        Antioxidant Properties of Peptides Extracted from Tenebrio molitor Larvae

        본 연구는 기능성 식품의 개발을 위해 갈색거저리 유충 유래 추출물에서 새로운 생리활성 펩타이드를 확인하는 것이다. 동결건조된 T. molitor 유충에서 다양한 용매로 시간에 따라 추출하였다. T. molitor 유충추출 결과 물 추출물에서 5시간과 10시간에서 총 단백질과 고형분 함량당 각각 가장 높은 2,2,1-diphenyl-1-picrylhydrazyl (DPPH) 활성을 보였다. 물 추출물을 메탄올 농도에 따라 분획하였을 때, 높은 메탄올 농도는 상등액 중에 고분자량 단백질의 수준을 감소시키는 반면, 상등액의 DPPH 활성 수준을 증가시켜 저분자량 펩타이드가 항산화 활성을 매개하는 것으로 나타났다. 유기용매 분배에서 부탄올을 제외한 유기용매 분배에서 물층에서 유사한 활성을 보였다. 유기용매분획물을 열처리 후 활성이 28~44% 감소하여 열에 대한 불안정한 성분이 존재하는 것으로 추정되었다. 물 추출물에 프로테아제 K를 첨가하면 DPPH활성이 10~20% 증가하여 전체 단백질에서 방출되는 펩타이드가 항산화 활성을 부분적으로 증가시켰음을 알 수 있다. 그러므로, T. molitor 유충 추출물은 항산화제로써 기능을 수행할 수 있는 물질이 존재하는 것으로 제의할 수 있다. The goal of this study was to identify new bioactive peptides in extracts derived from Tenebrio molitor (T. molitor) larvae for the development of functional foods. After extraction from freeze-dried T. molitor larvae with various solvents on time course, the extracts showed the highest 2,2,1-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity at 5 and 10 hr per total protein and solid contents, respectively. When the water extract was fractionated, a high methanol concentration led to a reduced level of high-molecular-weight proteins in the centrifugal supernatant, whereas increased DPPH activity in the supernatants suggests low-molecular-weight peptides may mediate antioxidant activity in the supernatant. Most of the organic solvent partitions, excluding butanol, showed similar activities in the water phases, and the organic solvent partition fraction exhibited a 28~44% decrease in activity following heat treatment, implying that some components in the fraction become unstable in the presence of heat. The addition of proteinase K to the water extract increased DPPH activity by 10~20%, suggesting that peptides, when released from total proteins, partially increase antioxidant activity. Therefore, we suggest that the antioxidants in T. molitor larval extracts make them a potential source of functional animal food.

        The explanation of isolatedorgan tuberculosis rests on the assumption that in the course of the lymphatic or hematogenous dissemination of bacilli, organisms may be rapidly destroyed in all other sites save for the particular tissue involved in the isolated tuberculous process. Tuberculosis can arise in all tissues having lymphatics or blood supply, but the disease causing biliary tract obstruction has been known to be rare. Recently, we experienced a case of isolated-organ tuberculosis causing common bile duct obstruction and periductal lymph node enlargement in a 46-year-old Korean male. An ultrasonographyguided percutaneous needle biopsy revealed a granulomatous inflammation of the lymph node. After 7 months of antituberculous medication, the common bile duct obstruction and periductal lymph node enlargement disappeared completely in a follow up abdominal CT and ERCP.

      • Isolation and characterization of the novel antibacterial peptide “Coprisin” from a dung beetle, Copris tripartitus

        To find some antibacterial peptides responsible for bacterial resistance, we performed differential hybridization with total cDNA probes which synthesized from normal and immunized larvae. Thirteen individual cDNA transcripts were expressed differentially in a total 1,862 random cDNA clones. One of upregulated genes is a novel member of the insect defensin-like peptide(Coprisin), a family of antibacterial peptide. Northern blot analysis showed that Coprisin was up-regulated at 4h and reached the highest point level at 16h after injection of E.coli. The deduced amino acid sequence of Coprisin was composed of 80 amino acids with predicted molecular weight of 8.6 kDa and PI of 8.72. Comparison of the deduced amino acid mature portion of Coprisin with defensin-like peptide of other insect indicated that it has 79.1% and 67.4% identity with Anomala cuprea and Allomyrina dichotoma, respectively. To find antibacterial active region of Coprisin, we synthesized four peptides corresponding to amino acid residues 1V-43N-NH2(CopN1), 5-16(CopN2), 19-30(CopN3) and 31-43(CopN4) of coprisin having amidated amino acid residues at their Cterminal. A 12-mer amidated at its C-terminus, ACALHCIALRKK-NH2 (Ala19-Lys30-NH2) was synthesized based on the deduced amino acid sequence, assumed to be an active site sequence. This peptides showed antibacterial activity against E.coli, Staphylococcus aureus, MRSA, Psedomonas syringae, and Pectobacterium carotovorium. Modified 9-mer peptide, LRCIALRKK-NH2, showed strong antibacterial activity than mellitin peptide used as a positive control against gram-negative and gram-positive bacteria. This peptide showed no haemolytic activity and quite stable at 100℃ for several hours of incubation and in a wide pH range(pH2-12). Therefore, this peptide may be a good candidate for the development of new drug with potent antibacterial activity without cytotoxicity.

        Coronary vasospasm has been proposed as a mechanism of muocardial infarction, particularly in variant angina pectoris with normal coronary arteries. Plaque rupture or prolonged stasis facilitating thrombus formation has been postulated as the mechanisms of vasospasm induced myocardial infarction, Recently some reports decribed effects of thrombolytic therapy in patients with variant angina and chest pain with long duration to prevent or reduce the amount of myocardial infarction. We report two cases of patients with myocardial infarction in normal coronary artery, who were proved coronary artery spasm and were successfully managed by thrombolytic therapy.

        LW-RCP와 Odroid 기반의 Machine Vision을 이용한 Ball and Plate 시스템 구현에 관한 연구

        In this paper, we propose the architecture of a ball-and-plate system as an educational platform that can help students comprehensively study control engineering, robotics, machine vision, and embedded systems. In addition, we describe how to implement the sub-systems and present an example of an education plan using the proposed system. In the proposed system, we use an LW-RCP(Light Weight-Rapid Control Prototyping) system, which is lab-built, to help students implement the control algorithm. An RCP system makes it easy for students who are not familiar with u-Controller or the C language to implement controllers. We also use Odroid, a low-cost SBC (single-board computer), and a single camera to calculate the position of ball using a machine vision algorithm. In this structure, the measurement subsystem is more durable compared with contact-based sensing systems and student can test their own machine vision algorithms for better measurements. We adopt the Stewart platform, a type of parallel robot, to generate roll-and-pitch actuation for the ball-and-plate system. By adopting the Stewart platform, we can handle more complicated control problems for movement that requires more than two degrees of freedom. Additionally, we implement an LQ controller, which is a typical model-based controller, and apply it to the ball-plate system. Finally, through experiments, we illustrate the usefulness of the proposed system.

        한국어판 Cigarette Dependence Scale-12(CDS-12)의 신뢰도와 타당도 연구

        Objectives:This study aimed to assess the reliability and val-idity of the Korean version of the Cigarette Dependence Scale-12 (CDS-12). Methods:Participants were 189 male and 8 female smokers from a public health center. We administered the Korean Version of the Fagerström Test for Nicotine Dependence (FTND), the CDS-12, and two shorter versions of the same measures [the CDS-5 and the Heavy Smoking Index (HIS)] to the participants, who started smoking cessation attempts after this initial assessment. At their first visit, we measured the participants’ carbon monoxide (CO), and participants revealed their cessation state through self-report at four weeks, six weeks, and six months. Results:Cronbach’s alpha, which indicates internal reliability, was 0.889 for the CDS-12 and 0.741 for the CDS-5. Split-half reliability was 0.882 for the CDS-12 and 0.650 for the CDS-5. The Pearson correlation coefficients, indicating the relationships between the FTND and either the CDS-12 or the CDS-5, were 0.675 and 0.746, respectively. The Pearson correlation coefficients between the HSI and the CDS-12 or the CDS-5 were 0.662 and 0.756, respectively. The CDS-12 score at baseline predicted smoking cessation after 4 weeks. Conclusion:The CDS-12 is a valid and reliable measure of nicotine dependence.

        양단자유공진주 및 초음파속도법으로 획득한 압축파 속도를 이용한 암석시편의 전단파 속도 도출

        암석의 강도 및 건전성 평가에 있어 전단파를 사용하는 것이 압축파에 비해 높은 신뢰성과 정확도를 제공한다.암편의 VS 도출을 위해 양단자유공진주기법을 수행할 시 비틀림파에 의한 공진주파수를 구분해야 하나 쉽지 않은 상황이 자주 발생한다. 또한, 초음파속도기법에서는 P파에 비해 S파 도달 시점이 모호하여 암편의 VS를 객관적으로 산출하는것이 쉽지 않다. 반면에 초음파 속도법을 통해서는 VP 값을, 양단자유공진주기법을 통해서는 VL 값을 안정적으로 획득할수 있는데 탄성계수간의 관계식을 이용하여 포아송비를 계산할 수 있게 되며 VS 값을 산출할 수 있다. 알루미늄, 모노캐스트 등 다른 재질과 다른 길이를 가지는 모형 시편을 이용하여 검증 실험을 수행하였고 국내 여러 지역에서 채취한 암석시편에 대해서 제안된 방법을 적용하여 본 결과 제안된 방법의 유용함을 확인할 수 있었다. Using shear wave velocity is more reasonable to estimate strength and integrity of rock compared with using compressional wave. It is often ambiguous to pick the dominant frequency caused by torsional wave when evaluating VS of rock specimen from FFRC method. It is also sometimes ambiguous to pick the first arrival point of S wav ecompared with P wave in the signals acquired from ultrasonic velocity method. Otherwise, the procedure of evaluating VP using ultrasonic velocity method and VL using FFRC method is relatively stable. Through the relationship between elastic modulus, poisson`s ratio and VS can be obtained from VP, VL. Applicability was checked using model specimens having different material property and length and rock specimens sampled in mine area, and usefulness of proposed procedure was verified.
