        Naringin and Naringenin Relax Rat Tracheal Smooth by Regulating BKCa Activation

        Naringin and its aglycone, naringenin, occur naturally in our regular diet and traditional Chinese medicines. This study aimed to detect an effective therapeutic approach for cough variant asthma (CVA) through evaluating the relaxant effect of these two bioactive herbal monomers as antitussive and antiasthmatic on rat tracheal smooth muscle. The relaxant effect was determined by measuring muscular tension with a mechanical recording system in rat tracheal rings. Cytosolic Ca2+ concentration was measured using a confocal imaging system in primary cultured tracheal smooth muscle cells. In rat tracheal rings, addition of both naringin and naringenin could concentration dependently relax carbachol (CCh)-evoked tonic contraction. This epithelium-independent relaxation could be suppressed by BaCl2, tetraethylammonium, and iberiotoxin (IbTX), but not by glibenclamide. After stimulating primary cultured tracheal smooth muscle cells by CCh or high KCl, the intracellular Ca2+ increase could be inhibited by both naringin and naringenin, respectively. This reaction was also suppressed by IbTX. These results demonstrate that both naringin and naringenin can relax tracheal smooth muscle through opening big conductance Ca2+-activated K+ channel, which mediates plasma membrane hyperpolarization and reduces Ca2+ influx. Our data indicate a potentially effective therapeutic approach of naringin and naringenin for CVA.

      • Synthesis and characterization of monodisperse yttrium aluminum garnet (YAG) micro-crystals with rhombic dodecahedron

        <P><B>Abstract</B></P> <P>Uniform, monodisperse yttrium aluminum garnet (YAG) rhombic dodecahedron micro-crystals were synthesized using coprecipitation and hydrothermal methods. The Na<SUB>2</SUB>SO<SUB>4</SUB> as a surfactant was helpful in improving the dispersity of the crystallites and forming well-faceted, micro-sized dodecahedral YAG crystallites. The effect of the reaction time and the solvent has also been studied. The crystal growth mechanism called dissolution/crystallization was determined by analyzing the experimental and simulated results. And the specific well-developed crystal facets in the {110} family have been demonstrated by the theoretical calculation. A higher integrated emission intensity can be achieved for the YAG:Ce crystallites by improving the phase purity, morphology with better developed dodecahedrons, and dispersion. Furthermore, a rather high luminous efficacy (LE = 104.14 lm/W) for the white LED fabricated using the YAG:Ce<SUP>3+</SUP> phosphor prepared in this work was realized.       • A Discovery of Low Hydraulic Resistance Channel Along Meridians

        A hydro-mechanic model was put forward to study the fundamental nature of acupuncture meridians. The basic state of low hydraulic resistance was tested on humans and mini pigs using three methods. The first, a modified Guyton’s method, proved that there was lower hydraulic resistance on meridians compared with nonmeridians. The second scanning method involved a single pressure transducer that can find the lowest resistance point in tissue, and the third method used two transducers and provided a more stable measurement. Using the latter method, low hydraulic resistance points were found very close to low impedance points along meridians. The transmission of artificial interstitial fluid pressure waves was measured to examine their connection to the low resistance points, with the result that a good connection between the points was confirmed. This means the points form channels along the meridians that we refer to as low hydraulic resistance channels. The channel was imaged through isotopic tracing and a migration of isotope 99mTe could be found along the channel. The layer of the channel was detected by injecting Alcian blue and the track was found beneath the skin. All of the above experiments suggest the existence of a new type of channel in living tissues that has not yet been described in modern science, but coincides quite well with the Qi channel theory of traditional Chinese medicine.       • Activin pathway enhances colorectal cancer stem cell self-renew and tumor progression

        <P><B>Abstract</B></P> <P>Activin belongs to transforming growth factor (TGF)-β super family of growth and differentiation factors and activin pathway participated in broad range of cell process. Studies elaborated activin pathway maintain pluripotency in human stem cells and suggest that the function of activin/nodal signaling in self-renew would be conserved across embryonic and adult stem cells. In this study, we tried to determine the effect of activin signaling pathway in regulation of cancer stem cells as a potential target for cancer therapy in clinical trials. A population of colorectal cancer cells was selected by the treatment of activin A. This population of cell possessed stem cell character with sphere formation ability. We demonstrated activin pathway enhanced the colorectal cancer stem cells self-renew and contribute to colorectal cancer progression in vivo.         An automatic 3D CAD model errors detection method of aircraft structural part for NC machining

        Feature-based NC machining, which requires high quality of 3D CAD model, is widely used in machining aircraft structural part. However, there has been little research on how to automatically detect the CAD model errors. As a result, the user has to manually check the errors with great effort before NC programming. This paper proposes an automatic CAD model errors detection approach for aircraft structural part. First, the base faces are identified based on the reference directions corresponding to machining coordinate systems. Then, the CAD models are partitioned into multiple local regions based on the base faces. Finally, the CAD model error types are evaluated based on the heuristic rules. A prototype system based on CATIA has been developed to verify the effectiveness of the proposed approach.

      • Design of a Bidirectional DC/AC Converter with Battery Charging/Discharging/Standing Balance Control

        Battery cells connected in series have been widely used in high-voltage and high-power applications. In this paper, a single-phase battery energy storage system with battery balance charging, battery balance discharging, and power factor correction capabilities was developed. A prototype suitable for a single-phase 110V power supply was designed and implemented for verification. To verify its performance, three 48V/7Ah battery modules were used as normal batteries, and a 48V/5Ah battery module was used as a retired battery. The experimental results showed that the battery energy storage system has excellent battery balancing capability. Compared with that of the conventional system without balanced control, the balancing performance of the proposed system is increased by about 15.25% and 26.92%, respectively, when the system was operated in the converter and rectifier modes. In addition, the proposed system also had battery fault tolerance, and was compatible with recycled batteries and an independent power supply.

        The Distribution of Transcutaneous CO2 Emission and Correlation With the Points Along the Pericardium Meridian

        This study aimed to understand energy metabolism distribution along the pericardium meridian and verify the correlation between the body surface (points), and classic meridian theory. A highly sensitive CO2 instrument was used to measure the transcutaneous CO2 emission at 13 points along the pericardium meridian line (12 points on the line and one point beyond the line) and 13 control points beside them. Results showed that the distribution of transcutaneous CO2 emission is highly related to the position on the body. Transcutaneous CO2 emission is significantly higher at P7 and P3, than the control points beside them. The points along the meridian and the points beside them were clustered with relative distance by SAS statistics software. Two distance matrixes were then obtained. The correlation coefficients between the points along the line and between the control points were calculated. The results showed that the 13th point beyond the line was far from the 12 points on the line (distance, 0.24), while acupoints on the line clustered earlier when compared with the non-acupoints. The average correlation coefficients among the acupoints was 0.65 which was significantly higher than 0.56, among the non-acupoints. No such characteristics were found among the control points. It was concluded that there is a strong correlativity of energy metabolism activity between the body surfaces along the meridian, and an even stronger correlativity between the acupoints on the meridian.         Cloning, Heterologous Expression, and Characterization of Novel Protease- Resistant α-Galactosidase from New Sphingomonas Strain

        The α-galactosidase-coding gene agaAJB13 was cloned from Sphingomonas sp. JB13 showing 16S rDNA (1,343 bp) identities of ≤97.2% with other identified Sphingomonas strains. agaAJB13 (2,217 bp; 64.9% GC content) encodes a 738-residue polypeptide (AgaAJB13) with a calculated mass of 82.3 kDa. AgaAJB13 showed the highest identity of 61.4% with the putative glycosyl hydrolase family 36 α-galactosidase from Granulicella mallensis MP5ACTX8 (EFI56085). AgaAJB13 also showed <37% identities with reported protease-resistant or Sphingomonas α-galactosidases. A sequence analysis revealed different catalytic motifs between reported Sphingomonas α-galactosidases (KXD and RXXXD) and AgaAJB13 (KWD and SDXXDXXXR). Recombinant AgaAJB13 (rAgaAJB13) was expressed in Escherichia coli BL21 (DE3). The purified rAgaAJB13 was characterized using p-nitrophenyl-α-D-galactopyranoside as the substrate and showed an apparent optimum at pH 5.0 and 60oC and strong resistance to trypsin and proteinase K digestion. Compared with reported proteaseresistant α-galactosidases showing thermolability at 50oC or 60oC and specific activities of <71 U/mg with or without protease treatments, rAgaAJB13 exhibited a better thermal stability (half-life of >60 min at 60oC) and higher specific activities (225.0-256.5 U/mg). These sequence and enzymatic properties suggest AgaAJB13 is the first identified and characterized Sphingomonas α-galactosidase, and shows novel protease resistance with a potential value for basic research and industrial applications.

        Synthesis, Antibacterial Activity, and Structure–Activity Relationship of Fusaric Acid Analogs

        Forty-one fusaric acid analogs possessing a pyridine carboxylic acid scaffold have been synthesized. The antibacterial activity results demonstrated that compounds 5b, 7b, 8c, and 8d displayed strong antibacterial activities against Staphylococcus aureus ATCC25923 with minimum inhibitory concentrations (MICs) of 4–16 μg/mL. Molecular docking study indicated that these compounds have strong hydrogen-bonding interactions with TyrRS. Meanwhile, 8c and 8d showed promising antibacterial activities against Pseudomonas aeruginosa ATCC9027. Compound 4 exhibited pronounced antibacterial activities against a clinically isolated multidrug-resistant strain of Escherichia coli (MIC: 64 μg/mL as compared 64 μg/mL of levofloxacin and 1024 μg/mL of ceftriaxone sodium). Moreover, compound 17e displayed strong synergistic antibacterial effect with levofloxacin against the multidrug-resistant strain, decreasing the MIC value of levofloxacin to 1/16 of its original MIC. No obvious cytotoxic activities against LO2 was observed for compounds 4, 5b, 8c, 8d, 17d, and 17e at 50 μM. The preliminary structure–activity relationship of fusaric acid analogs was also discussed.

        LncRNA TMPO-AS1 promotes esophageal squamous cell carcinoma progression by forming biomolecular condensates with FUS and p300 to regulate TMPO transcription

        Esophageal squamous cell carcinoma (ESCC) is one of the most life- and health-threatening malignant diseases worldwide, especially in China. Long noncoding RNAs (lncRNAs) have emerged as important regulators of tumorigenesis and tumor progression. However, the roles and mechanisms of lncRNAs in ESCC require further exploration. Here, in combination with a small interfering RNA (siRNA) library targeting specific lncRNAs, we performed MTS and Transwell assays to screen functional lncRNAs that were overexpressed in ESCC. TMPO-AS1 expression was significantly upregulated in ESCC tumor samples, with higher TMPOAS1 expression positively correlated with shorter overall survival times. In vitro and in vivo functional experiments revealed that TMPO-AS1 promotes the proliferation and metastasis of ESCC cells. Mechanistically, TMPO-AS1 bound to fused in sarcoma (FUS) and recruited p300 to the TMPO promoter, forming biomolecular condensates in situ to activate TMPO transcription in cis by increasing the acetylation of histone H3 lysine 27 (H3K27ac). Targeting TMPO-AS1 led to impaired ESCC tumor growth in a patient-derived xenograft (PDX) model. We found that TMPO-AS1 is required for cell proliferation and metastasis in ESCC by promoting the expression of TMPO, and both TMPO-AS1 and TMPO might be potential biomarkers and therapeutic targets in ESCC.
