      • Effects of Wushu-Sanda Player`s Different Exercise Intensity on the Fatigue Substance, Oxidative Stress and Antioxidant Enzyme

        Purpose: The purpose of this study was designed to examine the activation of Fatigue Substance, Oxidative Stress and Antioxidant Enzyme in Wushu Sanda players according to the different exercise intensity. Methods & Materials: Subjects of this four-week-long Wushu exercise program were divided into 2 groups: high intensity group (n=8), and low intensity group (n=8). Subjects participated in this program once a day (high intensity group:60mins, low intensity group:90mins) and 5 times per week (Monday to Friday). Every subjects’ fatigue substance (LAC, LDH), oxidative stress (MDA) and antioxidant enzyme (GPx, SOD, CAT) were measured before the program, immediately after exercise, after 30mins recovery and after 60mins recovery. All these parameters were measured again four-week latter. Statistics: ANOVA, paired t-test and repeated ANOVA were carried out to analyze the changes before and after the exercises, changes in terms of fatigue substance, oxidative stress and antioxidant enzyme, respectively. Statistical significance is established at the .05 level (p<.05). Result: 1. Change of LAC and LDH Pre and Post exercises, analysis of the LAC have statistically significant changes in recovery 30 and 60minutes than immediately after within each group (p<.05). But according to the time of measurement there are no statistically significant difference (p>.05). And, after four weeks, LAC in both groups are lower than the concentration of before exercise. Especially, the low intensity exercise group are lower than the high intensity exercise group. But all variables are no statistically significant differences(p<.05). Pre and Post exercises, analysis of the LDH was statistically significant changes in immediately after and recovery 60minutes within each group(p<.05). And according to the time of measurement were only significantly difference in the rest (p<.05). What’s more, the high intensity exercise group are lower than the low intensity exercise group (p>.05). 2. Change of MDA MDA in low intensity group was significantly higher after exercise and 30mins recovery than before exercise (p<.05). In high intensity group, no significant difference was found. 3. Change of GPx, SOD, CAT After the four weeks exercise, GPx in high intensity group shows significant differences after 30mins and 60mins recovery compared with the value of pre-exercise (p<.05). No significant difference was found in low intensity group. Compared to the value of SOD pre-exercise, the SOD of post-exercise shows significant differences after 30mins and 60mins recovery in high intensity group ( p<.05) while only immediately after exercise in low intensity group (p<.01 ). In CAT, a significant difference was found in high intensity group after the four weeks exercise (p<.05). No significant difference was found in low intensity group. Conclusion: After four weeks exercise, some differences were found in fatigue substance(LAC, LDH), oxidative stress(MDA) and antioxidant enzyme(GPx, SOD, CAT) in both groups: LDH was found to be increased in high intensity group after the four-week exercise; MDA was increased in low intensity group after exercise and 30mins recovery than before exercise; compared with the value of pre-exercise, GPx and SOD in high intensity group shows significant differences after 30mins and 60mins recovery; while in low intensity group, the SOD difference only occurred immediately after exercise; In CAT, a significant difference was found in high intensity group but no significant difference was found in low intensity group.

        A Study of the Efficiency of Futures Research Institutes of China

        The purpose of this study is to analyze the efficiency of research institutes of futures companies, and to promote the development of futures market and real economy. This study employs DEA-solver software to conduct super-efficiency data envelopment analysis (SE-DEA), and also selects 40 representative futures research institutes in China as decision-making units (DMUs). For data of input and output indicators, we collect from the China Futures Association, Futures Daily, Hexun.com and Webstock.com respectively, and the time duration is the 103 trading days between from October 2019 to February 2020. Then the indicator for the strategy accuracy rate is calculated separately by analyzing the strategies published by each DMUs in public media. In conclusions, most institutes have excessive investment in human resources, and also have insufficient strategy accuracy rate and insufficient published research reports. The findings of this study suggest that Chinese futures companies need to improve the efficiency of research institutes, and better meet the demand of the financial market. In fact, the analysis of the efficiency of the futures company research institute has not been found in the literature worldwide, Application of DEA model in efficiency analysis of securities and futures research institutions and establishment of indicators are the innovations of this paper.

      • Screening of MicroRNA in Patients with Esophageal Cancer at Same Tumor Node Metastasis Stage with Different Prognoses

        Patients at the same pathological stage of esophageal cancer (EC) that received the same surgical therapy by the same surgeon may have distinct prognoses. The current study aimed to explore the possibility of differentially-expressed microRNAs (miRNAs) underlying this phenomenon. Samples were collected from EC patients at the same tumor node metastasis (TNM) stage but with different prognoses. Paracancerous normal tissues were taken as controls. The specimens were histopathologically analyzed. Differentially-expressed miRNAs were analyzed using real-time quantitative reverse transcription polymerase chain reaction. Compared with patients with poor prognosis, those with good prognosis exhibited 88 two-fold or more than two-fold increased miRNA fragments and 4 half-decreased miRNAs. The most noticeably up-regulated miRNAs included hsa-miR-31, hsa-miR-196b, hsa-miR-652, hsa-miR-125a-5p, hsa-miR-146b, hsa-miR-200c, hsa-miR-23b, hsa-miR-29a, hsa-miR-186, hsa-miR-205, hsa-miR-376a, hsa-miR-410, hsa-miR-532-3p, and hsa-miR-598, whereas the most significantly-downregulated miRNAs were hsa-let-7e, hsa-miR-130b, and hsa-miR-103. EC patients at same TNM stage but with different prognoses show differentially-expressed miRNAs.

      • Apoptotic Effects of psiRNA-STAT3 on 4T1 Breast Cancer Cells in Vitro

        Background: The aim of this study was to investigate the effect of a Lipofectamine2000 (Life2000) Transfection Reagent transfected psiRNA-STAT3 plasmid on 4T1 breast cancer cells. Materials and Methods: MTT was used to detect the cell proliferation of breast cancer 4T1 cells at different periods (0h, 6h, 8h, 10h); the cell cycle was assessed by flow cytometry; variation of apoptosis and mitochondrial membrane potential was observed under a fluorescence microscope; immunohistochemical staining was used to determine the expression of caspase-3 and cyclin-D1 protein. Results: An obvious effect of inhibition to 4T1 cancer cells could be observed at 8h after the psiRNA-STAT3 was transfected. Typical alterations of apoptotic morphological features were visible in the psiRNA-STAT3 treatment group. Mitochondrial membrane potential decreased significantly, the number of cells was increased in G0/G1 phase, and the number of cells was decreased in S phase, and the data were statistically significant (p<0.05), compared with the Scramble and Mock groups. Expression of caspase-3 protein was increased significantly, while that of cyclin D1 was significantly decreased. Conclusions: Life2000 transfected psiRNA-STAT3 plasmid can inhibit 4T1 tumor cell proliferation and promote apoptosis of 4T1 tumor cells, which process depends on the regulation of expression of cyclin D1 and caspase-3 protein.

        Association between FGFRs and the susceptibility of digestive and reproductive system cancers in Chinese population

        Fibroblast growth factors(FGFs) and their receptors (FGFRs) modulate a wide range of biological functions, especially tumor genesis. The aim of this study was to investigate the possible association of FGFR expression with the susceptibility of digestive system and reproductive system cancers in Chinese population. In total, 343 patients with digestive or reproductive system cancers were enrolled in this study. The expression levels of four highly-conserved FGFRs including FGFR1, FGFR2, FGFR3 and FGFR4 were measured by immunohistochemistry (IHC). The expression levels of FGFRs were compared between carcinoma and para-carcinoma tissues. FGFR1 expression significantly differed between carcinoma and para-carcinoma tissues in colon and gastric cancers. FGFR2 expression significantly differed between carcinoma and para-carcinoma tissues in esophageal cancer. FGFR3 expression was significantly different between carcinoma and para-carcinoma tissues in liver and gastric cancers. FGFR2 showed the highest expression probability in all the selected cancers and FGFR4 showed the lowest expression probability. FGFR1 and FGFR3 showed comparable moderate expression probabilities. Our findings have demonstrated significant differences regarding FGFR expression levels between carcinoma and para-carcinoma cells in digestive or reproductive system cancer patients. The data also implicated that FGFR2 and FGFR4 could serve as two prominent factors closely related to the susceptibility of digestive and reproductive system cancers.

        Effect of Surface Modification on Thermal Stability, Flexural Properties, and Impact Strength of Epoxy/Graphene Nanocomposites

        The surface of graphene (GE) was modified with a silane coupling agent, and its surface characteristics were investigated. Epoxy resin/GE nanocomposites were prepared via solution blending. The effect of the GE content on the thermal stability, flexural properties, impact strength, and morphology of the nanocomposites was studied. The thermal stability of the epoxy resin increased with the addition of pristine GE and surface-modified GE. The flexural strength, elastic modulus, and impact strength of the nanocomposites with surface-modified GE increased significantly with increasing GE content. Scanning electron microscopy images of the nanocomposites with surface-modified GE show a ductile deformation morphology, which is consistent with their high impact strength.

      • Involvement of MicroRNA-198 Overexpression in the Poor Prognosis of Esophageal Cancer

        Objective: This study aimed to investigate whether the miR-198 expression level is related to clinicopathological factors and prognosis of esophageal cancer. Methods: MicroRNA was extracted from esophageal cancer patients who underwent surgery for assessment using the Taqman@ MicroRNA assay. The correlation between miR-198 expression and clinicopathological features was analyzed, and the significance of miR-198 as a prognostic factor and its relationship with survival was determined. Results: MicroRNA-198 (miR-198) expression was higher in patients with poor prognosis than those with good prognosis (P<0.05). Kaplan-Meier analysis results showed that the miR-198 expression level had a significant correlation with survival time (P=0.030) and that patients with a higher expression of miR-198 had a shorter survival time. Cox multi-factor model analysis showed that patient prognosis (P=0.014), tumor length (P=0.040) and expression (P=0.012), and survival time had a significant correlation; the corresponding risks were 7.268, 1.246, and 3.524, respectively. Conclusion: miR-198 overexpression is involved in the poor prognosis of esophageal cancer and can be used as a biomarker for selection of cases requiring especial attention.

      • Tumor Markers for Diagnosis, Monitoring of Recurrence and Prognosis in Patients with Upper Gastrointestinal Tract Cancer

        To evaluate the value of combined detection of serum CEA, CA19-9, CA24-2, AFP, CA72-4, SCC, TPA and TPS for the clinical diagnosis of upper gastrointestinal tract (GIT) cancer and to analyze the efficacy of these tumor markers (TMs) in evaluating curative effects and prognosis. A total of 573 patients with upper GIT cancer between January 2004 and December 2007 were enrolled in this study. Serum levels of CEA, CA19-9, CA24-2, AFP, CA72-4, SCC, TPA and TPS were examined preoperatively and every 3 months postoperatively by ELISA. The sensitivity of CEA, CA19-9, CA24-2, AFP, CA72-4, SCC, TPA and TPS were 26.8%, 36.2%, 42.9%, 2.84%, 25.4%, 34.6%, 34.2% and 30.9%, respectively. The combined detection of CEA+CA199+CA242+CA724 had higher sensitivity and specificity in gastric cancer (GC) and cardiac cancer, while CEA+CA199+CA242+SCC was the best combination of diagnosis for esophageal cancer (EC). Elevation of preoperative CEA, CA19-9 and CA24-2, SCC and CA72-4 was significantly associated with pathological types (p<0.05) and TNM staging (p<0.05). Correlation analysis showed that CA24-2 was significantly correlated with CA19-9 (r=0.810, p<0.001). The levels of CEA, CA19-9, CA24-2, CA72-4 and SCC decreased obviously 3 months after operations. When metastasis and recurrence occurred, the levels of TMs significantly increased. On multivariate analysis, high preoperative CA72-4, CA24-2 and SCC served as prognostic factors for cardiac carcinoma, GC and EC, respectively. combined detection of CEA+CA199+CA242+SCC proved to be the most economic and practical strategy in diagnosis of EC; CEA+CA199+CA242+CA724 proved to be a better evaluation indicator for cardiac cancer and GC. CEA and CA19-9, CA24-2, CA72-4 and SCC, examined postoperatively during follow-up, were useful to find early tumor recurrence and metastasis, and evaluate prognosis. AFP, TPA and TPS have no significant value in diagnosis of patients with upper GIT cancer.

        Neutrophil-inspired photothermo-responsive drug delivery system for targeted treatment of bacterial infection and endotoxins neutralization

        Background P. aeruginosa, a highly virulent Gram-negative bacterium, can cause severe nosocomial infections, and it has developed resistance against most antibiotics. New therapeutic strategies are urgently needed to treat such bacterial infection and reduce its toxicity caused by endotoxin (lipopolysaccharide, LPS). Neutrophils have been proven to be able to target inflammation site and neutrophil membrane receptors such as Toll-like receptor-4 (TLR4) and CD14, and exhibit specific affinity to LPS. However, antibacterial delivery system based on the unique properties of neutrophils has not been reported. Methods A neutrophil-inspired antibacterial delivery system for targeted photothermal treatment, stimuli-responsive antibiotic release and endotoxin neutralization is reported in this study. Specifically, the photothermal reagent indocyanine green (ICG) and antibiotic rifampicin (RIF) are co-loaded into poly(lactic-co-glycolic acid) (PLGA) nanoparticles (NP-ICG/RIF), followed by coating with neutrophil membrane to obtain antibacterial delivery system (NM-NP-ICG/RIF). The inflammation targeting properties, synergistic antibacterial activity of photothermal therapy and antibiotic treatment, and endotoxin neutralization have been studied in vitro. A P. aeruginosa-induced murine skin abscess infection model has been used to evaluate the therapeutic efficacy of the NM-NP-ICG/RIF. Results Once irradiated by near-infrared lasers, the heat generated by NP-ICG/RIF triggers the release of RIF and ICG, resulting in a synergistic chemo-photothermal antibacterial effect against P. aeruginosa (~ 99.99% killing efficiency in 5 min). After coating with neutrophil-like cell membrane vesicles (NMVs), the nanoparticles (NM-NP-ICG/RIF) specifically bind to inflammatory vascular endothelial cells in infectious site, endowing the nanoparticles with an infection microenvironment targeting function to enhance retention time. Importantly, it is discovered for the first time that NMVs-coated nanoparticles are able to neutralize endotoxins. The P. aeruginosa murine skin abscess infection model further demonstrates the in vivo therapeutic efficacy of NM-NP-ICG/RIF. Conclusion The neutrophil-inspired antibacterial delivery system (NM-NP-ICG/RIF) is capable of targeting infection microenvironment, neutralizing endotoxin, and eradicating bacteria through a synergistic effect of photothermal therapy and antibiotic treatment. This drug delivery system made from FDA-approved compounds provides a promising approach to fighting against hard-to-treat bacterial infections.
