The changes in the proteolysis activity and the accumulation of free amino acids during chinese traditional dry-cured loins processing

        Chang-Yu Zhou,Ying Wang,Dao-Dong Pan,Jin-Xuan Cao,Yin-Ji Chen,Yuan Liu,Yang-Ying Sun,Chang-Rong Ou 한국식품과학회 2017 Food Science and Biotechnology Vol.26 No.3

Twelve pieces of longissimus dorsi were processed into Chinese traditional dry-cured loins. The changes in the proteolylic enzymes activities, myofibrillar proteins degradation, and free amino acids content were investigated during processing. Compared with fresh piece (0 day), the cathepsin B ? L and calpains activities decreased after dry-curing and maintained potential activities values of 23.25 and 15.04% in the final products, respectively. The myosin heavy chain (MHC) and C protein were intensely degraded at the dry-ripened stage; the 50 kDa desmin increased at day 2 and then disappeared at day 11. The total free amino acids content increased from 333.18 mg/100 g in the raw to 1096.54 mg/100 g at the end of the dry-ripening. This work provided a mechanism for the accumulation of free amino acids and predicted the proteolysis extent of myofibrillar proteins by monitoring the changes of three marker proteins (MHC, C protein and 50 kDa desmin) during Chinese traditional dry-cured loins processing. Recent advances of bioactive proteins/polypeptides in the treatment of breast cancer

        Qi-Zhang Li,Ze-Rong Zhou,Cui-Yu Hu,Xian-Bin Li,Yu-Zhou Chang,Yan Liu,Yu-Liang Wang,Xuan-Wei Zhou 한국식품과학회 2023 Food Science and Biotechnology Vol.32 No.3

Proteins do not only serve as nutrients to fulfill the demand for food, but also are used as a source of bioactive proteins/polypeptides for regulating physical functions and promoting physical health. Female breast cancer has the highest incidence in the world and is a serious threat to women’s health. Bioactive proteins/polypeptides exert strong anti-tumor effects and exhibit inhibition of multiple breast cancer cells. This review discussed the suppressing effects of bioactive proteins/polypeptides on breast cancer in vitro and in vivo, and their mechanisms of migration and invasion inhibition, apoptosis induction, and cell cycle arrest. This may contribute to providing a basis for the development of bioactive proteins/polypeptides for the treatment of breast cancer.

      • Thermus amyloliquefaciens sp. nov., isolated from a hot spring sediment sample.

        Yu, Tian-Tian,Ming, Hong,Yao, Ji-Cheng,Zhou, En-Min,Park, Dong-Jin,Hozzein, Wael N,Kim, Chang-Jin,Wadaan, Mohammed A M,Li, Wen-Jun Society for General Microbiology 2015 International journal of systematic and evolutiona Vol.65 No.8

<P>A Gram-stain-negative, aerobic bacterium, designated strain YIM 77409T, was isolated from the Niujie hot spring in the Eryuan county of Dali, Yunnan province, south-west China. Cells of the strain were rod-shaped and colonies were yellow and circular. The strain grew at pH?6.0-8.0 (optimum, pH?7.0) and 50-70C (optimum, 60-65C). The predominant menaquinone was MK-8 and the DNA G+C content was 66.4?mol%. Major fatty acids (>10?%) were iso-C15?:?0 and iso-C17?:?0.The polar lipids consisted of one aminophospholipid, one phospholipid and two glycolipids. 16S rRNA gene sequence analysis showed that strain YIM 77409T formed a cluster with Thermus scotoductus DSM 8553T, Thermus antranikianii DSM 12462T, Thermus caliditerrae YIM 77925T and Thermus tengchongensis YIM 77924T, with highest 16S rRNA gene sequence similarity to T. scotoductus DSM 8553T (97.57?%). However, DNA-DNA hybridization indicated that strain YIM 77409T should be viewed as a representative of a novel species, as there was only 30.6??1.6?% reassociation with T. scotoductus DSM 8553T. On the basis of the morphological and chemotaxonomic characteristics, as well as the genotypic data, it is proposed that strain YIM 77409T represents a novel species of the genus Thermus, with the name Thermus amyloliquefaciens sp. nov. The type strain is YIM 77409T (?=?DSM 25898T?=?KCTC 32024T).</P>

Geometrical and Magnetic Properties of Vanadium Clusters Supported on Graphene

Yu Zou,Chang Yong Zhan,Jian-Chun Wu,Li-Ping Zhou,Hai-Xia Da 한국물리학회 2013 THE JOURNAL OF THE KOREAN PHYSICAL SOCIETY Vol.63 No.2

We report ab-initio calculations of vanadium-cluster Vn(n = 2−5) adsorption on graphene sheets. Geometrical and magnetic properties of various adsorption configurations are studied using firstprinciples density-functional theory with the generalized gradient approximation. The geometrical and magnetic properties of vanadium clusters are found to be size-dependent, and the supported graphene sheet could influence the formation of the vanadium clusters. Low-dimensional Vn cluster configurations could be easily formed when they are absorbed on a graphene sheet, and the combined Vn-graphene systems exhibit a nonmagnetic state, which is the most stable magnetic configuration. Our calculations for the geometrical and the magnetic moment properties of Vn-graphene systems may be of interest for some nanotechnological applications.

Lithium chloride’s inhibition of 3T3-L1 cell differentiation by regulating the Wnt/β-catenin pathway and enhancing villin 2 expression

        Yu-Yang Chi,Jing-Lin Shen,Jing Zhang,Anshan Shan,Shu-Ling Niu,Chang-Hai Zhou,이홍구,Yong-Cheng Jin 한국식품과학회 2016 Food Science and Biotechnology Vol.25 No.4

The aim of this study is to reveal the relation among villin 2, Wnt/β-catenin, andadipogenesis by adding appropriate lithium chloride (LiCl). The study comprises three parts: theselection of LiCl concentration, the effect of LiCl on adipocyte differentiation during and afterdifferentiation induction. By comprehensively analyzing the results of the experiments, we proved thatLiCl can inhibit adipocyte differentiation and enhance villin 2 and β-catenin expressions not only duringdifferentiation induction but also after it. Moreover, villin 2 has a significant impact on β-catenin. Wesuggest that villin 2 may participate in Wnt/β-catenin signaling.

Expression analysis of HSP70 gene in response to environmental stress in Spodoptera frugiperda (Lepidoptera: Noctuidae)

        Zhou Lv,Meng Jian-Yu,Ruan Hong-Yun,Zhang Chang-Yu 한국응용곤충학회 2023 Journal of Asia-Pacific Entomology Vol.26 No.3

This study aimed to investigate the role of heat shock proteins in the response of Spodoptera frugiperda to environmental stress. Four Hsp70 genes of S. frugiperda were cloned using reverse transcription–polymerase chain reaction (RT-PCR) and their sequence characteristics were characterized using bioinformatics analysis. RTqPCR was used to detect Hsp70 at different developmental stages, in different tissues, and under varying environmental stresses (45 ◦ C, 36 ◦ C, 4 ◦ C, ultraviolet-A, and emamectin benzoate) to quantify the relative expression of this gene. Four Hsp70 genes were obtained by cloning, namely SfHsp70-1, SfHsp70-2, SfHsp70-3, and SfHsp70-like, which encode 653, 686, 638, and 627 amino acids, respectively, and all contain the three Hsp70 family signature sequences. Neighbor-joining (NJ) method was used to construct phylogenetic tree, and four Hsp70 of the S. frugiperda species were clustered with Lepidoptera. The results of real-time PCR showed that the SfHsp70s gene was specifically expressed in different tissues at different developmental stages and its expression increased considerably under nearly all environmental stresses in adults. This study shows that the SfHsp70 gene contributes to a large extent to the resistance of S. frugiperda to environmental stress.

The Porcine FoxO1, FoxO3a and FoxO4 Genes: Cloning, Mapping, Expression and Association Analysis with Meat Production Traits

        Yu, Jing,Zhou, Quan-Yong,Zhu, Meng-Jin,Li, Chang-Chun,Liu, Bang,Fan, Bin,Zhao, Shu-Hong Asian Australasian Association of Animal Productio 2007 Animal Bioscience Vol.20 No.5

FoxO1, FoxO3a and FoxO4 belong to the FoxO gene family, which play important roles in the PI3K/PKB pathway. In this study, we cloned the porcine FoxO1, FoxO3a and FoxO4 sequences and assigned them to SSC11p11-15, SSC1p13 and SSC xq13 using somatic cell hybrid panel (SCHP) and radiation hybrid panel (IMpRH). RT-PCR results showed that these three genes are expressed in multiple tissues. Sequencing of PCR products from different breeds identified a synonymous T/C polymorphism in exon 2 of FoxO3a. This FoxO3a single nucleotide polymorphism (SNP) can be detected by AvaII restriction enzyme. The allele frequencies of this SNP were investigated in Dahuabai, Meishan, Tongcheng, Yushan, Large White, and Duroc pigs. Association of the genotypes with growth and carcass traits showed that different genotypes of FoxO3a were associated with carcass length and backfat thickness between 6th and 7th ribs (BTR) and drip loss (p<0.05).

      • Effects of the Hippo Signaling Pathway in Human Gastric Cancer

        Zhou, Guang-Xi,Li, Xiao-Yu,Zhang, Qi,Zhao, Kun,Zhang, Cui-Ping,Xue, Chang-Hu,Yang, Kun,Tian, Zi-Bin Asian Pacific Journal of Cancer Prevention 2013 Asian Pacific journal of cancer prevention Vol.14 No.9

Background/Aim: The Hippo signaling pathway is a newly discovered and conserved signaling cascade, which regulates organ size control by governing cell proliferation and apoptosis. This study aimed to investigate its effects in human gastric cancer. Methods: Tumor tissues (n=60), adjacent non-tumor tissues (n=60) and normal tissues (n=60) were obtained from the same patients with primary gastric cancer (GC). In addition, 70 samples of chronic atrophic gastritis (CAG) tissues were obtained from patients with intestinal metaplasia (IM) by endoscopic biopsy. Hippo signaling molecules, including Mst1, Lats1, YAP1, TAZ, TEAD1, Oct4 and CDX2, were determined by quantitative polymerase chain reaction (qPCR). Protein expression of Mst1, Lats1, YAP1, TEAD1 and CDX2 was assessed by immunohistochemistry and Western blotting. Results: Mst1, Lats1 and Oct4 mRNA expression showed an increasing tendency from GC tissues to normal gastric tissues, while the mRNA expression of YAP1, TAZ and TEAD1 was up-regulated (all P<0.01). Mst1 and Lats1 protein expression presented a similar trend with their mRNA expression. In addition, YAP1 and TEAD1 protein expression in GC was significantly higher than in the other groups (all P<0.01). CDX2 mRNA and protein expression in the CAG group were higher than in the other groups (all P<0.01). In GC, mRNA expression of Mst1, Lats1, Oct4, YAP1, TAZ, TEAD1 and CDX2 had a close correlation with lymphatic metastasis and tumor TNM stage (all P<0.01). Furthermore, protein expression of Mst1, Lats1, YAP1, TAZ, TEAD1 and CDX2 had a close correlation between each other (P<0.05). Conclusion: The Hippo signaling pathway is involved in the development, progression and metastasis of human gastric cancer. Therefore, manipulation of Hippo signaling molecules may be a potential therapeutic strategy for gastric cancer.

Molecular cloning, chromosomal localization and expression profiling of porcine selenoprotein M gene

        Ji-Chang Zhou,Hua Zhao,Jia-Yong Tang,Jun-Gang Li,Xiao-Li Liu,Yu-Mei Zhu 한국유전학회 2011 Genes & Genomics Vol.33 No.5

Selenoprotein M may regulate a myriad of biological processes through its redox function. In pigs, neither the nucleotide sequence nor the amino acid sequence is known. Furthermore, patterns of tissue expression and regulation by dietary selenium (Se) have not been examined. We determined the full coding sequence (CDS) and the chromosomal location of the porcine gene, SELM, and described its expression profile in vivo under different dietary Se concentrations. The cDNA sequence of porcine SELM from the start codon to the poly(A) tail was cloned by reverse transcription PCR. The CDS contained 429bases with a typical mammalian selenocysteine insertion sequence of form 2 (F2) located in the 3′-untranslated region. The gene was mapped to chromosome 14q21, where porcine SELM and its neighboring genes exhibited a similar organization to human homologues on chromosome 22q12.2. The expression pattern of SELM mRNA in muscle, thyroid, cerebral cortex, pituitary, testis, liver, and kidney was analyzed with real-time quantitative PCR in young male pigs fed a Se-deficient corn-soybean meal basal diet supplemented with 0.0, 0.3,or 3.0 mg Se/kg in the form of Se-rich yeast. Though the SELM mRNA abundance in each of the 7 tissues was not affected by the dietary Se concentrations, it was significantly higher in thyroid (P < 0.01) than in cerebral cortex, pituitary, testis, liver, and kidney at all of the 3 dietary Se concentrations.

Role of folP1 and folP2 Genes in the Action of Sulfamethoxazole and Trimethoprim Against Mycobacteria

( Tian Zhou Liu ),( Bang Xing Wang ),( Jin Tao Guo ),( Yang Zhou ),( Mugweru Julius ),( Moses Njire ),( Yuan Yuan Cao ),( Tian Wu ),( Zhi Yong Liu ),( Chang Wei Wang ),( Yong Xu ),( Tian Yu Zhang ) 한국미생물 · 생명공학회 2015 Journal of microbiology and biotechnology Vol.25 No.9

The combination of trimethoprim (TMP) and sulfamethoxazole (SMX) has been shown to be active against Mycobacterium tuberculosis (Mtb) in clinical tuberculosis (TB) treatment. However, the mechanism of action of TMP-SMX against Mtb is still unknown. To unravel this, we have studied the effect of TMP and SMX by deleting the folP2 gene in Mycobacterium smegmatis (Msm), and overexpressing the Mtb and Msm folP1/2 genes in Msm. Knocking out of the folP2 gene in Msm reduced the minimum inhibitory concentration of SMX 8-fold compared with wild type. Overexpression of the folP1 genes from Mtb and Msm increased the MICs by 4- and 2-fold in Msm for SMX and TMP, respectively. We show a strong correlation between the expression of folP1 and folP2 genes and TMP-SMX resistance in mycobacteria. This suggests that a combination of FolP2 inhibitor and SMX could be used for TB treatment with a better outcome.
