      • Characterization and Resistance Mechanisms of A 5-fluorouracil-resistant Hepatocellular Carcinoma Cell Line

        Purpose: The chemoresistance of human hepatocellular carcinoma (HCC) to cytotoxic drugs, especially intrinsic or acquired multidrug resistance (MDR), still remains a major challenge in the management of HCC. In the present study, possible mechanisms involved in MDR of HCC were identified using a 5-fluorouracil (5-FU)-resistant human HCC cell line. Methods: BEL-7402/5-FU cells were established through continuous culturing parental BEL-7402 cells, imitating the pattern of chemotherapy clinically. Growth curves and chemosensitivity to cytotoxic drugs were determined by MTT assay. Doubling times, colony formation and adherence rates were calculated after cell counting. Morphological alteration, karyotype morphology, and untrastructure were assessed under optical and electron microscopes. The distribution in the cell cycle and drug efflux pump activity were measured by flow cytometry. Furthermore, expression of potential genes involved in MDR of BEL-7402/5-FU cells were detected by immunocytochemistry. Results: Compared to its parental cells, BEL-7402/5-FU cells had a prolonged doubling time, a lower mitotic index, colony efficiency and adhesive ability, and a decreased drug efflux pump activity. The resistant cells tended to grow in clusters and apparent changes of ultrastructures occurred. BEL-7402/5-FU cells presented with an increased proportion in S and G2/M phases with a concomitant decrease in G0/G1 phase. The MDR phenotype of BEL-7402/5-FU might be partly attributed to increased drug efflux pump activity via multidrug resistance protein 1 (MRP1), overexpression of thymidylate synthase (TS), resistance to apoptosis by augmentation of the Bcl-xl/Bax ratio, and intracellular adhesion medicated by E-cadherin (E-cad). P-glycoprotein (P-gp) might play a limited role in the MDR of BEL-7402/5-FU. Conclusion: Increased activity or expression of MRP1, Bcl-xl, TS, and E-cad appear to be involved in the MDR mechanism of BEL-7402/5-FU.

      • Inventory Prediction Research Based on the Improved BP Neural Network Algorithm

        Since the idea of the supply chain management is proposed, many enterprises have attached great importance to the supply chain management and pay a lot of manpower and resources to study. It is also the focus to study the inventory in the field of the supply chain. Quantity of the inventory is not only related to the profit of the enterprises, but also related to the survival of the entire supply chain. Predicting the inventory can improve the ability of enterprises to prevent risk, increase the profits and reduce the losses. In order to predict better on inventory, we propose an improved BP neural network algorithm. In the algorithm, we use the improved GSA algorithm to optimize the parameters of BP neural network algorithm and improve the BP neural network algorithm aiming at its deficiency. The experimental results show that this method has good prediction effect.

      • Understanding Users’ Continuance Intention in Cloud Computing Services : An Integrated Model

        The Cloud Computing has become one of the hottest issues in the world of information technology (IT). However, the understanding of the determinants of user’s continuance intention in the context of cloud computing services is still limited. To fill the gap, this article attempts to demonstrate the factors that influence individuals’ continuance intention of cloud computing services. In this study, we adopted the Stimulus-Organism-Response (SOR) framework as research framework. And then, on the basis of the IS success model, UTAUT model and TTF, this research proposed an integrated model to investigate the factors that affect the cloud computing continuance intention. Furthermore, perceived functionality fit is regarded as a significant intervening variable and tested in the research..

        한국과 중국의 실화영화의 특징에 대한 비교 분석

        한국과 중국에서는 실화를 바탕으로 관객들에게 많은 사랑을 받을 만한 영화 작품들이 쏟아져 나오기 했다. 한중 양국의 사회 제도와 영화 심사 체제의 차이로 인해 한중 양국의 영화는 현저한 차이를 보이게 되었다. 연구에 보다 믿을 수 있는 데이터 근거를 제시하기 위해 인용한 데이터는 전부 “도우반망”의 데이터를 사용할 예정이다. 세 가지 방법으로 분석한다: 관객의 영화 관심도와 인정도 피드백, 인물 캐릭터 구축특성, 영화 결말.이것은 중국 영화 관객의 피드백에서 한국 실화영화와 중국 실화영화의 차이점을 분석하며, 한국의 실화 영화가 보다 정확하게 중국 관객들의 수요를 충족시키고 있으며 상대적으로 중국의 실화영화는 대중들의 수요 욕구를 정확히 반영하지 못하고 있다. 동시에 한국과 중국의 실화영화의 내러티브 특성에 있어서의 몇 가지 서사적 측면에서의 차이점을 반영 할 것이다. 한국의 경우 대중들이 관심을 갖는 해결되지 않은 사회적 문제를 소재로 하는 영화가 대부분이었으나 중국의 경우 중국 사회관리계층들이 대중들의 애국심을 고취시키고자 하는 관점에서 선택한 소재의 영화가 대부분이었다는 것을 알 수 있었다. This analyzes in the feedback of Chinese moviegoers, and more precisely South Korean true story films are meeting Chinese audiences' needs, while relatively Chinese true story movies do not accurately reflect the needs of the public. At the same time, it will reflect differences narrative Characteristics of the true films of Korea and China. In Korea, most movies narrative were based on unresolved social issues that the public is interested in , but in China, most of them were made narrative from the perspective of Chinese social officials wanting to inspire the public's patriotism.

        한국과 중국의 현실 소재 영화 대한 비교 분석 - 영화 “도가니”와 “천사가 흰옷을 입다” 중심으로 -

        최근 한국과 중국에서 현실 소재를 다룬 유형의 영화들은 사회적으로 매우 큰 영향을 미치고 있다. 특히 한국에서 제작된 사회 비판적인, 사회의 아픔을 직면하며 이를 대담하게 드러내고 있는 영화들이 특히 사회적으로 더 강력한 영향을 끼치고 있다. 중국의 현실 소재 영화의 경우에도 중국만의 독특한 예술 스타일을 자랑하며 영향력을 높이고 있다. 따라서 본 논문에서는 중국과 한국에서 가장 대표성을 띠는 영화 작품을 각각 한 작품씩 선정하여 이에 관한 비교 연구를 진행해보고자 한다. 비교 대상인 두 영화는 스토리 소재, 사회적 영향, 관객들의 동일시와 같은 측면에서 유사성을 띠고 있다. 하지만 영화 표현 측면과 관객이해 측면에서 어느 정도 차이를 보인다. 본 논문에서는 기호학적 관점에서 연출자의 코딩부터 관객의 이해에까지 이르는 과정에 대해 자세하고 구체적인 인물 및 은유 분석을 진행하였다. 한국 영화에서는 대담한 전달 기법과 정교하고 시청각적 언어 활용이 돋보이며 막대한 사회적 영향력을 지니고 있다. 중국 영화에서는 전반적으로 직접적인 표현을 극도로 자제하고 있으며, 다양한 은유 기법과 간접적이고 부드러운 사회적 피드백을 고수하고 있다. In recent years, both Korean and Chinese Realistic theme category movies have exerted great influence, especially those movies in South Korea that awaken the world to criticize the society, face up to social pain and illness, and express themselves boldly. At the same time, Chinese films also have their own unique artistic styles. Therefore, this thesis makes a comparative study of the most representative films from both countries. These two films have similar story materials, social influence and audience identification, as well as different film expression and audience understanding. From the perspective of semiotics, this paper makes a detailed and specific analysis of the characters and metaphors in the process from the director's coding to the audience's decoding. China is characterized by rich metaphors, restrained expression, indirect and gentle social feedback, while Korea is characterized by bold expression, profound audio-visual language use and great social influence.

        An efficient protocol for Agrobacterium-mediated transformation of the biofuel plant Jatropha curcas by optimizing kanamycin concentration and duration of delayed selection

        Jatropha curcas is considered a potential biodiesel feedstock crop. Currently, the value of J. curcas is limited because its seed yield is generally low. Transgenic modification is a promising approach to improve the seed yield of J. curcas. Although Agrobacterium-mediated genetic transformation of J. curcas has been pursued for several years, the transformation efficiency remains unsatisfying. Therefore, a highly efficient and simple Agrobacterium-mediated genetic transformation method for J. curcas should be developed. We examined and optimized several key factors that affect genetic transformation of J. curcas in this study. The results showed that the EHA105 strain was superior to the other three Agrobacterium tumefaciens strains for infecting J. curcas cotyledons, and the supplementation of 100 mM acetosyringone slightly increased the transient transformation frequency. Use of the appropriate inoculation method, optimal kanamycin concentration and appropriate duration of delayed selection also improved the efficiency of stable genetic transformation of J. curcas. The percentage of b-glucuronidase positive J. curcas shoots reached as high as 56.0 %, and 1.70 transformants per explant were obtained with this protocol. Furthermore, we optimized the root-inducing medium to achieve a rooting rate of 84.9 %. Stable integration of the T-DNA into the genomes of putative transgenic lines was confirmed by PCR and Southern blot analysis. Using this improved protocol, a large number of transgenic J. curcas plantlets can be routinely obtained within approximately 4 months. The detailed information provided here for each step of J. curcas transformation should enable successful implementation of this transgenic technology in other laboratories.

        Fulvifomes nonggangensis and F. tubogeneratus (Hymenochaetales, Basidiomycota): Two New Species from Southern China Based on Morphological and Molecular Evidences

        Two new species of Fulvifomes are described from specimens collected in rainforests of Nonggang Nature Reserve of southern China, based on morphological characteristics and molecular phylogenetic analysis of the internal transcribed spacer (ITS) and nuclear large subunit ribosomal DNA (nLSU) sequences. Fulvifomes nonggangensis sp. nov. is characterized by perennial, sessile and solitary basidiocarps, applanate pileus, small cystidioles of 9.9-15.4×2.9-3.5 lm, large pores of 5-6 per mm, a dimitic hyphal system, and broadly ellipsoid basidiospores of 4.3-5.3×3.3-4.2 lm. F. tubogeneratus sp. nov. is characterized by perennial, sessile, and imbricate basidiocarps, a duplex context, small pores of 7-8 per mm, a dimitic hyphal system, and ovoid to subglobose basidiospores of 5.72×5.00 lm.

        遼西地區漢以前文化發展序列的建立及文化縱橫關系的探討

        位於醫巫閭山以西幷以西拉木倫河、老哈河、敎來河和大、小凌河流域爲重心的遼西山地地區, 行政區화爲中國遼寧省的西部和內蒙古自治區的東南部。這一地區目前已識別出來的漢以前考古學文化共有15種, 有6種爲新石器時代的文化遺存, 9種爲夏至戰國時期的文化遺存。屬於新石器時代的6種考古學文化, 年代最早的是小河西文化, 最晩的是小河沿文化, 兩者之間依次是``興隆와文化→趙寶구文化→富河文化→紅山文化``。遼西地區已發現的處於夏至戰國時期的9種考古學文化,分別爲夏家店下層文化、魏營子類型(或稱魏營子文化)、夏家店上層文化、凌河類型(或稱凌河遺存)、水泉類型(或稱水泉遺存)、井구子類型(或稱井구子遺存)、鐵匠구類型(或稱鐵匠구遺存)、五道河子遺存和戰國燕文化。而將此遼西地區諸考古學文化統一화分爲12個大的發展階段。關於這些遺存的文化性質,絶大多數學者認爲타們都是各自獨立的考古學文化, 筆者也基本贊同這樣的宏觀認識。但在遼西地區, 文化發展存在著明顯承襲關系的和不明顯承襲關系的。只有小河西文化、興隆와文化、趙寶구文化、紅山文化、小河沿文化、夏家店下層文化6種是明顯承襲關系的。這6種考古學文化處在第1至第8階段, 彼此之間構成了一個較長的文化련條, 盡管在各個不同的發展階段上타們各自對外、對內有過文化成빈上的輸出與輸入, 但是從主體文化因素的傳承上看應該同屬於一個大的文化系統。與此相反, 有早晩關系但不存在承襲關系的文化有7種, 타們分別爲魏營子類型、夏家店上層文化、水泉類型、井구子類型(含鐵匠구類型)、五道河子類型、凌河類型和燕文化。遼西地區漢以前文化在與周邊文化關系方面所表現出的交流方式是多樣的。時間不同, 方式也有所不同。具體說來, 可分爲三個時期。早期以1~3段的小河西文化、興隆와文化、趙寶구文化爲代表,文化的交流方式是向外輸出式的。中期以4~7段的紅山文化和小河沿文化爲代表, 文化的交流方式是向內吸入式的。晩期以8~12段的夏家店下層文化、魏營子類型、凌河類型、水泉類型、井구子類型(含鐵匠구類型)、五道河子類型、燕文化爲代表, 文化的來源和去向均表現爲外地文化的遷入(簡稱遷入式)或本地文化的徙出(簡稱徙出式)。

        Amylosporus sulcatus sp. nov. (Russulales, Basidiomycota) from Southern China

        Amylosporus sulcatus sp. nov. is described from Nonggang Nature Reserve, southern China, on the basis of morphological and molecular data. The morphological description and illustrations for the new species are provided. The species is characterized by pileate and stipitate basidiocarps. The pileus surface is obviously concentrically and radiately sulcate and tomentum, and the pore surface is snow white. Phylogenetic analyses based on sequences of the internal transcribed spacer and nuclear large subunit ribosomal DNA confirmed it to be a new species.
