      • 향류식 역삼투공정에 의한 상용막의 에탄올 농축 특성에 관한 연구

        0The concentration characteristics for reverse osmosis and countercurrent reverse osmosis process with varing concentration of ethanol solution and applied pressure were investigated in this study. Also, the theoretical value derived for countercurrent reverse osmosis(Osmotic sink reverse osmosis) were compared with the experimental results. In RO and CCRO experiments, three kinds of commercial membrane(TFCL, UOP Co., U.S.A./FT-30, Film Tec Co., U.S.A. / Cellulose Acetate blend, UOP Co., U.S.A.), applied pressure of 3.92, 4.9 MPa and feed concentration of 3.9, 10.2 wt% were used. From the results of this study. DC(Degree of Concentration) of ethanol by TFCL membrane was the highest value of three kinds of membrane, and DC of ethanol by OSRO was the highest value of two types of ethanol concentration process. In addition, DC of OSRO more increased greately than that of RO as applied pressure difference increased. In comparison with experimental and numrical results, the numerical results were accorded with experimental results within 5-13% of error.

        Il Woong Cleft lip and palate foundation was founded in 1968 by Professor emeritus Byoung-Il Min. Since then the foundation has operated numerous cleft lip and palate patients not only in Korea but also in under-developed countries such as Vietnam, China, and Cambodia. In December 2005, the foundation was allowed as an incorporated association by Ministry of Foreign affairs and Trade, so that it could have official status. From March 6th 2009 to March 14th 2009, we conductp,d charity cleft lip and palate operations of 23 patients in areas of Deozhou, China. Hereby we present the results of operations.

        호흡기 ; 만성기도질환에서 호기말 흉부 CT를 이용한 기관협착에 관한 연구

        목적: 기관과 기관지와 같은 대기도는 호기시에 어느 정도의 협착이 정상적으로도 발생하게 된다. 이와 같은 호시기 대기도의 협착이 만성 기도질환에서 정상인과 차이가 있는지에 대하여는 알려진 바가 많지 않다. 따라서 저자들은 만성 기도질환으로 흉부 CT 촬영을 하였던 환자에서 호기말 CT 영상을 이용한 기관협착 양상을 알아보고자 하였다. 방법: 만성 기도질환으로 흉부 고해상 CT와 폐활량검사를 시행받았던 모든 경우에서 호기말 scan을 확인할 수 있었던 환자 45명과 정상 대조군 22명을 대상으로 하였다. 호기말 CT에서 대동맥궁 위치의 스캔을 선택하고 흡기말 스캔 중 같은 위치의 스캔을 선정하여 의료용 영상 프로그램 내 측정도구로 각각의 기관내부 면적과 전후 직경을 측정하였고, 측정된 기관내부 면적과 전후 직경의 호기시에 줄어 드는 정도를 백분율로 표시하여 이를 폐활량 검사 등의 임 상지표와 비교분석 하였다. 결과: 호기말 평균 기관내부 면적 감소율은 환자군에서 13.3%, 정상 대조군에서 9.0%이었으며, 호기말 기관 전후직경 감소율은 각각 11.8%와 9.6%였다. 기관내부 면적 감소율은 기관지확장증에서 평균 14.3%로 가장 많았으나 질환군에 따른 호기시 기관내부 면적 감소율에 유의한 차이는 없었다. 기관내부 면적 감소율 28% 이상의 협착소견이 있었던 경우는 3명(6.7%)이었으며 이들의 기저질환은 만성폐쇄성폐 질환이 2명, 기관지확장증이 1명이었다. 환자군과 정상 대조군을 포함하는 전체인원을 대상으로 하여 분석하였을 때 기관내부 면적 감소율은 강제폐활량 예측치 %(r =-0.33, p=0.01), 1초간 강제날숨량 예측치 %(r =-0.27, p=0.03)와 상관관계를 보였으나, 전후직경 감소율은 폐활량지표와 의미 있는 관련성을 보이지 않았다. 결론: 흉부 CT를 이용하여 측정된 호기말 기관 내부면적의 감소는 만성 기도질환에서 정상군에 비하여 심하였으며 이는 폐기능과 관련이 있었다. Background/Aims: Partial tracheal narrowing can occur during expiration in the normal population. It is not certain whether the trachea collapses more readily in chronic airway disease. We evaluated the tracheal narrowing at end-expiration using computed tomography (CT). Methods: We investigated 45 patients and 22 normal subjects who underwent high-resolution CT and pulmonary function tests. In each subject, two CT images at the same level of the aortic arch were compared: one at end-inspiration and the other at end-expiration. The cross-sectional area and sagittal diameter of the trachea were measured using a hand-tracing method, using the in-program measuring tools of Medical Image Viewer, and the percentage changes of each value were calculated. Results: Of the 45 patients with chronic airway disease, 21 had chronic obstructive pulmonary disease, 16 had bronchial asthma, and 8 had bronchiectasis. The mean change in the cross-sectional area was 13.3% in the patients and 9.0% in the normal subjects (p<0.05), and the mean change in sagittal diameter was 11.8 and 9.6%, respectively (p>0.05). The decrease in tracheal cross-sectional area was greatest in bronchiectasis, while the greatest decrease in sagittal diameter was in bronchial asthma. There was no significant difference in tracheal collapsibility among the disease groups. The percent change in the tracheal cross-sectional area was correlated with the % predicted FVC (r = -0.033, p<0.05) and FEV1 (r = 0.277, p<0.05) in both the patients and normal controls. Conclusions: The decrease in tracheal cross-sectional area at end-expiration measured using chest CT was greater in chronic airway disease than in normal subjects, and was associated with ventilatory function.         Systemic immune modulation using chemokine receptor 7 expressing porcine Sertoli cells

        <P>Abstract: </P><P>Background: </P><P>Sertoli cells (SC) are known to have active mechanism for evading humoral immune response and are known to have immune modulatory effects in the presence of other antigens. This has led us to hypothesize that systemic immune modulating effect of SC might be optimized by their residence on peripheral lymph node. This study was designed to evaluate our new strategy to promote preventive or therapeutic effects of SC in systemic immune modulation for organ transplantation. </P><P>Methods: </P><P>For this purpose, we created chemokine receptor 7 (CCR7) expressing porcine SC (NPSCi-CCR7) to facilitate their migration into lymphoid organ in vivo and their potential to modulate systemic immune responses was evaluated using mouse allogeneic skin graft model. </P><P>Results: </P><P>Directed migration of NPSCi-CCR7 cells from periphery into lymphoid organs was dramatically promoted compared to control NPSCi cells. Also pre-transplantation of NPSCi-CCR7 significantly suppressed lymphocyte proliferation and prolonged the allogeneic skin graft survival. </P><P>Conclusion: </P><P>These results suggest that our new strategy to traffic SC to lymphoid organs using CCR7 is very effective and can be extended to traffic other immune modulatory cells or proteins to primary and secondary lymphoid structures to augment their therapeutic potential.</P>

      • NecroX-5 suppresses sodium nitroprusside-induced cardiac cell death through inhibition of JNK and caspase-3 activation.

        <P>Although sodium nitroprusside (SNP) is an effective hypotensive drug and is often used in pediatric intensive care units and to treat acute heart failure, clinical application of SNP is limited by its cardiotoxicity. NecroX-5 (NX-5) was recently developed and has the capacity to inhibit necrotic cell death. No current literature addresses whether NX-5 suppresses SNP-induced cell death or its mechanism of action. We have investigated the protective role of NX-5 against SNP-induced cell death in cardiomyocyte-like H9c2 cells. SNP treatment induced severe cell death, possibly through phosphorylation of stress-activated protein kinase/c-Jun NH2-terminal kinase (JNK) and activation of the apoptotic signaling pathway, including downregulation of Bcl-2 and cleavage of caspase-3. However, NX-5 suppresses SNP-induced cell death through inhibition of JNK activation and suppression of both downregulation of Bcl-2 protein expression and caspase-3 cleavage. These findings will provide insights and facilitate development of antidotes to SNP toxicity in cardiac cells.</P>

        건일로딘 정(미결정에토돌락 200 ㎎)에 대한 에토돌 정의 생물학적동등성

        A bioequivalence of Etodol™ tablets (Yuhan corporation) and Kuhnillodine™ tablets (Kuhnil Pharm, Co., Ltd.) was evaluated according to the guideline of Korea Food and Drug Administration (KFDA). Single 200 ㎎ dose of etodolac of each medicine was administered orally to 24 healthy male volunteers. This study was performed in a 2×2 cross-over design. Concentrations of etodolac in human plasma were monitored by a high-performance liquid chromatography. AUCt (the area under the plasma concentration-time curve from time zero to 24 hr) was calculated by the linear trapezoidal rule method. C_(max) (maximum plasma drug concentration) and T_(max) (time to reach C_(max)) were compiled from the plasma concentration-time data. Analysis of variance was performed using logarithmically transformed AUCt and C_(max). No significant sequence effect was found for all of the bioavailability parameters. The 90% confidence intervals of the AUCt ratio and the C_(max) ratio for Etodol™/Kuhnillodine™ were 1.01 - 1.10 and 0.87 - 1.06, respectively. This study demonstrated a bioequivalence of Etodol™ and Kuhnillodine™ with respect to the rate and extent of absorption.

        Establishment and characterization of porcine Sertoli cell line for the study of xenotransplantation

        <P>Abstract: </P><P>Background: </P><P>An understanding of the main mechanism that determines the ability of immune privilege related to Sertoli cells (SC) will provide clues for promoting a local tolerogenic environment. In this report, we established neonatal porcine SC line and evaluated their characteristics.</P><P>Methods: </P><P>SC line was established following the transfection of primary SC (NPSC) from the testis of neonatal pig with plasmid pRNS-1 carrying genes for neomycin resistance and the SV40 large T antigen. Immunohistochemistry and RT-PCR were performed to evaluate the character of immortalized SC lines.</P><P>Results: </P><P>Our immortalized SC line (iPS) proliferated stably and had a phenotype similar to NPSC, as indicated by the immunoexpression of follicle stimulating hormone receptor (FSHR), and mRNA expression of androgen receptor (AR), and Wilms’ tumor antigen (WT1). Interestingly, NPSC and iPS expressed mRNA of complement regulatory proteins (CRP) such as membrane cofactor protein (CD46), decay accelerating factor (DAF or CD55), and protectin (CD59), but CD59 mRNA expression was negligible in iPS.</P><P>Conclusion: </P><P>These results suggest that iPS, immortalized by the introduction of SV40 T, retain their original characteristics, except for the relatively low expression of CD59, and that they may be useful for future in vitro and in vivo studies of immune privilege mechanisms related to SC.</P>

      • Age-dependent expression of immune-privilege and proliferation-related molecules on porcine Sertoli cells

        <P>AbstractBackground</P><P>The immunoprotective nature of the testes has prompted numerous investigations into their supportive roles during allogeneic or xenogeneic cellular grafts. However, the optimal developmental stage of these cells in terms of maximum efficacy for cellular grafts has not been elucidated. In this study, the time-dependent expressions of immune-privilege- and proliferation-related molecules in Sertoli cells were determined.</P><P>Methods</P><P>To investigate the time course of the expression of proteins related to immune privilege and proliferation, immunohistochemistry and reverse transcriptase-polymerase chain reaction (RT-PCR) were performed using testes of 18 Yorkshire pigs from birth to 20 weeks of age. We included fas ligand (FasL), transforming growth factor β1 (TGF-β1) and clusterin as the immune-protective molecules, and follicle stimulating hormone receptor (FSHR), p27<SUP>Kip1</SUP>, GATA-4 and Wilm's tumor antigen (WT1) as Sertoli cell proliferation markers. RT-PCR was used to complement histological assessment.</P><P>Results</P><P>The expression of FasL in Sertoli cells gradually increased with age, whereas TGF-β1 showed the reverse pattern, and clusterin expression showed no age-related difference. p27<SUP>Kip1</SUP> showed a gradual increase in its expression from week 12, but GATA-4 showed earlier postnatal expression from birth to week 12. WT1 expression gradually increased from week 16.</P><P>Conclusion</P><P>We confirmed the age-dependent expression of immune-privilege- and proliferation-related molecules in porcine Sertoli cells. These data may be useful for determining the optimal time for harvesting Sertoli cells with respect to maximal immunoprotection and proliferative activity.</P>
