD(2.06). Therefore, the vacuum drying with moderate stirring could improve qualities of tea drying. Based on the results of this study, tea farmers or makers can choose the proper dryer, and tea equipment manufacturers can improve and enhance the performance of tea dryers

Dietary Supplementation of Magnesium Sulfate during Late Gestation and Lactation Affects the Milk Composition and Immunoglobulin Levels in Sows

        Hou, W.X.,Cheng, S.Y.,Liu, S.T.,Shi, B.M.,Shan, A.S. Asian Australasian Association of Animal Productio 2014 Animal Bioscience Vol.27 No.10

This experiment was conducted to investigate the effects of dietary supplementation of magnesium sulfate ($MgSO_4$) during late gestation and lactation on sow and litter performance, fecal moisture, blood biochemistry parameters, immunoglobulin levels and milk composition in sows. Forty-eight sows ($Yorkshire{\times}Landrace$, 4th to 5th parity) were randomly allocated to 1 of 4 dietary treatments supplemented with 0, 200, 400, or 600 mg/kg $MgSO_4$ (n = 12). The experiment started on day 90 of gestation and continued through day 21 of lactation. Blood samples were collected on day 107 of gestation, day 0 (farrowing) and 21 (weaning) of lactation for the analyses of the blood biochemistry parameters and immunoglobulin levels. The colostrum and milk samples were obtained on day 0 and 14 of lactation, respectively. Fecal samples were collected from the sows on day 107 of gestation as well as day 7 and 20 of lactation to determine fecal moisture content. The results showed that the survival percentage of piglets and the litter weight at weaning were decreased linearly (p<0.05) and other parameters of the sow or litter performance were not influenced (p>0.05) by $MgSO_4$ supplementation. The fecal moisture content of the sows were increased (p<0.05) linearly as dietary $MgSO_4$ increased on day 7 and 20 of lactation. Supplementation with $MgSO_4$ increased the plasma magnesium (Mg) level linearly (p<0.05) and had a trend to increase total protein level (p>0.05 and p<0.10). However, an increase in the dietary $MgSO_4$ level resulted in a linear decrease in the colostrum fat content (p<0.05). Dietary $MgSO_4$ supplementation enhanced the immunoglobulin G (IgG) level (linear, p<0.05) in plasma on day of farrowing and immunoglobulin A (IgA) level in colostrum (quadratic, p<0.05) and milk (linear, p<0.05) of the sows. These results indicated that supplementation with $MgSO_4$ during late gestation and lactation may have the potential to prevent sow constipation, but may also result in some negative effects.
